Water Extract from Spent Mushroom Substrate of Hericium erinaceus Suppresses Bacterial Wilt Disease of Tomato

  • Culture filtrates of six different edible mushroom species were screened for antimicrobial activity against tomato wilt bacteria Ralstonia solanacearum B3. Hericium erinaceus, Lentinula edodes (Sanjo 701), Grifola frondosa, and Hypsizygus marmoreus showed antibacterial activity against the bacteria. Water, n-butanol, and ethyl acetate extracts of spent mushroom substrate (SMS) of H. erinaceus exhibited high antibacterial activity against different phytopathogenic bacteria: Pectobacterium carotovorum subsp. carotovorum, Agrobacterium tumefaciens, R. solanacearum, Xanthomonas oryzae pv. oryzae, X. campestris pv. campestris, X. axonopodis pv. vesicatoria, X. axonopodis pv. citiri, and X. axonopodis pv. glycine. Quantitative real-time PCR revealed that water extracts of SMS (WESMS) of H. erinaceus induced expressions of plant defense genes encoding ${\beta}$-1,3-glucanase (GluA) and pathogenesis-related protein-1a (PR-1a), associated with systemic acquired resistance. Furthermore, WESMS also suppressed tomato wilt disease caused by R. solanacearum by 85% in seedlings and promoted growth (height, leaf number, and fresh weight of the root and shoot) of tomato plants. These findings suggest the WESMS of H. erinaceus has the potential to suppress bacterial wilt disease of tomato through multiple effects including antibacterial activity, plant growth promotion, and defense gene induction.

큰느타리 수확 후 배지 물 추출물의 토마토 풋마름병 억제 (Suppressive Effect of Water Extract from Spent Mushroom Substrate of Pleurotus eryngii against Tomato Bacterial Wilt Disease)

  • 큰느타리버섯(Pleurotus eryngii) 수확 후 배지(spent mushroom substrate, SMS)의 물 추출액(WESMS)을 처리한 토마토 식물체는 토마토 풋마름병을 70% 이상 억제하였으며 페놀 성분(3%)과 salicylic acid 함량이 증가되었다. 또한 큰느타리 WESMS 처리 토마토는, 초장, 엽폭, 입장, 입수, 줄기와 뿌리 생체량 등에서 물 처리 및 큰느타리버섯 배지만을 사용한 대조군에 비하여 높은 생육 촉진 효과를 보였다. 이는 큰느타리 WESMS가 병 저항성유도와 생육촉진의 토마토 복합기능성으로 환경친화적 풋마름병 방제에 유용하게 활용될 수 있음을 나타낸다.

Changes in the Composition and Microbial Community of the Pepper Rhizosphere in Field with Bacterial Wilt Disease

  • Bacterial wilt caused by Ralstonia solanacearum is considered one of the most harmful diseases of pepper plants. Recently, research on plant disease control through the rhizosphere microbiome has been actively conducted. In this study, the relationship with disease occurrence between the neighboring plant confirmed by analyzing the physicochemical properties of the rhizosphere soil and changes in the microbial community. The results confirmed that the microbial community changes significantly depending on the organic matters, P2O5, and clay in the soil. Despite significant differences in microbial communities according to soil composition, Actinobacteriota at the phylum level was higher in healthy plant rhizosphere (mean of relative abundance, D: 8.05 ± 1.13; H: 10.06 ± 1.59). These results suggest that Actinobacteriota may be associated with bacterial wilt disease. In this study, we present basic information for constructing of healthy soil in the future by presenting the major microbial groups that can suppress bacterial wilt.

Plant-derived Antibacterial Metabolites Suppressing Tomato Bacterial Wilt Caused by Ralstonia solanacearum

  • Ralstonia solanacearum species complex (RSSC) causes bacterial wilt, and it is one of the most important soil-borne plant pathogenic bacteria. RSSC has a large host range of more than 50 botanical families, which represent more than 200 plant species, including tomato. It is difficult to control bacterial wilt due to following reasons: the bacterial wilt pathogen can grow inside the plant tissue, and it can also survive in soil for a long period; moreover, it has a wide host range and biological diversity. In most previous studies, scientists have focused on developing biological control agents, such as antagonistic microorganisms and botanical materials. However, biocontrol attempts are not successful. Plant-derived metabolites and extracts have been promising candidates to environmentally friendly control bacterial wilt diseases. Therefore, we review the plant extracts, essential oils, and secondary metabolites that show potent in vivo antibacterial activities (in potted plants or in field) against tomato bacterial wilt, which is caused by RSSC.

코퍼 하이드록사이드를 이용한 토마토 풋마름병 방제 (Control of Bacterial Wilt of Tomato using Copper Hydroxide)

  • 국내에서 Ralstonia solanacearum에 의한 풋마름병은 토마토 재배에 심각한 피해를 주고 있다. R. solanacearum에 의한 풋마름병을 방제하기 위한 약제를 선발하기 위하여 5종 항생제를 이용하여 균에 대한 생장 억제 효과와 포장에서의 토마토 풋마름병 방제효과를 조사하였다 R. solanacearum에 대한 생장억제효과를 조사한 결과, streptomycin 수화제, oxytetracyclin streptomycin sulfate 수화제, oxolinic acid 수화제는 병원균에 대한 생육억제 효과가 우수하였다. 포장에서 토마토 풋마름병에 대한 방제효과 시험을 실시한 결과, copper hydroxide 수화제가 62.5%의 가장 높은 방제효과를 나타내었다. Copper hydroxide 수화제는 친환경유기농자재에 등록된 약제로서 관행 재배뿐만 아니라 토마토 친환경 재배시에도 풋마름병 방제에 사용할 수 있을 것이다.

First Report of Bacterial Wilt Caused by Ralstonia solanacearum Biovar 2 Race 1 on Tomato in Egypt

  • This study aims to isolate and identify the causal pathogen of tomato bacterial wilt in Egypt. In 2008, tomato plants showing typical symptoms of bacterial wilt disease with no foliar yellowing were observed in Minia, Assiut and Sohag governorates, Egypt. When cut stems of symptomatic plants were submerged in water, whitish ooze was evident and longitudinal sections showed a brown discoloration in the vascular tissues. Bacteria were isolated on triphenyl tetrazolium chloride medium and fifteen isolates shown typical morphological and cultural characteristics were confirmed as Ralstonia solanacearum biovar 2 race 1. Pathogenicity tests showed that all isolates proved to be pathogenic to tomato plants, varied from 52 to 97% wilting. This is the first report of R. solanacearum biovar 2 race 1 causing bacterial wilt in tomato crop in Egypt.

Chemical Pesticides and Plant Essential Oils for Disease Control of Tomato Bacterial Wilt

  • Efficacy of different control methods was evaluated for disease management of tomato bacterial wilt caused by $Ralstonia$ $solanacearum$. All six chemical pesticides applied to the bacterial suspension showed $in$ $vitro$ bactericidal activities against $R.$ $solanacearum$. Minimal inhibitory concentrations (MICs) of copper hydroxide (CH), copper hydroxide-oxadixyl mixture (CH+O), and copper oxychloride-dithianon mixture (CO+D) were all 200 ${\mu}g/ml$; MIC of copper oxychloride-kasugamycin (CO+K) mixture was 100 ${\mu}g/ml$; MICs of both streptomycin- validamycin (S+V) and oxine copper-polyoxine B mixture (OC+PB) were 10 ${\mu}g/ml$. Among these chemical pesticides, treatment of the detached tomato leaves with the 5 pesticides (1 mg/ml), except for OC+PB delayed early wilting symptom development caused by the bacterial inoculation ($10^6$ and $10^7$ cfu/ml). Four pesticides, CH, CH+O, CO+K and S+V, showed disease protection in pot analyses. Six plant essential oils, such as cinnamon oil, citral, clove oil, eugenol, geraniol and limonene, differentially showed their antibacterial activities $in$ $vitro$ against $R.$ $solanacearum$ demonstrated by paper disc assay. Among those, cinnamon oil and clove oil exert the most effective activity for protection from the wilt disease caused by the bacterial infection ($10^6$ cfu/ml). Treatment with cinnamon oil and clove oil also suppressed bacterial disease by a higher inoculum concentration ($10^7$ cfu/ml). Clove oil could be used for prevention of bacterial wilt disease of tomato plants without any phytotoxicity. Thus, we suggest that copper compounds, antibiotics and essential oils have potency as a controlling agent of tomato bacterial wilt.

Hydrogen Peroxide- and Nitric Oxide-mediated Disease Control of Bacterial Wilt in Tomato Plants

  • Reactive oxygen species (ROS) generation in tomato plants by Ralstonia solanacearum infection and the role of hydrogen peroxide ($H_2O_2$) and nitric oxide in tomato bacterial wilt control were demonstrated. During disease development of tomato bacterial wilt, accumulation of superoxide anion ($O_2{^-}$) and $H_2O_2$ was observed and lipid peroxidation also occurred in the tomato leaf tissues. High doses of $H_2O_2$ and sodium nitroprusside (SNP) nitric oxide donor showed phytotoxicity to detached tomato leaves 1 day after petiole feeding showing reduced fresh weight. Both $H_2O_2$ and SNP have in vitro antibacterial activities against R. solanacearum in a dose-dependent manner, as well as plant protection in detached tomato leaves against bacterial wilt by $10^6$ and $10^7$ cfu/ml of R. solanacearum. $H_2O_2$- and SNP-mediated protection was also evaluated in pots using soil-drench treatment with the bacterial inoculation, and relative 'area under the disease progressive curve (AUDPC)' was calculated to compare disease protection by $H_2O_2$ and/or SNP with untreated control. Neither $H_2O_2$ nor SNP protect the tomato seedlings from the bacterial wilt, but $H_2O_2$ + SNP mixture significantly decreased disease severity with reduced relative AUDPC. These results suggest that $H_2O_2$ and SNP could be used together to control bacterial wilt in tomato plants as bactericidal agents.

2006년도 황색종 담배 병해발생 상황<단보> (Disease Severities of Flue-Cured Tobacco Plants Surveyed in Korea, 2006)

  • Main tobacco diseases were surveyed on major flue-cured leaf tobacco fields throughout South Korea in 2006. Mosaic caused by potato virus Y and bacterial wilt caused by Ralstonia solanacearum were most severe during harvest season. During last ten years, the damage by tobacco mosaic virus was reduced but the incidence of bacterial wilt increased. These changes of the disease incidences coincide with release time of the tobacco cultivar resistant to the tobacco mosaic virus but susceptible to bacterial wilt pathogen. Wild fire(Pseudomonas syringae pv. tabaci strain ungulate) occurred severely at Kangwon province, though the symptom of wild fire with yellow halo was not observed.

Relationship between the Population of Ralstonia solanacearum in Soil and the Incidence of Bacterial Wilt in the Naturally Infested Tobacco Fields

  • The population of Ralstonia solanacearum (Rs) in soil is very important as a primary inoculum source of bacterial wilt in tobacco fields. To investigate the population of Rs, physical properties and chemical components during the tobacco growing season, soil samples were taken from the fifteen fields which were located in the flue-cured tobacco growing area, Ansung, Kyunggi province and Wonju, Kangwon province. Two fields of the fifteen were bacterial wilt free. Six fields had less than 10% plants being diseased and seven over 10%. The Rs population level determined by using SMSA medium generally showed an up-and-down pattern being low in May, high in Jun and July and low in August. The soil population in May and June showed a positive correlation with the incidence of bacterial wilt (r=0.571$^*$, r=0.688$^{**}$), but P$_2O_5$, content of soil was negatively correlated with the disease incidence (r=-0.539$^*$). These results suggest that Rs population in soil examined in May or in June, and the P$_2O_5$ content in soil should be key factors to determine the bacterial wilt potential of tobacco fields.
