Optimization and Scale-up of Fish Skin Peptide Loaded Liposome Preparation and Its Storage Stability (어피 펩타이드 리포좀 대량생산 최적 조건 및 저장 안정성)

  • Fish skin peptide-loaded liposomes were prepared in 100 mL and 1 L solution as lab scales, and 10 L solution as a prototype scale. The particle size and zeta potential were measured to determine the optimal conditions for the production of fish skin peptide-loaded liposome. The liposome was manufactured by the following conditions: (1) primary homogenization at 4,000 rpm, 8,000 rpm, and 12,000 rpm for 3 minutes; (2) secondary homogenization at 40 watt (W), 60 W, and 80 W for 3 minutes. From this experimental design, the optimal conditions of homogenization were selected as 4,000 rpm and 60 W. For the next step, fish peptides were prepared as the concentrations of 3, 6, and 12% at the optimum manufacturing conditions of liposome and stored at $4^{\circ}C$. Particle size, polydispersion index (pdI), and zeta potential of peptide-loaded liposome were measured for its stability. Particle size increased significantly as manufacture scale and peptide concentration increased, and decreased over storage time. The zeta potential results increased as storage time increased at 10 L scale. In addition, 12% peptide showed the formation of a sediment layer after 3 weeks, and 6% peptide was considered to be the most suitable for industrial application.

Influence of heading date difference on gene flow from GM to non-GM rices (GM벼에서 non-GM벼로 유전자 이동에 대한 개화기 차이의 영향 분석)

  • Genetically modified (GM) crops have been increased continuously over the world and concerns about the potential risks of GM crops have also been increasing. Even though GM crops have not been cultivated commercially in Korea, it should be necessary to develop the safety assesment technology for GM crops. In this study, we investigated the influence of heading date difference on gene flow from GM to non-GM rice. In the experimental plot design, The PAC GM rice was placed in the center as a pollen donor and non-GM rice were placed in eight directions as pollen receivers. Five pollen receiver rice cultivars were Unkawng, Daebo, Saegyejinmi, Nakdong-byeo, and Ilmi which had different flowering times. A total of 266,436, 300,237, 305,223, 273,373, and 290,759 seeds were collected from Unkawng, Daebo, Saegyejinmi, Nakdong, and Ilmi, respectively, which were planted around PAC GM rice. The GM${\times}$non-GM hybrids were detected by repeated spraying of herbicide and PAT immunostrip assay. Finally, the hybrids were confirmed by PCR analysis using PAC gene specific primer. The hybrids were found in Nakdong-byeo which had the same heading date with PAC GM rice. The hybridization rate was 0.0007% at Nakdong-byeo plot. All of GM${\times}$non-GM hybrids were located within 2 m distance from the PAC GM rice zone. The physiological elements including rice heading date were found to be important factors to determine GM?rice out crossing rate with GM rice. Consideration should be taken into for many factors like the physiological elements of field heading date of rice cultivars to set up the safety management guideline for prevention of GM rice gene flow.

Determination and Validation of an Analytical Method for Dichlobentiazox in Agricultural Products with LC-MS/MS (LC-MS/MS를 이용한 농산물 중 Dichlobentiazox 시험법 개발 및 검증)

  • BACKGROUND: Dichlobentiazox is a newly registered pesticide in Korea as a triazole fungicide and requires establishment of an official analysis method for the safety management. Therefore, the aim of this study was to determine the residual analysis method of dichlobentiazox for the five representative agricultural products. METHODS AND RESULTS: Three QuEChERS methods were applied to establish the extraction method, and the EN method was finally selected through the recovery test. In addition, various adsorbent agents were applied to establish the clean-up method. As a result, it was found that the recovery of the tested pesticide was reduced when using the d-SPE method with PSA and GCB, but C18 showed an excellent recovery. Therefore this method was established as the final analysis method. For the analysis, LC-MS/MS was used with consideration of the selectivity and sensitivity of the target pesticide and was operated in MRM mode. The results of the recovery test using the established analysis method and inter laboratory validation showed a valid range of 70-120%, with standard deviation and coefficient of variation of less than 3.0% and 11.6%, respectively. CONCLUSION: Dichlobentiazox could be analyzed with a modified QuEChERS method, and the method determined would be widely available to ensure the safety of residual pesticides in Korea.

A Study on the Vascular Flora and its Management Plan at The Forest Genetic Resource Reserve of Mt. Munsu (Gimpo) (문수산(김포) 산림유전자원보호구역 관속식물상 변화 및 관리방안)

  • This study was investigated to find out the distribution of vascular flora and remarkable plants and its factors and management plan in the forest genetic resource reserve of Mt. Munsu, located in the DMZ and DMZ border area in Gimpo, Gyeonggi-do province. The survey was carried out 17 times from April 2019 to October 2020. First of all, in the forest genetic resource reserve of the Mt. Munsu, a total of 444 taxa in 95 families, 276 genera, 395 species, 13 subspecies, 33 varieties and 3 forms. This result was found to be about 9.09% of the total 4,881 taxa of vascular plants in Korea. In addition, endemic plants were classified as 6 taxa. Floristic special and rare plants were identified as 39 taxa and 3 taxa, respectively. Lastly, the invasive alien plants that appeared in Mt. Munsu of the forest genetic resource reserve were observed in 58 taxa. Moreover, the naturalization rate was 13.1% and the urbanization index was calculated to be 18.0%. As a result of comparing the vascular flora at the forest genetic resource reserve on Mt. Munsu identified in this study with previous studies, it was found the number of taxa was decreased compared to the previous studies, despite the fact that the recent survey section was wider and more varies routes were investigated. In particular, it was confirmed that the number of rare and endemic plants decreased significantly, and the number of invasive plants greatly spread to forest roads and hiking trails. Therefore, it is considered that the forest rest year system should be introduced for the conservation and management of native plants in the forest genetic resource reserve.

Association between Perceived Susceptibility to Fine Dust Exposure and Wearing Masks, Attitude toward Respiratory Disease Prevention Education in Farmers (농업인의 미세먼지 노출에 대한 인지된 감수성과 마스크 착용 및 호흡기질환 예방교육 참여 태도와의 연관성)

  • Objectives: This study aimed to investigate health risk awareness pertaining to fine dust exposure and the use of face masks in farmers, as well as their attitude toward education regarding fine dust-related respiratory disease prevention. Methods: In total, 295 farmers were interviewed in a survey using a structured questionnaire to obtain data on general characteristics, farming-related characteristics, health risk awareness pertaining to fine dust exposure, attitude toward education on fine dust-related respiratory disease prevention and the use of face masks. This study was analyzed the correlation between the perceived susceptibility to fine dust exposure and willingness to participate in education on fine dust-related respiratory disease prevention. Results: The mean score for perceived susceptibility to fine dust exposure was 3.8 (out of 5), and the participants were highly willing to receive education on fine dust-related respiratory disease prevention. In Multiple response analysis of reactions to exposure to fine dust generated during work, 221 participants responded that they practiced at least one preventive action; participants gave a positive response to "wearing masks" (56.1%), "personal hygiene, such as hand washing." (52.9%). In terms of education methods, 94 (33.6%) participants preferred to learn online or via text messages. Conclusions: The significant correlation between the perceived susceptibility to fine dust exposure and willingness to participate in education on fine dust-related respiratory disease prevention shows the importance of promoting education on prevention. The results of this study can help understand as reference for education on fine dust-related respiratory disease prevention.

Comparison of Particulate Matter and Ammonia Emission in Different Types of Laying Hen Poultry Houses during Spring (봄철 산란계사 사육형태별 미세먼지 및 암모니아 농도 비교)

  • This study was conducted to determine the concentrations of particulate matter (PM) and NH3 emissions from different types of laying hens poultry houses during spring. The concentrations of PM and NH3 were measured three times (2-week intervals; March to May) in Floor-pen-, Aviary-, and Cage-type poultry houses. Overall, PM10 and PM2.5 concentrations were found to be low from 22:00 to 04:00. The PM10 and PM2.5 concentrations in Floor-pen and Cage houses were similar with no significant daily deviation. NH3 concentrations measured over 24 h at the center and end of Floor-pen house were relatively constant. Irrespective of measurement location, NH3 concentrations were the lowest in Floor-pen house. Moreover, NH3 concentrations were higher at the end of Floor-pen and Aviary houses than that at the center; however, lower concentrations of NH3 were detected at the end of Cage house. The concentrations of PM10 and PM2.5 around the poultry houses were 57.5 and 34.0 ㎍/m3, respectively, with the daily average PM10 and PM2.5 concentrations (4,730 and 447.7 ㎍/m3, respective) being the highest in Aviary house. The concentrations of NH3 at the center and end of Cage house were the highest at 12.0 and 9.31 ppm, respectively. Furthermore, in Cage house, the emission factor of NH3 was the lowest, whereas there was no significant difference on that of NH3. In conclusion, among the three types of poultry houses assessed, PM (PM10, PM2.5) concentrations were higher in Aviary house, whereas NH3 concentrations were higher in Cage house.

The Memorial Park Planning of 5·18 Historic Sites - For Gwangju Hospital of Korea Army and 505 Security Forces - (5·18 사적지 기념공원화 계획 - 국군광주병원과 505보안부대 옛터를 대상으로 -)

  • This study presents a plan for a memorial park that respects the characteristics based on the historical facts for the concept of space of the Gwangju Hospital of Korea Army and the location of the 505 Security Forces, which were designated as historic sites after the 5-18 Democratization Movement. The Gwangju Metropolitan City as it is the location of the 5-18 historic sites, is taking part in the 5-18 Memorial Project, and plans to establish a city park recognizing the historic site of the 5-18 Democratization Movement, which has been preserved only as a memory space to this point. The park is promoting a phased development plan. This study suggests that the 5-18 historic sites can be modernized and that social consensus can establish the framework of the step-by-step planning and composition process to ensure the plans for the space heals wounds while preserving the history. In this paper, we propose a solution to a problem. We solve the approach for space utilization through an analysis of precedent research and planning cases related to park planning at historical sites. In addition to exploring the value of the site, we also describe the space utilization strategy that covers the historical characteristics and facts while maintaining the concept of park planning. As a result of the research, the historic site of the Gwangju Hospital of Korea Army is planned as a park of historical memory and healing in order to solve the problems left behind by the 5-18 Democratization Movement. The historic site of the 505 Security Forces was selected as an area for historical experiences and a place for learning that can be sympathized with by future generations of children and adolescents in terms of expanding and sustaining the memory of the 5-18 Democratization Movement. In the planning stage, the historical sites suggested the direction of space utilization for representation as did the social consensus of citizens, related groups, and specialists. Through this study, we will contribute to construction of a memorial park containing historical values in from 5-18 historic sites. It is meaningful to suggest a direction that can revitalize the life of the city as well as its citizen and can share with the history with future generations beyond being a place to heal wounds and keep alive the memory of the past.

Development of an Automatic Grafting Robot for Fruit Vegetables using Image Recognition (영상인식 기술 이용 과채류 접목로봇 개발)

  • This study was conducted to improve the performance of automatic grafting robot using image recognition technique. The stem diameters of tomatoes and cucumber at the time of grafting were $2.5{\pm}0.3mm$ and $2.2{\pm}0.2mm$ for scions and $3.1{\pm}0.7mm$ and $3.6{\pm}0.3mm$ for rootstocks, respectively. The grafting failure was occurred when the different height between scions and rootstocks were over 4 mm and below 2 mm due to the small contact area of both cutting surface. Therefore, it was found that the height difference at the cutting surface of 3 mm is appropriate. This study also found that grafting failure was occurred when the stem diameters of both scions and rootstocks were thin. Therefore, it was suggested to use at least one stem with thicker than the average stem diameter. Field survey on the cutting angle of stems by hand were ranged from 13 to 55 degree for scions and 15 to 67 degree for rootstocks, respectively, which indicates that this could cause the grafting failure problem. However, the automatic grafting robot developed in this study rotates the seedlings 90 degree and then the stems are cut using a cutting blade. The control part of robot use all images taken from grafting process to determine the distance between a center of both ends of stem and a gripper center and then control the rotation angle of a gripper. Overall, this study found that The performance of automatic grafting robot using image recognition technique was superior with the grafting success rates of cucumber and tomato as $96{\pm}3.2%$ and $95{\pm}4%$, respectively.

Evaluations of Growth and Forage Quality of Sesbania Accessions Adaptable to Korean Environments (국내적응 세스바니아의 생육특성 및 사료가치 평가)

  • Sesbania, an annual herb, is known for its high forage value and salt tolerance. It has merits as a forage crop that is adaptable to reclaimed land in the Republic of Korea. Therefore, we collected Sesbania genetic resources from the Republic of Korea and other countries, and conducted experiments to evaluate their potential as a forage crop in Korean climate and soil conditions. In the preliminary experiments, 15 genetic resources which were able to set seeds in Korean environment were selected out of a total of 46 collected genetic resources. Among 15 genetic resources, SL13 was the tallest and it was followed by that of SC04, SR01 and SE07. The accessions with the earliest flowering started flowering 101 days after sowing and set seed in early August. Fifteen accessions were evaluated for their salt tolerance at germination stage based on germination rate and growth of germinated seedlings at 0 mM, 150 mM and 300 mM NaCl concentrations. Five genetic resources like SC04, SL13, SS20, SS24 and SR01 were selected to be tolerant to NaCl treatment. Forage value was evaluated based on crude protein, acid detergent fiber, neutral detergent fiber and in vitro dry matter digestibility. The forage value of leaves was significantly higher than that of stems, and the forage value of the stem was slightly better than that of rice straw. The forage value of leaves of all the genetic resources was higher than grade 1 by the American Forage and Grassland Council grade. Among five selected genetic resources, the relative feed value of SC04 was the highest and it was followed by that of SS20, SL13, SS24 and SR01.

Wetland Habitat Assessement Utilizing TDI(Trophic Diatom Index) (부착돌말영양지수(TDI)를 활용한 습지환경 평가)

  • The purpose of this study was to analyze the habitat status and species diversity of benthic diatoms and estimate the applicability of TDI (Trophic Diatom Index) to obtain the basic data for the identification and management of created wetlands in the Nakdong River. We observed a total of 38 families and 173 species of benthic diatom during the survey period, and spring and autumn showed a similar number of species of 156 and 154, respectively. The result of the SOM (Self-Organizing Map) analysis showed that the distribution of benthic diatom was sensitive to environmental factors such as nutrient concentration and rainfall in each wetland. The cluster 1 was characterized by the survey sites of autumn mostly and consisted of points of high TDI, although the nutrients such as total phosphorus and total nitrogen were low, and the species number and abundance of diatoms were low. Conversely, cluster 4 was characterized by the survey sites of spring mostly and consisted of points of low TDI, even though total nitrogen was high. Considering that most of the created wetlands had the reduced inflow and outflow, the increased flow rate in the summer lowers nutrient values in autumn, and the species number and abundance of benthic diatom decreases due to the increase of turbidity, which reduces the light penetrations to the substrates. On the contrary, the TDI value is low in spring because the low water level causes insufficient substrate surface to the benthic diatoms, and it is too early for the establishment and development of saprophilous species. Although various studies have used TDI as an indicator for evaluating the habitat environment and water quality, it is not a good evaluation indicator in this study since the nutrient concentration in the wetlands mostly high as they have a low flow rate and are close to the stagnant area. Nevertheless, additional periodic surveys that comprehensively reflect the fact that the summer rainfall and inflow/outflow regulating function might affect the species diversity and distribution of benthic diatoms are necessary.
