Selective Homocysteine Assay with Cucurbit[7]uril by pH Regulation

  • We report the effect of pH on the supramolecular complexation of two biothiols, viz., homocysteine (Hcy) and cysteine (Cys), with cucurbit[7]uril (CB[7]). Under basic pH conditions, Cys did not complex with CB[7], whereas Hcy efficiently complexed with CB[7], as confirmed by 1H NMR spectroscopy and Ellman's reagent (5,5'-dithio-bis(2-nitrobenzoic acid), DTNB) assay. 1H NMR and Raman spectroscopic studies revealed that, in the absence of CB[7], Hcy auto-oxidized slowly (~36 h) to homocystine (HSSH) under basic pH conditions. However, the rate of Hcy oxidation increased by up to 150 fold in the presence of CB[7], as suggested by the DTNB assay. Thus, supramolecular complexation under basic pH conditions led to the formation of a HSSH-CB[7] complex, and not Hcy-CB[7]. The results indicate that Hcy is rapidly oxidized to HSSH under the catalysis of CB[7], which acts as a reaction chamber, in basic pH conditions. Our studies suggest that Hcy concentration, a risk factor for cardiovascular disease, can be selectively and more easily quantified by supramolecular complexation with CB [7].

본태성 고혈압 쥐에서 복분자 완숙과 추출물의 혈압 개선 효과 (Anti-Hypertensive Effects of Black Raspberry (Rubus occidentalis) in Spontaneously Hypertensive Rats (SHR))

  • 본 연구는 본태성 고혈압 쥐인 SHR을 이용하여 복분자 완숙과 에탄올 50% 추출물의 혈압 강하 효과 및 그와 관련된 인자들[endothelial nitric oxide synthase(eNOS), homocysteine, plasminogen activator inhibitor-1(PAI-1), albumin, creatinine, blood lipid 등]에 미치는 영향을 조사하였다. 본태성 고혈압 쥐에게 복분자 완숙과 에탄올 50% 추출물을 12주간 투여한 결과 수축기 혈압이 고혈압대조군에 비하여 약 12% 억제되었다. 또한 eNOS 유전자 발현을 유의하게 증가하였으며 혈청 homocysteine 및 PAI-1의 활성이 눈에 띄게 감소됨을 확인하였다. 또한 심근 호산성 세포수를 감소시켜 심장의 손상 및 병변을 경감시키는 효과가 있음을 확인하였다. 그러나 혈중 지질 및 혈청 albumin, creatinine의 농도에는 각 군 간의 변화가 뚜렷하게 나타나지 않았다. 본 연구 결과 복분자 완숙과 에탄올 50% 추출물은 혈관의 손상을 야기할 수 있는 homocysteine이나 PAI-1과 같은 염증성 인자로부터 혈관을 보호하고 이를 통해 eNOS의 유전자 발현을 개선시킴으로써 혈압의 조절에 영향을 미치는 것으로 나타났으며, 향후 심혈관계 질환의 개선에 도움이 되는 식품소재로서의 가능성이 있음을 확인하였다.

노년기 여성들의 혈장 타우린의 농도와 혈장 호모시스테인 및 혈청 지질농도와의 상관관계 (Correlations of Plasma Taurine, Homocysteine and Serum Lipid Levels of Elderly Women)

  • This study was conducted to assess blood components caused by metabolic syndrome increasing in postmenopausal elderly women. The blood samples of these subjects were analyzed to investigate the correlation of plasma taurine levels and plasma homocysteine levels, and serum lipid profiles. The subjects were 33 elderly women($72.8{\pm}4.4$ years). Their mean height, weight and BMI were $150.5{\pm}5.7\;cm$, $57.5{\pm}6.3\;kg$ and $25.4{\pm}2.5\;kg/m^2$. 16 women of this study subjects have been chronic diseases such as hypertension or diabetes. Their fasting blood glucose was $98.2{\pm}24.0\;mg/d{\ell}$, and their plasma total cholesterol (TC), HDL-C, LDL-C, triglyceride(TG) were $216.5{\pm}29.9$, $52.1{\pm}10.7$, $145.7{\pm}27.9$ and $141.2{\pm}59.6\;mg/d{\ell}$, respectively. Their blood lipid profiles were higher than the standard levels of metabolic syndrome, thus these levels of lipid profiles may play a role as risk factors on the elderly person. Plasma taurine level of the subjects was $278.5{\pm}48.1\;{\mu}mol/{\ell}$, and their plasma homocysteine level was $12.8{\pm}2.9\;{\mu}mol/{\ell}$. The concentration of plasma vitamin $B_{12}$ was significantly decreased by aging(p<0.05). The correlation of plasma homocysteine and plasma folate showed significantly negative(p<0.05). Thus, the decreased levels of plasma vitamin $B_{12}$ and folate by aging might affect on the plasma homocyteine concentration acting as a risk factor of cardiovascular diseases for elderly person. The correlation of plasma taurine and hemoglobin, and their platelet showed significantly positive(p<0.05). In conclusion, the diet on the elderly person is one of the important factors to prevent their health from chronic diseases. This study recommends that well balanced diets are needed for elderly person to keep their health and prevent from metabolic syndrome.

구기엽 에탄올 추출물이 고지방 식이 유도 비만쥐의 비만과 혈중 생화학적 지표 조절에 미치는 영향 (Effects of the Ethanol Extract from Lycii folium Leaves on Obesity and Blood Biochemical Indices in High-fat Diet Induced Obese Rats)

  • 13주 동안 고지방식이로 비만을 유도한 비만쥐에게 10% 구기엽 에탄올 추출물을 1 mL/day로 8주간 경구투여한 후비만도, 혈중 지질, homocysteine, leptin, ghrelin, glucose 농도를 분석한 결과를 요약하면 다음과 같다. 체중증가와 식이효율은 구기엽 추출물의 유의한 영향이 없었다. 그러나 비만도의 경우 일반식이 비만군의 Rohrer index와 Lee index는 FO 군과 유의한 차이가 없었고 T.M. index는 FO 군보다 유의하게 낮았다. 또한 FLEO 군은 모든 비만지수가 FO 군보다 유의하게 낮아 구기엽 추출물이 비만도를 낮추는 것으로 나타났다. 한편 BO 군의 혈중 중성지질은 FO 군보다 유의하게 낮은 반면, 구기엽 에탄올 추출물은 혈청 지질, homocysteine, leptin, ghrelin, glucose 농도에 유의한 영향을 미치지 못한 것으로 나타났다. 이상의 결과를 종합할 때 비만쥐에서 구기엽 에탄올 추출물의 섭취는 혈중 지질이 나 비만관련 생화학적 지표에는 영향을 미치지 않지만 비만도를 낮춰 비만치료 효과의 가능성을 볼 수 있었다.

경기도 일개 지역사회 대표집단에서 고호모시스테인혈증의 유병률 및 관련요인에 대한 단면조사연구 (Prevalence of Hyperhomocysteinemia and Related Factors in a Community-based Health Examination Survey: A Cross-sectional Study)

  • Background : Many previous studies have shown that elevated homocysteine in the serum is a well known risk factor for cardiovascular disease and this is associated with other risk factors for cardiovascular disease, but any Korean data on this is limited. Objectives : This study aimed to calculate the prevalence of hyperhomocysteinemia and to analyze the relation between elevated homocysteine and the lifestyle factors of Korean adults. Methods : We conducted a cross-sectional survey that included 650 men and 743 women (age range, 20 to 79 years) who were residents of Gwangju City in Gyeonggi-do. These subjects participated in the health interview and examination survey from November to December 2005. The total homocysteine, total cholesterol, HDL-cholesterol, LDL-cholesterol, and triglyceride in the serum were measured. All the participants had their body composition measured such as height and weight, and we obtained health-related behavioral information through the self-entry questionnaire. Results : Very right-handed skewed distributions of homocysteine were shown in men and women. The prevalence of hyperhomocysteinemia was 22.6% in men and 13.7% in women in Gwangju city. On the multiple logistic regression analysis, hyperhomocysteinemia was associated with age (OR=1.02, 95% CI=1.01-1.04), male gender (OR=1.60, 95% CI=1.02-2.52), severe general physical activity (OR=0.32, 95% CI=0.15-0.69) and nutrient consumption (OR=0.49, 95% CI=0.31-0.76). Conclusions : There is a great prevalence of hyperhomocysteinemia in adults of Gwangju City, Korea and it was associated with both genetic factors and lifestyle risk factors. This study can suggest that comprehensive lifestyle modification is needed in order to diminish the prevalence of hyperhomocysteinemia and to prevent CVD.

Characterization of Homocysteine ${\gamma}$-Lyase from Submerged and Solid Cultures of Aspergillus fumigatus ASH (JX006238)

  • Among 25 isolates, Aspergillus fumigatus ASH (JX006238) was identified as a potent producer of homocysteine ${\gamma}$-lyase. The nutritional requirements to maximize the enzyme yield were optimized under submerged (SF) and solid-state fermentation (SSF) conditions, resulting in a 5.2- and 2.3-fold increase, respectively, after the last purification step. The enzyme exhibited a single homogenous band of 50 kDa on SDS-PAGE, along with an optimum pH of 7.8 and pH stability range of 6.5 to 7.8. It also showed a pI of 5.0, as detected by pH precipitation with no glycosyl residues. The highest enzyme activity was obtained at $37-40^{\circ}C$, with a $T_m$ value of $70.1^{\circ}C$. The enzyme showed clear catalytic and thermal stability below $40^{\circ}C$, with $T_{1/2}$ values of 18.1, 9.9, 5.9, 3.3, and 1.9 h at $30^{\circ}C$, $35^{\circ}C$, $40^{\circ}C$, $50^{\circ}C$, and $60^{\circ}C$, respectively. Additionally, the enzyme $K_r$ values were 0.002, 0.054, 0.097, 0.184, and 0.341 $S^{-1}$ at $30^{\circ}C$, $35^{\circ}C$, $40^{\circ}C$, $50^{\circ}C$, and $60^{\circ}C$, respectively. The enzyme displayed a strong affinity to homocysteine, followed by methionine and cysteine when compared with non-S amino acids, confirming its potency against homocysteinuria-related diseases, and as an anti-cardiovascular agent and a specific biosensor for homocysteinuria. The enzyme showed its maximum affinity for homocysteine ($K_m$ 2.46 mM, $K_{cat}\;1.39{\times}10^{-3}\;s^{-1}$), methionine ($K_m$ 4.1 mM, $K_{cat}\;0.97{\times}10^{-3}\;s^{-1}$), and cysteine ($K_m$ 4.9 m M, $K_{cat}\;0.77{\times}10^{-3}\;s^{-1}$). The enzyme was also strongly inhibited by hydroxylamine and DDT, confirming its pyridoxal 5'-phosphate (PLP) identity, yet not inhibited by EDTA. In vivo, using Swiss Albino mice, the enzyme showed no detectable negative effects on platelet aggregation, the RBC number, aspartate aminotransferase, alanine aminotransferase, or creatinine titer when compared with negative controls.

항산화성 비타민 보충 급여가 경기지역 일부 대학생 흡연자와 비흡연자의 혈압과 혈장지질 및 엽산과 호모시스테인에 미치는 영향 (The Effect of Antioxidant Vitamins Supplementation on Blood Pressure, Plasma Lipids, Folate, and Homocysteine Levels in Smokers and Non-Smokers of College Students in the Gyeonggi Area)

  • This study was performed to investigate the effect of vitamin C and E supplementation on blood pressure, plasma lipids, folate, and homocysteine levels in smokers and non-smokers of college male students in Gyeonggi Area. The nutrient intakes were determined by a 24hr-recall method. The subjects were divided into six groups: vitamin C sup-plementation group (n: smokers = 10, nonsmokers = 10), vitamin E supplementation group (n: smokers = 10, nonsmokers = 10), vitamin C and E supplementation group (n: smokers = 10, nonsmokers = 10), respectively. There were no significant differences between the smokers and nonsmokers in terms of anthropometric measurements. Systolic and diastolic blood pressure were significantly higher (p < 0.05) in smokers than that of non-smokers. There was no significant difference in energy and other nutrients intakes between smokers and non-smokers. In plasma lipids levels, smokers had higher plasma triglyceride, LDL-cholesterol, VLDL-cholesterol, total cholesterol concentration than that of non-smokers (p < 0.05). HDL-cholesterol level of smokers had a tendency to be lower than that of non-smokers. In smokers, AI, TPH, LPH were significantly higher than that of non-smokers (p < 0.01). Plasma folate, homocysteine levels were not significantly different between smokers and non-smokers. The effect of antioxidant vitamins supplementation in smokers: In vitamin C supplementation group, HDL-cholesterol level was significantly in-creased (p < 0.01) and AI, TPH, LPH were significantly decreased (p < 0.01). In vitamin E supplementation group, HDL-cholesterol level was significantly increased (p < 0.05). In vitamin C and E supplementation group, LPH was significantly decreased (p < 0.05). The effect of antioxidant vitamins supplementation in non-smokers: HDL-cholesterol level was significantly increased (p <0.05) and AI, TPH, LPH were significantly decreased (p <0.05) by vitamin C supplementation group. Plasma homocysteine level was decreased by vitamin E supplementation group in non-smokers (p < 0.01). The results of this study showed that smoking had a tendency to increase plasma lipids levels that factor into the risk of coronary heart disease. It is considered that antioxidant vitamin supplementation in smokers had a tendency to decrease cardiovascular disease than in nonsmokers.

The role of methylenetetrahydrofolate reductase C677T polymorphism on the peripheral blood natural killer cell proportion in women with unexplained recurrent miscarriages

  • Objective: To examine the association between methylenetetrahydrofolate reductase (MTHFR) C677T polymorphism and hyperhomocysteinemia in women with unexplained recurrent miscarriages (RM) and to investigate the association between MTHFR genotype variants and alloimmune activation, proportion of peripheral blood natural killer (pbNK) cells. Methods: A total of 39 patients with a history of two or more unexplained miscarriages were recruited to this study. The controls were women who had a live birth without a history of RM (n=50). The proportion of pbNK cells was measured by flow cytometry. Plasma homocysteine levels and the incidence of the MTHFR variant of the RM and control groups were compared. The proportion of pbNK cells was compared to the MTHFR variants in the RM group. Results: No differences were found between the two groups' mean plasma homocysteine levels ($7.6{\pm}1.5{\mu}mol$/L vs. $7.1{\pm}2.1{\mu}mol$/L) or incidence of the MTHFR genotype variant (CC, 35% vs. 33%; CT, 40% vs. 53%; and TT, 25% vs. 14%). In the RM group, individuals with the TT variant ($7.7{\pm}1.1{\mu}mol$/L) had higher homocysteine levels than those with the CC and CT variants ($7.4{\pm}1.9{\mu}mol$/L and $7.4{\pm}1.2{\mu}mol$/L) and those with the CT variant ($19.2{\pm}8.1%$) had a higher proportion of CD3-/CD56+ pbNK cells than those with the CC and TT variants ($17.7{\pm}6.6%$ and $17.9{\pm}7.0%$), but the results of both comparisons were statistically insignificant. Conclusion: These preliminary results show no difference in plasma homocysteine levels between the RM and control groups or among MTHFR genotype variants in the RM group, which may suggest that the plasma homocysteine level is difficult to use as a predictive marker of RM in the Korean population. A study of a larger number of patients is needed.

국화 기형화 발생과 S-adenosyl-L-homocysteine hydrolase 유전자 발현 (S-adenosyl-L-homocysteine hydrolase gene is down-regulated in abnormal flower inducing environment in chyrsanthemum)

  • 본 시험은 국화에서의 기형화 발생과 DNA 메틸레이션에 관여하는 S-adenosyl-L-homocysteine hydrolase (SAHH) 유전자 발현과의 관련성을 검토하기 위해 수행하였다. 스프레이 국화 'Lerbin'은 단일후 14일에서 27일 사이에 고온과 장일 조건에서 기형화가 발생되어, $35/20^{\circ}C$의 고온과 14시간의 장일 처리 할 경우 $25/20^{\circ}C$(12 h/12 h)에 비해 설상화가 2배 이상 증가하는 양상을 보였다. 스프레이 국화 'Lerbin'에서 분리한 전장 cDNA (DgSAHH)는 크기가 1455 bp로 다른 작물과 90%이상의 높은 상동성을 나타내었다. 국화의 DgSAHH 유전자 발현은 기형화 발생을 유도하는 고온과 장일 조건에서 크게 감소하였다. 이러한 결과를 통해 국화의 화아 발달에 있어서 DgSAHH 유전자 발현은 온도와 일장의 영향을 받으며, 억제된 이 유전자의 발현이 기형화 발생에 관여할 수 있을 것으로 판단된다.

임신초기 모체의 엽산영양상태와 동 영향인자 (Maternal Folate Status and Its Influencing Factors in Early Pregnancy)

  • 임신초기 모체의 엽산영양은 태아의 신경관 손상을 예방하는 측면에서는 물론이고 엽산 필요량이 증가하는 임신후기를 대비한다는 점에서도 중요하다. 그러므로 본 연구에서는 임신초기에 있는 151명의 임신부들을 대상으로, 이들이 식사를 통해 얼마의 엽산을 섭취하는지를 조사하고, 혈청과 적혈구의 엽산 농도와 혈장 호모시스테인 농도를 분석하여 엽산영양상태를 평가하고, 이들의 엽산영양상태에 영향을 미치는 인자들이 무엇인지 분석하고자 하였다. 본 연구대상자의 조사당시 임신기간은 9.1$\pm$2.3주이었으며, 초산부가 46%이었고 나머지는 경산부이었다. 에너지 및 엽산 섭취량은 각각 1559$\pm$589 ㎉/day 및 230.8$\pm$145.2 $\mu$g/day로 각 권장량의 72.5%및 46.2%로 낮았다 혈청과 적혈구의 엽산 농도는 각각 5.5$\pm$1.9 ng/mL 및 266.6$\pm$75.0 ng/mL으로, 연구 대상자의 7.8%가 혈청 엽산 수준이 3 ng/mL미만인 결핍상태에 해당하였고 60.3%는 경계역(3∼5.9 ng/mL)에 속하였으며, 적혈구 엽산 농도가 l57 ng/mL미만인 결핍상태에는 4.3%가 해 당하였다. 그러나 혈청 호모시스테인 농도는 정상범위이었다. 따라서 본 연구대상자의 엽산영양상태는 비교적 양호하다고 판정되었다. 혈청 엽산 농도에 영향을 끼칠 수 있는 유의한 인자는 건강상태점 수(+), 혈장 호모시스테인 농도(-), 연령 (-), 임신기간(-) 및 엽산 섭취량(+) 순으로 그 영향력이 높았고, 적혈구 엽산 농도의 영향인자는 교육수준(-), 혈장 호모시스테인 농도(-), 지난번 출산 영아의 출생시 신장(+),연령(-)및 에너지 섭취량(+)이었고, 혈장 호모시스테인 농도의 영향인자는 지난번 출산 영아의 출생시 신장(-), 경제수준(+),혈청 엽산 농도(-)와 임신전 체중(+)이었다. 이러한 결과는, 혈장이나 적혈구의 엽산 농도나 혈장 호모시스테인 농도를 분석하지 않고도, 임신초기에 있는 모체의 엽산영양상태를 개략적으로 판정하는데 있어 주관적으로 평가한 불량한 건강상태, 높은 교육수준 또는 경제수준, 지난번 출산에서 작은 체위의 영아를 출산한 경력, 고연령 또는 엽산이나 에너지섭취량 부족을 위험 인자로 활용할 수 있음을 시사한다. 아울러 임신기간이 진행되었을수록 엽산영양상태가 저하되었을 가능성이 있음도 나타내준다. 앞으로 임신초기 임신부의 엽산영양상태를 어떤 위험인자를 몇 가지나 가졌느냐에 따라 평가하는 연구결과가 나와야 이들 위험 인자의 실용성 을 확인할 수 있을 것이다. 그러나 우선 이들 위험 인자를 많이 지닌 임신부라면 임신초기부터 엽산 섭취량을 늘리거나 엽산보충제 섭취를 일찍 시작하는 등의 방법으로 엽산영양상태를 양호하게 유지하도록 하는 것이 바람직할 것이다.
