Percoll 분리된 미니돼지 정액에서 LEY와 Triladyl을 이용한 동결융해후의 정자 성상 비교 (The Comparison of Triladyl and LEY for Cryosurvival Improvement of Sperm Separated by Percoll in Miniature Pig)

  • The objective of this study was to evaluated the efficiency of sperm cryosurvival using each extenders Triladyl and LEY containing egg yolk to the cryopreservation of separated sperm by percoll in miniature pig. The ejaculated semen from miniature pig was separated by 65% percoll and un-separated sperm as a control before freezing. The freezing of diluted semen added with Triladyl containing egg yolk and LEY solution (solution I: 11% Lactose or Triladyl + egg yolk; solution II: solution I + glycerol + OEP). Analysis of sperm ability was estimated by viability, capacitation acrosome reaction using chlortetracycline (CIC) the morphologic abnormality and hypoosmotic swelling test(HOST). The groups were designed that as separated sperm by Percoll with Triladyl(ST) or LEY(SL) for cryopreservation. And unseparated sperm with Triladyl(UT) or LEY(UL). As a results, the viability was higher significantly(p<0.05) in ST, SL, UT than UL extender. The morphologic abnormality was measured significantly (p<0.05) lower in ST than other extenders. The AR-patterned in CTC analysis was measured significantly(p<0.05) lower in SL and UL than other extenders. In conclusion, using Triladyl extender resulted in viability and morphology of separated sperm by percoll that were effective than using LEY extender, but it resulted in capacitation acrosome reaction was lower than using LEY extender.

Effect of Optixcell and Triladyl extenders on frozen-thawed sperm motilities and calving rates following artificial insemination in Hanwoo

  • In this study, we examined the effect of a liposome-based extender (Optixcell) and a tris-citric egg-yolk extender (Triladyl) on the frozen-thawed spermatozoa characteristics and the calving rate. The percentages for the total motility of the frozen-thawed spermatozoa were similar in the Optixcell and Triladyl groups. However, among the motile spermatozoa with a straight line velocity (VSL) ${\geq}25{\mu}m/sec$, the curvilinear velocity (VCL, ${\mu}m/sec$), VSL (${\mu}m/sec$), average path velocity (VAP, ${\mu}m/sec$), amplitude of lateral head displacement (ALH, ${\mu}m$), beat cross frequency (BCF, Hz), and plasma membrane integrity of the frozen-thawed spermatozoa for the Optixcell group were significantly higher than those for the Triladyl group. Furthermore, the calving rate in the Optixcell group (79.0%) was higher than that of the Triladyl group (62.8%). However, the acrosomal membrane integrity of the frozen-thawed spermatozoa in the Optixcell and Triladyl groups was not significantly different. These results indicate that semen freezing with Optixcell improved the motility and plasma membrane integrity of frozen-thawed spermatozoa and the calving rate of Hanwoo cows (native Korean cattle). In conclusion, our results suggest that semen freezing with the liposome-based extender Optixcell is more efficient than with the tris-citric egg-yolk extender Triladyl for improved offspring production.

희석액의 종류가 재래 흑염소 액상 정액의 생존율에 미치는 영향 (Comparison of Diluents on Liquid Storage of Korean Native Goat Spermatozoa)

  • 본 연구는 염소 액상 정액의 활용에 적합한 희석액을 검토하여, 국내 염소 인공수정 실용화를 위한 기초 자료를 확보하기 위하여 실시되었다. 두 마리의 흑염소 수컷으로부터 전기 자극법으로 정액을 채취하였다. 채취된 정액은 BTS로 희석하여 500g에서 5분간 원심분리하여 정장을 제거하고, BTS, Modena, Triladyl$^{(R)}$로 희석하여 $4^{\circ}C$ 냉장고나 $17^{\circ}C$ 온장고에서 8일간 보관되었다. BTS와 Modena로 희석된 정자의 운동성은 하루 만에 소멸되었으며, BTS로 희석된 정액을 $17^{\circ}C$에 보관하였을 때 2일째에 30$\sim$45%로 떨어졌으며, Modena에서는 3일째까지 인공수정에 가능한 활력을 유지하였다. 한편 Triladyl$^{(R)}$로 희석하여 $4^{\circ}C$$17^{\circ}C$에서 보관한 경우에는 좀더 좋은 생존성을 나타내었으며, 4일까지도 인공수정이 가능한 운동성을 보여주었다.

  • The objective of this study was to evaluated the efficiency on sperm cryosurvival and ability of in vitro fertilization using Triladyl and Lactose Egg-Yolk(LEY) as extenders for cryopreservation of separated sperm by 65% percoll in miniature pig. Sperm viability was measured with SYBR-14/PI double stained sperm by flow cytometry. Ability on embryo cleavage rate and blastocyst development were observed by in vitro fertilization after frozen-thawing of sperm separated by 65% percoll. The experimental groups were designed that separated sperm by 65% percoll with Triladyl (ST) or LEY(SL) and unseparated sperm with Triladyl(UT) or LEY(UL) for cryopreservation. As a results, the viability was significantly(p<0.05) higher in ST(55.1%), SL(63.1%), UL(58.8%) than UT(38.2%) group. Sperm viability in SL(63.1%) group was significantly(p<0.05) higher than other experimental groups. On the other hand, embryo cleavage rate was significantly(p<0.05) higher in ST(79.1%), SL(83.2) than UT(74.1) and UL(75.7%) groups at 96h after in vitro fertilization. Blastocyst development was also significantly(p<0.05) higher in ST(21.5%), SL(20.9%) than UT(17.0%) and UL(18.8%) groups. In conclusion, cryopreservation of miniature boar sperm separated by 65% percoll were beneficial to viability and capacity on in vitro fertilization.

제주흑우 정액 동결을 위한 AndreoMed와 Triladyl 희석제의 동결성 비교 (Comparison of AndroMed and Triladyl Extender for Freezing of Jeju Native Black Bull Semen)

  • 본 연구는 제주흑우의 정액의 동결과 보존기술 개발을 위해 AndroMed와 Trialdyl를 사용하여 보다 우수한 희석제와 우수 종모우 선정을 위해 실시되었다. 종모우 1번부터 4번까지 개체 정자의 성상비교에서 수정능 획득과 첨체반응의 비율은 36.8%, 26.8%, 37.8% 그리고 38%의 결과를 나타내었다. 2번 개체 정자에서 수정능 획득과 첨체반응의 비율이 유의적으로(p<0.05) 낮은 결과를 보였다. 제주흑우 정액을 동결하기 전 정자의 생존율은 약 $93.27{\pm}1.62$, 동결 융해 후 생존율은 $73.34{\pm}3.27%$로 나타나 동결전 정액의 생존율이 유의적으로(p<0.05) 높은 결과를 나타내었으나, 사멸 정자의 비율은 약 $7.35{\pm}2.63%$$13.71{\pm}2.85%$의 결과를 나타내었다. 그리고 $Triladyl^{(R)}$ 동해 방지제를 사용하였을 때, 정자의 운동성은 $72.86{\pm}2.83%$, $AndorMed^{(R)}$를 사용하였을 때 운동성은 $81.47{\pm}2.48%$$AndorMed^{(R)}$ 동해방지제를 사용하였을 때 운동성이 다소 높은 경향을 보였으나, 유의적인 차이는 나타나지 않았다. 마찬가지로 정자의 사멸율에서도 $Triladyl^{(R)}$ 동해 방지제는 $18.41{\pm}3.42%$, $AndorMed^{(R)}$ 동해방지제는 $17.26{\pm}4.25%$의 결과를 보여 AndroMed를 사용한 동해방지제가 정자의 생존율을 향상시키는 것으로 나타났다. 결론적으로 제주흑우 정액의 동결 보존을 위해 보다 향상된 동결보호제 선정과 동결보존 기술 개발이 필요할 것으로 사료된다.

Triladyl-난황 희석제가 한국 재래 흑염소의 정소상체 및 전기자극 유래 정자의 융해 후 생존성에 미치는 영향 (Effects of Triladyl-egg Yolk Diluents on the Viability of Frozen Korean Black-goat Spermatozoa from Cauda Epididymis and Electro-ejaculated Semen)

  • 가축유전자원으로서 흑염소 정액은 가축의 증식 및 유전적 개량을 위하여 동결정액으로서 보존될 필요성이 높으나 아직까지 연구 결과가 많이 누적되어 있지 않은 것으로 판단된다. 국내의 흑염소 동결유전자원의 생산성과 효율성 분석을 위하여 가축유전자원센터에서 보유 중인 흑염소 3 계통을 이용하여 전기자극방법에 의하여 채취된 정자와 정소상체유래 정자를 동결 보존하였다. 동결 전 Triladyl 난황희석액을 이용하여 흑염소의 정액과 혼합하여 $17^{\circ}C$에서 2시간 보존하면 응고현상을 관찰할 수 있었으며 42.9%의 비율로 난황이 변화한다는 것을 육안으로 관찰할 수 있었다. 정장 제거용 배양액으로 전기자극 유래 정자를 세정 후 동결 보존하면 융해 후 정자 생존율이 정액 세정용 배양액 $53.8{\pm}5.2%$로 나타났으며 정소상체 정자는 $74.6{\pm}10.6%$로 유의적으로 높게 관찰되었다. 융해된 정자의 장수성을 $17^{\circ}C$에서 조사해 보면 정소상체 유래 정자가 전기자극 유래 정자보다 우수한 것으로 조사되었다. 그러므로, 염소 동결 유전자원을 보존하는 방법으로 정소상체 정자는 활용성이 인정되며, 염소의 개량과 선발을 위하여 농가에서 활용 가능한 동결정액의 생산과 융해 후 생존성 증진을 위하여 더 많은 동결 보존 연구가 필요하다고 판단된다.

자기활성수가 소 정액 동결에 미치는 영향 (Effect of Magnetized Water on Cryopreservation in Bovine Spermatozoa)

  • The purpose of this study was to improve of frozen-thawed sperm using magnetized water in Korean native cattle. Before cryopreservation, without egg-yolk Triladyl$^{(R)}$ solution was flowed though magnet [0, 2000, 4000 and 6000 Gauss(G)] for S min. The freezing of dilluted semen added with Triladyl containing 20% egg-yolk. Analysis of frozen-thawed sperm was estimated viability with SYBR14/PI double stain, membrane intact with hypoosmotic swelling test (HOST), acrosome reaction with FITC-PNA, mitochondria membrane function with Rhodamin 123 by flow- cytometry. Sperm viability was significantly higher in 4000G group than other groups (p<0.05). However, the Hypoosmotic Swelling Test(HOST) was significantly higher in fresh, 4000 and 6000G than 0 and 2000G (p<0.05). In addition, mitochondria membrane damage and acrosome damage were significantly lower in 6000G group than other groups (p<0.05). in conclusion we suggest that magnetized water could be improve ability of sperm on cryopreservation in Korean native cattle.

소 정자의 동결 및 수정능력 향상을 위한 정액운반법의 개발 (Development of Semen Transport System for Cryopreservation and Fertility in Bull Sperm)

  • The objective of this study was to develop of semen transport system for cryopreservation and fertility in bull sperm. The ejaculated semen were diluted with Triladyl containing 20% egg-yolk for transportation. Diluted semen was transported by three methods that there were wrapping tissue (Tissue), sinking under $30^{\circ}C$ water (Water) and sinking between warm water and air (Air) methods. Semen was transported within 2 hours in $0.3^{\circ}C$. For this study, the freezing of diluted semen were added with Triladyl containing 20% egg-yolk. And frozen-thawed sperm were estimated with SYBR14/PI double stain for viability, FITC-PNA/PI double stain for acrosome reaction analysis and Rhodamine123 double stain for mitochondrial intact assessment. In results, live sperm (SYBR+/PI-) in Air treatment group ($43.3{\pm}4.7%$) was significantly (p<0.05) higher than other treatment groups (Tissue: $16.3{\pm}2.7%$ and Water: $27.5{\pm}3.1%$), dying sperm (SYBR+/PI+) in Air treatment group ($55.6{\pm}4.7%$) was significantly lower than other treatment groups (Tissue: $77.6{\pm}3.2%$ and Water: $67.6{\pm}3.3%$) (p<0.05). Acrosome reaction in Air treatment group ($0.2{\pm}0.1%$) within live sperm (PI negative region) was significantly (p<0.05) lower than other treatment groups (Tissue: $0.7{\pm}0.2%$ and Water: $0.5{\pm}0.1%$), the acrosome reaction in Air treatment group ($28.6{\pm}2.8%$) within all sperm also was significantly lower than other treatment groups (Tissue: $44.2{\pm}1.8%$ and Water: $36.2{\pm}2.0%$) (p<0.05). And mitochondrial intact in Air treatment group within live ($97.1{\pm}0.4%$) and all ($61.9{\pm}3.3%$) sperm were significantly higher than other treatment groups (Tissue: $85.2{\pm}3.3%$, Water: $87.8{\pm}2.9%$ within live sperm and Tissue: $49.28{\pm}3.7%$, Water: $42.0{\pm}3.1%$ within all sperm) (p<0.05). Therefore, we suggest that transportation by sinking method between warm water and air was beneficial to improvement of fertility in frozen-thawed in bull semen.

칡한우 정액 동결에 있어서 희석액과 수소의 연령이 정자의 동결성, 체외수정란 발달 및 인공수정 임신율에 미치는 효과 (Effect of Freezing Buffers and Age of Bulls on Freezability of Semen, $In$ $Vitro$ Embryo Development and the Pregnancy Rate after Artificial Insemination of Korean Native Stripped Bull)

  • The purpose of this study was to examine the effect of growing stages of the Korean Native Striped Bull (KNSB) on the freezability and fertility of frozen-thawed semen. First, we investigated the total motility (TM) and progressive motility (PM) according to the diluent used for semen freezing. Second, we examined the effect of the age of KNSB on semen volume, TM and PM of fresh and frozen-thawed semen. Third, we examined the effect of frozen semen from the different age of KNSB on the $in-vitro$ fertilization rate, and the artificial insemination pregnancy rate. The diluents used in this experiment were Triladyl$^{(R)}$ and Tris-egg yolk extender (EYE). Semen was collected from 5 KNSB in the growing stage (15 months) and 5 adult KNSB (36 months). When Triladyl or Tris-EYE extender was used for semen freezing, there was no difference of the mean TM and the mean PM. However, the mean TM was significantly higher in Bull No. 1885 than Bull No. 4283 ($p$ <0.05). The mean volume of semen collected from the 15-month-old bulls (2.3 ml) was significantly lower ($p$ <0.05) than that from the 36-month-old bulls (5.0 ml). The mean semen concentration was similar for the 15-month-old ($2.1{\times}10^9$ spermatozoa/ml) and 36-month-old ($1.8{\times}10^9$ spermatozoa/ml) bulls. For the 15-month-old and 36-month-old bulls, the mean TM of fresh semen were 93.7% and 88.3%, respectively, and the mean PM were 97.0% and 88.3%, respectively; the 15-month-old bulls showed a particularly high PM ($p$ <0.05). For the 15-month-old and 36-month-old bulls, the mean TM (56.0% and 58.0%, respectively) and the mean PM (64.0% and 70.7%, respectively) of frozen-thawed semen did not differ. The development rates of embryos after $in-vitro$ fertilization and the pregnancy rate after artificial insemination using frozen-thawed semen did not differ according to the bull's age. In summary, semen volume differed according to the bull's age, but semen concentration and survival rate, the $in-vitro$ fertilization rate, and the pregnancy rate did not differ according to the stripe bull's age. Accordingly, semen from bulls in the growing stage can be collected and frozen for the preservation and multiplication of rare livestock.

소 정액의 동결 보존시 Methyl-Beta-Cyclodextrin (MBCD)의 영향 (Effect of Methyl-Beta-Cyclodextrin (MBCD) on Cryopreservation of Bull Spermatozoa)

  • The objective of this study was to evaluate efficiency of methyl-beta-cyclodextrin (MBCD) in the sperm preservation of bull. For this study, the freezing of diluted semen were added with Triladyl containing 20% egg-yolk and/or 0, 1, 5, 10 and 20 mM MBCD before freezing process. Analysis of viability in frozen-thawed sperm was estimated by SYBR14/PI double stain, hypoosmotic swelling test(HOST) and acrosome damage with FITC-PNA, and mitochondria activation with Rhodamin123 by flow-cytometry. The sperm viability was significantly in 0 mM and 5 mM concentrations of MBCD than other groups (p<0.05). However, the HOST was significantly lower at 20 mM concentration of MBCD than other concentrations (p<0.05). In addition, acrosome damage and mitochondria activation rates were significantly lower at 20 mM concentration of MBCD than other groups (p<0.05). In conclusion, the viability of sperm was not significantly different among concentrations of MBCD 0, 5 and 10 mM, but MBCD 20 mM was significantly lower than other groups. In addition, as concentrations of MBCD was high, HOST, acrosome damage and mitochondria activation rates had a negative effect in bull sperm.
