In order to develop a antimicrobial agent, dried Sophora flavescens Ait. was extracted with several solvents, and than antimicrobial activity was investigated. The minimal inhibitory concentration (MIC) of the extracted substance against microorganisms were also examined. Antimicrobial activity of the initial methanol extract from the sample was the strongest compared to those of other solvent extracts such as $H_2O$, n-hexane, chloroform, ethyl acetate and methanol. The methanol extract had strong growth inhibition activity against gram-postive bacteria (MIC, $25~50{\;}{\mu\textrm{g}}/ml$) such as B. subtilis. Among gram-positive bacteria tested, B. subtilis was the most susceptible to the extracted substance. The antimicrobial activity of the methanol extract from the sample had strong growth inhibition activity against gram-negative bacteria such as P. aeruginosa (MIC, $25{\;}{\mu\textrm{g}}$/ml).
산마늘 에탄올 추출물과 각종 유기용매 분획물에 대하여 항돌연변이원성과 세포독성을 검토하였다. 시료자체는 돌연변이원성이 없는 것으로 나타났다. 직접 변이원인 MNNG에 대한 항돌연변이 효과에서 S. typhimurium TA100 균주에 대해 산마늘 에탄올 추출물(200$\mu\textrm{g}$/plate)에서 88.2%의 억제효과를 나타내었다. 동일 시료 농도에서 4NQO에 대해서는 S. typhimurim TA98과 TA100 두 균주 모두에서 각각 76.4%와 83.0%의 비교적 높은 억제효과를 나타내었다. 암세포 성장 억제 효과를 검토한 실험에서는 에탄올 추출물이 다른 분획물보다 높은 억제활성을 나타내었다. 시료농도의 증가와 함께 억제활성도 증가하는 경향을 나타내었으며 시료농도 50 $\mu\textrm{g}$/plate에서 A549가 74.2%, MCF-7이 71.3% 그리고 KATOIII에 대하여 67.4%의 암세포 성장억제효과를 나타내었다.
유색미 쌀겨가 화학적 변이원에 의한 유전독성을 억제하는 작용기작을 유색미 품종인 수원415호 쌀겨의 유기용매추출물과 변이원인 mitomycin C를 이용하여 조사하였다. 70%에탄올 분획 및 클로로포름분획에서 수원 415호는 대조구로 사용된 추청에 비하여 변이원에 대한 억제활성이 월등히 높았음에도 불구하고 항산화활성은 추청보다 다소 낮은 결과로 보아 항산화작용과는 다른 억제기작이 존재함을 추정할 수 있었다. E. coli를 지시세포로 유색미 쌀겨 추출물의 mitomycin C에 대한 변이원성 억제기작을 조사한 결과, desmutagen으로 작용할 가능성이 크다는 사실을 알았고, 쌀겨 추출물과 mitomycin C를 반응시킨 후 반응액 중에 존재하는 유리상태의 mitomycin C를 정량한 결과, 유색미의 쌀겨 추출물이 mitomycin C에 직접 결합하여 변이원을 흡착함으로써 세포에 대한 유전독성을 억제할 것으로 생각되었다.
Stimulation of AMP-activated protein kinase (AMPK) signaling followed by increase of glucose uptake in L6 myotubes were studied with organic solvent extract of Malva verticillata (MV) seeds. Ethanol extract of M. verticillata seeds (MVE) significantly increased the phosphorylation level of AMPK, acetyl-CoA carboxylase (ACC), and glucose uptake in L6 myotube cells. The MVE was fractionated with n-hexane (MVE-H), chloroform (MVE-C), ethylacetate (MVE-E), n-butanol (MVE-B), and water (MVE-W). MVE-H (150 ${\mu}g$/ml) showed the highest phosphorylating activity and increased glucose uptake by 2.3-fold. Oral administration of MVE-H (40 mg/kg) for 4 weeks to type 2 diabetic (db/db) mice reduced non-fasting and fasting blood glucose levels by 17.1% and 23.3%, respectively. Phosphorylation levels of AMPK and ACC in the soleus muscle and liver tissue of db/db mice were significantly increased by the administration of MVE-H. MVE-H was further fractionated using preparative HPLC to identify the AMPK-activating compounds. The NMR and GC-MS analyses revealed that ${\beta}$-sitosterol was a major effective compound in MVE-H. Phosphorylation levels of AMPK and ACC, and glucose uptake were significantly increased by the treatment of MVE-S (${\beta}$-sitosterol) isolated from M. verticillata to L6 cells, and these effects were attenuated by an AMPK inhibitor (Compound C) pretreatment. These results, taken together, demonstrate that increased glucose uptake in L6 myotubes by MVE-H treatment is mainly accomplished through the activation of AMPK. Our finding suggests that the extract isolated from M. verticillata seed would be beneficial for the treatment of metabolic disease including type 2 diabetes and hyperlipidemia.
Deer antler has been widely prescribed in Chinese and Korean pharmacology. Although there have been several reports concerning the effects of deer antler, such as anti-aging action, anti-inflammatory activity, antifungal action and regulatory activity of the level of glucose, the effect on bone has not determined yet. The purpose of this study was to examine the effect of deer antler on osteoblast differentiation. Hexane extract(CN-H) and chloroform extract(CN-C) were acquired from deer antler(Cervus nippon) and MC3T3-E1 pre-osteoblasts were cultured in the presence or absence of each extract. Osteoblast differentiation was estimated with the formation of mineralized nodules and the mRNA expression of alkaline phosphatase(ALP), osteocalcin(OC) and bone sialoprotein(BSP) which are markers of osteoblast differentiation. Non-treated group did not show mineralized nodule. CN-C or CN-H-treated group showed minerlaized nodules in 16 days. In northern blot analysis, CN-C or CN-H-treated group showed the elevated expression of ALP, BSP and OC in 16 days. These results suggest the possibility to develop deer antler as a bone regenerative agent in periodontal therapy by showing the stimulating activity of deer antler on differentiation of osteoblast.
The antioxidant potential of a $75\%$ methanolic extract of highbush blueberry (Vaccinium corymbosum L.) and its different fractions was investigated using different reactive oxygen species (ROS), nitric oxide (NO.), metal chelating and lipid peroxidation assays. Methylene chloride and $75\%$ methanol fractions showed equally high activities $(IC_{50} 0.010 mg/mL)$ for hydroxyl radical (HO) scavenging. Higher hydrogen peroxide $(H_2O_2)$ scavenging values were reported for the ethyl acetate and methylene chloride fractions and their $IC_{50}$ values were 0.20 and 0.15 mg/mL, respectively. Nitric oxide (NO.) and 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) scavenging activities were higher in ethyl acetate and methylene chloride fractions. Chloroform and water fractions showed higher activities in superoxide $(O_2.)$ scavenging. All fractions showed strong metal chelating capacities compared with the commercial antioxidants tested. The $0.1\%$ ethyl acetate fraction showed notable capacity to suppress lipid peroxidation in both fish oil and linoleic acid. Phenolic content was measured in all the fractions and methanolic extract. Among the fractions, ethyl acetate fraction showed the highest phenolic content.
Background: The objective of this study were to determine the antibacterial activity and antibiotic activity-enhancing effect of 70% ethanol extract of the root of Rumex japonicus Houtt. and its fractions when used in combination with gentamicin against aerobic skin flora. Methods and Results: The antibacterial activity and antibiotic (gentamicin) activity enhancing effect against aerobic skin flora were determined using the disc diffusion assay. Chloroform fraction (CF) and ethyl acetate fraction (EF) showed higher activities against Staphylococcus aureus and Staphylococcus epidermidis than those shown by other fractions. Regarding the antibiotic (gentamicin) activity-enhancing effect against aerobic skin flora, the n-hexane fraction (HF) and CF showed strong activity. The combination of HF and CF with gentamicin was evaluated using the broth dilution assay to determine the inhibitory effect on the growth of aerobic skin flora. The combination of CF with gentamicin exhibited the highest inhibitory effect on the growth of S. aureus and S. epdermidis. MTT assay performed to determine the viability of L929 cells revealed that EF treatment resulted in viability of 33.96 - 116.76% at the tested concentration. The combination of 70% ethanol extract and its other fractions with gentamicin showed low cell toxicity. Conclusions: Appropriate use of antimicrobial agents is important prior to the development of new antibiotics. The 70% ethanol extract of the root of R. japonicus Houtt. and its fractions showed significant synergism with gentamicin when used in combination against S. aureus and S. epdermidis. Thus, R. japonicus Houtt. could be used as a functional materials in antimicrobial-related fields.
To search of more selective vasculogenic relaxation activity, the antioxidant activity of silkworm male pupae was determined by measuring its radical scavenging effect on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radicals, and anticoagulant activity of them was measured clotting time in both activated partial thromboplastin time (aPTT). Because, most of cGMP-enhancing agent such as, sildenafil, promotes thrombin-induced platelet aggregation, developed unexplained thrombic conditions including heart attack. To search more suitable and safe drug for vasculogenic relaxation, we purified silkworm pupae male extract. The ethyl acetate extract of silkworm male pupae showed strong scavenging activity in both DPPH and aPTT anticoagulant activity. The antioxidant activity potential of the individual fraction was in order of ethyl acetate > n-butanol > chloroform > n-hexane. The ethyl acetate soluble fraction exhibiting strong anti-oxidant and anticoagulant activity was further purified by repeated silica gel and Sephadex LH-20 column chromatography. Cholesterol was isolated as one of the active principles from ethyl acetate fraction, together with, minor portion, ${\beta}-sitosterol$.
The nutritional composition of the white lotus(Nelumbo nucifera Gaertn) leaf are moisture($1.87{\pm}0.06%$), crude ash($9.57{\pm}0.07%$), crude protein($15.53{\pm}0.4%$) and crude lipid($2.42{\pm}0.1%$) per 100g. The highest mineral content was K and Ca. The principal free sugars in the white lotus leaf was sucrose, fructose and glucose. The total phenolics compounds evidenced maximal levels of $8,842.73{\pm}20.57\;g/100g$. DPPH and hydroxyl radical of scavenging ability of each sample tended to increase with increasing degrees of the sample concentration. The $IC_{50}$ values of the ethanol extract, n-hexane, chloroform, ethylacetate, butanol and water fraction from the white lotus leaf to DPPH radicals were 5.65, 8.5, 8.27, 2.03, 2.3 and $13.7\;{\mu}g/mL$ respectively. The $IC_{50}$ values of the hydroxyl radical scavenging ability of the ethanol extract, ethylacetate, butanol and water fraction were 390, 574, 327 and $378\;{\mu}g/mL$ respectively.
This study was to isolate an active component of the chloroform fraction from the methanol extract of Ruta chalepensis leaves and to measure inhibitory effects against ${\alpha}$-glucosidase or ${\alpha}$-amylase. The inhibitory compound of R. chalepensis leaves was isolated using chromatographic methods and identified as quinoline. Quinoline and its structurally related derivatives were tested for their inhibitory activities by evaluating the $IC_{50}$ values against ${\alpha}$-amylase or ${\alpha}$-glucosidase and were compared with that of acarbose. Based on the $IC_{50}$ values, quinazoline exhibited the greatest inhibitory activity ($20.5{\mu}g/mL$), followed by acarbose ($66.5{\mu}g/mL$), and quinoline ($80.3{\mu}g/mL$) against ${\alpha}$-glucosidase. In case of ${\alpha}$-amylase, quinazoline had potent inhibitory activity, followed by quinoline ($179.5{\mu}g/mL$) and acarbose ($180.6{\mu}g/mL$). These results indicate that R. chalepensis extract, quinoline, and quinazoline could be useful for inhibiting ${\alpha}$-glucosidase or ${\alpha}$-amylase.
