Purine catabolite screening enables reliable diagnosis of certain diseases. In this regard, the development of a facile detection strategy with high sensitivity and selectivity is demanded for point-of-care applications. In this work, the simultaneous detection of uric acid (UA), xanthine (XA), and hypoxanthine (HX) was carried out as model purine catabolites by surface-enhanced Raman Spectroscopy (SERS). The detection assay was conducted by employing high-aspect ratio Au nanopillar substrates coupled with in-situ Au electrodeposition on the substrates. The additional modification of the Au nanopillar substrates via electrodeposition was found to be an effective method to encapsulate molecules in solution into nanogaps of growing Au films that increase metal-molecule contact and improve substrate's sensitivity and selectivity. In complex solutions, the approach facilitated ternary identification of UA, XA, and HX down to concentration limits of 4.33 𝜇M, 0.71 𝜇M, and 0.22 𝜇M, respectively, which are comparable to their existing levels in normal human physiology. These results demonstrate that the proposed platform is reliable for practical point-of-care analysis of biofluids where solution matrix effects greatly reduce selectivity and sensitivity for rapid on-site disease diagnosis.
Chaff-vinegar is known for having a variety of useful purposes in the fields of health and lifestyles. In a previous study we isolated and identified the active fractions of the polyphenol compound 7 species as a potential biomaterial for cosmeceuticals. To further test for its potential use as a functional material, we carried out an MTT assay, collagenase inhibition assay, elastase inhibition assay, tyrosinase inhibition assay, DPPH free radical scavenging assay, SOD-like activity assay and a xanthine oxidase inhibition assay. Chaff-vinegar exhibited potent collagenase and elastase inhibitory activities in a concentration dependent manner, indicating that the agent has the potential to alleviate the skin wrinkling process. Chaff-vinegar also showed 80% tyrosinase inhibition at a concentration of $100{\mu}L/mL$. DPPH radical scavenging, xanthine oxidase inhibition, and SOD-like activity results for each activity were 80%, 80%, and 100%, respectively. Taken together, the present study suggests that chaff-vinegar is a good candidate for use as an anti-wrinkling and/or whitening agent.

We investigated the applicability of the TEM-l ${\beta}$-lactamase fragment complementation (BFC) system to develop a strategy for the screening of protein-protein interactions in bacteria. A BFC system containing a human Fas-associated death domain (hFADD) and human Fas death domain (hFasDD) was generated. The hFADD-hFasDD interaction was verified by cell survivability in ampicillin-containing medium and the colorimetric change of nitrocefin. It was also confirmed by His pull-down assay using cell lysates obtained in selection steps. A coiled-coil helix coiled-coil domain-containing protein 5 (CHCH5) was identified as an interacting protein of human uracil DNA glycosylase (hUNG) from the bacterial BFC cDNA library strategy. The interaction between hUNG and CHCH5 was further confirmed with immunoprecipitation using a mammalian expression system. CHCH5 enhanced the DNA glycosylase activity of hUNG to remove uracil from DNA duplexes containing a U/G mismatch pair. These results suggest that the bacterial BFC cDNA library strategy can be effectively used to identify interacting protein pairs.
Peroxisome proliferator-activated receptors (PPARs) are a subfamily of nuclear receptors (NRs). Human peroxisome proliferator-activated receptor gamma (hPPAR${\gamma}$) has been implicated in the pathology of numerous diseases, including obesity, diabetes, and cancer. ELISA-based hPPAR${\gamma}$ activation assay showed that biapigenin increased the binding between hPPAR${\gamma}$ and steroid receptor coactivator-1 (SRC-1) by approximately 3-fold. In order to confirm that biapigenin binds to hPPAR${\gamma}$, fluorescence quenching experiment was performed. The results showed that biapigenin has higher binding affinity to hPPAR${\gamma}$ at nanomolar concentrations compared to indomethacin. Biapigenin showed anticancer activity against HeLa cells. Biapigenin was noncytotoxic against HaCa T cell. All these data implied that biapigenin may be a potent agonist of hPPAR${\gamma}$ with anticancer activity. We will further investigate its anticancer effects against human cervical cancer.
Ambient air particulate matters are classified into two distinct modes in sire distribution, namely the coarse and fine particles. Correlation between high particulate concentration and adverse effect on human populations has long been recognized. However, the toxicology of these adverse efforts has not been clarified. We investigated the genotoxic effect of PM 2.5 collected from urban area in Seoul by comet assay (A549 cells), CBMN assay (CHO-K1 cells) and EROD-microbioassay (H4IIE cells). Results from in vitro micronucleus assay and comet assay showed that PM 2.5 samples collected from traffic area, residential area and indoor air induced chromosomal damage and DNA breakage in a non-cytotoxic dose. The complex mixture effect of these PM 2.5 extracts was quantified by EROD-microbioassay in terms of its bio-TEQ (biologiral -TCDD equivalent concentration) which was 70.87$\pm$28.07, 93.55$\pm$21.80 and 14.31 $\pm$ 1.10 ng/g-PM 2.5 in traffic area, residental area and indoor air samples, respectively. Conclusively, we suggested that PM 2.5 collected from traffic area and residential area contains CYPIA inducer and genotoxic materials.
Gastrodia elata Blume is a major imp0l1ant medicinal resource in Korea. In order to confirm the biological activities of Gastrodia elata Blume, we carried out various in vitro assays. Of them, anti-oxidant and anti-tumor activities were detected from assays. The prototype of Gastrodia elata Blume extracts was used for 1he evaluation of DPPH, FRAP, hydroxyradical scavenging assay as anti-oxidant assays, as well as anti-tumor asctivities as wound assay and invasion assay. As a result, the prototype of Gastrodia elata Blume extracts showed potent anti-oxidative activity and anti-tumor activity in vitro. These above results suggest that 1he Gastrodia elata Blume extracts could have potential to alleviate oxidation process, cell motility activity, and tumorigenesis.

Purpose: Rapid detection of etiologic organisms is crucial for initiating appropriate therapy in patients with central nervous system (CNS) infection. This study aimed to evaluate the diagnostic value of the BioFire® Meningitis/Encephalitis (ME) panel in detecting etiologic organisms in cerebrospinal fluid (CSF) samples from febrile infants. Methods: CSF samples from infants aged <90 days who were evaluated for fever were collected between January 2016 and July 2019 at the Seoul National University Children's Hospital. We performed BioFire® ME panel testing of CSF samples that had been used for CSF analysis and conventional tests (bacterial culture, Xpert® enterovirus assay, and herpes simplex virus-1 and -2 polymerase chain reaction) and stored at -70℃ until further use. Results: In total, 72 (24 pathogen-identified and 48 pathogen-unidentified) CSF samples were included. Using BioFire® ME panel testing, 41 (85.4%) of the 48 pathogen-unidentified CSF samples yielded negative results and 22 (91.7%) of the 24 pathogen-identified CSF samples yielded the same results (enterovirus in 19, Streptococcus agalactiae in 2, and Streptococcus pneumoniae in 1) as those obtained using the conventional tests, thereby resulting in an overall agreement of 87.5% (63/72). Six of the 7 pathogen-unidentified samples were positive for human parechovirus (HPeV) via BioFire® ME panel testing. Conclusions: Compared with the currently available etiologic tests for CNS infection, BioFire® ME panel testing demonstrated a high agreement score for pathogen-identified samples and enabled HPeV detection in young infants. The clinical utility and cost-effectiveness of BioFire® ME panel testing in children must be evaluated for its wider application.

Purpose: Immunosorbent assay most commonly used in the laboratory and commercial third-party quality control material is a substance that provided by BIO-RAD. However, in Reference Sheet by radioimmunoassay test kit or a measuring device for the mention of Acceptable Range is somewhat lacking. Radioimmunoassay for the inspection of test results by setting Acceptable Range to increase the objectivity of the recommendations on the data accumulated by the manufacturer listed in the Reference Sheet is to be issued. Materials and Methods : In our hospital since 2009 partially BIO-RAD using quality control materials in 2011, excluding certain items, some items were most of the BIO-RAD third-party quality control materials are used. Thus, internal quality control data accumulated BIO-RAD's Unity Real Time program by using the items were measured. Results : BIO-RAD using quality control material items were about 50 of the 20 Point Data averages, standard deviations, variation coefficients were calculated to measure the Acceptable Range of kit, automated immunoassay attributed Roche Elecsys / E170 / cobas e Systems the measures and compared. Conclusions : BIO-RAD QC materials commonly used hospital and peer group by setting the measurement kit, suitable for laboratory equipment for radioimmunoassay of Acceptable Range manufacturer recommendation to increase the objectivity of the test results by national and international recognition for radioimmunoassay should seek to increase.
Head and neck cancers (HNC) are extremely complex disease types and it is likely that chromosomal instability is involved in the genetic mechanisms of its genesis. However, there is little information regarding the background levels of chromosome instability in these patients. In this pilot study, we examined spontaneous chromosome instability in short-term lymphocyte cultures (72 hours) from 72 study subjects - 36 newly diagnosed HNC squamous cell carcinoma patients and 36 healthy ethnic controls. We estimated chromosome instability (CIN) using chromosomal aberration (CA) analysis and nuclear level anomalies using the Cytokinesis Block Micronucleus Cytome Assay (CBMN Cyt Assay). The proliferation rates in cultures of peripheral blood lymphocytes (PBL) were assessed by calculating the Cytokinesis Block Proliferation Index (CBPI). Our results showed a significantly higher mean level of spontaneous chromosome type aberrations (CSAs), chromatid type aberration (CTAs) dicentric chromosomes (DIC) and chromosome aneuploidy (CANE UP) in patients (CSAs, $0.0294{\pm}0.0038$; CTAs, $0.0925{\pm}0.0060$; DICs, $0.0213{\pm}0.0003$; and CANE UPs, $0.0308{\pm}0.0035$) compared to controls (CSAs, $0.0005{\pm}0.0003$; CTAs, $0.0058{\pm}0.0015$; DICs, $0.0005{\pm}0.0003$; and CANEUPs, $0.0052{\pm}0.0013$) where p<0.001l. Similarly, spontaneous nuclear anomalies showed significantly higher mean level of micronuclei (MNi), nucleoplasmic bridges (NPBs) and nuclear buds (NBUDs) among cases (MNi, $0.01867{\pm}0.00108$; NPBs, $0.0156{\pm}0.00234$; NBUDs, $0.00658{\pm}0.00068$) compared with controls (MNi, $0.00027{\pm}0.00009$; NPBs, $0.00002{\pm}0.00002$; NBUDs, $0.00011{\pm}0.00007$).The evaluation of CBPI supported genomic instability in the peripheral blood lymphocytes showing a significantly lower proliferation rate in HNC patients ($1.525{\pm}0.005552$) compared to healthy subjects ($1.686{\pm}0.009520$) (p<0.0001). In conclusion, our preliminary results showed that visible spontaneous genomic instability and low rate proliferation in the cultured peripheral lymphocytes of solid tumors could be biomarkers to predict malignancy in early stages.

In this study, the anti-oxidative and anti-obesity activities of two medicinal herb extracts, Tetrapanax papyriferus (TP) and Siegesbeckia pubescens (SP), were evaluated using DPPH radical scavenging activity assay, lipase enzyme inhibition assay, and the cell culture model system. Both methanol extracts of TP and SP showed DPPH radical scavenging activities dose-dependently, and the $IC_{50}$ of DPPH radical scavenging activities of the two medicinal herbs were 65.23 and 47.79 ${\mu}g/ml$, respectively. Furthermore, both extracts suppressed effectively lipase enzyme activity dose-dependently. Moreover, TP and SP extracts significantly suppressed adipocyte differentiation, lipid accumulation, triglyceride (TG) contents on 3T3-L1 preadipocytes in a dose-dependent manner without cytotoxicity. Their anti-obesity effect was modulated by cytidine-cytidine-adenosine-adenosine-thymidine (CCAAT)/enhancer binding proteins ${\alpha}$ ($C/EBP{\alpha}$), $C/EBP{\beta}$ and peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$) gene and protein expressions. Furthermore, TP and SP possessed a synergistic effect on anti-obesity activity. The identification of the active compounds that confer the anti-obesity activity of TP and SP might be needed.
