Cultivation of Paecilomyces tenuipes using Mini-kit, small culture container (소규모 재배상을 이용한 생동충하초 재배)

  • Cordyceps and its allies fungi has been described as a secret medicine that gives eternal youth and a long life. Some species of Cordyceps are sources of biochemicals, such ascordycepin, with interesting biological and pharmacological properties. Hence, it has been studied to uncover its pharmacological effect. We attempted to study the formation of fruiting bodies and to develop means of mass production Korean isolate of Paecilomyces tenuipes has been inoculated into silkworms, where it reproduced using culture container, mini-kit successfully. Culture container, mini-kit is composed of a cylinder-shaped body and lid. The container is made of translucent polyethylene terephthalate. The size of the container is $82{\times}75mm$, reduced by 10 times as compared with the conventional culture kit. The mini kit has many advantages - high culture amount, ability of maintaining optimal humidity, parasite-free cultivation and high-end appearance. With the kit, the optimal cultivation condition is under $22^{\circ}C$, culture period of 53 days. And synnemata of P. tenuipes could be kept fresh for 14 days at the temperature of under $10^{\circ}C$. Therefore, the Min-kit can be used in both ways as a culture container and a packing kit for end-user customers.

Development of New Strains of Wolfiporia cocos for Sclerotium Formation by 2-Way Cross-Breeding (이원교배에 의해 균핵 형성하는 복령 균주 개발)

  • Wolfiporia cocos is an important medicinal fungus that has been used in regions of Northeast Asia including Korea, Japan, and China. W. cocos is classified in Korea into two types (red bokryeong and white bokryeong) based on the internal colors (yellow orange-pale pink and white) of the sclerotium. Generally, the W. cocos type cultivated on farms produces white sclerotium. In this study, we endeavored to select strains that form sclerotium in sawdust medium using 2-way cross-breeding among two cultivated strains and three wild strains. Monospores were isolated from the fruiting bodies of cultivated and wild strains on potato dextrose agar. Thirty-nine strains of 338 hybrid strains isolated formed sclerotia with white or yellow colors upon culture for 3 months in Pinus densiflora sawdust medium. Selection for sclerotium forming strains using sawdust culture follows a very simple and easy procedure that is presented for the first time in this paper. We plan to test selected strains in the field to aid in developing new varieties for the future.

Selection of Functional Lentinula edodes Strains with High Ergothioneine Content using Mono-mono Hybridization (단핵균주간 교잡에 의한 에르고티오네인 함량이 높은 기능성 표고 균주 선발)

  • Lentinula edodes have been cultivated extensively not only in South Korea but also in other Asian countries. In terms of taste and nutrition, it is a valuable mushroom that is comparable to other mushrooms. L. edodes contains various physiologically active compounds that promote human health. One compound, ergothioneine, exerts a powerful antioxidant function that inhibits cellular senescence. As it is not biosynthesized by plants or animals, L. edodes is very important for ergothioneine supply. The L. edodes cultivars Bambithyang (NIFoS 3404) and Sansanhyang (NIFoS 3420) of the National Institute of Forest Science (NIFoS) had higher ergothioneine content than the other cultivars, and 11 strains were obtained using mono-mono hybridization between them. The strains were inoculated into sawdust media to obtain fruiting bodies, and the ergothioneine content of each hybrid strain was confirmed using high-performance liquid chromatography (HPLC) analysis. Among the 11 strains, NIFoS 5108 had a higher ergothioneine content than the parental strains, Bambithyang (NIFoS 3404) and Sansanhyang (NIFoS 3420). This study revealed that it is possible to develop cultivars with improved specific functions using mono-mono hybridization.

Development of a safe culture technique for Agrocybe cylindracea (버들송이버섯 봉지재배 안정생산 기술)

  • Agrocybe cylindracea was cultured in a bag, in which sawdust culture medium (1 kg) is put in a plastic bag (PE), with poplar sawdust, rice bran, wheat bran, and dried bean curd refuse in the ratio of 70:10:10:10 (v/v). 2% of the culture medium was inoculated with the liquid starter of Agrocybe cylindracea, and this was incubated at $25^{\circ}C$. After incubation, the A. cylindracea was further cultured by cutting the top vinyl portion of the bag down to the level of the culture medium surface of the first inoculation part. The cut culture medium was placed in a growth room at $25^{\circ}C$, and pin-heading was induced under light irradiation at 99% humidity and 1,000 ppm $CO_$ level for 3days. When the grow the environment was controlled at 95% humidity and $21^{\circ}C$, the bending of the stem was less as compared to that when the cap of the bag had been removed. The number of effective fruiting bodies per bag increased by 140% (28.8), the quantity per bag increased by 29.5%, and 148.5 g A. cylindracea could be potentially harvested.

Comparison of the cultural characteristics and productivity of Lentinula edodes cultivated in different types of containers (재배용기별 표고의 배양특성 및 생산성 비교)

  • This study aimed to develop a new method of Lentinula edodes cultivation to reduce labor and waste disposal. To identify the optimum container for L. edodes cultivation, box, bottle, and bag containers were assessed for their effects on the cultural characteristics and mushroom yield. Among the three approaches, box-cultivation with 7 kg of substrate had the shortest browning period (42 days) and resulted in the highest mushroom yield (1,253 g). Contrastingly, bottle-cultivation was unsuitable for cultivation due to low production of the fruiting bodies and poor quality. Further studies on the box-cultivation approach will be required for commercial applications of L. edodes.

Sawdust Substitution in Growth Medium of Oyster Mushroom for Using Its By-product Spent Mushroom Substrates as Ruminant Feed (수확 후 배지의 가축 사료화를 위한 느타리 생육배지 톱밥 대체재료 선발 연구)

  • To replace the sawdust in the growth medium of oyster mushroom to utilize its by-product spent mushroom substrates (SMS) as feed for ruminant, we performed cultivation test using cotton seed hull pellet (CSHP), corn stalk pellet (CSP), corncob (CC), and analyzed the feed chemical properties of those SMS. As a result of cultivation test, CC and CSHP treatment took 27 days for spawn run, 4 days for primordium formation, and 3 days for development fruiting bodies, resulting in a total cultivation period of 34 days. The yield per bottle was 134 g for CC treatment, similar to 130 g for control, while CSHP treatment (112 g) and CSP treatment (68 g) were lower than that of control. The highest biological efficiency (BE) was shown in CC treatment as 80.1%, which was 11.4% higher than 68.7% of control. The SMS of CC treatment had a relatively low content of neutral detergent fiber and acid detergent fiber, and in particular, lignin content was the lowest and crude protein content was the highest among other treatments. Therefore, CC as a substitute material for sawdust was capable of stable mushroom production and excellent nutritional value as a feed for its by-products.

Effect of LED LightIrradiation on the Mycelial Growth and Fruit Body Development of Hypsizygus Marmoreus (LED 광원이 느티만가닥버섯 균의 균사 생장과 자실체 생육에 미치는 영향)

  • A edible mushroom, Hypsizygus marmoreus is commercially cultivated. However, the researches of cultivation and physiological characteristics were not conducted in Korea. In this study, we conducted on artificial cultivation of H. marmoreus and elucidated the effect of light on the mycelial growth and fruit body development using LED light sources with different wavelength; blue (peak wave length 460nm), green(peak wave length 530nm), yellow(peak wave length 590nm), red(peak wave length 630nm), and white as positive control. Mycelial growth of H. marmoreus strains were inhibited about 30~40% in inhibition ratio under the illumination with blue, green, yellow LED light. However, red LED light was not inhibited. Elongation of stipe was effective under the long wave length such as yellow and red light. Abnormal fruit body was produced under the long wavelength and dark. However, development of pileus was effective under the short wavelength such as green and blue light. Also, as a result of cultivation with mixed light for high quality and harvest, many effective numbers and yields of fruiting bodies were obtained in the mixed treatment of blue and white light, and pileus developed well.

Growth characteristics of oyster mushroom following the addition of wheat straw pellet as a substitute for beet pulp (비트펄프 대체 밀짚펠렛 첨가에 따른 느타리 생육 특성)

  • The present study investigated the optimum additive ratio of wheat straw pellet as a substitute for beet pulp during oyster mushroom cultivation. The chemical properties across treatments were pH of 4.8-5.5, total carbon content of 45.9-46.5%, total nitrogen content of 1.5-1.7%, and carbon-to-nitrogen ratio of 27.8-31.0. Mycelial growth was the fastest in a medium containing 20% wheat straw pellet (at 88.2 mm in 'Heuktari' and 70.3-79.6 mm in 'Suhan-1Ho'); however, there were no significant differences in mycelial density among the treatments. The characteristics of fruiting bodies by variety were as follows. In 'Heuktari', the valid stipe number per bottle (1,100 cc) was 22.5 and yield was 177.1 g/1,100 cc in the mixed medium containing 10% wheat straw pellet, with the values being comparable to those of controls. In 'Suhan-1Ho', the valid stipe number per bottle (1,100 cc) was 14.0 and yield was 151.2 g/1,100 cc in the mixed medium containing 10% wheat straw pellet, with the values being comparable to those of controls.

  • The objective of this study was to achieve biological control of green mold disease in Pyogo mushrooms using antagonistic microorganisms. Bacillus subtilis BSM320 cells inhibited mycelial growth by 48-60% against three Trichodermaisolates including T. hazianumisolated from the substrates of Lentinula edodes, showing their antifungal activity.The bacteria were cultured to a high density of 4.2 × 109±113.7 cfu/mlin aqueous extract of composted spent mushroom substrates of L. edodes containing 1% glucose and showed a higher growth rate than that observed when using the commercial medium, Luria-Bertani broth. The bacterial culture showed a 75% protective effect without damaging the mushroom fruiting bodies. These results suggest that B. subtilis BSM320culture is suitable for biological control of green mold disease during mushroom cultivation.

Vertical Transmission of RNA Mycoviruses in Lentinula edodes (표고에서의 RNA 바이러스 수직감염)

  • Lentinula edodes is an important commercial mushroom and there have been several reports of viral infections in L. edodes. Two mycoviruses (LeV-HKB and LeNSRV1) were detected in Sanbaekhyang (NIFoS 2778) and Taehyanggo (NIFoS 4317), the sawdust-cultivated commercial strains. The vertical transmission rates of the viruses were investigated by detecting the viruses in 80 monokaryotic strains derived from basidiospores isolated from the fruiting bodies of each strain. Most of the monokaryotic strains were infected with the virus and the two viruses showed different levels of meiotic stability, with LeV-HKB showing higher meiotic stability than LeNSRV1. Therefore, it seems that the vertical transmission mechanism of mycoviruses is different depending on the virus species. We also examined the mycelial growth rate of the monokaryotic strains and compared the growth rate according to virus infection status. Although there was no statistically significant correlation between viral infection and mycelial growth rate, we found that the average growth rate was reduced by additional virus infection. We expect our data to contribute to a greater understanding of the mechanism of the vertical transmission of mycoviruses, and promoting breeding using virus-free monokaryotic strains.
