Inhibition of Growth and Microcystin Toxicity, and Characterization of Algicidal Substances from Lactobacillus graminis against Microcystis aeruginosa (Microcystis aeruginosa에 대한 Lactobacillus graminis의 성장 억제능, microcystin 분해 및 살조 물질의 특성)

  • For several decades, lactic acid bacterium (Lactobacillus graminis: LAB) has been generally recognized as safe. To develop the pan-environmental bio-control agent, algicidal activity of the live LAB cell and its culture filtrate (CF) was examined against Microcystis aeruginosa. LAB cells perfectly lysed M. aeruginosa within 3 days, while the CF had a less effect than the live cells, approximately 78% inhibition of algal growth during a same culture period. The concentration of microcystin in alone culture of M. aeruginosa was $7.1{\mu}gL^{-1}$, but gradually increased and leach $158.5{\mu}gL^{-1}$ on 10 days. However, LAB cells clearly decreased the microcystin by $10.3{\mu}gL^{-1}$ in the same period, approximately 93.5%. CF of LAB showed a strong algicidal activity over 75% between pH 2-7, 91.3% by the treatment of proteinase K, 87.8% by below 3 kDa in particle size, and 75.3% by heat treatment, respectively. Of five solvents, fractions of CF passed through solvents diethyl ether and ethyl acetate showed an obvious algicidal activity in the algal-lawn test. Among 5 fractions purified by silica-gel TLC plate, two spots showed a most strong removal activity on M. aeruginosa. Another analysis of GC indicate that CF contained six representative fatty acids. Even though most of these substance have been known as an anti-algal substance against M. aeruginosa, oleic acid is the most effective. These results suggested that the culture filtrate or specific substances, like a fatty acids, in comparison with live L. graminis can be a successful and eco-friendly agent to control Microcystis bloom.

Potential in the Application for Biological Control of Winter Diatom Bloom Caused by Stephanodiscus hantzschii (겨울철 녹조발생 원인종 Stephanodiscus hantzschii의 생물학적제어를 위한 미소생물제재의 적용실험)

  • To examine the algicidal effect of co-treatment of biological control agent on centric diatom Stephanodiscus hantzschii in the filtered water, an endemic algicidal bacterium (Pseudomonas putida) and a heterotrich ciliate (Stentor roselli) were iso-lated from Pal'tang reservoir, Korea. Bacterial isolate and ciliate removed 98% and 80% of the diatom for 7 days of cultivation. Co-treatments of these two agents perfectly inhibited the diatom growth, compared to the single treatment of each agent. This synergistic interaction of the bacterium and ciliate could provide an effective tool in the biomanipulation to control the diatom bloom in freshwater lakes and streams.

Improved Dissolution of Poorly Water Soluble TD49, a Novel Algicidal Agent, via the Preparation of Solid Dispersion

  • The objective of this study was to improve the extent of drug release as well as the dissolution rate of TD49, a novel algicidal agent, via the preparation of solid dispersion (SD). Among the various carriers tested, $Solutol^{(R)}$ HS15 was most effective to enhance the solubility of TD49. Subsequently, SDs of TD49 were prepared by using $Solutol^{(R)}$ HS15 and their solubility, dissolution characteristics and drug crystallinity were examined at various drug-carrier ratios. Solubili ty of TD49 was increased significantly in accordance with increasing the ratio of $Solutol^{(R)}$ HS15 in SDs. Compared to untreated powders and physical mixtures (PMs), SDs facilitated the faster and greater extent of drug release in water. Particularly, SD having the drug-carrier ratio of 1:20 exhibited approximately 90% of drug release within 1 hr. Differential scanning calorimetry (DSC) thermograms and X-ray diffraction (XRD) patterns suggested that SDs might enhance the dissolution of TD49 by changing the drug crystallinity to an amorphous form in addition to the increased solubilization of drug in the presence of $Solutol^{(R)}$ HS15. In conclusion, SD using $Solutol^{(R)}$ HS15 appeared to be effective to improve the extent of drug release and the dissolution rate of poorly water soluble TD49.

Application Possibility of Naphthoquinone Derivative Nq 4-6 for Mitigation of Winter Diatom Bloom (겨울철 규조류 대발생 제어를 위한 Naphthoquinone 유도체 Nq 4-6의 적용 가능성)

  • We conducted the algicidal activity screening tests using 10 L microcosm to investigate the possibility of the field application of naphthoquinone derivative Nq 4-6 compound as an algicide. We determined its application range to assess its algicidal effects on the phytoplankton and to evaluate the response of the planktonic community and the water environment to this chemical. From results of the microcosm experiments, Nq 4-6 compound showed high algicidal activity on the centric diatoms such as Stephanodiscus hantzschii and Cyclotella meneghiniana, but it had no effect on other phytoplankton. The abundance of S. hantzschii continuously increased in the control, but its cell density decreased 1 day after the Nq 4-6 treatment. In particular, Nq 4-6 showed algicidal activity of 94.4% against S. hantzschii 7 days after the treatment. The dominance index of phytoplankton community was lower in the treatment than in the control. The diversity index, richness index and evenness index of phytoplankton community was higher in the treatment. Environmental factors and biological factors did not show specific changes after the Nq 4-6 compound treatment. Therefore, the results of this study demonstrates that Nq 4-6 is an effective agent for the control of S. hantzschii blooms, and that the microcosm tests play a crucial role when assessing field application.

Development of Immobilized Naphthoquinone for Effective Algicidal Activity under Various Environmental Conditions and It's Ecological Changing Monitoring (다양한 환경에서의 효율적 녹조 저감을 위한 Naphthquinone 물질의 담체화 기술 개발 및 이에 따른 생태계 변화 모니터링)

  • Bloom of small centric diatom Stephanodiscus is quite occasional in winter season in temperate freshwater ecosystems. It often leads to degradation of water quality and affects the quality of supplied drinking water. In a previous study, we have found that naphthoquinone (NQ) 4-6 derivate is an effective tool for efficient mitigation of natural S. hantzschii blooms. In the present research, polylactide (PLA) and agar foam were used as immobilized agent for NQ 4-6 to improve the efficiency of NQ 4-6 compound releasing process for its application under various field conditions. Mesocosm experiments at 10 ton scale suggested that the abundance of S. hantzschii was continuously increased in the control and upon treatment of the mesocosm with immobilized NQ 4-6 from PLA and agar foam. Their algicidal activities were 78.8% and 77.1%, respectively, on S. hantzschii after 10 days. In the mesocosm experiments, the dynamics of biotic (bacteria, HNFs, ciliates, zooplankton) and abiotic (water temperature, dissolved oxygen, pH, conductivity, nutrients) factors remained unaffected. They exhibited similar trends in the control and treatment groups. Therefore, the immobilized NQ 4-6 from PLA and agar foam has potential to be used as an alternative algicidal substance to effectively mitigate natural S. hantzschii blooms under various field conditions. In addition, it not only can be used to control S. hantzschii, but also is an effective technique. The immobilized NQ 4-6 showed stable controlled release in desired system.

Effects of Biological Control Agent Algicidal Bacterium on the Phytoplankton Community and Microcystin-LR Contents in a Mesocosm Experiment (살조세균 적용이 식물플랑크톤 군집과 조류독소 분포에 미치는 영향)

  • Biological control agents (BCA; algicidal bacterium Xantobacter autotrophycus) plus casitone media, strongly changed physicochemical variables, standing crops of phytoplankton and microcystin-LR phytoplankton in 100-L mesocosm constructed in a small hexagonal pond (3.5 m ${\times}$ 5 m). No M. aeruginosa showed by 8 days, and 60% of total standing crops of phytoplanktons were decreased by the BCA treatment. BCA treatment also induced a strong decline of cellular extracted microcystin-LR (MCLR) and a remarkable increase of dissolved MCLR with the decrement in standing crops of cyanobacteria. In addition, BCA strongly increased all nutrients, but new outbreak of phytoplanktons hardly showed in the experimental mesocosm. The field application of BCA to controling the cyanobacterial bloom in large lakes and reservoirs is not relevant due to high concentration of nutrients and toxins. Thus, a further study is needed to diminish the adverse effects after BCA treatment for water quality preservation.

The difference of photosynthetic efficiency and electron transport rate by control of the red tide organism using algicidal substance and yellow clay (살조물질과 황토를 이용한 적조생물 제어에 따른광합성 효율 및 전자전달율의 차이)

  • The development of worldwide harmful algal blooms(HAB) is a serious problem for public health and fisheries industries. To evaluate the algicidal impact on the HAB species, algicide thiazolidinedione derivative (TD49) and yellow clay were examined, which is focus on assess the algicidal effects and inhibition to photosynthesis of HAB species. To obtain the detailed information, we analyzed the viability of target species related to activity Chl. a, photosynthetic efficiency($F_v/F_m$), and electron transport rate(ETR). Culture experiment was conducted to evaluate the algicidal effects of three harmful species(raphidophyceae Heterosigma akashiwo, Chattonella marina, and dinophyceae Heterocapsa circularisquama) and one non-harmful species (cryptophyceae Rhodomonas salina). Our experiments revealed that three HAB species were easily destroyed of the cell walls after TD49 dosing. Also, they had significantly reducing values of active Chl. a, $F_v/F_m$, and ETR, due to the damage of photosystem II by inter-cellular disturbance. As a result, the algicidal effect(%) for the three HABs were as follows, in the order of greatest to the least: H. circularisquama> C. marina> H. akashiwo. However, the algicidal effect for yellow clay remained to be <30% (p>0.01), implying that it may not have damaged the photosystem II. On the other hand, non-HAB R. salina was promoted at both TD49 and yellow clay treatments. Our results demonstrated that the TD49 is a good agent for the control of HABs H. akashiwo, C. marina, and H. circularisquama, whereas the yellow clay would not be suitable for the field application based on our experimental results.

  • CT(Sodium N-chloro-para toluenesulfonamide, Chloramine-T) known to be a strong oxidative agent was investigated to use as a candidate of disinfectant in the marine aquaculture industry by the analysis of the bactericidal activity against different aquatic microorganisms. One hundred percent mortality appeared at and above 16 ppm CT in sea water in flounder, the predominant species in the marine aquaculture of Korea, when exposed for 48hr. However, bactericidal activity was appeared to be very effective, and all different species of the fish pathogenic bacteria exposed to CT of less than 2 ppm in sea water were dead within 15 min. It allowed us to confirm that CT could be a very effective disinfectant to protect the spread of fish pathogenic bacteria derived from diseased fish or sea water in marine aquaculture. High concentration(10 ppm) with longer exposing time(24 hrs) was required for the algicidal activity of CT, at which concentration might induce the acute toxicity against fish, however, restricted the expanded use of CT for the elimination of zooplanktons at phytoplanktons in marine farms. Moreover the bactericidal activity of CT inhibited almost completely in the present of more than 10 ppm of organic materials pointed out that quality of the culturing sea water should be considered carefully for the application CT to the aquatic farms as a therapeutic agent.

Analysis of a Prodigiosin Biosynthetic Gene Cluster from the Marine Bacterium Hahella chejuensis KCTC 2396

  • Marine bacterium Hahella chejuensis KCTC 2396 simultaneously produced red antibiotic prodigiosin and undecylprodiginine. A complete set of the prodigiosin biosynthetic gene cluster has been cloned, sequenced, and successfully expressed in a heterologous host. Sequence analysis of the gene cluster revealed 14 ORFs showing high similarity to pig and red genes from Serratia spp. and Streptomyces coelicolor A3(2), respectively, and the gene organization was almost: similar to that of pig genes. These genes were named hap for Hahella prodigiosin, and determined to be transcribed as a single operon, by RT-PCR experiment. Based on the hap gene mutagenesis experiments and comparative analysis with pig and red genes, we propose a prodigiosin-biosynthetic pathway in KCTC 2396.

Growth Suppression of Microcystis aeruginosa by Pseudomonas aeruginosa AJ1 (Pseudomonas aeruginosa AJ1에 의한 Microcystis aeruginosa의 성장제어)

  • Among total 176 strains with antialgal effects isolated from So-ok stream in Korea, Pseudomonas aeruginosa AJ1 showed the highest removal efficiency for an algal species Microcystis aeruginosa (clear zone of diameter 50.0 mm on algal lawn after 20 days). The algal growth was suppressed even when the supernatant of AJ1 culture was applied, suggesting that extracellular substances are responsible for its antialgal activity. The removal activity of AJ1 was optimal under the following condition: pH 8, $30^{\circ}C$, and mannitol as a carbon source. The antialgal activity of AJ1 appeared to be dependent of the growth phase of M. aeruginosa, i.e., the highest at the early phase, but not its own phase. As expected, the algicidal effect was improved as the amount of the treated supernatant was increased; the highest removal efficiency (80.3%) was achieved when 40 ml/L of the supernatant was used. Interestingly, however, the removal rate was opposite. The highest removal rate ($8.2{\mu}g$ chl-a/ml supernatant/day) was achieved when low concentration (10 ml/L) was applied. These results suggest that P. aeruginosa AJ1 is a promising biological agent to control the problematic algal bloom.
