Quality Characteristics and Antioxidant Activities of Sulgidduk Added with Almond Powder (아몬드 첨가 설기떡의 품질 특성 및 항산화성)

  • The purposes of this study were to evaluate the quality characteristics and antioxidant activities of Sulgidduk added with almond powder (0, 6, 12, or 15%). As the amount of almond powder increased, soluble solid content ($^{\circ}Brix$) and reducing sugar content (%) decreased, whereas pH and acidity were unchanged. The L (lightness) value in the Hunter color system of Sulgidduk added with almond powder decreased, whereas a (redness) and b (yellowness) values increased. Total phenolic content of Sulgidduk increased according to the amount of almond powder. The antioxidant activities, including 1,1-diphenyl-2-picrylhydrazyl and hydroxy radical scavenging activities, increased in Sulgidduk with almond powder compared with that of the control. It is concluded that Sulgidduk with almond powder may be of high quality with antioxidant activity. In the sensory test, Sulgidduk added with 12% almond powder showed the highest score in terms of overall preference.

  • This study aimed to obtain high Fischer's ratio oligopeptides from goat whey (HFO) and investigate antioxidant property of it. Hydrolysis of goat whey was done with the approach of sequential digestion of pepsin and flavourzyme. With the adsorption of aromatic amino acids by activated carbon, HFO with a Fischer's ratio of 27.070 and a molecular weight of 200-1,000 Da were obtained, and the branched-chain amino acids accounted for 22.87%. Then the antioxidant activity of HFO was evaluated. At the concentrations of 2.0 mg/mL and 0.50 mg/mL, HFO scavenged 77.27% and 99.63% of 1,1-diphenyl-2-picrylhydrazyl and 3-ethylbenzthiazoline-6-sulphonate free radicals respectively. The scavenging rate of HFO against hydroxyl radicals reached 92.31% at the concentration of 0.25 mg/mL. Animal experiments demonstrated that HFO could moderate the changes of malondialdehyde, superoxide dismutase and glutathione peroxidase caused by CCl4-induced oxidative stress in vivo. This study indicated that HFO from goat whey was capable of oxidation resistance both in vivo and in vitro, which provided a scientific basis for the high-value processing and application of goat milk whey.

Antioxidant Activities of Glycyrrhizin and its Effect on Renal Expression of Na,K-ATPase in Gentamicin-induced Acute Renal Failure Rats (Glycyrrhizin의 항산화 활성 및 Gentamincin 유도 급성 신부전 백서 신장의 Na,K-ATPase 발현에 미치는 영향)

  • The present study was aimed to investigate whether glycyrrhizin, which is the major component of Glycyrrhiza uralensis, has an antioxidant effect and regulatory effect on Na,K-ATPase in gentamicin-induced acute renal failure (ARF) rats . It is well known that reactive oxygen species (ROS), such as superoxide anion and hydroxyl radical, are main pathophysiological factor in gentamicin-induced ARF. Glycyrrhizin showed potent in vitro antioxidant activity, especially superoxide scavenging activity, in a dose-dependent manner. Plasma lipid peroxide level was restored to normal level by oral administration of glycyrrhizin (200 mg/kg) in the gentamicin-induced ARF rats. The expression of Na,K-ATPase α1 subunit was restored in the gentamicin-induced ARF rats by administration of glycyrrhizin, whereas β1 subunit was not restored. The renal functional parameters including urine volume, cleatinine clearance, urine osmolality, solute-free water reabroption were also partially restored in gentamicin-ARF rats by administration of glycyrrhizin. Taken together, the amelioration of renal functions and the expression of sodium pump by administration of glycyrrhizin in the gentamicin-induced ARF was appear to be mediated by the scavenging of ROS.

Biological activity of water-soluble polysaccharides from Cedrela sinensis according to extraction methods (참죽의 추출방법에 따른 수용성 다당류의 생리기능성)

  • The biological activity of water-soluble polysaccharide (WSP) fractions extracted from Cedrela sinensis was examined in this study. Cedrela sinensis was extracted using hot water, ultrasonication, and enzymes (Viscozyme, Shearzyme) and precipitated using ethanol to produce crude polysaccharides. The yield (3.51%) and total polysaccharide content (28.03 g/100 g) of WSP extracted using Shearzyme (WSPs) were highest compared to other extracts. The antioxidant activity of WSP extracted using hot water was highest and had the lowest $IC_{50}$ values in DPPH, ABTS, hydroxyl radical scavenging activity, reducing power, and superoxide dismutase-like activity. Tyrosinase inhibitory activity increased as the concentration increased. All extracts showed higher retardation effects on glucose and bile acid compared to the control; particularly, WSPs showed a similar glucose retardation effect to carboxymethyl cellulose. This study suggests that WSP from C. sinensis can be used as a functional food material.

Antioxidative effects of fermented sesame sauce against hydrogen peroxide-induced oxidative damage in LLC-PK1 porcine renal tubule cells

  • BACKGROUND/OBJECTIVES: This study was performed to investigate the in vitro antioxidant and cytoprotective effects of fermented sesame sauce (FSeS) against hydrogen peroxide ($H_2O_2$)-induced oxidative damage in renal proximal tubule LLC-PK1 cells. MATERIALS/METHODS: 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical ($^{\bullet}OH$), and $H_2O_2$ scavenging assay was used to evaluate the in vitro antioxidant activity of FSeS. To investigate the cytoprotective effect of FSeS against $H_2O_2$-induced oxidative damage in LLC-PK1 cells, the cellular levels of reactive oxygen species (ROS), lipid peroxidation, and endogenous antioxidant enzymes including catalase (CAT), superoxide dismutase (SOD), and glutathione peroxidase (GSH-px) were measured. RESULTS: The ability of FSeS to scavenge DPPH, $^{\bullet}OH$ and $H_2O_2$ was greater than that of FSS and AHSS. FSeS also significantly inhibited $H_2O_2$-induced ($500{\mu}M$) oxidative damage in the LLC-PK1 cells compared to FSS and AHSS (P < 0.05). Following treatment with $100{\mu}g/mL$ of FSeS and FSS to prevent $H_2O_2$-induced oxidation, cell viability increased from 56.7% (control) to 83.7% and 75.6%, respectively. However, AHSS was not able to reduce $H_2O_2$-induced cell damage (viability of the AHSS-treated cells was 54.6%). FSeS more effectively suppressed $H_2O_2$-induced ROS generation and lipid peroxidation compared to FSS and AHSS (P < 0.05). Compared to the other sauces, FSeS also significantly increased cellular CAT, SOD, and GSH-px activities and mRNA expression (P < 0.05). CONCULUSIONS: These results from the present study suggest that FSeS is an effective radical scavenger and protects against $H_2O_2$-induced oxidative damage in LLC-PK1 cells by reducing ROS levels, inhibiting lipid peroxidation, and stimulating antioxidant enzyme activity.

Establishment of Optimal Fermentation Conditions for Steam-dried Ginseng Berry via Friendly Bacteria and Its Antioxidant Activities (생체친화성 균주에 의한 인삼열매증포 추출물의 최적발효조건 및 항산화활성)

  • In this study, we observed optimal conditions and suitable bacteria for the fermentation of steam-dried ginseng berry extracts (SGB) and determined antioxidant effects of the fermented extracts. Five bacteria (Lactobacillus fermentarum, L. plantarum, L. brevis, L. casei, Bacillus subtillis) were examined on their growth activities and viabilities in various culture temperatures ($25-35^{\circ}C$) and concentrations (25-100%). L. plantarum was considered to be the most suitable bacteria for the fermentation in both growth activity and viability. Moreover, the extracts fermented with L. plantarum showed more potent antioxidant efficacy in both 1,1-diphenyl-2-picrylhydrazyl radical and hydroxyl radical scavenging assay. High performance liquid chromatography analysis revealed that fermentation with L. plantarum changed the contents and components of ginsenosides. In conclusion, these data suggest that L. plantarum efficiently ferment SGB and the fermented extracts may have therapeutical values against oxidative stress and be a good candidate in adjuvant therapy where ginsenoside would be the main composition.

Quality Characteristics and Antioxidant Activities of Black Garlic Jam Prepared with Fructooligosaccharide (프락토올리고당 첨가 흑마늘 잼의 품질 특성 및 항산화성)

  • The quality characteristics and antioxidant activities were evaluated for jams containing fructooligosaccharide (FTO) and FTOS (sucrose 50%+FTO 50%) substituted for sucrose. The endpoint of jam preparation was determined to be sweetness of $64^{\circ}Brix$. Moisture contents differed among the treatments, whereas acidity did not. Reducing sugar content and viscosity were highest in the FTO (0.144%, 126,800 cP), and the lowest in control (0.074%, 101,600 cP). Lightness (L value), redness (a value), and yellowness (b value) values of the Hunter color system were highest in the FTO sample. Hardness and springiness of textural properties were the highest in the FTO sample as well. Antioxidant activities were the highest in the FTO sample with the lowest $IC_50$ values (42.3 mg/g for DPPH and 22.4 mg/g for hydroxyl radical scavenging activities). Total phenolic content was the highest in the FTO sample among all treatments. The overall acceptance score of black garlic jam containing FTO was the highest. Based on these results, it is suggested that FTO was appropriate for achieving good qualities such as antioxidative activity in black garlic jam.

  • The free radical theory of aging was introduced in 1956 by Denham Harman. This aging theory proposed that normal aging results from random deleterious damage to tissues by free radical and supplying antioxidant lead to decrease oxidative damage, inhibit aging process. In this study, we investigated antioxidantive effects of four Korean constitutional prescriptions for 'Soum' constitution - Palmulgunjatang(Y1), Sipyimiguanjungtang(Y2), Osuyubujayijungtang(Y3) and Seungyangyikkibujatang(Y4). Antioxidative activity of this prescriptions was examined by 1,1-diphenyl-2-picrylhyrdazyl radicals, superoxide anion radicals, peroxyl radical, hydroxyl radical scavenging effects and erythrocyte hemolysis inhibitory effects. Y2 and Y3 were shown to have relatively high antioxidative activity on this methods. In additions, result of the cytoprotective effects of Korean constitutional prescriptions agianst 2,2'-azobis(amidinopropane) dihydrochloride (AAPH), a free radical initiator, induced cytotoxcity in human hepatoblastoma cell line was similarly obtained. On the basis of this result, we assayed the antioxidative effects of Y2 and Y3 on experimental oxidative damage, induced in mouse by 100mg/kg AAPH. Male ICR mouse were given oral administration of 500mg/kg Y2 and Y3 for 4 weeks. Thiobarbuturic acid reactive substance (TBARS) and protein degradation level in liver, plasma and brain as index of oxidative damage were decreased and thiol compound, total antioxidant status in plasma were increased by Y2 administration. But, Y3 injected group was decreased only protein degradation level in brain. Also, glutathione, a potent water-soluble endogenous antioxidant, concentration was increased by Y2 and Y3 administration in liver and brain. However, superoxide dismutase and catalase activity as a major antioxidative enzyme in vivo were not shown change by Y2 and Y3 administration. On the basis of these result, Y2 have an antioxidative effects on both water-soluble fraction and lipid-solube fraction in cell and tissues. But, Y3 has a lower antioxidative effects on lipid-soluble fraction than Y2 in cell and tissues. These results suggest that Y2 has a antioxidative effects by protect the tissue against oxygen free radical mediated oxidative damage and Y3 has a limited antioxidaitve effects on water-soluble fraction in vivo. Therefore, we make report that Y2 is more effective prescriptions for anti-aging or therapeutics of diseases.

Antioxidant Activities of Methanol Extracts from Prunella vulgaris (하고초 메탄올 추출물의 항산화 활성)

  • The aim of the present study was to investigate the antioxidant activities of methanol extracts from whole plant, flower stalk and stem of Hagocho (Prunella vulgaris). Content of total phenolic compound was the highest in flower stalk (77.1 mg/100 g) and those of others were below 54.0 mg/100 g. Flavonoid contents was the highest in stem (36.1 mg/100 g) compared to other samples. Electron donating ability of Prunella vulgaris was activated at over 70% in all samples at $500{\mu}g/mL$ concentration, especially, the activity was the highest (92.1%) in flower stalk extracts. Reducing power showed similar tendency to electron donating ability, which was significantly higher flower stalk ($0.3{\sim}1.9$), whole plant ($0.2{\sim}1.6$) and stem ($0.2{\sim}1.5$). Hydroxyl radical was scavenged over 80% in $100{\mu}g/mL$ concentration and was not significantly different between parts. Antioxidant activity in $\beta$-carotene-linoleic acid system was $47.5{\sim}84.6%$ when $1,000{\mu}g/mL$ methanol extracts was added to reaction mixtures, and flower stalk showed the highest activity. Ability of ABTs cation decolorization from Prunella vulgaris was activated over 50% in all samples when $250{\mu}g/mL$ of methanol extracts was added to reaction mixtures and $500{\mu}g/mL$ were the most suitable concentration for its activation. Nitric oxide scavenging activity was lower under 20%, but its activity was significantly higher in flower stalk than other parts. The results indicate that flower stalk from Prunella vulgaris has potent antioxidant activities.

Effect of Garlic and Medicinal Plants Composites on Antioxidant Activity and Lipid Levels of Liver in Hypercholesterolemic Rats (마늘과 한약재 복합물의 항산화 활성 및 고콜레스테롤 급이 흰쥐의 간장 지질 함량에 미치는 영향)

  • The effects of combined garlic and medicinal plant extracts such as Gyeolmyeongja (Cassia obtusifolia Linne), Hasuo (Polygoni multiflori Radix), Youngji (Ganoderma lucium) and Sansayuk (Crataegus pinnatifida Bunge) on the antioxidant activity and lipid levels in the livers of rats fed a high cholesterol diet were analyzed. Total phenol and flavonoid contents were the highest in the Gyeolmyeongja by $151.02{\pm}5.20\;mg$/100 g and $43.69{\pm}5.58\;mg$/100 g. Electron donating ability, reducing power and hydroxyl radical scavenging activity were significantly increased when over 0.3% garlic extract was added. The antioxidant activity of linoleic acid in $\beta$- carotene increased in a dose dependant manner in response to the concentration of garlic extract. In livers of rats, the content of total lipids was significantly decreased by feeding garlic and medicinal plants composites; in particular, the group in which 0.7% garlic extract was added was the lowest. Total cholesterol was 14.95 mg/g in the control group; its level was lower in the groups fed garlic and medicinal plants composites, ranging from 11.47 to 11.86 mg/g. Triglyceride concentration was significantly decreased in the group fed 0.7% garlic extracts, with 46.42 mg/g compared to groups fed 0.3% and 0.5%. TBARS content showed a 15.8~17.6% decrease in groups fed 0.5~0.7% garlic extract and medicinal plants composites. Antioxidant activity was significantly increased in groups fed over 0.5% garlic extract compared to the control group. This study shows that garlic and medicinal plant composites intake is able to reduce the levels of liver lipids in hypercholesterolemic rats.
