신결석 치료에 사용되는 단방용법에 대한 결석성분의 용해 실험 (The Study on the Solubility of the Ingredients of the Kidney Stone In the Traditional Oriental Medicines)

  • This study was carried out to investigate the solubility of the ingredients of the kidney stone in the solution of the traditional oriental medicines. Calcium hydroxide, apatite and uric acid were chosen as the ingredients of the kidney stone. Plantaginis Semen, Lysimachiae Herba, Saururi Herba seu Rhizoma, Imperatae Rhizoma, Allium tuberosum Rottler were studied as the oriental medicines for the kidney stone. Calcium hydroxide had showed the very good solubility in the solution of Imperatae Rhizoma, the apatite had showed the good solubility in the solutions of Saururi Herba seu Rhizoma and Allium tuberosum Rottler. Uric acid had showed the mild solubility in the solution of Lysimachiae Herba and Saururi Herba seu Rhizoma.

Samsoeum inhibits systemic anaphylaxis and release of histamine, cytokine in vivo and in vitro

  • Samsoeum (SSE) is used in traditional oriental medicine for various medicinal purposes. However, the exact mechanism that accounts for the anti-allergy and anti-inflammatory effects of the SSE is still not fully understood. The aim of the present study is to elucidate whether and how SSE modulates the allergic reactions in vivo, and inflammatory reaction in vitro. In this study, we showed that SSE significantly decreased compound 48/80-induced systemic anaphylaxis, ear-swelling response, histamine release from preparation of rat peritoneal mast cells and anti-dinitropheny IgE-induced passive cutaneous reaction. Also, SSE inhibited the expression of inflammatory cytokine and cyclooxygenase-2 in PMA plus A23187-stimulated human mast cells (HMC-1). In addition, we showed that anti-inflammatory mechanism of SSE is through suppression of nuclear factor-${\kappa}B$ activation and $I{\kappa}B-{\alpha}$ phosphorylation/degradation in HMC-1. These results provided new insight into the pharmacological actions of SSE as a potential molecule for therapy of inflammatory allergic diseases.

한방의료기관 한약제제 구비 현황 조사 연구 (The Analysis of Herbal Medicine Preparations Equipped in Traditional Korean Medical Clinics)

  • Objective : There is a growing need to expand or adjust insurance coverage of herbal prescriptions in National Health Insurance. The purpose of this study was to provide basic data for expansions or adjustments of 56 herbal prescriptions in the National Health Insurance. Method : We surveyed lists of 56 insurance-covered herbal prescriptions, non-covered company-produced herbal medicines and self-prepared herbal medicines in 531 traditional korean medical clinics by mail from 6/1/2008 to 12/9/2008. Result : Among the 56 Prescriptions, Ojeok-san was stocked in 66.7% of clinics (1st), Samso-eum ranked 2nd, Socheongnyong-tang ranked 3rd, Gumiganghwal-tang was 4th, Hyangsapyeongwi-san ranked 5th. Among the non-covered company-produced herbal medicines, Danggwisu-san ranked 1st, Gwakyangjeonggi-san ranked 2nd, Oryeong-san was 3rd, Bangpungtongseong-san was 4th, Maengmundong-tang ranked 5th. Among the self-prepared herbal medicines, Gongjin-dan ranked 1st, Soche-hwan was 2nd, Gyejibongnyeong-hwan was 3rd, Yeongsin-hwan was 4th, Palmi-hwan ranked 5th. Conclusions : It is reasonable to select frequently used company-produced or self-prepared herbal prescriptions in this stock rate survey for improvement of National Health Insurance coverage.

중국 약용식물 추출물의 알도즈 환원 효소 억제 효능 검색 (V) (Screening of Chinese Herbal Medicines with Inhibitory Effect on Aldose Reductase (V))

  • Aldose reductase (AR), the key enzyme in the polyol pathway, plays an important role in the development of the diabetic complications. None of Aldose reductase Inhibitor (ARI) has achieved worldwide use because of limited efficacy or undesirable side effects. Therefore, evaluating natural sources for ARI potential may lead to the development of safer and more effective agents against diabetic complications. Fifty two Chinese herbal medicines have been investigated for inhibitory activities on AR. Among them, twelve herbal medicines, Artemisia anomala (aerial part), Centella asiatica (aerial part), Scutellaria baicalensis (root), Senecio chrysanthermoides (whole plant), Gleditsia japonica (twig), Zizyphus jujube (twig, leaf), Citrus aurantium (fruit), Hydnocarpus anthelmintica (fruit), Potaninia mongolica (aerial part), Tribulus terrestris (fruit), Artemisia apiacea (aerial part) and Eclipta prostrate (aerial part) exhibited a significant inhibitory activity against AR. Particularly, Artemisia anomala, Centella asiatica and Scutellaria baicalensis showed four times more potent inhibitory activity than the positive control, 3,3-tetramethyleneglutaric acid (TMG).

독성학적 측면에서의 한약에 의한 간독성 유발과 기전 (Inducible Mechanisms for Hepatotoxicity caused by Traditional Korean Medicines in a View of Toxicology)

  • Objectives: There has been a lot of controversy about whether Korean traditional medicines cause drug-induced hepatotoxicity or not. However, this controversy has not been examined from a scientific approach, especially in terms of toxicology. Thus, this article for traditional Korean medicine-induced hepatotoxicity was reviewed on a basis of toxicological assessment. Methods: Toxicological assessments were carried out in a view of reactive intermediates formation from biotransformation in animals based on cytochrome P450 activity inducing oxidative stress, and case reports in world-wide. Results and Conclusions: Several herbal materials used for traditional Korean medicines were identified as possible agents causing drug-induced hepatotoxicity. Most of these agents seem to produce toxic reactive intermediates caused by cytochrome P450. Oxidative stress followed by cytochrome P450 activation could also be a reason for herb-induced hepatotoxicity. Plausible prevention of herb-induced hepatotoxicity was suggested.

한방혼합액 APA-01의 면역 증강 효과 (Enhancement of Immune Response by New Herb Mixture, APA-01, in Mice)

  • APA-01, which is an aqueous extract of five Chinese herbs, is a modified formula of Huoxiang-Zhengqi-San. The effect of new herb extract on immune response was investigated. The parameter examined to assess apparent immune response of APA-01 in mice included changes of body weight, relative weight of immune organs, cell proliferation and cytokine gene expression. The body weight and relative weight of immune organs were not significantly changed among the tested groups. In the spleen cell prolijeration assay, APA-01 increased the cell proliferation in a dose-dependent manner. Methotrexate (MTX), an agent of immune suppression, inhibited the spleen cell proliferation (IC/sub 50/: 800㎍/㎖). However, APA-01 significantly inhibited the suppression of mouse spleen cell proliferation. Therefore, it seems that APA-01 has a reducing effect of immune suppression. Immunomodulatory effect of APA-01 was further investigated using reverse transcription polymerase chain reaction (RT-PCR) in mouse spleen cells. In RT-PCR test, APA-01 enhanced the expression of cyclooxygenase-2 (COX-2) mRNA in a dose-dependent manner. In spite of immune suppression by MTX, COX-2 mRNA was induced by co-treatment with APA-01. These results suggest that APA-01 stimulates the proliferation of spleen cells, regulates the expression of COX-2 mRNA, and accelerates the recovery of inhibition of spleen cell proliferation induced by MTX, thus providing the immunological basis for clinical benefit of APA-01.

한국약용식물의 최종당화산물 생성저해활성 검색 (III) (Screening of Korean Herbal Medicines with Inhibitory Activity on Advanced Glycation End Products (AGEs) Formation (III))

  • Enhanced formation and accumulation of advanced glycation end products (AGEs) have been implicated as a major pathogenesis process leading to diabetic complications, normal aging, atherosclerosis and Alzheimer's disease. In our ongoing project to discover novel treatments for diabetic complications from natural sources, we have investigated on the inhibitory activity of 67 ethanol extracts from 57 Korean herbal medicines against the formation of AGEs in vitro. Of these, 22 extracts were found to have a significant AGEs inhibitory activity ($IC_{50}$<50 ${\mu}g$/ml) compared with aminoguanidine ($IC_{50}$=75.98 ${\mu}g$/ml). Particularly, 6 extracts from 3 herbal medicines, Castanea crenata (flower, leaf, bark-twig), Acer tatarium subsp. ginnala (fruit) and Sapium japonicum (leaf, twig) showed (approximately 8-17 fold) stronger inhibitory activity than that of aminoguanidine.

전통 음료의 한의학적(韓醫學的) 고찰(考察) (The Oriental Medicine the Traditional Drink Study on the Herbal)

  • The review of the literature had been done to investigate the mechanism of the herbal medicines contained in traditional Korean medical books. We had thought the results could provide the theoretical basis of adding herbal medicies to functional drink. This study is a document research in cluding a historical review of traditional beverages from after the period of the Joseon Dynasty, analysis of the characteristics of tradition the herbal medicine of the Joseon entered the new stage at the era of King Sejong. In the reign of Sejong, there had been a noticeable increase in kinds of herbs. Besides understanding the quality of herbs by provinces, the Joseon government also had pushed ahead with increase policy of herbs. The more the standard of living and incomes have increased, the more the concern of our health has been raised. This social phenomenon has affected on the food& beverage industry. The food& beverage industry of Korea has been already reorganized like the ones in developed countries in the early 21st century. Consumers are particular choosing their beverages; they concern their health, convenience, and taste more than ever. We had selected several herbal medicines contained in functional drink, and investigated the mechanism in field of oriental medicine and pharmacology.

수종 한약제의 위암세포에 대한 항암작용 효능 검색 및 약리작용에 관한 분자생물학적 연구 (Molecular analysis on the pharmacological actions of oriental medicines in stomach cancer cells)

  • Traditional oriental medicines have been used for treatment of various kinds of human cancers for long times and some of them proven to be effective clinically. However, the pharmacological actions and mechanisms related to cancer treatment are generally unknown. In an effort to clarify the action mechanisms of several oriental medicines used for cancer treatments, we planned this experimental procedures. We selected Cordyceps sinensis (冬蟲下草), Punellae Herba (夏枯草), Rehmanniae Radix (熟地黃), Paeoniae Radix (白芍藥), Oldenlandiae Herba (白花蛇舌草), Partulaceae Herba (馬齒? ), Scdopendra subspinipes mutilans (蜈蚣), Mylabris Phalerara (班蟄), Phellinus igniarius(桑黃), Ganodermae Lignum(靈芝) for evaluation, which have been used for patients of gastric cancers. The twenty grams of medicines were boiled in 100ml of water for 1 hour and filtered with $0.2\;{\mu}m$ pore-sized filter unit to remove insoluble particles. Initially we evaluated the effects oriental medicines on growth inhibition in stomach cancer cells. The gastric cancer cell line, AGS, was cultured in RPMI 1640 supplemented with l0% heat-inactivated fetal bovine serum and treated with $10{\mu}l$ oriental medicines per 1ml of medium up to 48 hours. The specimens were subjected to MTT assay for evaluation of growth inhibition. We found mat Mylabris phalerata (班蟄) markedly suppressed the growth of cancer cells as shown in results. Next, we checked the effects of oriental medicines on cancer on cell cycles and apptosis. Mylabrls phalerata (班蟄) inhibited cell cycle progression of cancer cells a compared with control cells and cells treated with other medicines. In addition, Mylabri phalerata (班蟄) induced apoptosis in 30-40% of stomch cancer cells within 24 hours. Other oriental medicines used for this experiments did not show apoptosis-inducing effects on cancer cells. Finally, we determined the expression levels of genes associated with cell cycle and apoptosis. The expressions of Bcl-2 and bcl-XL were downregulated by the treatment of Mylabris phalerata (班蟄). However, the expression levels of genes related to cell cycles were not altered significantly. In conclusion, we found that Mylabris phalerata (班蟄) has in vivo gowth-inhibiting and apptosis-inducing effects on stomach cancer cells. However, we think that at least animal experiments are necessay for evduations.

무동무언증과 인지기능장애를 주소로 하는 급성 백질뇌증 환자 치험 1례 (A Case of Acute Leukoencephalopathy Patient with Akinetic Mutism and Cognitive Dysfunctions)

  • This case report presents a 34-year-old female patient. Her chief complaints are akinetic mutism and cognitive dysfunctions. We have diagnosed her with leukoencephalopathy. She was treated with Korean traditional herbal medicines (Joojackwhan) for 33 days continuously. The effects of treatment were measured by the Korean Mini - Mental State Examination and Modified Bathel Index. After treatment, her chief complaints have been improved. Furthermore, the K-MMSE, MBI scores increased. From this case, we deduced that the Korean traditional herbal medicines (Joojackwhan) could be effective for cognitive dysfunctions and akinetic mutism caused by leukoencephalopathy.
