A pH Optosensor Based on Fluoresence from Nile Blue Encapsulated within Silica Sol-Gel Film (실리카 졸-겔막을 이용한 pH 광학센서)

  • A fiber optic pH sensor has been fabricated using nile blue entrapped in an ammonia catalyzed silica sol-gel film coated on glass substrate by dip-coating. The sensor was fixed on the end of an optical fiber. The sensor showed pH sensitivity when dipped into liquids at different pHs. Linear and reproducible responses were obtained in standard buffer solutions in the pH range $6.0{\sim}8.5$, which encompasses the clinically-relevant range. The effects of interferences on the determination of pH were also investigated. The sensors were successfully applied to the determination of pH in different commercial ionic drinks.

Effect of Cordycepin on the Expression of the Inflammatory Cytokines TNF-alpha, IL-6, and IL-17A in C57BL/6 Mice

  • Culture supernatants of splenocytes from C57BL/6 mice were exposed to 0.3, 1.0, and 3.0 ${\mu}g/ml$ cordycepin plus 3.0 ${\mu}g/ml$ lipopolysaccharide (LPS) to investigate the effects of cordycepin (3'-deoxyadenosine) on the production of inflammatory cytokines. Coadministration of 3.0 ${\mu}g/ml$ cordycepin with LPS in cultured murine spleen cells significantly diminished expression of the inflammatory cytokines tumor necrosis factor-${\alpha}$ and interleukin-6 (IL-6) in a time-dependent manner. Expression of the inflammatory cytokine IL-17A was substantially downregulated in a timeand concentration-dependent manner at all cordycepin concentrations. These findings suggest that cordycepin downregulates the immediate hypersensitivity reaction stimulated by LPS.

Effect of Clove Extracts on Tyrosinase Gene Expression (정향(Clove) 추출물이 티로시나아제 유전자의 발현에 미치는 효과)

  • Clove extract by methanol increased expression of the tyrosinase gene on B16 mouse melanoma cells containing tyrosinase promoter. $10{\mu}g/mL$ and $100{\mu}g/mL$ of the extract showed expression rate of the tyrosinase gene about 138% and 245%, respectively, compared with control. At $500{\mu}g/mL$, expression rate of the extract was impossible to measurement by high cytotoxicity. The solvent fraction of methylene chloride also exhibited highly expression rate as methanol extract. However, the solvent fractions of butyl alcohol and water showed repressive effect on expression of tyrosinase gene at $500{\mu}g/mL$. In MTT assay, cell survival rate of the extract exhibited similar to expression rate of tyrosinase gene. That is, $10{\mu}g/mL$ and $100{\mu}g/mL$ of the extract showed the cell survival rate about 128% and 187%, respectively.

Rahnella aquatilis Strain AY2000 Produces an Anti-Yeast Substance

  • To screen for an anti-yeast substance (AYS), many bacteria were isolated from soil and a strain AY2000 was selected. The strain AY2000 was identified as Rahnella aquatilis by morphology, biochemical properties, and 16S r-RNA nucleotide sequence analyses. The strain AY2000 showed anti-yeast activity against Candida albicans and Saccharomyces cerevisiae, whereas R. aquatilis ATCC33071 as a type strain did not show the activity against the yeasts under the same condition. The growth of yeast cell was significantly inhibited by AYS produced by the strain AY2000, as shown by optical density and MTT assay. The minimum inhibitory concentration (MIC) of the AYS against S. cerevisiae and C. albicans at $28^{\circ}C\;was\;20{\mu}g/ml\;and\;60{\mu}g/ml$, respectively. The MIC of AYS against hyphae of C. albicans at $37^{\circ}C\;was\;600{\mu}g/ml$. Scanning electron microscopic analysis revealed that yeast cells treated with AYS had an irregular form with a wrinkled and rough surface.

A Study on a Solution For Growth of Beauty Salon Franchise - around an influence of a conflict upon a solidarity -

  • A purpose of this study is to measure degree of solidarity and conflict of the beauty salon franchisees according to an influence strategy of franchiser and grasp characteristics revealed from managing a franchise system. This will also contribute to improving results of franchise systems and considering a direction for development. This study examined existing studies relevant with conflict and solidarity and analysed relativity between relevant variables and grasped the flow of studies and then set hypotheses in order to present a device for decreasing conflict within a franchise system of beauty shops and for increasing a solidarity and subsequently analysed samples and tested data. The data for this study are researched by sending questionnaires to beauty salons which participated in one of franchise systems and they are analyzed by various methods in a statistics. After studying some relevant theories, this study used the definition of the franchise system in beauty salons as an entity of a political-economy paradigm. A franchiser desires to influence member shops with an influence-strategy. Therefore member shops which perceived it go through a conflict and then such a process is revealed as making significant influence on solidarity. Beauty salon franchise exists by reciprocal confidence and transaction. If an opportunism gain in influence owing to an conflict intensification, there would be no steady formation of relationship. Therefore the head office should develop a member-shop-management-system by more scientific and more technical methods and prevent, in advance, member stores from seeking for negative opportunism.

Expression of Streptodornase by Use of Streptokinase Promoter in Streptococcus equismilis H46A. (Streptococcus equisimilis H46A에서 Streptokinase 프로모터를 이용한 Streptodornase 발현 증진)

  • A gene encoding streptodornase(sdc) from Streptococcus equisimilis H46A was expressed in S. equisimilis H64H sdc under the control of the streptokinase gene promoter. Secretion of the streptodornase was directed by the signal sequences of streptokinase or streptodornase. The expressed streptodornase activity from S. equisimilis H46A sdc transformant with streptokinase promoter - streptodornase coding sequence fusion vector was 2.3 fold higher than that from wild type. Construct of signal sequence region replaced by streptokinase ones was similarly expressed as a wild type. But constructs of skc or lrp core regions of streptokinase promoter streptodornase fusion were similarly expressed as in sdc mutant. In conclusion, improved expression of streptodornase by use of streptokinase promoter required the full length of promoter.

Protective Effect of Crataegi Fructus Extract on the Neurotoxicity Induced by Reactive Oxygen Species in Cultured C6 Glioma Cell

  • To clerify the antioxidant effect of Crataegi Fructus (CF) extract on reactive oxygen species (ROS), The C6 glioma cells were treated with various concentrations of hydrogen peroxide ($H_2O_2$). The $H_2O_2$-induced neurotoxicity was measured by XTT assay for the cell viability. For the protective effect of CF extract on the cytotoxicity induced by $H_2O_2$, cell viability, lactate dehydroganase (LDH) activity, and the inhibitive activity of lipid peroxidation of CF extract were performed. In this study, $H_2O_2$ decreased cell viability dose- and time-dependent manners and increased LDH activity compared with the control. In the protective effect on $H_2O_2$, CF extract increased cell viability and decreased LDH activity on $H_2O_2$-induced cytotoxicity, lipid peroxidation by FTC assay. From these results, It is suggested that $H_2O_2$ was highly toxic on cultured C6 glioma cells, and also, CF extract showed the protective effect on $H_2O_2$-mediated cytotoxicity.

Isolation of Lipase Producing Bacillus subtilis and Some Characteristics of the Enzyme (중성 Lipase를 생산하는 Bacillus subtilis JKA-3의 분리 동정 및 효소 특성)

  • As part of an investigation to identify microorganisms that are biotechnologically interesting for industrial application, we isolated a bacterial strain from Chungkookjang that produces extracellular neutral lipase. In addition, the crude enzyme was characterized. This isolated strain, designated as JKA-3 was identified as Bacillus subtilis JKA-3 based on morphological, physiological and biochemical characteristics, as well as phylogenetic analysis using 16S rRNA gene sequence. The cells were rod-shaped and $0.6-0.8{\times}2.0-2.3\;{\mu}m$ in size. Optimal growth conditions were $35-40^{\circ}C$ and pH 6.0-8.0. The isolate was able to grow in up to 0-10.0% (w/v) NaCl. Optimal activity conditions of the crude lipase fraction of B. subtilis JKA-3 were pH of 7.0 at $35^{\circ}C$. This enzyme was stable in the pH ranging 6.0-8.0.

  • Periodically, harmful algal blooms (HABs) have occurred, with impacts on various areas including public health, tourism, and aquatic ecosystems, especially aquacultured and caged fisheries. To prevent or manage invasions of HABs into fish farms on an emergency basis, many methods have been proposed. Frequently over the past 30 years in coastal countries, treatments of clay and clay mixed with polyaluminum chloride (PAC) and chitosan have been tested for HAB-removal effectiveness in both the laboratory and the field. In Korea, yellow loess clay (hwangto) has been dispersed using electrolytic clay dispensers, both to decrease the amount of yellow loess clay's usage in containers and enhance HAB-removal efficiency. However, this emergency method has limitations, among which is the requirement for more effective controlling agents for field applications. Thus, in this paper, we review technologies for clay-based red tides prevention and control and their limitations, and, further, introduce next-generation algicidal technologies for the emergency protection of fish farms.

The Effect of Experience Marketing on Brand Attitude and Brand Loyalty of Beauty Salon Franchise Stores (프랜차이즈 미용실에서의 체험마케팅이 브랜드 태도와 브랜드 충성도에 미치는 영향)

  • This study examined the relative importance level of experience marketing on brand attitude and brand loyalty in beauty salon franchise stores, and attempted to raise the competitiveness of beauty salon services by presenting this elements as experience marketing strategy directions for beauty salon franchise store management. A questionnaire was used for this study on 350 adults residing in Gwangju Metropolitan City and SPSS 20.0 statistics package was used for analysis. In summary, the results of this study are as follows. First, five factors that make up experience marketing in beauty salon franchise stores were found. They were sensory marketing, relative marketing, emotional marketing, cognitive marketing and behavior marketing. Brand loyalty was categorized into the two factors, attitude loyalty and behavioral loyalty. Also, we found that experience marketing had a statistically positive impact on brand loyalty. Secondly, experience marketing of beauty salon franchise stores was found to have a statistically significant positive effect on brand attitude. Thirdly, we found that the brand attitude of beauty salon franchise stores had a statistically significant positive effect on brand loyalty.
