3차원 Co-culture 시스템을 통한 BMSC의 NP-like Cell로의 분화 (The Differentiation of bone Marrow Stromal Cells into NP-like Cells through 3-Dimensional Co-culture System)

  • The goal of this study is to investigate the effect and potential of three-dimensional Co-culture of BMSCs (bone marrow stromal Cells) and NP (nucleus pulposus) Cells on the differentiation of BMSCs into NP-like Cells. The NP Cells and BMSCs were isolated and cultured from New Zealand White rabbits. The isolated NP Cells and BMSCs were prepared in different alginate beads. Those two types of beads were separated by a track-etched membrane of $3\;{\mu}m$ pore in a 6-well culture plate. No growth factors were used. In addition to these, NP and BMSC were cultured in the beads independently for control. The number of Cells in Co-culturing system was half of those in two control groups. Proliferation and production of glycosaminoglycan (GAG) were evaluated along with histological observation. The GAG production rate(GAG contents/Cell) of Co-cultured BMSCs were much higher than that of BMSCs cultured alone. The total amounts of GAG produced by BMSCs in Co-culturing system were larger than those produced by BMSCs in control group and were comparable with those produced by NP alone even the number of each Cell was half of BMSCs in Co-culturing system. This study showed the potential of differentiation of BMSCs into NP-like Cells through three-dimensional Co-culture system even without any chemical agents.

AFC용 Raney 니켈 촉매의 크롬 첨가 특성 (Characteristics of Chromium Addition on Raney Nickel Catalyst for AFC)

  • 크롬의 첨가에 따른 Raney 니켈의 특성을 파악하여 우수한 성능을 나타낼 수 있는 알칼리형 연료전지용 수소극의 촉매를 제조하였다. 또한 제조된 촉매를 이용, 실제 전극으로 제작하여 전류밀도를 측정한 뒤 최적의 전극재료의 제조조건을 확립하였다. 먼저 Ni-Al합금 제조시 크롬의 첨가량을 달리하여 Raney 니켈을 제조한 뒤 이를 이용하여 수소극을 제작, 전기화학적 특성을 고찰한 결과 크롬 첨가량이 증가함에 따라 mass activity도 증가하여 크롬 첨가량이 1 wt% 인 경우 최고 3.688 A/g을 나타내었다. 전기화학적 측정시 half cell의 운전온도는 8$0^{\circ}C$이었으며 전해질은 6N KOH이었다. Raney 니켈 촉매의 입자크기와 비표면 적을 조사한 결과 크롬의 첨가량이 증가함에 따라 Raney 니켈 촉매의 입자크기는 12.11 $\mu\textrm{m}$에서 크롬의 첨가량이 1.25 wt%인 경우 11.07 $\mu\textrm{m}$로 감소하였고 비표면적은 0.653 $\m^2$/g에서 0.685 $\m^2$로 중가함을 알 수 있었다. 평균입자 크기가 커짐에 따라 잔존 알루미늄의 함량은 증가함을 알 수 있었다.

Bioluminescent Determination of Lactose Secretion: A Measure of the In Vitro Performance of Mammary Acini from Lactating Rats

  • A culture system for lactating rat mammary acini was evaluated, where the primary indicator of performance was lactose secretion, measured by a sensitive bioluminescence assay. Lactose secretion was reduced by half (p<0.01) over the first 6 h of culture by overnight feed withdrawal (FW) from tissue donors but was sensitive to increased glucose concentration in the culture media (p<0.001) up to 30 mM. Lactose production of cells from fed donors over the first 6 h in culture in 30 mM glucose was 8.9 fmol/cell/h - a rate calculated to be about half that in vivo. No significant difference was shown in lactose secretion by cells from fed or FW rats over 6-24 h. Lactose secretion was 3.6 fmol/cell/h by cells from fed animals in 40 mM glucose concentration media over the 6-24 h culture period. Addition of insulin to the culture media had no effect on rates of lactose secretion while addition of prolactin and hydrocortisone, with or without insulin, significantly (p<0.001) decreased lactose production over both 0-6 h and 6-24 h culture periods. Lactose synthesis in vitro was significantly enhanced by aeration of the media during collagenase digestion of mammary tissue (p<0.05). No improvement in lactose secretion was effected by shaking of cells during culture, Matrigel coating of culture dishes or change in cell density over a range up to 2.5 million cells per ml.

Establishment of a novel plant regeneration system from suspension-derived callus in the halophytic Leymus chinensis (Trin.)

  • The establishment of cell suspension culture and plant regeneration of the halophytic Leymus chinensis (Trin.) are described in this study for the first time. Callus induction solid medium containing Murashige and Shoog (MS) basic salt, $2.0\;mg\;l^{-1}$ 2,4-dichlorophenoxyacetic acid (2,4-D), and $5.0\;mg\;l^{-1}$ L-glutamic acid with $30.0\;g\;l^{-1}$ sucrose and $4.0\;g\;l^{-1}$ gelrite for solidification induced the highest rate of cell division in Type 1 callus among calli of various types. Liquid medium with the same hormone distribution was therefore, used for cell suspension culture from Type 1 callus. Over a 30 d suspension culture at 100 rpm, great amounts of biomass were accumulated, with 71.07% average daily increment and 22.32-fold total fresh weight increment. Comparison of before and after suspension culture, the distribution of different size callus pieces and the maintenance of callus type were basically unaltered, but a slight increase in relative water contents was observed. To induce the potential of plant regeneration, the directly transferring on plant regeneration solid medium containing MS basic salt, $0.2\;mg\;l^{-1}$ $\alpha$-naphthalene acetic acid (NAA), $2.0\;mg\;l^{-1}$ kinetin (Kn), and $2.0\;g\;l^{-1}$ casamino acid and indirectly transferring were simultaneously performed. Even now growth rates of suspension-derived callus on solid medium were approximately half of those of Type 1 callus, but faster somatic embryogenesis was observed. Rooting of all regenerated shoots was successfully performed on half-strength MS medium. All plants appeared phenotypically normal.

와편모조류 Alexandrium affine의 생장에 미치는 용존태 무기 및 유기 영양염(질소와 인)의 영향 (Effects of Dissolved Inorganic and Organic Nutrient (Nitrogen and Phosphorus) on the Growth of Dinoflagellate Alexandrium affine)

  • 본 연구에서는 와편모조류 Alexandrium affine(LIMS-PS-2345)의 생장에 미치는 용존태 무기 및 유기 영양염의 영향을 조사하였다. 영양염 흡수 동력학 실험에서 A. affine의 최대흡수속도(ρmax)와 반포화상수(Ks)는 질산염에서 77.0 pmol/cell/hr과 17.6 μM, 인산염에서 15.5 pmol/cell/hr과 3.88 μM로 산출되어, 무기영양염에 대하여 높은 요구량 및 낮은 친화성을 가지고 있는 것으로 나타났다. 유기 영양염에 따른 A. affine의 생장속도를 확인한 결과, 유기 질소 urea, glycine와 유기 인 adenosine triphosphate(ATP), glycerol phosphate(Glycerol-P) 첨가구에서 무기 영양염 첨가구의 70 % 이상 생장속도를 보였다. 따라서 낮은 무기 영양염 환경에서 A. affine의 우점화와 종간경쟁에서 우위를 위해서는 용존태 유기 영양염의 이용이 필요할 것으로 생각된다.

Some of the Food Color Additives Are Potent Inhibitors of Human Protein Tyrosine Phosphatases

  • Synthetic color additives approved for general food use are sixteen in European Union, seven in U. S. A. and twelve in Japan. Twelve food dyes were examined for their inhibitory potency against human protein tyrosine phosphatases (PTPases). Half of the food colorants inhibited PTPases significantly and three of them were potent inhibitors with low micromolar IC50 values. Also examined were the synthetic dyes structurally similar but not allowed in food. Some of them were potent inhibitors of PTPases. Considering the importance of PTPases in cellular signal transduction, inhibition of PTPases by food colorants might cause harmful effects in human health.

연료전지발전용 부스터의 출력전압제어 연구 (Control of Booster Output Voltage in Fuel Cell Power Plant)

  • Booster is used widely as one of the step-up DC/DC power converter in power conversion process for fuel cell power plant which have the electrical characteristic of the high current density and low cell voltage. In view of control system, booster can be unstable when it is operated in broad operation range because the transfer function of booster has zero in right half plane of s-domain. So for reliable operation, controller must make the system stable in whole working range. In this paper, the two control method such as digital PID control and fuzzy control is studied for booster output voltage regulation in fuel cell plant. The design procedure of PID control and fuzzy control is described. And the experiment of designed controller action is performed in various operation points for controller performance test.

EEPROM 셀에서 폴리실리콘 플로팅 게이트의 도핑 농도가 프로그래밍 문턱전압에 미치는 영향 (Effects of Doping Concentration in Polysilicon Floating Gate on Programming Threshold Voltage of EEPROM Cell)

  • We have investigated the effects of doping concentration in polysilicon floating gate on the endurance characteristics of the EEPROM cell haying the structure of spacer select transistor. Several samples were prepared with different implantation conditions of phosphorus for the floating gate. Results show the dependence of doping concentration in polysilicon floating gate on performance of EEPROM cell from the floating gate engineering point of view. All of the samples were endured up to half million programming/erasing cycle. However, the best $program-{\Delta}V_{T}$ characteristic was obtained in the cell doped at the dose of $1{\times}10^{15}/cm^{2}$.

MOCVD를 이용한 GAs/Si 태양전지의 제작과 특성에 관한 연구 (A Study on Fabrication and Properties of the GaAs/Si Solar Cell Using MOCVD)

  • In this paper, the current status of manufacturing technologies for GaAs/Si solar cell were revived and provied new MOCVD. In the manufacturing process of GaAs/Si solar cells and an experiment to get the high efficiency GaAs solar cells, we must investigate the optimum growth conditions to get high quality GaAs films on Si substrates by MOCVD. The GaAs on Si substrates has been recognized as a lightweight alternative to pure substrate for space applicaton. Because its density is less the half of GaAs or Ge.So GaAs/Si has twofold weight advantage to GaAs monolithic cell. The theoretical conversion efficiecy limit of tandem GaAs/Si solar cell is 32% under AM 0 and $25^{\circ}C$ condition. It was concluded that the development of cost effective MOCVD technologies shoud be ahead GaAs solar cells for achived move high efficiency III-V solar cells involving tandem structure.

아동용 휴대전화 중독 도구 개발 및 타당화 연구 (Development and Validation Study of a Cell Phone Addiction Scale for Korean Children)

  • Purpose: This study was done to develop a cell phone addiction scale for Korean children. Methods: The process included construction of a conceptual framework, generation of initial items, verification of content validity, selection of secondary items, preliminary study, and extraction of final items. The participants were 268 children from grades 4 to 6 in two urban elementary schools. Construct validity, internal consistence reliability, and split-half reliability were used to analyze the data. Results: Nine items were categorized into 2 factors explaining 61.29% of total variance. The factors were labeled as obsessive compulsion (6 items) and dependency (3 items). Cronbach's alpha coefficient for the 9 items was .87, and Spearman-Brown coefficient was .83. Scale scores identified children as high risk users, at risk users, or average users by standard scores. Conclusion: The above findings indicate that the cell phone addiction scale for children has good validity and reliability when used with Korean children.
