The sinus node (SN) is located at the apex of the cardiac conduction system, and SN dysfunction (SND)-characterized by electrical remodeling-is generally attributed to idiopathic fibrosis or ischemic injuries in the SN. SND is associated with increased risk of cardiovascular disorders, including syncope, heart failure, and atrial arrhythmias, particularly atrial fibrillation. One of the histological SND hallmarks is degenerative atrial remodeling that is associated with conduction abnormalities and increased right atrial refractoriness. Although SND is frequently accompanied by increased fibrosis in the right atrium (RA), its molecular basis still remains elusive. Therefore, we investigated whether SND can induce significant molecular changes that account for the structural remodeling of RA. Towards this, we employed a rabbit model of experimental SND, and then compared the genome-wide RNA expression profiles in RA between SND-induced rabbits and sham-operated controls to identify the differentially expressed transcripts. The accompanying gene enrichment analysis revealed extensive pro-fibrotic changes within 7 days after the SN ablation, including activation of transforming growth factor-β (TGF-β) signaling and alterations in the levels of extracellular matrix components and their regulators. Importantly, our findings suggest that periostin, a matricellular factor that regulates the development of cardiac tissue, might play a key role in mediating TGF-β-signaling-induced aberrant atrial remodeling. In conclusion, the present study provides valuable information regarding the molecular signatures underlying SND-induced atrial remodeling, and indicates that periostin can be potentially used in the diagnosis of fibroproliferative cardiac dysfunctions.
글루코코르티코이드는 해마 조직에서 대사, 스냅신 형성, apoptosis, 신경세포 생성과 세포에 있어서 수지상의 형태에 영향을 준다. 글루코코르티코이드 호르몬에 의한 해마조직의 생리학적 조절을 이해하기 위하여, CA3와 DG (dentate gyrus)에서 유전자 발현에 대하여 조사하였다. Lewis 쥐에 9.5mg의 코르티코스테론 알약 또는 플라시보 알약을 20일 동안 처리한 후에 올리고머 유전자 칩을 이용하여 유전자 발현을 조사 하였다 (Rat Neurobiology U34 Arrays, Affymetrix). 플라시보 알약을 처리한 쥐에서 32 유전자들이 DG보다 CA3에서 발현이 높았으며, 3개 유전자는 CA3보다 DG에서 높은 발현을 보였다. 코르티코스테론 호르몬 처리에 의한 해마조직의 유전자 발현 형태는 해부학적 구조의 차이를 보였다. 특히, CA3에서 6개의 유전자와 DG에서 41 개의 유전자가 호르몬에 의하여 조절 받았으며, 이중 43개의 유전자가 상승 발현하였으며, 4개의 유전자가 하강 발현 하였다. 이들 유전자를 기능에 의해 분류하면, 13개의 신경전달물질관련 유전자, 5개의 이온채널,4개의 전사인자, 3개의 neurotrophic인자, 1개의 각 사이토카인과 apoptosis관련 유전자, 그리고 5개의 스냅신형성관련 유전자가 해마조직에서 발현의 변화를 보였다. 특히, 스트레스 호르몬에 의하여 CA3에서 BDNF의 감소를 볼 수 있었다. 이러한 결과는 호르몬에 의하여 해마구조의 생리학적인 다양성을 내포 하고 있다.
대장균 야생주와 프로판올 내성 변이주에서 프로판올 스트레스에 의해 발현이 크게 변화하는 유전자를 DNA microarray 기술을 이용하여 비교 분석하였다. 프로판올 첨가한 야생주와 무첨가한 야생주 사이의 RNA 발현 연관값은 0.949이며, 50개의 유전자 발현이 2배 이상 변화하였다. 프로판올을 첨가한 내성변이주와 무첨가한 변이주 사이의 연관값은 0.952이며, 71개의 유전자 발현이 크게 변화하였다. 그러나, 야생주와 변이주 사이의 연관값은 프로판올 무첨가한 조건과 첨가한 조건에서 각각 0.992 및 0.974로 매우 높았으며, 2배 이상의 발현차이를 나타내는 유전자는 각각 1개 및 2개로, 두 균주는 매우 유사한 발현양상을 보였다. 야생주 또는 변이주에서 프로판올 스트레스에 반응하는 대표적인 유전자들의 특징은 많은 열충격 반응에 관여하는 유전자들의 발현이 크게 증가하였으며, 리보소옴 합성에 필요한 많은 유전자들의 발현이 감소하였다. 또한, 전사조절인자들인 ArcA, CRP, FNR, H-NS, GatR, PurR에 의해 조절받는 유전자들의 발현이 크게 변화하였으며, 시그마인자들 중에서는 RpoH에 의해서 발현되는 유전자들의 발현이 크게 증가하였다. rpoH가 정상적으로 발현되지 못하는 변이주와 야생주를 이용한 프로판올 내성정도를 측정한 결과, RpoH는 대장균에서 프로판올 스트레스에 적응하는데 중요한 기능을 하는 것으로 확인되었다.
배스(Micropterus salmoides)는 수생태계에서 최상위단계에 위치하는 생태계교란 어종으로 심각한 담수생태계의 불균형을 초래하고 있다. 배스의 퇴치 및 관리를 위한 다양한 시도를 하고 있지만 효과적인 방안은 없는 상황이므로 배스의 고유한 특성에 기반한 개체군 감소의 효율성을 극대화할 수 있는 방식을 모색하였다. 본 연구에서는 배스의 Transcriptom 분석으로 Unigene contigs는 182,887개, 그리고 정자-난자 인식 단백질인 IZUMO1과 Zona pellucida sperm-binding protein의 유전자에서 CRISPR/Cas9 system을 적용할 최종 Target sequence는 12종을 산출하였다. 각 Target sequence를 인식할 수 있는 12종의 sgRNA를 합성한 후 후속 연구에 사용할 12종의 Cas9-sgRNA ribonucleoprotein (RNP) complex를 제작하였다. 본 연구에서는 차세대염기서열 분석법으로 정자-난자 인식 단백질을 암호화하는 유전자를 탐색하였고, CRISPR/Cas9 system으로 유전자를 편집하여 번식행동은 하지만 수정란을 형성하지 못하는 생식세포를 생산하는 불임개체를 유도하기 위한 조성물 개발 과정을 확립하였다. 그리고 배스와 동일한 수계에 있는 고유 생물종의 서식에는 영향을 미치지 않는 생태교란종 관리 방안으로서의 유용성을 검증하기 위한 후속 연구의 귀중한 기초 자료를 확보하는데 기여했다고 판단된다.
Autosomal dominant polycystic kidney disease (ADPKD), one of the most common human monogenic diseases (frequency of 1/1000-1/400), is characterized by numerous fluid-filled renal cysts (RCs). Inactivation of the PKD1 or PKD2 gene by germline and somatic mutations is necessary for cyst formation in ADPKD. To mechanistically understand cyst formation and growth, we isolated RCs from Korean patients with ADPKD and immortalized them with human telomerase reverse transcriptase (hTERT). Three hTERT-immortalized RC cell lines were characterized as proximal epithelial cells with germline and somatic PKD1 mutations. Thus, we first established hTERT-immortalized proximal cyst cells with somatic PKD1 mutations. Through transcriptome sequencing and Gene Ontology (GO) analysis, we found that upregulated genes were related to cell division and that downregulated genes were related to cell differentiation. We wondered whether the upregulated gene for the chemokine CXCL12 is related to the mTOR signaling pathway in cyst growth in ADPKD. CXCL12 mRNA expression and secretion were increased in RC cell lines. We then examined CXCL12 levels in RC fluids from patients with ADPKD and found increased CXCL12 levels. The CXCL12 receptor CXC chemokine receptor 4 (CXCR4) was upregulated, and the mTOR signaling pathway, which is downstream of the CXCL12/CXCR4 axis, was activated in ADPKD kidney tissue. To confirm activation of the mTOR signaling pathway by CXCL12 via CXCR4, we treated the RC cell lines with recombinant CXCL12 and the CXCR4 antagonist AMD3100; CXCL12 induced the mTOR signaling pathway, but the CXCR4 antagonist AMD3100 blocked the mTOR signaling pathway. Taken together, these results suggest that enhanced CXCL12 in RC fluids activates the mTOR signaling pathway via CXCR4 in ADPKD cyst growth.
한우의 성장단계별 부위 근육발달을 이해하는 것은 도체율 개선에 따른 소득증대와 증체율 증가에 따른 생산효율 향상에 긍정적인 영향을 미친다. 본 연구에서는 한우의 등심과 사태 유래 근육위성세포를 분리 후 세포단위의 발달 및 분화를 비교하여 transcriptome 단위의 작용기전을 제시하였다. 한우의 부위별 근육 유래 근육위성세포의 근섬유의 양은 4일에 가장 높게 나타났고 이후 감소하였다. 한우의 근육위성세포의 발달 단계에 따라 발현되는 총 전사체 유전자의 종류는 사태근육 위성세포에서 높게 나타났다. 등심과 사태 근육 유래 위성세포의 발달단계에 따라 유의적인 차등 유전자 453개를 찾아냈고 이를 이용한 기능유 전체 분석이 필요하다. 등심과 사태유래 근육위성세포를 이용한 동일조건 분화 비교에서 사태유래 근육 위성세포의 분화 시 myosin complex, skeletal muscle contraction, troponin complex, skeletal muscle tissue development 와 같은 근섬유 형성관련 유전자의 발현이 높게 나타나는 것으로 보아 같은 개체의 근육조직에서도 부위별로 차등 발달이 되고 있다는 것을 알 수 있다. 기존 연구에서는 근육의 성장에 대한 이해를 위해 사양과 영양관련 시험이 많이 이루어졌다. 향후 세포단위의 연구들이 많이 이루어져 작용기작에 대한 생물정보 자료를 추가로 적용한다면 한우의 정밀사양을 적용할수 있는 바탕이 마련될 것이다. 또한 근육위성세포의 연구는 추후 동물실험 윤리제도 강화에 따른 비동물 전임상 screening 시험 활용과 대체단백질 산업의 주요 이슈인 배양육 소재 개발 연구와 같이 축산시험연구의 지속적인 확장성에 많은 영향을 미칠 것으로 생각된다.
수산업은 수산물의 남획, 국가 간의 제한, 기후변화로 인하여 수산물 개체 수 감소 등으로 전 세계 수산물 생산량이 정체되면서, 양식업은 항상 세계 식량 소비를 위해 오랫동안 지속되어온 패러다임을 뒤집고 인류가 바다, 호수, 강의 풍부함을 새로운 방식으로 확장할 수 있도록 하는 '푸른 혁명'의 가능성을 제시하고 있다. 양식산업은 이제 인간 소비를 위한 해산물의 원료로서 해양에서 얻는 어업생산량의 절반이상을 넘어섰다. 지속 가능한 양식산업의 발전을 위하여 다양한 최신 생물학적 연구 방법들이 적용되고 있다. 유전체학은 2011년 대서양 대구의 염기서열이 규명된 이래 최근 상당한 진전을 이루었으며 더 많은 종의 유전체가 해독되어 우수 형질의 육종 및 번식 기술, 질병 저항성 품질 개량, 양식 사료 및 사료방식의 최적화 등 양식산업을 위한 보다 견고하고 생산적인 지식 기반을 제공한다. 본 리뷰는 수산양식종의 유전체 연구를 위한 유전체 분석 기술과 수산양식종의 유전체 연구의 현황에 대해서 살펴보았다. 이와 같이 유전체 분석 기술과 유전체학의 발전은 수산양식산업의 과제를 해결하는 데 중요하며 지속 가능한 어업과 양식에 도움을 줄 것이다.

Objective: Vanin-1 (VNN1) is a pantetheinase that catalyses the hydrolysis of pantetheine to produce pantothenic acid and cysteamine. Our previous studies have shown that the VNN1 is specifically expressed in chicken liver which negatively regulated by microRNA-122. However, the functions of the VNN1 in lipid metabolism in chicken liver haven't been elucidated. Methods: First, we detected the VNN1 mRNA expression in 4-week chickens which were fasted 24 hours. Next, knocked out VNN1 via CRISPR/Cas9 system in the chicken Leghorn Male Hepatoma cell line. Detected the lipid deposition via oil red staining and analysis the content of triglycerides (TG), low-density lipoprotein-C (LDL-C), and high-density lipoprotein-C (HDL-C) after VNN1 knockout in Leghorn Male Hepatoma cell line. Then we captured various differentially expressed genes (DEGs) between VNN1-modified LMH cells and original LMH cells by RNA-seq. Results: Firstly, fasting-induced expression of VNN1. Meanwhile, we successfully used the CRISPR/Cas9 system to achieve targeted mutations of the VNN1 in the chicken LMH cell line. Moreover, the expression level of VNN1 mRNA in LMH-KO-VNN1 cells decreased compared with that in the wild-type LMH cells (p<0.0001). Compared with control, lipid deposition was decreased after knockout VNN1 via oil red staining, meanwhile, the contents of TG and LDL-C were significantly reduced, and the content of HDL-C was increased in LMH-KO-VNN1 cells. Transcriptome sequencing showed that there were 1,335 DEGs between LMH-KO-VNN1 cells and original LMH cells. Of these DEGs, 431 were upregulated, and 904 were downregulated. Gene ontology analyses of all DEGs showed that the lipid metabolism-related pathways, such as fatty acid biosynthesis and long-chain fatty acid biosynthesis, were enriched. KEGG pathway analyses showed that "lipid metabolism pathway", "energy metabolism", and "carbohydrate metabolism" were enriched. A total of 76 DEGs were involved in these pathways, of which 29 genes were upregulated (such as cytochrome P450 family 7 subfamily A member 1, ELOVL fatty acid elongase 2, and apolipoprotein A4) and 47 genes were downregulated (such as phosphoenolpyruvate carboxykinase 1) by VNN1 knockout in the LMH cells. Conclusion: These results suggest that VNN1 plays an important role in coordinating lipid metabolism in the chicken liver.
Objective: The objective of this study was to identify candidate genes that play important roles in skeletal muscle development in ducks. Methods: In this study, we investigated the transcriptional sequencing of embryonic pectoral muscles from two specialized lines: Liancheng white ducks (female) and Cherry valley ducks (male) hybrid Line A (LCA) and Line C (LCC) ducks. In addition, prediction of target genes for the differentially expressed mRNAs was conducted and the enriched gene ontology (GO) terms and Kyoto encyclopedia of genes and genomes signaling pathways were further analyzed. Finally, a protein-to-protein interaction network was analyzed by using the target genes to gain insights into their potential functional association. Results: A total of 1,428 differentially expressed genes (DEGs) with 762 being up-regulated genes and 666 being down-regulated genes in pectoral muscle of LCA and LCC ducks identified by RNA-seq (p<0.05). Meanwhile, 23 GO terms in the down-regulated genes and 75 GO terms in up-regulated genes were significantly enriched (p<0.05). Furthermore, the top 5 most enriched pathways were ECM-receptor interaction, fatty acid degradation, pyruvate degradation, PPAR signaling pathway, and glycolysis/gluconeogenesis. Finally, the candidate genes including integrin b3 (Itgb3), pyruvate kinase M1/2 (Pkm), insulin-like growth factor 1 (Igf1), glucose-6-phosphate isomerase (Gpi), GABA type A receptor-associated protein-like 1 (Gabarapl1), and thyroid hormone receptor beta (Thrb) showed the most expression difference, and then were selected to verification by quantitative real-time polymerase chain reaction (qRT-PCR). The result of qRT-PCR was consistent with that of transcriptome sequencing. Conclusion: This study provided information of molecular mechanisms underlying the developmental differences in skeletal muscles between specialized duck lines.
본 연구의 목적은 유묘기 TO1000DH3와 Early big에서 MeJA 처리에 의해 글루코시놀레이트 함량 변화 및 유전자의 발현 변화를 분석하기 위하여 수행되었다. $200{\mu}M$ 농도의 MeJA를 처리하여 글루코시놀레이트 함량을 분석한 결과, 글루코시놀레이트 총 함량이 처리 전보다 TO1000DH3에서 1.3~1.5배, Early big에서 1.3 ~ 3.8배 증가하였다. 알리패틱 글루코시놀레이트인 progoitrin과 gluconapin은 TO1000DH3에서만 검출되었으며, neoglucobrassicin 성분의 함량 변화가 MeJA 처리 48시간 후 TO1000DH3와 Early big에서 가장 크게 증가되었다. 전사체 분석을 통해 TO1000DH3에서는 stress나 defense 반응에 관여하거나, 생장과 관련된 전사체가 특이적으로 발현하고, Early big에서는 nucleoside 또는 ATP 생합성 관련 전사체가 특이적으로 발현하는 것을 알 수 있었다. MeJA를 처리함에 따라 발현이 2배 이상 변한 전사체를 TO1000DH3에서 12,020개, Early big에서 13,510개를 선발하여 GO 분석한 결과 stimulus, chemical에 반응하는 전사체의 발현이 공통적으로 증가하였고, single-organism 및 ribosome 합성 관련 전사체의 발현이 공통적으로 감소하였다. 특히 glucobrassicin, neoglucobrassicin 함량과 연관되어 발현이 증가한 인돌릭 글루코시놀레이트 생합성 관련 전사체의 발현이 모두 증가하였다 (MYB34 (Bo7g098110), IGMT2 (Bo8g070650), CYP81D1 (Bo6g056440), CYP81D4 (Bo7g118500), CYP81F4 (Bo1g004730, Bo01007s020), CYP81G1 (Bo4g154660), CYP83B1 (Bo8g024390) 및 CYP91A2 (Bo1g003710)). 글루코시놀레이트 생합성 경로 관련 유전자를 대표하는 전사체 104개를 선발하여 발현 양상을 분석한 결과 transcription factor에 속하는 MYB28, MYB51의 발현은 MeJA 처리 전에 비해 처리 후 발현양이 감소하였지만, 대부분의 전사체의 발현은 MeJA 처리에 의해 증가하였다. MeJA 처리에 의해 AOP3 (Bo9g006220, Bo9g006240), TGG1 (Bo14804s010)는 TO1000DH3에서만 특이적으로 발현이 증가하였고, Dof1.1 (Bo5g008360), UGT74C1 (Bo4g177540), GSL-OH (Bo4g173560, Bo4g173550, Bo4g173530)는 Early big 특이적으로 발현이 증가하였다. MeJA 처리 전 두 계통에서 발현이 가장 높은 글루코시놀레이트 생합성 관련 유전자는 GSTU20이었고, MeJA 처리에 의해 12시간 후TO1000DH3에서 CYP79B2 (Bo7g118840), Early big에서는 CYP79B3 (Bo4g149550)의 발현이 가장 많이 증가하였다.

① 서비스 이용은 당 사이트의 업무상 또는 기술상 특별한 지장이 없는 한 연중무휴, 1일 24시간 운영을

② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
