한국산 약용식물의 항산화 효과 (Antioxidative Activities of Korean Medicinal Plants)

  • 천연 보존제, 기능성 식품 및 화장품 그리고 의약품 신소재의 발굴을 위한 전단계로서 국내산 약용식물 160종 209점에 대한 DPPH 라디칼 소거 및 linoleic acid 과산화 저해 등의 항산화능을 평가하였다. DPPH 라디칼 소거능에서는 $13.5{\mu}g/ml$$RC_{50}$을 보인 ${\alpha}-tocopherol$을 대조구로 하였을 때 쥐손이풀 지상부, 뱀무 지상부, 이질풀 지상부, 오동나무 잎이 각각 $19.3{\mu}g/ml,\;22.5{\mu}g/ml,\;23.9{\mu}g/ml,$$27.2{\mu}g/ml$로서 우수하였고, linoleic acid과산화 반응에 대해서는 고로쇠나무 잎 등 38점의 식물추출물이 90%이상의 강한 항산화력을 나타내었다.

독활의 항혈소판 및 항산화 효과 (Effects of Aralia continentalis Kitagawa on Antiplatelet and Antioxidative Activities.)

  • 독활 메탄올 추출물과 그 분획의 항혈전 및 항산화 효과를 확인하기 위하여 ADP를 이용하여 혈소판 응집억제효과를 탐색하고 DPPH 및 ABTS-radical 소거능을 측정하여 항산화능을 측정하였다. 그 결과 독활, 특히 EtOAc 분획은 ADP에 의해 유도된 혈소판응집과 트롬빈에 의한 혈소판 부착에 대하여 농도 의존적으로 저해효과를 나타내었으며, 높은 폴리페놀 함량과 가장 높은 라디칼 소거능을 통하여 독활의 항산화 효과를 확인하였다. 이상의 결과로 독활 EtOAc 분획의 높은 DPPH 및 ABTS radical 소거능이 항혈소판 효과에 영향을 미치는 것으로 사료된다.

  • The aim of this study was to investigate the potential of Acanthopanax sieboldianum (Makino) Koidz (ACS) as a potent antioxidant and antidiabetic agent. The antioxidative and alpha-glucosidase inhibitory activities were examined using the methanol extracts and solvent fractions from ACS-leaf and ACS-stem. Antioxidative activities were measured by in vitro methods such as DPPH and ABTS radical scavenging activity and superoxide dismutase (SOD) activity. When the chloroform and ethyl acetate fractions of ACS-leaf and ethyl acetate fractions of ACS-stem were compared with the control, the SOD-like activity was impaired even at the low treatment concentrations. In addition, the ethyl acetate fractions of ACS-leaf and ACS-stem showed alpha-glucosidase inhibition activities at low treatment concentrations. Analysis of the major components in the fractions of ACS-leaf and ACS-stem was also performed using HPLC. Finally, astragalin, isoqurecetin, chlorogenic acid and caffeic acid contents were measured. Based on this work, we propose that ACS-leaf and ACS-stem have great potential as natural antioxidant and antidiabetic materials related to health benefits.

Chemical composition, antioxidant and antifungal activities of rhizome essential oil of Kaempferia parviflora Wall. ex Baker grown in Vietnam

  • The aim of the present study was to evaluate the chemical composition and antioxidative activity of rhizome essential oil of Kaempferia parviflora Wall. ex Baker. The essential oil extracted by hydrodistillation was chemically profiled by GC/MS analysis. The antioxidative activity was determined and evaluated spectroscopically by the 2,2-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radical scavenging assays. According to the results, the major essential oil components were camphene (18.03%), β-pinene (14.25%), a-pinene (12.38%), endo-borneol (10.23%), β-copaene (8.38%), and linalool (8.20%). K. parviflora rhizome oil possessed antioxidant potential, exhibiting DPPH and ABTS radical scavenging activities as high as 80.90 and 94.04%, respectively, at a concentration of 10 mg/mL. The corresponding IC50 values were 0.451±0.051 and 0.527±0.022 mg/mL, respectively (IC50 values for ascorbic acid, as the standard, were 0.209±0.016 and 0.245±0.022 mg/mL, respectively). The mycelium of F. oxysporum was distorted and collapsed when treated with 0.5 mg/mL of the EO of K. parviflora rhizome for 3 days treatment, which may provide an important information for exploring the metabolism of the fungicide K. parviflora rhizome and its derived compounds against F. oxysporum. This study provides the chemical properties of the essential oil of K. parviflora rhizome grown in Vietnam and their potential antioxidant and antifungal activities.

Antioxidant and Antimicrobial Activities of the Ethanol Extract of Allium victoriallis L. var. platyphyllum

  • This study was conducted to evaluate in vitro antioxidative and antimicrobial properties of ethanolic extracts from Allium victoriallis L. var. platyphyllum (AVP) with 6 different parts. The antioxidative activities of these samples were determined using the 4 separate methods that involved reducing power, DPPH, hydrogen radical scavenging, and lipid peroxidation with use of a $\beta$-carotene/linoleic acid system. The leaf part ethanolic extracts (1,000 ppm) showed the strongest inhibitory potential for reducing power, DPPH, and hydroxyl radicals to 99.8, 49.4, and 52.8%, respectively. Inhibition values of linoleic oxidation were calculated as 58.0, 39.5, and 38.0% for seed, flower, and leaf ethanolic extracts (1,000 ppm), respectively, from AVP. In addition, the ethanolic extracts of the root part showed the most effective antimicrobial activity. The inhibition zones of the root ethanolic extracts ($200\;{\mu}g/disc$) of AVP against Bacillus cereus and Staphylococcus aureus were 17 and 14 mm, respectively. In a micro-dilution assay, B. cereus, S. aureus, and Vibrio parahaemolyticus exhibited sensitivity to root part ethanolic extracts with an minimum inhibition concentration (MIC) value of 20, 28, and 18 mg/mL, respectively. Therefore, the AVP extracts may be suitable as antimicrobial and antioxidative agents in the food industry.

Antioxidative Activity and Irritation Test of a Complex Herbal Bath Extracted from Korean Traditional Plants

  • The purpose of this study is to evaluate the free radical scavenging ability and xanthine oxidase inhibitory activity of a complex herbal bath consisted of Artemisiae argyi folium, Angelicae sinensis radix, Ligustici wallichii radix and Angelicae tuhuo radix, and its potential irritation response were also tested for safety use in the rabbits. For antioxidative activity, the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity of the complex herbal bath were examined at five different concentrations (0, 250, 500, 1000 and 2000 ㎍/ml). The concentration of the complex herbal bath required for scavenging DPPH free radical by 50% was 897.2 ㎍/ml. In the inhibition of xanthine oxidase (XO) activity, the concentration of the complex herbal bath required for 50% of inhibition was 221.4 ㎍/ml. In the skin irritation study in rabbit, all animals survived for the duration of the study and the examined skin exhibited no edema, erythema, and eschar formation. In the ocular irritation study in rabbit, after application of the sample to eyes, all of the eyes were normal. In summary, the complex herbal bath has potent antioxidant effects against the DPPH radical and XO and was considered to be a non-irritation bath for safety use.

가축사양에 있어서 감 과피와 감 과피탄닌 추출물의 이용 (Utilization of Persimmon Peel and Its Tannin Extracts for Animal Feeding)

  • Tannins are phenolic compounds that precipitate proteins and composed of a very diverse group of oligomers and polymers. Tannins are potential biological antioxidants, which are widely believed to be an important line of defense against oxidative damage and may participate in the prevention of cancer and cardiovascular disease. Persimmon(Diospyros kaki L.) has been cultivated in East Asia and is a good source of nutritional antioxidant vitamins, carotenoids and tannins. In general persimmon peel was regarded as a waste matter, although based on recent studies, the peel contains more carotenoids and polyphenols than pulp. Several investigation conducted in experimental animals have reported that dietary persimmon fruit and peel effectively lowered the levels of plasma total cholesterol and LDL-cholesterol. We conducted experiments to investigate in vitro antioxidative activities of persimmon peel powder (PP) and its soluble tannin extract (ST) and their dietary effects on productive performances and physiological responses in poultry. The PP and ST exhibited in vitro antioxidative activity in SOD - like activity model. The yolk color and eggshell color were significantly improved by the addition of PP and ST into layer diets. The contents of total cholesterol, triacylglycerol and phospholipid of liver in the groups fed diets containing PP and ST tended to be reduce as compared with those of control. With adding of PP and ST, Haugh unit was increased after 7 and 14 days of storage. In conclusion, PP and ST can be used as valuable feed additives for reducing hepatic lipid contents without harmful effects on overall productive performances and physiological responses in laying hens.

Antioxidant and Anti-Obesity Activities of Polygonum cuspidatum Extract through Alleviation of Lipid Accumulation on 3T3-L1 Adipocytes

  • Natural products are widely used due to their various biological activities which include anti-inflammatory, antioxidant, and anti-obesity effects. In this study, we determined the antioxidative and anti-obesity effects of Polygonum cuspidatum 50% ethanol extract (PEE). The antioxidative effect of PEE was evaluated using its radical scavenging activity, total phenolic content, and reducing power. The anti-obesity effect of PEE was investigated using 3T3-L1 adipocytes. The antioxidative activity of PEE was progressively increased in various concentrations, mainly due to the presence of phenolic compounds. PEE also alleviated lipid accumulation on 3T3-L1 adipocytes and downregulated the mRNA and protein production of adipogenesis-related (SREBP-1c, PPARγ, C/EBPα) and lipogenesis-related (aP2, FAS, ACC) markers. Furthermore, we found that the inhibitory effect on lipid accumulation via PEE was caused by the alleviation of NF-κB, p38 MAPK, ERK1/2, and JNK at the protein level. Taken together, our results imply that PEE is a potential antioxidant that can prevent obesity-associated disorders.

  • This study was conducted to develop and characterize of a high quality salted mackerel using enzymatic extracts of Ecklonia cava (EEC). In this study, potential antioxidative properties of EEC were evaluated by DPPH free radical scavenging activity, hydrogen peroxide scavenging activity, peroxide value, and fatty acid composition, and the antimicrobial properties were also measured by analysis for volatile basic nitrogen, pH, viable cells, Eschericia coli and biogenic amine. Compared to EEC-untreated salted mackerel, the salted mackerel with EEC was superior in antioxidative properties, while was negligible in the difference of antimicrobial properties. These results suggested that the high quality salted mackerel with antioxidative activity could be developed by treatment of EEC.

중국산 녹두의 항산화 활성 (Antioxidant Activity of Chinese Mung Bean)

  • 본 연구는 이용도가 높은 녹두의 국내 생산 부족으로 수입이 증가되고 있는 중국산 녹두의 천연 항산화성 소재로 활용시 기초자료를 제공하고자 하였으며, 녹두의 메탄올 추출물의 페놀성 화합물 함량과 in vitro 항산화 모델(DPPH 라디칼 소거능, ABTS 라디칼 소거능)을 통하여 항산화 활성을 측정하였다. 중국산 녹두 추출물의 총 페놀 및 플라보노이드 함량은 각각 $174.83{\pm}2.90GAE\;mg/g$$68.87{\pm}2.84QE\;mg/g$으로 나타났다. 중국산 녹두 추출물의 DPPH 라디칼 소거능과 ABTS 라디칼 소거능은 농도 의존적으로 유의적으로 증가하였으며(p<0.05), 초기 ABTS 라디칼의 50%를 소거하는데 필요한 농도($IC_{50}$)는 2.85 mg/mL로 나타났다. 이상의 결과로부터, 중국산 녹두는 페놀성 화합물이 풍부하며, 천연물 유래 항산화성 소재로서 활용 가능성이 높은 것으로 기대된다.
