Phenylpropanoid Glycosides of Paulownia coreana Uyeki Leaves

  • The leaves of Paulownia coreana Uyeki were collected, extracted with acetone-$H_2O$ (7:3, v/v), concentrated under reduced pressure and successively fractionated using n-hexane, methylene chloride, ethyl acetate and water on a separatory funnel. A portion of the ethyl acetate soluble powder was chromatographed on a Sephadex LH-20 column using aqueous methanol and ethanol-hexane as washing solvents. Two isomeric phenylpropanoid glycosides were isolated and elucidated as verbascoside and isoverbascoside by NMR and MS spectrometers.

Neuraminidase Inhibitors from the Culture Broth of Phellinus linteus

  • During the search for neuraminidase inhibitors from medicinal fungi, we found that the culture broth of Phellinus linteus exhibited potent inhibitory activity. Solvent partition, Sephadex LH-20 column chromatography, and high-performance liquid chromatography (HPLC) were performed for purification of two active substances from the culture broth. According to $^1H$ NMR measurements and comparison of HPLC retention times with those of authentic compounds, their chemical structures were identified as hispidin and hypholomine B. Compounds (hispidin) 1 and 2 (hypholomine B) inhibited neuraminidase, with $IC_{50}$ values of 13.1 and 0.03 ${\mu}M$, respectively.

Antioxidative Activity of Prunus sargentii Outer Bark Extractives

  • The outer bark of Prunus sargentii was collected, air-dried and extracted with 70% aqueous acetone. Then it was successively partitioned with n-hexane, dichloromethane ($CH_2Cl_2$), ethyl acetate (EtOAc) and $H_2O$. From the EtOAc soluble fraction, four compounds were isolated by the repeated Sephadex LH-20 column chromatography. The isolated compounds were determined as (+)-catechin (1), (-)-epicatechin (2), taxifolin (3), and neosakuranin (4) by the spectroscopic analysis including $^1H$, $^{13}C$-NMR, and 2D-NMR spectrometers. The antioxidative activities on the isolated compounds and the separated fractions were evaluated by DPPH radical scavenging assay. The crude, EtOAc, and $H_2O$ soluble fractions indicated good antioxidative potential compared to the $CH_2Cl_2$ and n-hexane soluble fractions.

  • An antifungal antibiotic was produced from Bacillus sp. YJ-63, and antibiotic was purified to a homogeneity by butanol extraction, Silica-gel column chromatography, Sephadex LH-20 gel filtration column chromatography and HPLC. The purified antibiotic showed one spot on Thin Layer Chromatography, which was negative to ninhydrin and positive to iodine. This finding indicated that the antibiotic was a cyclopeptide structure. UV absorption spectrum of the antibiotic in methanol was maximal at 277 nm, which corresponded to the spectrum for all of the Iturin antibiotic. The antibiotic was stable up to $121^{cdot}C$ and in the range of pH $3.O{\sim}l2.0$ with a stable of ${\alpha}-amino$ acid and ${beta}-amino$ acid, and showed high activity for fungi and yeasts,

Lipoxygenase Inhibitor from Defatted Nutmeg Seed

  • Lipoxygenase inhibitory acitivity of the methanolic extract of 60 different kinds of plant seeds was determined by a spectrophotometric method using a soybean lipoxigenase(SLO) and linolenic acid. Among the extracts examined, the methanolic extract of nutmeg(Myristical fragrans)seed showed the most potent SLO inhibitory activity. To isolate SLO inhibitor, hence, the defatted methanol extract was further partitioned with ether, ehtylacetate , and n-butanol , stepwise. The ether souble fraction was successively chromatographed on silica gel, Sephadex LH-20 and preparative TLC. Three phenolic compounds were isolated , and one of them showing a strong SLO inhibition activity was identified as a 2,6-dihydroxy-9-(3', 4', -dihydroxyphenyl)nonylphenone (IC50a=0.39$\mu\textrm{g}$/ml) by 1H-& 13C0NMR, IR, and MS spectroscopy.

Radical Scavenging Activity of Gallic Acid from Woodfordia Fruticosa Flowers

  • Woodfordia fruticosa Kurz (Lythraceae) is used in the treatment of various ailments in traditional medicines. DPPH activity guided fractionation and purification process was used to identify the free radical-scavenging components from the flowers of this plant. The methanolic extract of the plant was first fractionated into four extracts; namely, n-hexane, chloroform, ethyl acetate and water fractions. Among them, the ethyl acetate fraction was found to be the most effective and was further subjected to activity guided-fractionation and isolation procedures. After successive column chromatography on silica gel and Sephadex LH-20, gallic acid, which is responsible for the radical scavenging activity, was isolated and its structure was elucidated by spectral methods ($^1H$ NMR, $^{13}C$ NMR) and by comparison with literature.

  • The antioxidant activity of Paeonia suffruticosa (Paeoniaceae) was determined by measuring the radical scavenging effect on DPPH (1,1-diphenyl-2-picrylhydrazyl). The methanolic extract of P. suffruticosa showed the strong antioxidant activity. The antioxidant activity of ethyl acetate soluble fraction was stronger than that of the others ($IC_{50}$, $1.2\;{\mu}g/mL$), and the fraction was subjected to purification by repeated silica gel and Sephadex LH-20 column chromatography. Suffruticosol A, suffruticosol B, methyl gallate, and gallic acid, were isolated from the ethyl acetate soluble fraction. Their structures were elucidated by physico-chemical and spectroscopic studies.

  • $^1H$$^{13}C$ 핵자기(核磁氣) 공오(共嗚) 분광기(分光器), 적외선(赤外線) 흡광(吸光) 분광기(分光器)와 질량분석기(質量分析器)를 이용(利用)하여 설폰화(化) 반응(反應)의 한 반응물(反應物)이 단리(單離)되어 구조적(構造的)으로 규명(糾明)되었다. 그 화합물(化合物)은 에탄올 및 에탄올 수용액(水溶液)을 용매(容媒)로 사용(使用)하여 Sephadex LH-20상(上)에서의 반복적(反復的)인 컬럼 크로마토그래피로 단리 및 정제(精製)되었다. 규명된 화합물은 이전에 보고(報告)된 바 없는 새로운 구조의 disodium epicatechin-(4${\beta}$, 5')-disulfonate였다. 이 화합물은 온화(溫和)한 설폰화(化) 조건하(條件下)에서 최초(最初)의 친전자(親電子) 치환(置換)의 예(例)로서 catechol B 환구조(環構造)가 quinone methide 중간물(中間物)을 생성(生成)하는 한 반응기구(反應機構)가 제시(提示)되었다.

The $H_2O$ soluble fraction was chromatographed on a Sephadex LH-20 column using aqueous methanol and ethanol-hexane as washing solvents. Two isomeric phenylethanoid glycosides, verbascoside (1) and isoverbascoside (2), and one epimeric phenylethanoid glycoside, campneoside II (3), were isolated and their structures were elucidated on the basis of chemical and spectroscopical data.

애기땅빈대의 화학적 성분 (Chemical constituents from the whole plants of Euphorbia supina Rafin)

  • Eight compounds were isolated from the whole plants of Euphorbia supina (Euphorbiaceae) through repeated silica gel, YMC gel and Sephadex LH-20 column chromatography. Their chemical structures were elucidated as 7-hydroxy-6-methoxycoumarin (scopoletin) (1), p-hydroxybenzaldehyde (2), methyl gallate (3), gallic acid (4), quercetin (5), quercetin $3-O-{\alpha}-L-arabinofuranoside$ (avicularin) (6), kaempferol $3-O-{\alpha}-L-arabinofuranoside$ (juglanin) (7) and kaempferol $3-O-{\beta}-D-glucopyranoside$ (astragaline) (8) by spectroscopic (NMR and MS) analysis.
