Objectives : This study investigated the anti-oxidant activities and improving effect of Phellinus linteus and Glycyrrhiza uralensis Extract (PGE) on Atopic Dermatitis. Methods : 2,2′-diphenyl-1-picrylhydrazyl (DPPH) radical, 2,2′-azino-bis 3-ethylbenzothiazoline-6-sulphonic acid (ABTS) radical, Hydrogen peroxides scavenging activities and Superoxide dismutase (SOD)-like activities were used for the measurement of anti-oxidant ability. Cytotoxicity of PGE in Raw 264.7 cell was evaluated by MTT assay. To evaluate the anti-atopic dermatitis effect of PGE, a total of 33 patients with atopic dermatitis were observed trans epidermal water loss, skin moisture content, modified SCORAD index of atopic dermatitis and pruritic degree after applying the PGE for 4 weeks. Results : PGE scavenged DPPH ($IC_{50}=25ppm$) effectively, ABTS and Hydrogenperoxides scavenged similar to BHA. As for the SOD-like activity, it had lower effect than ascorbic acid, but it comparable activities in 500ppm. There was no cytotoxicity at PGE at concentrations of 10,000ppm. In clinical research about PGE on patients with atopic dermatitis, skin condition was improved. After 4 weeks, the application of PGE increased skin moisture content from 19.43 to 31.22. Moreover, it reduced the skin temperature (from 32.5 to 31.9), skin pH (from 5.39 to 5.22), trans epidermal water loss (from 39.03 to 24.46) and pruritus score (from 6.07 to 3.87). In addition, the Modified SCORAD index decreased from 31.28 to 20.3. Conclusions : In conclusion, PGE possesses anti-oxidant and anti-atopic dermatitis activities, thus it could be potentially valuable as anti-atopic dermatitis material.

Objectives : The purpose of this study was to investigate the anti-inflammatory and anti-oxidative effects of Osung-tang(OST) extract. Methods : MTT assay was performed to confirm the survival rate of RAW 264.7 cells treated with OST extract(50-400㎍/㎖) and the production of NO from LPS-induced RAW 264.7 cells was confirmed. The mRNA expression of iNOS, COX-2, and NF-κB were measured by real-time PCR. The protein expression of iNOS, COX-2 and p-IκB were measured by western blot and the anti-oxidant activity of OST extract(50-400㎍/㎖) was investigated by measuring DPPH scavenging activity. Results : OST extract showed a cell survival rate of 90% or more at 50-400㎍/㎖. The NO production was inhibited in a dose-dependent manner in RAW 264.7 cells treated with LPS on OST treated group. mRNA expression levels of iNOS, COX-2 and NF-κB decreased in a concentration-dependent manner after treatment with OST(50-400㎍/㎖). Protein expression levels of iNOS, COX-2 and p-IκB decreased in a concentration-dependent manner after treatment with OST(50-400㎍/㎖). It was found that OST has a high free-radical scavenging ability. Conclusions : These results suggest that OST extract can be used as a treatment for various skin diseases by demonstrating its anti-inflammatory and anti-oxidant effects.

Objectives : Agrimoniae Herba is a herbal medicine widely distributed in Asia and contains flavonoids including catechin, quercitrin, rutin, hyperoside, and quercetin. This study aimed to investigate the anti-oxidant activity and skin barrier function of different solvent fractions (Hexane; methylene chloride, MC; ethyl acetate, EA; n-butanol, Bu; Water) obtained from Agrimoniae Herba. Methods : Anti-oxidant activity of different solvent fractions obtained from Agrimoniae Herba was investigated through total polyphenol contents, total flavonoid contents, 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity measurements. Then, filament aggregating protein (Filaggrin), Type I collagen, ceramide synthase (CERS) 3, and CERS4 were analyzed to evaluate the skin barrier strengthening effect of different solvent fractions obtained from Agrimoniae Herba on UVB-stimulated HaCaT cells. Results : As a result of measuring total polyphenol contents, total flavonoid contents, DPPH free radical scavenging activity, and ABTS radical scavenging activity, antioxidant activity was found to be excellent in the order of EA > Bu > MC > Hexane > Water. As a result of measuring mRNA gene expression of Type I collagen, Filaggrin, CERS3, and CERS4 after UVB-stimulated was applied to HaCaT cells treated with different solvent fractions obtained from Agrimoniae Herba, it was found to increase significantly in the Bu-treated group. Conclusion : Our findings show that the Bu sample obtained from Agrimoniae Herba has excellent anti-oxidant ability, which increases Type I collagen, Filaggrin, and ceramide synthetase in UVB-stimulated HaCaT cells to control the skin barrier improvement effect.

Objectives : This study investigated the anti-oxidant and anti-inflammatory activities of Changchulgeumryeontang (CCGRT) extract. Methods : The macrophage cell line RAW 264.7 cells were used and MTT assay was performed to measure the cell viabilities at the various concentrations of CCGRT ($25-200{\mu}g/m{\ell}$). Nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) were measured in LPS-induced RAW 264.7 cells. Expressions of iNOS, $NF-{\kappa}B$, $IL-1{\alpha}$, $IL-1{\beta}$ and IL-6 were also performed by real-time PCR. The anti-oxidant activities of CCGRT was measured by DPPH radical scavenging activity. Results : 1. there was no cytotoxicity in RAW264.7 cells treated with CCGRT compared to the control. 2. CCGRT treated group significantly inhibited NO and $PGE_2$ production compared to the LPS treated group. 3. CCGRT treated group significantly decreased mRNA expressions of iNOS, $NF-{\kappa}B$, $IL-1{\alpha}$, $IL-1{\beta}$ and IL-6 compared to the LPS treated group. 4. CCGTR was found to have high DPPH free radical scavenging ability. Conclusions : According to the above results, CCGRT may be a potentional choice for the treatment of inflammatory skin disease. Objective : In this study, anti-oxidant and cosmeceutical activities of Isatis tinctoria L. extracted from water, ethanol and supercritical fluid condition were confirmed to investigate cosmeceutical activities for utilization as cosmetic ingredient. Methods: Anti-oxidant and cosmeceutical activities were investigated by using electron donating ability, xanthine oxidase, tyrosinase, astringent effect. Result : Isatis tinctoria L. extracts by supercritical fluid, water and ethanol showed good electron donating ability which were 82.7%, 62.6% and 44.8% at the concentration at 1,000ppm, respectively. Xanthine oxidase activity related with purine metabolism was inhibited by ethanol extract about 52.3% at the concentration at 1,000ppm. Tyrosinase inhibition effects, by supercritical fluid extract, ethanol extract and water extract, were 83.3%, 52.9% and 41.2% respectively at 1,000ppm. In the measurement of astringent effect, supercritical fluid extract at the concentration to 5,000ppm showed 85.7% in related activity. The water extract showed 95.9% nitrite scavenging activity at 5,000ppm. Conclusion : According to these results, it is possible that the extract of Isatis tinctoria L. can be used as a new natural material of cosmetic industry.

Objectives : This experiment study was performed to investigate the anti-inflammation and anti-oxidant effects of Indigo Naturalis(IN) and Rehmanniae Radix(RR) which were herbs for clearing heat. Methods : Anti-inflammation and anti-oxidation effects of IN and RR that were measured by the inhibitory ability of Nitric oxide(NO) production and the scavenging for 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical. Results : 1. IN and RR groups were not showed cell toxicity. 2. In the inhibitory ability of NO production, IN groups were better than RR groups, but there were no statistical significances among the groups. 3. In the scavenging for DPPH radical, IN groups were better than RR groups, but there were no statistical significances among the groups. Conclusions : These results showed that the IN group was better than the RR group in treating skin eruptions, especially those such as erythema, purple spot, papule, pustules which appear in acute inflammation.
Tremendous natural product extracts were used as a herb medicine remedy or therapy for centuries. Because these extracts have various biological activities, we examined the effects of Gryllotalpa orientalis extract for anti-oxidant and anti-inflammation activity by 1,1-diphenyl-2-picrylhydrazyl (DPPH), ferric reducing ability of plasma (FRAP), hydroxy radical scarvenging and cyclooxygenase-2 promoter assays. Gryllotalpa orientalis extracts were prepared from solvents such as distilled water (DW), dimethyl sulfoxide (DMSO), ethanol and methanol. The results showed that Gryllotalpa orientalis extracts have potent DPPH (methanol extract), FRAP (DW extract) and hydroxy radical scavenging (DW and methanol extracts) activity than any other extracts used. A significant inhibition of cyclooxygenase-2 (Cox-2) promoter activity was detected in the presence of DMSO extract or ethanol extract. Collectively, the present results suggested that Gryllotalpa orientalis extract could be used for anti-oxidant and/or anti-inflammation agent for human or agricultural purposes.
The solvent extracts of Prunus persica Flos were investigated for the activities of anti-oxidant and anti-inflammation to apply as a functional ingredient for cosmetic products. The electron donating ability of both ethanol (PPE) or acetone (PPA) extracts of P. persica Flos was above 90.0% at the concentration of 500ppm. The superoxide dismutase (SOD)-like activity of P. persica Flos extracts (PPE, PPA) were approximately 40.0% at 1,000 ppm. The xanthine oxidase inhibitory effect of P. persica Flos extracts (PPE, PPA) was approximately 30.0% at 1,000 ppm and equivalent to that of ascorbic acid. Hyaluronidase inhibition activity related to the anti-inflammation effect was 35.0% with the treatment of P. persica Flos extracts (PPW, PPE, PPA) at 1,000 ppm, respectively. In the experiment of anti-inflammation effect, P. persica Flos extracts (PPW, PPE, PPA) inhibited the generation of nitric oxide. In the antimicrobial activity test against the human skin-resident microflora such as Staphylococcus epidermidis and Propionibacterium acnes, a clear zone was identified from 4mg/disc in P. persica Flos (PPE) extract.

The purpose of this study is verification of the anti-oxidant effect and anti-inflammatory effect of Eggplant-cheongyeolsodokum composed of 8 herbs (Solanum melongena L., Lonicera japonica Thunb., Glycyrrhiza uralensis Fisch., Ligusticum chuanxiong Hort., Angelica gigas Nakai., Coptis deltoidea C. Y. Cheng et Hsiao., Gardenia jasminoides J. Ellis., Forsythia suspensa Vahl) to confirm the possibilities as useful cosmetic material. We used the modified prescription of 'cheongyeolsodokum' contained in Korean traditional medical book 'Donguibogam' as composition of Eggplant-cheongyeolsodokum and their proportions. Eggplant-cheongyeolsodokum were extracted with hot water, 70% ethanol and then powdered. To confirm anti-oxidant effect, we investigated radical scavenging ability (DPPH, $ABTS^+$, superoxide), superoxide dismutase (SOD)-like activity, total polyphenolic contents. Also to confirm anti-inflammatory effect, we investigated inhibition effect of nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages, and Inhibition effect of the expression of inflammatory-related proteins (iNOS, COX-2) by western blot analysis. As a result, Eggplant-cheongyeolsodokum showed good anti-oxidant and anti-inflammation effects, we suggest that it can be used as an active ingredient for cosmetics.
Objectives : JiaoTeng-Yuan(교등원) is said to be a prescription for preservation of health in $\ulcorner$HuaTuo.ZhongZangJing (화타.중장경)$\lrcorner$. It is known to have the effect of Bu-Shen (보신: strengthening kidney) and Yi-Shou (익수: prolonging the span of one's life). This study investigates whether JTY is effective on inhibition of oxidation stress. Methods : Sprague-Dawley rats (12-week-old, weight $300{\pm}20g$) were divided into 3 groups. The normal group (n=8) was injected with PBS (1ml/body, s.c) subcutaneously behind the neck. The control group (n=8) was injected with D-galactose (50mg/kg, 1ml PBS/body, s.c) to induce pathological animals. The JTY group was injected with the same treatment as the control group, and fed containing JTY (10%). All groups were treated 1 time per day for 6 weeks. After the rats were sacrificed anti-oxidant enzyme (SOD, CAT, G-px) activity, GSH quantity of RBC and tissue (heart, liver and kidney), and plasma Vit-C quantity were examined. Also, the MDA levels of liver and kidney, lipofuscin of heart and endurance of erythrocyte membrane were measured. Results : In the JTY group, RBC's SOD activity decline was halted by 21 % of the normal level, compared to the control group, G-px activity (unit/g of Hb) increased significantly, compared to the normal group, and the level of Vit-C in plasma increased by 16%. Heart's SOD activity was kept at the same level as that of the normal group; and CAT activity decline was halted by 26%. Kidney's CAT and G-px activities were kept at the same level as that shown in the normal group, implying the existence of a halting effect. Liver also showed a slight halting effect against the decline of anti-oxidant ability, but the effect was not significant (${\alpha}=0.05$). A comparison between the levels of peroxide in SD rats showed that the level of TBARS in plasma increased significantly in the control group and that it was normal in the JTY group. The livers in the JTY group, compared to those in the control group, showed 36% halting effect of the normal level while their kidney's indicated the level significantly lower than the normal level. Heart's lipofuscin increased significantly in the control group, but was alike in both the JTY and the normal groups. Endurance of erythrocyte membrane (%) decreased significantly in the control group while it was kept at the similar level in both the JTY and the normal groups, indicating the halting effect. Conclusions : This study suggests that JTY is effective to defend oxidation stress caused by D-galactose in the animals. It showed that the anti-oxidant ability was maintained and strengthened. On the other hand, it reduced the level of peroxide in animals. In sum, JTY appeared to have the equilibrium normal physiological function in SD rat.
