Characterization and Identification of Organic Selenium-enriched Bacteria Isolated from Rumen Fluid and Hot Spring Water

  • In the present study, the isolation of selenium (Se)-enriched bacteria from rumen fluid and hot spring water was carried out. Rumen fluid samples were taken from cannulated goats fed a basal diet and the water samples were collected from Selayang hot spring, Selangor-Malaysia. A total number of 140 Se-tolerant isolates were obtained aerobically using an Se-enriched medium and spread plate technique. All the isolates were initially screened for the ability to transform the Se-containing medium to a red-orange culture using a spectrophotometer. Twenty isolates of dark red-orange medium were selected for a screening of the highest Se-containing protein accumulating strains using the dialysis technique and icp.ms to measure the Se content. Four isolates, identified as Enterobacter cloacae (ADS1, ADS7, and ADS11), and Klebsiella pneumoniae (ADS2) from rumen fluid origin, as well as, one isolate from hot spring water (Stenotrophomonas maltophilia (ADS18)), were associated with the highest biomass organic Se-containing protein when grown in a medium enriched with $10{\mu}g/ml$ sodium selenite. In addition, around $50{\mu}g/100{\mu}g$ of the absorbed inorganic Se was accumulated as an organic form. Organic Se-containing protein in all the selected strains showed antioxidant properties in the range of 0.306 to 0.353 Trolox equivalent antioxidant capacity (TEAC) mg/ml. Therefore, these strains may offer a potential source of organic Se due to their Se-tolerant nature and higher biomass organic to inorganic Se ratio.

Photoelectrochemical Cell Study on Closely Arranged Vertical Nanorod Bundles of CdSe and Zn doped CdSe Films

  • Closely arranged CdSe and Zn doped CdSe vertical nanorod bundles were grown directly on FTO coated glass by using electrodeposition method. Structural analysis by XRD showed the hexagonal phase without any precipitates related to Zn. FE-SEM image showed end capped vertically aligned nanorods arranged closely. From the UV-vis transmittance spectra, band gap energy was found to vary between 1.94 and 1.98 eV due to the incorporation of Zn. Solar cell parameters were obtained by assembling photoelectrochemical cells using CdSe and CdSe:Zn photoanodes, Pt cathode and polysulfide (1M $Na_2S$ + 1M S + 1M NaOH) electrolyte. The efficiency was found to increase from 0.16 to 0.22 upon Zn doping. Electrochemical impedance spectra (EIS) indicate that the charge-transfer resistance on the FTO/CdSe/polysulfide interface was greater than on FTO/CdSe:Zn/polysulfide. Cyclic voltammetry results also indicate that the FTO/CdSe:Zn/polysulfide showed higher activity towards polysulfide redox reaction than that of FTO/CdSe/polysulfide.

Point defect for $AgGaSe_2$ epilayers grown by hot wall epitaxy (Hot Wall Epitaxy (HWE)법에 의해 성장된 $AgGaSe_2$ 에피레이어의 점결함 연구)

  • To obtain the single crystal thin films, $AgGaSe_2$ mixed crystal was deposited on thoroughly etched semi-insulating GaAs(100) substrate by the hot wall epitaxy (HWE) system. The source and substrate temperatures were $630^{\circ}C$ and $420^{\circ}C$, respectively. The temperature dependence of the energy band gap of the $AgGaSe_2$ obtained from the absorption spectra was well described by the Varshni's relation, $E_g$(T) 1.9501 eV - ($8.79\times10^{-4}$ eV/K)$T^2$/(T + 250 K). After the as-grown $AgGaSe_2$ single crystal thin films was annealed in Ag-, Se-, and Ga-atmospheres, the origin of point defects of $AgGaSe_2$ single crystal thin films has been investigated by the photoluminescence(PL) at 10K. The native defects of $V_{Ag}$, $V_{Se}$, $Ag_{int}$, and $Se_{int}$ obtained by PL measurements were classified as a donors or acceptors type. And we concluded that the heat-treatment in the Ag-atmosphere converted $AgGaSe_2$ single crystal thin films to an optical p-type. Also, we confirmed that Ga in $AgGaSe_2$/GaAs did not form the native defects because Ga in $AgGaSe_2$ single crystal thin films existed in the form of stable bonds.

Thermoelectric Properties of the n-type $Bi_2(Te,Se)_3$ Processed by Hot Pressing (n형 $Bi_2(Te,Se)_3$ 가압소결체의 열전특성)

  • The n-type $Bi_2(Te,Se)_3$ powders were fabricated by melting/grinding method and were hot-pressed in order to compare thermoelectric properties of the hot-pressed specimens with those of the $Bi_2(Te,Se)_3$ ingot. Effects of mechanical milling treatment of the $Bi_2(Te,Se)_3$ powders on thermoelectric characteristics of a hot-pressed specimen were also examined. The hot-pressed $Bi_2(Te,Se)_3$ exhibited power factors of $27.3{\sim}32.3{\times}10^{-4}W/m-K^2$ which were superior to $24.2{\times}10^{-4}W/m-K^2$ of the ingot. The $Bi_2(Te,Se)_3$, hot-pressed after mechanical milling treatment of the powders, possessed a non-dimensional figure-of-merit of 1.02 at $100^{\circ}C$ and exhibited extrinsic-intrinsic transition at $130^{\circ}C$.

Study on the Effect of a New Antiseptic Preparation ″Swi-Se-Yo″ on the Carry-Over Cocoons in Silk Reeling Process (새로 개발한 조월견방부제 ″쉬세요(Swi-Se-Yo)″의 효과에 관하여)

  • In silk reeling process the carry-over cocoons must be submerged in the reeling baths filled with reeling water and left until reopening the operation. Under the detention the carry-over cocoons are apt to decay without any antiseptic treatment. Thus an useful antiseptic for the cocoons is urgently needed, and various antseptic agents have been tested for their applicability to the process. However, such an useful agent has not been developed yet. Formalin has been the only chemical used for antisepticizing carry-over cocoons, although it has many defects as the antiseptic for the cocoons. In these circumstances, recently we newly prepared an antiseptic useful for preventing the carryover cocoons from decaying. We named the new antiseptic preparation "Swi-Se-Yo." The Korean term "Swi-Se-Yo" literally means "please take a rest". Through a series of experiments with Swi-Se-Yo we obtained the following results: 1) Swi-Se-Yo, in 0.05% aqueous solution, exerted a good antiseptic effect on the boiled Cocoons submerged in the reeling baths and the effect lasted for 45 hours. The duration of the effect is about two times longer than that of Formalin. 2) The percentage of cocoon reel ability of the carry-over cocoons treated with Swi-Se-Yo was 6% higher than that of Formalin and was equal to that of flowing cold water. 3) The percentage of raw silk yield of the carry-over cocoons treated with Swi-Se-Yo was almost equal to that of Formalin and to that of flowing cold water. 4) The quality of raw silk of the carry-over cocoons treated with Swi-Se-Yo is the same as that of flowing cold water. Besides the above favourable results, Swi-Se-Yo has many advantages as an antiseptic. Chemically it is very stable. Its antimicrobial action is very strong and the spectrum is very broad. It can be available in water-soluble powder and in small bulk. And it is not harmful to human and domestic animals. Considering these profitable properties of Swi-Se-Yo, it will have a good reputation as a carry-over cocoon antiseptic. (The chemical composition and manufacturing method of Swi-Se-Yo will be published in the near future.)

Evaluation of Anti-SE Bacteriophage as Feed Additives to Prevent Salmonella enteritidis (SE) in Broiler

  • This experiment was conducted to evaluate anti-Salmonella enteritidis (anti-SE) bacteriophage as feed additives to prevent Salmonella enteritidis in broilers. The experimental diets were formulated for 2 phases feeding trial, and 3 different levels (0.05, 0.1 and 0.2%) of anti-SE bacteriophage were supplemented in basal diet. The basal diet was regarded as the control treatment. A total of 320 1-d-old male broilers (Ross 308) were allotted by randomized complete block (RCB) design in 8 replicates with 10 chicks per pen. All birds were raised on rice hull bedding in ambient controlled environment and free access to feed and water. There were no significant differences in body weight gain, feed intake and feed conversion ratio (FCR) at terminal period among treatments (p>0.05). Relative weights of liver, spleen, abdominal fat and tissue muscle of breast obtained from each anti-SE bacteriophage treatment were similar to control, with a slightly higher value in anti-SE bacteriophage 0.2%. In addition, a numerical difference of glutamic-oxaloacetic transaminase (GOT), glutamic-pyruvic transaminase (GPT) and LDL cholesterol level was observed in the 0.2% anti-SE bacteriophage application even though blood profiles were not significantly affected by supplemented levels of anti-SE bacteriophage (p>0.05). In the result of a 14 d record after Salmonella enteritidis challenge of 160 birds from 4 previous treatments, mortality was linearly decreased with increasing anti-SE bacteriophage level (p<0.05), and Salmonella enteritidis concentration in the cecum was decreased with increasing levels of anti-SE bacteriophage (p<0.05). Based on the results of this study, it is considered that supplementation of 0.2% anti-SE bacteriophage may not cause any negative effect on growth, meat production, and it reduces mortality after Salmonella enteritidis challenge. These results imply to a possible use of anti-SE bacteriophage as an alternative feed additive instead of antibiotics in broilers diet.

Effect of Selenium-enriched Japanese Radish Sprouts and Rhodobacter capsulatus on the Cholesterol and Immune Response of Laying Hens

  • Immune response and yolk cholesterol are crucial factors for commercial chicken producers. The objectives of this study were to investigate the effect of selenium-enriched Japanese radish sprouts (Se-enriched JRS) and R. capsulatus synergistically on immune response and cholesterol in laying hens. A total of 50 laying hens (20-wk old) were assigned to 5 dietary treatment groups, and fed diets supplemented with 2.5 ${\mu}g/kg$, 5 ${\mu}g/kg$, 10 ${\mu}g/kg$ Se-enriched JRS and 5 ${\mu}g/kg$ Se-enriched JRS+R. capsulatus (0.02%). Egg production and yolk color were significantly improved by the supplementation of Se-enriched JRS+R. capsulatus in the layer diet (p<0.05). Compared to the control, serum cholesterol concentration and triglyceride levels were decreased by all the treatments (p<0.05). After 8-wk of the experiment, supplementation of 5 ${\mu}g/kg$, 10 ${\mu}g/kg$ and Se-enriched JRS+R. capsulatus significantly reduced yolk cholesterol and triglycerides, while the greatest reduction was observed when R. capsulatus was incorporated with Se-enriched JRS. Spleen, bursa and thymus weight were significantly increased by both the 5 ${\mu}g/kg$ and 10 ${\mu}g/kg$ Se-enriched JRS. Compared to the control, supplementation of 5 ${\mu}g/kg$ and 10 ${\mu}g/kg$ Se-enriched JRS significantly increased serum IgG and yolk IgY concentration and foot web index activity by Newcastle Disease Virus (p<0.05). After 4-wk and 8-wk of supplementation, the highest number of leukocytes was observed with Se-enriched JRS+R. capsulatus (p<0.05). The highest concentration of serum and yolk Se was found in Se-enriched JRS plus R. capsulatus treatment. Combined dietary supplementation of Se-enriched JRS and R. capsulatus might be beneficial for better health, disease protection and overall production performance.

Sorghum extract exerts an anti-diabetic effect by improving insulin sensitivity via PPAR-${\gamma}$ in mice fed a high-fat diet

  • This study investigated the hypothesis that a sorghum extract exerts anti-diabetic effects through a mechanism that improves insulin sensitivity via peroxisome proliferator-activated receptor gamma (PPAR-${\gamma}$) from adipose tissue. Seven C57BL/6 mice were fed an AIN-93M diet with fat consisting of 10% of total energy intake (LF) for 14 weeks, and 21 mice were fed a high-fat AIN diet with 60% of calories derived from fat (HF). From week 8, the HF diet-fed mice were orally administered either saline (HF group), 0.5% (0.5% SE group), or 1% sorghum extract (1% SE group) for 6 weeks (n = 7/group). Perirenal fat content was significantly lower in the 0.5% SE and 1% SE groups than that in the HF mice. Levels of total and low-density lipoprotein cholesterol, triglycerides, glucose, and the area under the curve for glucose were significantly lower in mice administered 0.5% SE and 1% SE than those in HF mice. Serum insulin level was significantly lower in mice administered 1% SE than that in HF mice or those given 0.5% SE. PPAR-${\gamma}$ expression was significantly higher, whereas the expression of tumor necrosis factor-${\alpha}$ was significantly lower in mice given 1% SE compared to those in the HF mice. Adiponectin expression was also significantly higher in mice given 0.5% SE and 1% SE than that in the HF mice. These results suggest that the hypoglycemic effect of SE may be related with the regulation of PPAR-${\gamma}$-mediated metabolism in this mouse model.

Rectangular Region-based Selective Enhancement (RSE) for MPEG-4 FGS Video (MPEG-4 FGS 비디오를 위한 사각영역 기반의 선택적 향상기법)

  • In MPEG-4 FGS (fine granular scalability) video, SE (selective enhancement) function is adopted to enhance the subject quality of the region of interest (ROI). However, it has the problem of excessive bit-rate increase in the enhancement layer. We present a new rectangular region-based SE (RSE) algorithm to significantly reduce the overhead bits resulting from the standard SE. The proposed RSE is based on two new algorithms. The first is to apply the SE function to a rectangular region. By doing so, we can reduce the required bits for describing the selectively enhanced region. The second is to use constrained bit-plane scanning (CBS) to encode bit-planes of the enhancement layer. By using CBS, we can efficiently encode the ALL-ZERO symbols that are generated by applying the SE. It Is shown by simulation that the proposed RSE can provide a good visual quality for the selected rectangular region with significantly reduced overhead bits.
