LC-MS/MS를 이용한 육류 중 아미노글리코사이드계 항생제 9종의 동시분석 및 적용성 검증 (Simultaneous Determination of Aminoglycoside Antibiotics in Meat using Liquid Chromatography Tandem Mass Spectrometry)

  • LC-MS/MS를 이용하여 아미노글리코사이드계 동물용의 약품 9종에 대하여 동시분석법을 확립하고 잔류실태조사를 수행하였다. 현재 식품공전에 수록된 시험법을 기본으로 분석법을 개선하였으며 국제식품규격위원회 기준에 따라 특이성, 직선성, 정확성, 정밀성, 정량한계 등을 검증하였다. 아미노글리코사이드 혼합표준용액을 잔류허용기준의 0.125배, 0.25, 0.5, 1, 2, 4배 농도에 따라 검량선을 작성한 결과 0.98 이상의 직선성을 확인하였다. 소고기의 회수율은 60.5~114%, 돼지고기의 회수율은 60.1~112%, 닭고기 회수율은 63.8~131%을 나타내었다. 또한 LOD는 0.001~0.009 mg/kg, LOQ는 0.006~0.03 mg/kg이었다. 국내유통 중인 축산물 (소고기, 돼지고기, 닭고기) 193건을 수거하여 개선된 분석법의 적용성 검증과 축산물의 안전성 확인을 위한 잔류조사를 수행한 결과 시험법의 적용성이 충분히 검증되었으며 모든 시료에서 아미노글리코사이드 계열의 항생제가 검출되지 않아 유통 축산물이 안전한 것으로 확인되었다. 확립된 분석법은 식육에 잔류할 수 있는 아미노글리코사이드계열 동물용의약품 안전관리에 활용할 수 있을 것으로 사료되는 바이다.

Microbiological and Chemical Detection of Antibiotic Residues in Livestock and Seafood Products in the Korean Market

  • The microbiological and chemical identification of antibiotic residues was attempted for livestock and seafood products including pork (n=34), beef (n=34), chicken (n=32), flatfish (n=37), armorclad rockfish (n=36), and sea bream (n=27). The meat (n=100) and seafood (n=100) samples were collected from 9 markets in 5 major Korean cities. Antibiotic substances were identified from the classes of tetracyclines, macrolides, penicillins, aminoglycosides, polyethers, peptides, sulfonamides, quinolones, chlorampenicols, and novobiocins using a microbiological assay, the Charm II test and high performance liquid chromatography (HPLC) with ultra violet (UV) and fluorescence detectors. The results showed that 2 tetracyclines (oxytetracycline and tetracycline) and 3 quinolones (ciprofloxacin, norfloxacin, and enrofloxacin) were detected in 4 samples of flatfish among all 100 seafood samples tested. No antibiotic residues were detected in the 100 livestock product samples tested. The amounts (min-max, mg/kg) of the residual antibiotics were as follows; tetracycline 0.78-0.85, oxytetracycline 0.49-0.74, ciprofloxacin 0.09-0.83, norfloxacin 0.01-0.21, enrofloxacin 0.12-2.98. These data indicate that the total detection rate of antibiotics in livestock and seafood products was approximately 2%.

우유(牛乳)의 잔류항생물질(殘溜抗生物質)에 관(關)한 연구(硏究) (Studies on Antibiotic Residues in Cow Milk)

  • The antibiotics are usually used for the medical treatments or the other purpose for cow and almost of them are transfered to milk. Moreover a serious problems on public health are caused by misuse or overuse of antibiotics. These studies were conducted to determine the rise and fall of antibiotics in the raw milk which is milked from the cow treated with antibiotics. The number of samples examined were 500 which were taken from 36 cows, and the antibiotics in milk were detected by cylinder plate assay method. The results were as follows: 1) When penicillin, chloramphenycol and streptomycin were infused into a udder, they were detected up to the period of 84 hours, 36 hours and 60 hours after injection in the milk from the same udder, however in the milk from the other udder streptomycin was not detected but penicillin and chloramphenycol were detected up to 72 hours and 24 hours after injection. When streptomycin and penicillin were injected intramusculary, streptomycin was not detected in the milk but penicillin was detected up to 60 hours after injcetion. 2) When penicillin, streptomycin and chloramphenycol were infused into the udder, the lesser amount of milk was produced, the more antibiotics were detected in the milk. 3) It is necessary policy for the public health to inhibit to be sold the milk up to 3 days or more after the injection on antibiotics or to add food blue to mastitis drugs.

우유내 잔류물질 검사방법인 TTC-II법 및 Disk assay법의 비교시험 (Comparision of TTC-II and disk assay method for the detection of antibiotic residues in milk)

  • This test process on screening method for the detection of residual antibiotics in milk is simple, economic, sensitive to residual antibiotics and was given approval international organs. Thus, this study was carried out the comparison of Disk assay method and TTC-II method for sensitivity and minimum detectable range of antibiotics in raw milk. The results of this study was summarized as follows ; 1. The number of samples requested for treatment of mastitis was 198 samples. Comparison or analytical results among the methods of TTC-II, disk assay and Delve sp was that TTC-II 37 samples(18.6% ), Disk assay 125samples(63.1%), Delve SP 130 samples(65.7% ) reacted positively. Conformity rate of Delve SP and Disk assay was 70%. 2. Detectable limits of disk assay method in some antibiotics were more sensitive than those of official method(0.05-0.0025ppm in the $\beta$-lactams, 1ppm in two aminoglycoside, 0.2 ppm in one tetracycline, similar in one macrolide) 3. For sensitivity of residual sulfonamides TTC-II was much more sensitive than disk assay. Detectable limits of sulfamethazine and sulfadimethoxine were 30 to 50ppm levels. 4. The best medium preservation period is 1-2 days. 5. Concentration of brome cresol purple related to resistance for B stearothermophilus culture was 24ppm/ml. These results show that disk assay method for screening detection of antibiotics residuces in milk is worthy of use.

시료 중 잔류 항생제 분석 방법: I. 고상 시료 전처리 방법 (Determination of Antibiotic Residues: I. Extraction and Clean-up Methods for Solid Samples_A Review)

  • Korea is one of the countries with a large veterinary antibiotics market, although antimicrobial resistance in bacteria is becoming a serious issue in many countries. The Korean government started to take interest in estimating the effects of livestock manure on rivers and agricultural soils and in monitoring of heavy metals, organic pollutants and antibiotics in the ambient water and soil. In this paper, pre-treatment methods to separate the selected antibiotics from solid samples were reviewed. It is essential to select an efficient and appropriate procedure for pre-treatment due to the high proportion of proteins and organics in biosolid samples. Pre-treatment consists of extraction followed by clean-up. Initially, homogenized samples were extracted by sonication, mechanical agitation or pressurized liquid extraction with methanol/acetonitrile/water mixture under acidic/basic conditions depending on the compound. However, aminoglycosides and colistin were extracted with 5% trichloroacetic acid and HCl, respectively. Since the ${\beta}-lactams$ are easily decomposed in acidic and basic conditions, they were extracted in neutral pH. Filtration with a membrane (pore size, $0.2{\mu}m$) or solid phase extraction with HLB and methanol, as eluents, was normally applied for the clean-up. At least, three different pre-treatment procedures should be adopted to screen all the selected antibiotics in solid samples.

소 및 돼지의 정육과 내부장기중의 항생물질 잔류 조사 (Detection of Antibiotic Residues in Meats and Internal Organs of Cattle and Pigs)

  • The purpose of the present survey was to evaluate the antibiotic residues in meats and internal organs such as muscle, liver, heart, kidney and spleen of cattle (n=59) and pigs (n=115). The EEC-4-plate-method were employed. The results were obtained as follows ; 1. In BS 6.0, BS 7.2 and BS 8.0 used as media to detect antibiotic residues, the zone($M{\pm}SD,$ cm) of bacterial growth inhibition was narrow($1.40{\pm}0$) in meats, whereas the zone was wide($1.69{\pm}0.25-1.88{\pm}0.23$ and $1.58{\pm}0.18-1.86{\pm}0.15$ in cattle and pigs, respectively) in internal organs. But in SL 8.0, it was difficult to detect the zones ($0-1.40{\pm}0$) of both meats and internal organs. 2. Residues of antibiotic in beef and pork were rarely detected in BS 6.0, BS 7.2 and BS 8.0 (range 1.7-11.9% and 2.6-4.3%, respectively), whereas residual percentages of internal organs were relatively higher(range 69.5-96.6% and 43.5-84.3%, respectively). But in SL 8.0, it was not detected in both beef or pork, whereas they were 0-13.6% and 0-4.3% in interanal organs.

  • Food safety is a term broadly applied to food quality that may adversely affect human health. These include zoonotic diseases and acute and chronic effects of ingesting natural and human-made xenobiotics. There are two major areas of concern over the presence of residues of antibiotics in animal-derived foodstuffs with regard to human health. The first is allergic reactions. Some antibiotics, such as penicillins can evoke allergic reactions even though small amounts of them are ingested or exposed by parenteral routes. The second is development of antibiotic resistance in gut bacteria of human. Recently multi-resistant pneumococcal, glycopeptide-resistant enterococci and gram negative bacteria with extended-spectrum $\beta$-lactamases have spread all over the world, and are now a serious therapeutic problem in human. Although it is evident that drugs are required in the efficient production of meat, milk and eggs, their indiscriminate use should never be substituted for hygienic management of farm. Drug should be used only when they are required. In addition to veterinary drugs, environmental contaminants that were contaminated in feed, water and air can make residues in animal products. Mycotoxins, heavy metals, pesticides, herbicides and other chemicals derived from industries can be harmful both to animal and human health. Most of organic contaminants, such as dioxin, PCBs and DDT, and metals are persistent in environment and biological organisms and can be accumulated in fat and hard tissues. Some of them are suspected to have endocrine disrupting, carcinogenic, teratogenic, immunodepressive and nervous effects. The governmental agencies concerned make efforts to prevent residue problems; approval of drugs including withdrawal times of each preparation of drugs, establishment of tolerances, guidelines regarding drug use and sanitation enforcement of livestock products. National residue program is conducted to audit the status of the chemical residues in foods. Recently HACCP has been introduced to promote food safety from farm to table by reducing hazardous biological, chemical and physical factors. Animal Production Food Safety Program, Quality Assurance Programs, Food Animal Residue Avoidance Databank are para- or non-governmental activities ensuring food safety. This topic will cover classification and usage or sources of chemical residues, their adverse effects, and chemical residue status of some countries. Issues are expanded to residue detection methodologies, toxicological and pharmacokinetic backgrounds of MRL and withdrawal time establishments, and the importance of non-governmental activities with regard to reducing chemical residues in food.

가축매몰지 및 인근 농경지의 축산용 잔류 항생제 모니터링 (Monitoring of Selected Veterinary Antibiotics in Animal Carcass Disposal Site and Adjacent Agricultural Soil)

  • 2010년 발생한 가축전염병인 구제역(FMD)으로 인해 전국에 약 4,700여개소에 가축 매몰지가 조성되었다. 매몰된 가축의 부패 과정에서 발생하는 침출수는 항생제와 같은 다양한 오염물질들을 함유하고 있어 주변 토양 및 수계로 유입되는 경우 환경에 악영향을 초래할 수 있다. 이와 같이 환경에 잔류하는 항생제는 내성박테리아 생성 등을 통해 인간건강 및 생태계 건전성을 위협할 수 있다. 이에 본 연구에서는 가축 매몰에 의한 항생제 오염수준을 평가하기 위해 가축 매몰지 및 인근 농경지 토양에서 항생제 모니터링을 실시하였다. 모니터링 대상 항생제로는 축산용 항생제로 사용량이 가장 많은 tetracycline 계열의 tetracycline (TC), chlortetracycline (CTC), oxytetracycline (OTC)와 sulfonamide 계열의 sulfamethazine (SMZ), sulfamethoxazole (SMX)을 선정하였다. 항생제의 잔류농도는 매몰지 (TC: $144.26-350.73{\mu}g/kg$, SMZ: $17.72-44.94{\mu}g/kg$)가 인근 농경지 (TC: $134.16-320.73{\mu}g/kg$, SMZ: $6.48-8.85{\mu}g/kg$)에 비해 높은 것으로 나타났으며 CTC, OTC, SMX는 검출되지 않았다. 연구결과를 통해 단정할 수는 없으나 매몰된 가축사체에 함유된 항생제가 매몰지 및 인근 토양에 축적될 수 있는 개연성이 있는 것으로 판단되었다. 특히, 농경지에 잔류하는 항생제는 농작물에 의해 흡수되어 인간 건강에 악영향을 미치므로 이에 대한 지속적인 모니터링이 요구된다.

  • Increased attention has been paid to the presence of veterinary antibiotics in various environmental matrices due to their toxicological behavior in the ecosystem and development of antibiotic-resistant strains of pathogenic bacteria. In the this review, 37 target antimicrobials were selected based on annual sales of antibiotics for livestock in South Korea 2014. Also, extraction and clean-up methods for the determination of the antibiotic residues in liquid samples including water, milk, and honey were comprehensively reviewed in the literature. Solid-phase extraction (SPE) was commonly used as a pre-treatment method for the samples. Most of the analytes were extracted in acidic conditions (2.5~4.0) except for aminoglycosides, which were extracted in neutral conditions (7.0~8.0). ${\beta}-Lactams$ showed the highest recoveries in neutral pH due to their degradation characteristics in acidic media. Starta-X, Oasis HLB, and Oasis MCX were frequently applied as an SPE cartridge and Oasis HLB showed the highest recoveries for the majority of antibiotic classes. The homogenized honey and milk were extracted by mixing with acids for deproteinization. Solids and other interfering substances in the extract were eliminated by centrifugation followed by membrane filtration or SPE before injection into HPLC.

Application of a solid-phase fluorescence immunoassay to determine streptomycin residues in muscle tissue of olive flounder (Paralichthys olivaceus), rockfish (Sebastes schlegeli), and red sea bream (Pagrus major)

  • Parallux (IDEXX Laboratories, USA), a solid-phase fluorescence immunoassay (SPFIA) developed for antibiotics residue detection in milk, was applied for analysis of antibiotics in muscle tissue of olive flounder (Paralichthys olivaceus), rockfish (Sebastes schlegeli), and red sea bream (Pagrus major). Fishes were dipped in streptomycin 20 g/ton water, the recommended therapeutic dose, for 3 consecutive days. Muscle was sampled after 1st, 2nd, 3rd, 4th and 5th day drug treatment. The concentration of streptomycin in muscle, determined by SPFIA, was compared with that of internal standard (200 ppb as streptomycin). The absorbance ratio of sample to internal standard (S/C) was employed as an index to determine the muscle residues in fishes. The standard solutions were added to muscle samples to give final concentrations in muscle of 0.2 and 0.5 mg/ml to investigate the recovery rate. The recovery rates of all spiked samples were > 84% of the spiked value. Streptomycin was detected in muscles of fishes treated after the 1st day of withdrawal period. The present study showed that the SPFIA can be easily adopted in predicting muscle tissue residues for streptomycin in farmed fishes.
