Volatile Flavor Constituents of Cooked Oyster Sauce Prepared from Individually Quick-frozen Oyster Crassostrea gigas Extract (IQF 굴(Crassostrea gigas) 복합엑스분을 이용한 굴 소스의 가열향기 성분)

  • The pacific oyster Crassostrea gigas has a desirable taste and flavor that differs from those of other fish and shellfish. In order to develop a high value-added product from individually quick-frozen oyster extract (IQFOE), we prepared an oyster sauce from IQFOE and characterized its volatile compounds using vacuum simultaneous steam distillationsolvent extraction / gas chromatography / mass spectrometry. The moisture, crude protein, crude ash, salinity, pH and volatile basic nitrogen contents of the oyster sauce were 60.6%, 8.2%, 9.2%, 9.3%, 5.7 and 21.0 mg/100 g, respectively. Seventy-six volatile compounds were detected in the cooked odor of the oyster sauce. These volatile compounds included 14 esters, including ethyl acetate, 13 nitrogen- containing compounds, including 2,4,6-trimethyl pyridine, 13 acids, including hexadecanoic acid, 12 alcohols, including ethyl alcohol and 6-methyl heptanol, 6 alkanes, 5 aldehydes, including benzaldehyde, 5 ketones, including 1-(2-furanyl)-ethanone, 4 furans, including 2-furancarboxaldehyde and 2-furanmethanol, 3 aromatic compounds, including d-limonene, and 1 miscellaneous compound. Esters, acids and nitrogen-containing compounds, and alcohols were the most abundant compounds in the odor of the cooked oyster sauce, with some aldehydes, ketones, and furans.

The Action of Extract of Aconitum koreanum R. Raymond on Isolated Clam Heart (우리나라 백부자의 적출 조개 심장운동에 대한 작용)

  • Korean aconitum (Aconitum koreanum R. Raymond) as one of the botanical crude drugs which pertain to helleboraceae has been extensively applied in Chinese medicine during the past decades. It has been particularly used in immortal tonic among the folk remedies in China, however, its general uses comprehend diuresis, cardiotonic, analgesia, neuralgia, gout and, furthermore, even neoplastic effect. The components of aconitum have been acknowledged as aconitine, mesaconitine, hypaconitine, aconine and so on. The main ingredient, aconitine has the advantage of causing the atrial fibrillation, but, its pharmacological research has not been fully elucidated. Although there are many reports with regard to the pharmacological effects on the motility of several animal hearts, their conclusions have not been regretfully coincided yet. The authors hereby paid attention to this point of view and made experiment to examine the relationship between the alcohol extract of Korean aconitum and the motility of the isolated clam heart, making the use of several drugs related to the heart such as serotonin, acetylcholine, pilocarpine, physostigmine, barium chloride, procaine and quinidine. The cardiac movement of the isolated clam (Meretrix lusoria) heart in the standard sea water solution was recorded with the electric kymograph according to the Magnus method. The results of the experiment are as follows. 1. The motility of the isolated clam heart represents the tendency of gradual inhibition in proportion to the concentration of AK-A $10^{-4}$, $5{\times}10^{-4}$, and $10^{-3}$. 2. The cardiac inhibitory effect of AK-A $10^{-3}$ antagonizes the motility of the isolated clam heart pretreated with serotonin $10^{-6}$. 3. The cardiac inhibitory effect of AK-A $10^{-3}$ antagonizes the systolic state appealed by barium chloride $10^{-3}$. 4. The systolic state caused by quinidine $10^{-4}$ is not inhibited by AK-A $10^{-3}$.

Analgesic Effect and Inhibition of Prostaglandin $E_2$ Activity and Pro-inflammatory Cytokines Production by Ethyl Alcohol Extract from New Herbal Formula (신규 생약 복합추출물에 의한 Prostaglandin $E_2$ 활성억제, 염증성 Cytokine 생성 저해 및 진통 작용에 관한 연구)

  • New herbal formula (NHF) is the ethanol extract mixture of Puerariae radix, Artemisia capillaries and Perilla frutescens. We have Investigated the effects on anti-inflammation by NHF and attempted acetic acid induced writhing to verify the analgesic effect. Macrophages and chondrocytes were obtained from mouse and rabbit. Inflammation was induced bγ interleukin-1, tumor necrosis $factor-{\alpha}$, $interferon-{\gamma}$, and lipopolysaccharide. NHF showed strong inhibitory efficacy against cytokine-induced proteoglycan degradation, $PGE_2$ production, NO production, and MMP-9 expression in rabbit articular chondrocyte. In the writhing test, NHF exhibited a dose-dependent inhibition of writhing. Futhermore, NHF increased the activity of SOD. NHF have anti-inflammatory and analgesic activities, and could be a good herbal medicine candidate for curing of osteoarthritis.

Optimization of Beverage Preparation from Schizandra chinensis Baillon by Response Surface Methodlogy (반응표면 방법에 의한 오미자 음료 제조의 최적화)

  • To develop the beverage from Schizandra chinensis Baillon, extraction condition was optimized, using the fractional factorial design with 3 variables and 3 levels, by a RSM computer program. The optimum extraction time, extraction temperature and alcohol concentration levels were 3 hrs, $80{\sim}85^{\circ}C$ and 0%(water) for a desired yield, color and non-volatile organic acid. And to obtain the optimum recipe, amounts of extract, sugar and citric acid were optimized, using the fractional factorial design with 3 variables and 3 levels, by a RSM program. The optimum extract, sugar and citric acid levels were 4%, 9.6%n and 0.06%, respectively, for a disired flavor and overall acceptance.

Chemopreventive Potential of an Ethyl Acetate Fraction from Curcuma Longa is Associated with Upregulation of p57kip2 and Rad9 in the PC-3M Prostate Cancer Cell Line

  • Background: Turmeric ($Curcuma$ $longa$) has been shown to possess anti-inflammatory, antioxidant and antitumor properties. However, despite the progress in research with $C.$ $longa$, there is still a big lacuna in the information on the active principles and their molecular targets. More particularly very little is known about the role of cell cycle genes $p57^{kip2}$ and Rad9 during chemoprevention by turmeric and its derivatives especially in prostate cancer cell lines. Methods: Accordingly, in this study, we have examined the antitumor effect of several extracts of $C.$ $longa$ rhizomes by successive fractionation in clonogenic assays using highly metastatic PC-3M prostate cancer cell line. Results: A mixture of isopropyl alcohol: acetone: water: chloroform: and methanol extract of $C.$ $longa$ showed significant bioactivity. Further partition of this extract showed that bioactivity resides in the dichloromethane soluble fraction. Column chromatography of this fraction showed presence of biological activity only in ethyl acetate eluted fraction. HPLC, UV-Vis and Mass spectra studies showed presence three curcuminoids in this fraction besides few unidentified components. Conclusions: From these observations it was concluded that the ethyl acetate fraction showed not only inhibition of colony forming ability of PC-3M cells but also up-regulated cell cycle genes $p57^{kip2}$ and Rad9 and further reduced the migration and invasive ability of prostate cancer cells.

  • In order to examine anti-inflammatory effect of Cinnamomi Ramulus (CR) alcohol extract on rheumatoid arthritis, the present study investigated the viability and TNF-${\alpha}$ production in Raw264.7 cells treated with CR and collagen induced arthritis in DBA/1J mice which were orally administered with CR prior to immunization. The results are as follows: CR extract at $20{\mu}g/ml$ inhibited the viability of Raw264.7 by 50%. CR at 15 and $20{\mu}g/ml$ showed a significant decrease in TNF-${\alpha}$ levels from Raw264.7 cells treated with LPS. CR administration decreased arthritic index in DBA/1J mice immunized with bovine collagen type II but it did not reach statistical significance. CR administration significantly decreased spleen weights obtained from mice in 6 weeks after immunization. CR administration significantly decreased serum anti-type II collagen antibody levels compared with control group. CR administration decreased serum IL-6 levels compared with control group but it did not reach statistical significance.

  • The fruit of Actinidia arguta (AA) has been used mainly for the treatment of skin diseases, diuresis, diabetes mellitus and osteoporosis in Korean traditional medicine. It is known that AA (hardy kiwi) fruit extract has an effect on 2-chloro-1,3,5-trinitrobenzene-induced atopic dermatitis-like skin lesions in NC/Nga mice. Mode of action for it is associated with the modulation of biphasic Th1/Th2 cytokines. Furthermore, DA9102 containing AA is a herbal medicine currently under phase II clinical trial for atopic dermatitis in Korea. However, no active principles of AA on the decrease of Th2 cytokines including IL-4 and IL-10 have been identified. In this study, bioactivity-guided fractionation of an alcohol extract from the dried fruits of AA using ELISA assay for IL-4 production led to the isolation of $\alpha$-linolenic acid (I), linoleic acid (II), ethyl linolenate (III), ethyl linoleate (IV) and ethyl stearate (V) as the major active components. These compounds showed the down-regulatory effects of IL-4 production in A23187-stimulated RBL-2H3 cells without cytotoxicity.

Enhancing Effect of Extracts of Phellodendri Cortex on Glucose Uptake in Normal and Insulin-resistant 3T3-L1 Adipocytes (3T3-L1 지방세포에서 황백 추출물의 Glucose Uptake 촉진 및 인슐린 저항성 개선 활성)

  • Anti-hyperglycemic effects of 17 medicinal plants that have been used for ameliorating diabetes in oriental medicine were evaluated using glucose transport assay in 3T3-L1 adipocytes. Higher activities were obtained by treating water or alcohol extract of Phellodendri Cortex (PC), which showed enhancing effects both on basal and insulin-stimulated glucose uptake. The latter effect of PC was completely inhibited by wortmannin, a specific inhibitor for phosphatidyl inositol 3-kinase (PI 3-kinase), but not affected by SB203580, A specific inhibitor for p38 mitogen-activatedprotein kinase(MAPK). Genistein, an inhibitor for tyrosine kinases, abolished the PC effects completely. Treatment of vanadate, an inhibitor for tyrosine phosphatases, together with PC showed no significant synergic enhancement in glucose uptake. The results of inhibitors associated with insulin signaling pathway indicated that extracts of PC enhance glucose uptake by PI-3 kinase activation which is an upstream event for GLUT4 translocation. Antidiabetic effects of PC extract might be also due to enhanced tyrosine phosphorylation and reduced tyrosine dephosphorylation. In addition, PC accelerated insulin-stimulated glucose uptake in insulin-resistant cells, recovering the uptake level close to that of normal cells. These findings may offer a new way to utilize extracts of PC as novel anti-hyperglycemic agents.

Scavenging Activities of Reactive Oxygen and Nitrogen Species by Junglans sinensis (호도(胡桃)의 활성산소 및 활성질소 제거 기전)

  • Reactive oxygen species (ROS) and reactive nitrogen species (RNS) are widely implicated in the aging process and age-related diseases. The present study was carried out to investigate scavenging activities of Junglans sinensis extract and its subfraction using fluorescent probes, DCF-DA, DAF-2 and DHR 123. Jungians sinensis was washed and crushed. The crushed Junglans sinensis was extracted 3times, each time with 3 volumes of methyl alcohol at $60^{\circ}C$ for 24 h. The extract was filtered and evaporated under a reduced pressure using a rotary evaporator to yield 16 g. Scavenging activities of $ONOO^-$ was measured by Kooy' method and ROS was measured by DCFDA assay. Junglans sinensis had the marked scavenging activites of $ONOO^-$, NO and $O_2^-$. Junglans sinensis scavenged $ONOO^-$ through electron donation and dose-dependently inhibited the nitration of bovine serum albumin by $ONOO^-$. Junglans sinensis also had ROS scavenging activity. Especially, ethylacetate fraction of Junglans sinensis showed the most effective scavenging activities for ROS and RNS. These results suggest that Junglans sinensis might be developed as an effective ROS and RNS scavenger Therefore, Junglans sinensis might be used as a preventive agent for the aging and relevant to aging of illness.

Effects of Scolopendra on Ethanol-Induced Erectile Dysfunction in Rats (Ethanol로 발기부전을 유도한 흰쥐의 성기능 개선에 오공(蜈蚣)추출물이 미치는 영향)

  • Scolopendra was known to cure erectile dysfunction. This study was aimed to investigate the effects of Scolopendra on the nitric oxide synthase activity, nitrite level, cyclic-GMP and erectile responses in rat's corpus cavernosum penis. The crushed Scolopendra was extracted 3 times, each time with 3 volumes of methyl alcohol at $60^{\circ}C$ for 24 h. The extract was filtered and evaporated under a reduced pressure using a rotary evaporator to yield 14.2 g. Scolopendra extract was oral-administered 50 mg per 1 kg of body weight for 30 days. First, samples were treated with Scolopendra, and then ethanol-treated rats and L-N-Nitroarginine methyl ester (L-NAME) treated rats were fed with the samples. The level of urethral nitrite and nitric oxide synthase activity in the ethanol-Scolopendra double administered rats were as high as in the normal group, while the one in the ethanol-treated group was decreased. The level of urethral cyclic-GMP and guanylate cyclase actiyity in the ethanol-Scolopendra double administered rats were as high as in the normal group, while the one in the ethanol-treated group was decreased. The level of urethral phosphodiesterase activity in the ethanol-Scolopendra double administered rats was as low as in the normal group, while the one in the ethanol-treated group was increased. The erectile response to a cavernous nerve stimulation in L-NAME $(10^{-4})-treated$ rats was restored by the Scolopendra to the similar level seen in the normal group. The electile response to cavernous nerve stimulation in the ethanol-Scolopendra double administered rats were increased as high as in the normal group while the one in the ethanol-treated group was decreased. Scolopendra was effective in restoring the ethanol-induced or L-NAME-induced erectile dysfunction in rats.
