Impacts of stream water quality and fish histopathology by effluents of wastewater treatment plant (하수종말처리장 배출수에 의한 하천 수질 특성 및 어류의 조직병리학적 영향)

  • In this study, the histological changes of Zacco platypus exposed to discharge from a Wastewater Treatment Plant (WTP), which is a point source, for a long time (2 to 3 years) were compared to the same species at a reference site (Ref.). Overall, tissues displayed various lesions in samples obtained at the point where discharge water from the point source was mixed. In the skin tissue, mucus cells from the epithelial layer expanded to the multilayered epithelium, indicating the immune system was activated. Epithelial cell detachment and proliferation were most prominent in the gills, which may have adversely affected circulation and respiration. Our data suggest immune system collapse was due to stimulation by aquatic substances. Both the fish phase analysis and the water quality analysis demonstrated depreciated conditions at the point source as compared to the reference stream, supporting the histological health evaluation results. These data together suggest a histological approach can also be used to assess water quality, and to an even higher degree when combined with other existing methods. Given the presented evaluation, improvement in the water quality of water discharged from WTP's is required.

Accumulation, Elimination and Cell Response in the Kidney of Juvenile Rockfish (Sebastes schlegeli) Exposed to Dietary Cadmium

  • Experiments were carried out to investigate Cd accumulation, elimination and cell response in juvenile rockfish (Sebastes schlegeli) exposed to sub-chronic dietary Cd (0, 0.5, 5, 25 and 125 mg/kg) for 60 days and depuration periods of 30 days. Cd accumulation in the kidney of cock fish increased with exposure periods and concentrations for the 60 days of dietary Cd exposure. After the end of the dietary Cd exposure, Cd accumulation values in the kidney were $52.9{\pm}9.94\;{\mu}g/g$ and $90.6{\pm}15.7\;{\mu}g/g$ for those exposed to 25 mg/kg and 125 mg/kg Cd, respectively. The accumulation factors increased with the exposure period in the kidney. Cd elimination in the kidney of rockfish did not vary significantly and remained constant after the cessation of the dietary Cd exposure. In the primary exposure periods, the effect of kidney tissue in the rockfish exposed to dietary Cd was observed the swelling of capillary of the glomerulus. In addition, there was also hydropic swelling within the pyknotic nuclei, some of hyaline droplet accumulation and the microvilli showed a positive reaction to alcian blue in the tubular cells. While exposure time and concentrations were increased, there was a lot of hyaline droplet accumulation and the microvilli showed a positive reaction to alcian blue in the tubular cells. Fused renal tubule and its necrosis were observed after 60 days at l25 mg/kg.

Histological Response of Kidney, Gill and Hepatopancreas of the Juvenile Olive Flounder, Paralichthys olivaceus after PCBs Exposure (PCBs 노출에 따른 넙치(Paralichthys olivaceus) 치어의 신장, 아가미 및 간췌장의 조직학적 반응)

  • The effect on kidney, gill, and hepatopancreas of juvenile olive flounder, Paralichthys olivaceus were investigated by histological method under limit concentration of effulent of PCBs for 60 days. The free surface of epithelial cell layer in the renal tubules of kidney showed a strong positive reaction in alcian blue during all exposure period. There were also observed swelling of hemocyte in glomerulus and macrophage. At 60 days after exposure, epithelium of the renal tubules was fused and some of organism $(20\%)$ showed damage of glomerulus and eosinophlic cell in epithelium of the renal tubules. The phenomena such as the activation and increase of the chloride cell, and swelling of hemocyte in the capillary of gill were observed during all exposure period. At 60 days after exposure, swelling of the gill filament and mucous cell appeared in gill and some of organism $(30\%)$ showed swelling of the gill lamellar. Swelling of hemocyte in the capillary of hepatopancreas was observed and the free surface of epithelial layer of the bile duct showed positive reaction in alcian blue during all exposure period. Degenerated zymogen in the pancreas and swelling of the hepatocyte were occurred at 60 days after exposure.

Effects of Acute Toxicity of Chemical Treatments on the Cultured Oliver Flounder, Paralichthys olivaceus (화학제 처리가 성장기 넙치 (Paralichthys olivaceus)에 미치는 급성독성 효과)

  • This study was carried out for the purpose of developing environmental friendly and effective chemical treatment method for the disease control in the land-based flounder culture which is industrially popular in the coastal area in Korean. The chemicals such as flounder, Paralichthys olivaceus and their effects on the fish based on the 24hr-$LC_{50}$, $LT_{50}$, 24-hour survival rate at each experimental concentration, recovery rate of the survived individual from chemical treatment, and the histological change of the gill after chemical treatment were investigated and analyzed. The 24hr-$LC_{50}$ was 321.65 ppm for formalin, 419.62 ppm for chlorine dioxide, and 395.97 ppm for hydrogen peroxide, respectively. The $LT_{50}$ was 15-hour for formalin, 17-hour for chlorine dioxide and 24-hour for hydrogen peroxide, respectively. Fishes exposed to the experimental concentration of three chemicals were quickly susceptible in the order of formalin, chlorine dioxide and hydrogen peroxide with a trend of shorter half lethal time at higher concentration. Initial survival rate of the flounder soon after chemical treatment was the highest in the hydrogen peroxide treatment compared with the other two chemicals. The histological damage by the hydrogen peroxide treatment was negligible compared with the other two chemicals. Accordingly, hydrogen peroxide treatment showed the lowest toxicity compared with the other two chemicals to the experimental fishes.

Quality Improvements of Breast Specimen Fixation for Histopathology Test (병리검사를 위한 유방검체 고정의 질 향상 활동)

  • To improve accuracy of the immunohistochemical testing and fluorescence in situ hybridization (FISH) study as well as a routine histology diagnosis in breast cancer, quality improvement for optimal tissue handling is mandatory. We evaluate fraction defective of 7107 blocks from 349 breast cancer patients, who underwent surgical treatment at Samsung Medical Center Seoul, Korea from January 1, 2009 to March 31, 2010. We decided pre-improvement period from January, 2009 to June, 2009. In the first quality improvement period (July, 2009 to September, 2009) we made improvements in protocol of gross examination. In the second quality improvement period (October, 2009 to December, 2009) we attempted more effective formalin fixation such as frequent exchange of formalin and use of separate fixation container for each case. In the third quality improvement period (January, 2010 to March, 2010) improvement of tissue processor was performed. We achieved a marked reduction of fraction defective (9-16%) through efforts to improve quality of formalin-fixed, paraffin-embedded blocks when compared to pre-improvement period.

A Case Study of Mortality Caused by Viral Encephalopathy and Retinopathy (VER) in Cultured Sevenband Grouper, Epinephelus septemfasciatus during Winter (겨울철 양식 능성어의 바이러스성뇌망막증(VER) 감염사례)

  • In 2015, a nervous necrosis virus (NNV) was isolated from sevenband grouper, Epinephelus septemfasciatus, maintained in land-based aquaculture system at below $12^{\circ}C$ in winter. Mortality was up to 30% in brood fish, over 4 kg of body weight. Moribund fish showed clinical sings typical of viral encephalopathy and retinopathy (VER), also called viral nervous necrosis (VNN), such as uncoordinated, corkscrew-like swimming behavior, belly-up at rest, darkening of body, cloudy eyeball and hyperinflation of the swim bladder. Aetiology of the disease was confirmed by gross observation of clinical signs, histopathology and molecular diagnosis. Histological studies revealed severe vacuolation and necrosis in the brain. Molecular diagnosis by revere transcription-polymerase chain reaction (RT-PCR) specific to batanodavirus yielded a positive result. The nucleotide sequences of the PCR-amplified fragment were 99.48~100% similar to barfin flounder nervous necrosis virus (BFNNV) genotype and most closely aligned with Pacific cod betanodavirus (PCNNV). This is the first report of natural batanodavirus, NNV infection in sevenband grouper reared in low water temperature during winter (below $12^{\circ}C$) in Korea.

Effect of Disease Resistance on Oral Administration of Lightyellow Sophora Extract in Olive Flounder (생약제 고삼 뿌리 열수추출물의 넙치 투여시 질병 저항성에 미치는 영향)

  • The effects of lightyellow sophora, Sophora flavescens extract were tested on non-specific immune response and a disease resistance of olive flounder, Paralichthys olivaceus. For feeding trial, weight gain of fish fed different treated groups were not significant differences. Serum analysis of treated and control group did not demonstrated any differences. Lysozyme activity in the kidney and spleen of the 0.05% treated group on $4^{th}$ and $8^{th}$ week showed significant increase, respectively. In addition, phagocytic activity of the 0.05% treated group on $4^{th}$ and 8th week showed significant increase compared to the control group. Histopathology of the liver and kidney tissue of the administrated group for $4^{th}$, $8^{th}$ and $12^{th}$ week showed no particular signs of tissue degeneration. The treated group was higher than control group by analyzing the relative percent survival (RPS) of the experimental challenge of Edwardsiella tarda. Therefore, the lightyellow sophora showed effective in preventing fish-pathogenic edwardsiellosis for a certain period of time.

  • A flowerhorn fish with a subopercular mass, which had been reared in one of the private commercial aquaria was sent to the laboratory just before death. This fish began to show symptoms such as lethargy, anorexia, depression and dyspnea, and eventually died. The mass and intestinal organs were examined histopathologically. Thyroid showed variably sized follicles that contained abundant colloid and liver revealed steatosis. This report presents a case of goiter in the flowerhorn cichlid and its effect on the liver.

  • To understand responses of bivalve tissues to various injurious agents, histopathological observation was done in the Manila clam, Ruditapes hilippinarum, during 240 hours after an intramuscular injection of N-methyl-N-nitro-N-nitrosoguanidine (MNNG). Some histopathological changes were observed with the epithelia of stomach, midgut, hindgut, digestive diverticula and gonads after injecting MNNG. The earliest degenerative changes were recognized in the epithelium of midgut 12 hours after the injection. Partial or complete, epithelial destruction was constantly accompanied by the massive infiltration of a mononuclear, necrotic cell group including hemocytes. At 144 hours later, gonads showed the necrotic desquamation of their germinal epithelia. Nearly all of the infiltrated cells within the destructive epithelium were suggested to be hemocytes, from the binding property with a hemocyte-specific lectin, Ricinus communis (RCA-1). From these results it was concluded that the epithelium of digestive system is vulnerable to parenteral MNNG and hemocytes might be deeply involved in this MNNG-induced destruction of the digestive epithelium.

Histopathological features of pacific whiteleg shrimp, Litopenaeus vannamei, infected with Infectious Myonecrosis Virus (IMNV) with an emphasis on micro-traumas and inflammatory responses in muscle tissues (전염성근괴사증바이러스(IMNV)를 인위감염 시킨 흰다리새우(Litopenaeus vannamei)의 근육에서 나타난 미세 손상과 염증반응에 대한 조직병리학적 특성 연구)

  • We injected infectious myonecrosis virus (IMNV) to pacific whiteleg shrimp, Litopenaeus vannamei, and observed closely with using light microscope and transmission electron microscope (TEM) for 4-8 days post infection (dpi). As clinical signs, abdominal bodies had mild opaque muscles at 5 dpi. And the mortality was shown at 6 dpi. At 8 dpi, most injected shrimps had severe opaque muscles and humped back that cause of movement disorder. As results of histopathological examinations, local parts of abdominal body muscle had muscle fiber hyalinization, muscle fiber atrophy, rounded muscle fibers, myofibrillar hypertrophy in size, a decrease in number of myofibrils and phagocytosis from the sarcolemmas by multiple hemocytes at 4 dpi. Especially, myofibrillar hypertrophy appeared at the whole or random part of single muscle fiber not in specific locations like the center or edge of muscle fiber. At 6-7 dpi, multiple muscle necrosis, muscle fiber segmentation, myofibril lysis ap- peared and a few hemocytes were infiltrated at lesions. At 8 dpi, extensive muscle necrosis, multiple myofibril lysis and muscle fiber atrophy were shown, and very few hemocytes were infiltrated. In early stage of infection, local viral myositis with zenker's degeneration were shown. These lesions appeared multiply after the early stage. In late stage of infection, extensive coagulative muscle necrosis appeared with few of inflammatory response such as hemocytes infiltration. The lack of hemocytes infiltration response at the late stage might be disadvantage for Litopenaeus vannamei to defense against IMNV and to recover, because hematocytes (granulocyte, semi-granulocyte) eliminate pathogen and damaged tissues from infection sites and help recover. As results of the TEM observation, IMNVs that had nonenveloped icosahedral capsid which was 30-40 nm diameter were in myofibril and beside tubules of sarcoplasmic reticulum and moved to the certain direction. The micro-tears and micro-trau- mas in myofibrils caused muscle fiber necrosis. And semi-granulocytes engulfed IMNVs to eliminate virus.
