Anti-microbial Activities of Ten Lauraceae Species against Propionibacterium acnes (여드름 유발균 Propionibacterium acnes에 대한 녹나무과 10종의 항균활성)

  • This study was performed to develop a new natural antimicrobial materials by analyzing the effect of extracts obtained from Ten Lauraceae Species on the inhibitory activity against Propionibacterium acnes. The plant materials were collected from Wando and Jeju islands, and the antimicrobial activity of the crude extracts was examined by the agar diffusion method with different part (i.e., leaf and branch), solvents (i.e., distilled water, 80% ethanol, and 100% methanol) and at different ultrasonic extracting times (i.e., 15, 30, and 45 minutes). The control agents used were synthetic antimicrobials, methylparaben and phenoxyethanol, at concentrations of 0.4, 1, 2, and 4 mg/disc. Altogether, extracts of 10 species used in the study showed inhibitory activity, which confirmed their antimicrobial action against acnes. Among these, leaves of Laurus nobilis L. which was extracted in 80% ethanol for 45 min showed the largest clear zone (19.8 mm). Leaves of L. nobilis L., showing highest antimicrobial activities among 10 species, were successively reextracted with n-hexane, chloroform, ethylacetate and n-butanol. As a results, in all fractions except butanol, clear zone above 10 mm were formed. The ethyl acetate fraction showed the highest inhibitory activity (13.3 mm) and the inhibitory activity was significantly higher than that of crude extract (10.2 mm) and phenoxyethanol as a control (12.5 mm).

Quality Characteristics of Defatted Soybean Grits Fermented by Bacillus subtilis NUC1 (고초균에 의한 탈지대두 Grits(Defatted Soybean Grits) 발효물의 품질 특성)

  • This study was conducted to investigate the quality characteristics of fermented-defatted soybean grits (DSG). Fermented-DSG was prepared by inoculating Bacillus subtilis NUCI into steamed DSG at 2% (v/w) concentration and fermenting at $40^{\circ}C$ for 24 hr. The protein contents of DSG and fermented-DSG were higher by $57.2{\sim}61.0%$ than those of Chungkukjang, but lipid contents of these samples were not detected. The protease and $\alpha$-amylase activities, and solid contents in DSG were not shown. However, the activities of protease and $\alpha$-amylase, tyrosine contents, fluid consistency, and viscous substance contents of fermented-DSG were higher than those of Chungkukjang. The levels of free amino acids and isoflavone of fermented-DSG tended to increase more than those of DSG. The sensory score of fermented-DSG were higher in odor and taste than those of Chungkukjang groups, while lower in color and overall acceptability.

Antioxidant Effects of Red Ginseng Powder on Liver of Benzo(α)Pyrene-Treated Mice (벤조피렌을 투여한 생쥐의 간 조직에서의 홍삼분말의 항산화 효과)

  • The effects of red ginseng powder on hepatotoxicity in $benzo(\alpha)pyrene\;[B(\alpha)P]$-treated mice were investigated. Male ICR mice were pretreated with red ginseng powder (50 or 100 mg/kg/day, for 5 days, intraperitoneally) before treatment with $B(\alpha)P$ (0.5 mg/kg, i.p, single dose). The ability of red ginseng powder to protect against oxidative damage to the mouse liver was examined by determining the level of lipid peroxide, glutathione, and the antioxidant enzyme activities. The glutathione content depleted by $B(\alpha)P$ were significantly increased by red ginseng powder, but elevation of lipid peroxide content induced by $B(\alpha)P$ was decreased by red ginseng powder. The increased activities of superoxide dismutase, catalase and glutathione peroxidase after $B(\alpha)P$-treatment were decreased by the treatment of red ginseng Powder; however, glutathione S-transferase activity depleted by $B(\alpha)P$ were significantly increased. These results suggest that red ginseng powder can protect against $B(\alpha)P$ intoxification through its antioxidant properties.

Antioxidant Properties of Proanthocyanidin Fraction Isolated from Wild Grape (Vitis amurensis) Peel (산머루 과피에서 분리한 proanthocyanidin 획분의 항산화 특성)

  • The proanthocyanidin fraction was isolated from the wild grape (Vitis amurensis) peel and its antioxidant capacities were examined to promote the utilization of wild grape by-products. The 70% acetone crude extract of the wild grape peel was fractionated with hexane, ethyl acetate, and water. The ethyl acetate fraction was applied to a Sephadex LH-20 column chromatograph, which was eluted with 50% methanol, 75% methanol, and 75% acetone. The proanthocyanidin characteristics and contents of the isolated fractions were investigated by the vanillin-$H_2SO_4$ and BuOH-HCl methods. Fraction 6 had the highest proanthocyanidin content ($49.35{\pm}2.75\;g%$) among the isolated fractions. The antioxidant activities of the proanthocyanidin fraction were examined by DPPH radical scavenging, FRAP assay, and total phenolic contents. The FRAP values and total phenolic contents of the fractions ranged from 3.54 to 32.25 mmol/kg and from 4.48 to 50.80 g/100 g, respectively. The proanthocyanidin contents was strongly correlated with DPPH radical scavenging activities, FRAP values, and total phenolic contents.

Enhanced Immune Activity and Cytotoxicity of Artemisia capillaris Thunb. Extracts against Human Cell Lines (사철쑥 추출물의 면역세포의 생육증진 및 세포독성)

  • The immune activation and anticancer activities of the water and ethanol extracts from Artemisia capillaris Thunb. were studied. The growth of human hepatocarcinoma and human gastric cancer cell was inhibited by the addition of $1.0\;mg/m{\ell}$ of the water extract, by about 77% and 95%, respectively. The growth of human breast cancer cells was also inhibited by addition of $0.5\;mg/m{\ell}$ of both water and ethanol extracts by 88%. The growth of human normal lung cell, HEL299 was inhibited by 15% indicating very low cytotoxicity of both extracts. Overall selectivity of the both extracts on several human cancer cell line was over 2.5. The growth of both human B and T cells was enhanced up to 1.6 to 2.1 times by adding the ethanol extracts. The secretion of cytokines, $TNF-{\alpha}$ and IL-6, from human B cells was also increased showing $68\;pg/m{\ell}$ and $67\;pg/m{\ell}$, respectively, compared to $35{\sim}40\;pg/m{\ell}$ of the control. In terms of the immune activity, there was not much difference between water and ethanol extracts of Artemisia capillaris Thunb. It implies that the extraction solvent could not differ the biological activities of the extracts. Based on these results, Artemisia capillaris Thunb. can be developed into a potentially useful cancer chemoprentive agent.

Application of Isolated Tyrosinase Inhibitory Compounds from Persimmon Leaves (감나무 잎으로 부터 분리한 tyrosinase 억제물질의 응용)

  • Total phenolic content was the highest in 60% ethanol extracts at 21.91 mg/g, and inhibitory activity against tyrosinase of 60% ethanol extracts was higher than ethanol extracts of other concentration. The inhibitory compounds against tyrosinase from Persimmon leaves were purified using Sephadex LH-20, MCI-gel CHP-20 column chromatography with gradient elution. Two purified compounds were isolated as a result. The chemical structures of each compound were determined and identified using $^1H$-NMR and $^{13}C$-NMR, FAB-Mass. The compounds were confirmed as (+)-gallocatechin and prodelphinidin B-3. The tyrosinase inhibitory activities of purified (+)-gallocatechin and prodelphinidin B-3 were 29.5 and 40.2%, respectively. The inhibitory activities of (+)-gallocatechin and prodelphinidin B-3 against melanin biosynthesis in melanoma cell were 32.5 and 46.7%. The safety of essence with tyrosinase inhibitory compounds from persimmon leaves was also assessed by various safety profiles. First, changes in pH (4.90~4.95) and viscosity (23,000~26,000 cP) was not detected for 60 days. Essence also showed stability against temperature and light for 60 days. All these findings suggest that extracts from persimmon leaves have a great potential as a cosmetical ingredient with a potent whitening effect.

Phenolic Content, DPPH Radical Scavenging, and Tyrosinase Inhibitory Activities of Ecklonia cava Extracted with the Ultrasonic Wave Method (초음파 추출법에 의한 감태 추출물의 페놀성 화합물 함량, DPPH 라디칼 소거 활성 및 tyrosinase 저해 활성 분석)

  • The applicability of the ultrasonic wave method to the extraction of useful components from seaweeds was investigated. Extracts from freeze-dried Ecklonia cava powder were prepared with hot water ($65^{\circ}C$), water ($24^{\circ}C$), 50% ethanol, and 100% ethanol, and ultrasonic extraction was also performed. The content of phenolic compounds and the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and tyrosinase inhibitory activity of the extracts were analyzed, and differences in the data obtained by the ultrasonic extraction and the traditional extraction methods were compared. The phenolic content in the E. cava extract by ultrasonic extraction (142.80 mg/g) was approximately 14 times higher than the phenolic content in the hot water extract (10.03 mg/g). The DPPH radical scavenging and the tyrosinase inhibitory activities of the ultrasonic extract were approximately 4 times and 14 times higher than the hot water extracts, respectively. The correlation between the phenolic content and the DPPH radical scavenging activity ($R^2$=99.47) and between the phenolic content and the tyrosinase inhibitory activity ($R^2$=99.99) was very high. These results indicate that ultrasonic extraction is more suitable than traditional extraction for the extraction of useful components from E. cava.

Physicochemical Characteristics and Antioxidant Activities of Freezing Pretreated Black Garlic (동결 전처리한 숙성 흑마늘의 이화학적 특성 및 항산화 활성)

  • Freezing pretreatment can destroy the cell membrane of garlic and may improve some food-quality of garlic. Therefore we investigated the effect of freezing pretreatment at $-20^{\circ}C$ and $-70^{\circ}C$ on quality of aged black garlic, compared with traditional processing methods. Our results showed that freezing pretreatment at $-70^{\circ}C$ had the greatest impact on qualities and antioxidant activities of black garlic. Browning degree and pH of black garlic after both the freezing pretreatment and aging process were 3.14 and 3.55, respectively. Furthermore, 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity and reducing power of aged black garlic can be enhanced by pre-freezing processing. Reducing sugar and 5-hydroxymethyl-2-furaldehyde (5-HMF) contents of freezing pretreated and aged black garlic were increased by 1.69 and 1.14 fold, respectively, compared with the control samples. The results indicated that freezing pretreatment had improved the overall qualities (such as browning degree, pH, reducing sugar) and functional materials of black garlic.

Empirical Analysis of Export Structure Change and the Role of R&D in the Field of Information Technology (정보통신산업 수출성과변화 및 R&D 역할에 대한 실증분석)

  • This study empirically investigates the export structure change of OECD countries in the field of information technology from 1995 to 2005. We used Between and Within Entropy index to estimate the difference of export structure change rather than RCA index(simple export structure change). In addition, we also estimate the relationship between R&D activities and export difference. First, empirical findings showed that IT export market concentration is decreasing gradually, and changing to more competitive market structure. Second, compared to other OECD countries, Korean exports weight of information and communication industry can be seen higher than that of bio-industry. Third, Entropy index was increased from 1995 and then gradually decreased as the starting point of 2000. Finally, the relationship between R & D activities in the telecommunications industry and impact of the export structure changes was a positive.

Antioxidant Activity and DNA Protective Effect against Oxidative Stress of Pinus rigida × taeda Cone (리기테다 소나무 솔방울의 항산화 활성 및 산화적 DNA 손상에 대한 억제 효과)

  • Reactive oxygen species (ROS) damage DNA and cause cancer. Therefore, the research is being conducted on the development of antioxidants for the removal of ROS. This study was performed to investigate antioxidant activity and protective effect against oxidative DNA damage using ethyl acetate fractions from the cone of Pinus rigida × taeda (ERT). The antioxidant activity was evaluated using the DPPH, ABTS radical scavenging assay, reducing power assay, and Fe2+ chelating assay. Also, the contents of phenolic compounds and vitamin C related to antioxidant activity were analyzed to confirm phytochemicals. The DNA protective effect against oxidative stress was confirmed by the φX-174 RF I plasmid DNA cleavage assay. As a result, ERT showed DPPH and ABTS radical scavenging activities in a concentration-dependent manner. The results of reducing power and Fe2+ chelating activities were 77.32 ± 2.28% and 64.09 ± 1.01% at 200 ㎍/㎖. Also, ERT showed a DNA protective effect against oxidative stress.
