솔잎 열수추출물이 카드뮴으로 유도한 흰쥐의 산화적 손상에 미치는 영향 (Effect of Pine Needle Water Extract on Cadmium-Induced Oxidative Stress in Rats)

  • 솔잎 열수추출물이 카드뮴으로 유도한 흰쥐의 산화적 손상에 미치는 영향을 살펴보고자 체중 kg당 5mg의 카드뮴$(CdCl_2)$을 매주 1회 경구투여하였다. 솔잎은 매일 일정시각에 체중 kg당 1.26g수준이 되도록 4주간 경구투여 사육한 결과 카드뮴과 솔잎 열수추출은 체중에는 영향을 미치지 않았으며 솔잎 열수추출물은 식이섭취량을 감소시켰다. 체중 100g당 간조직 무게는 카드뮴 대조군이 정상군에 비하여 유의적으로 증가하였으며 솔잎 열수추출물은 간조직 무게를 감소시키는 경향이었다 혈장 중의 AST와 ALT활성은 카드뮴 투여에 의해 유의적으로 증가되었으나 솔잎 열수추출물에 의해 ALT활성이 카드뮴 대조군에 비하여 유의적으로 감소되었다. 혈장 알부민 함량은 실험군간의 차이가 없었으나 크레아티닌 함량은 솔잎 열수추출물 급여시 낮았다. 카드뮴 투여시 간조직의 CYP함량은 유의적으로 감소되었으며 솔잎 열수추출물에 의한 영향은 없었다. 한편 XO와 ADH활성은 솔잎 열수추출물군이 정상군과 카드뮴 대조군에 비하여 유의적으로 높았다. 또한 카드뮴 투여에 인해 유의적으로 높아진 SOD, MAO, CAT 및 GSH-Px 활성도는 솔잎 열수추출물에 의해 정상수준으로 회복되었다. 간조직 중의 글루타티온 함량은 솔잎 열수추출물 급여에 의한 차이가 발견되지 않았으나 MDA함량은 솔잎 열수추출물 급여군에서 카드뮴 대조군에 비하여 유의적인 감소를 보임으로써 카드뮴에 의해 유도된 지질과산화가 솔잎 열수추출물 급여로 개선될 수 있음을 제시한다.

tert-Butyl Hydroperoxide로 산화 스트레스가 유도된 HepG2 세포에서 말채나무 열수추출물의 항산화 활성 (Antioxidant Activities of Hot Water Extract from Cornus walteri Wanger against Oxidative Stress Induced by tert-Butyl Hydroperoxide in HepG2 Cells)

  • 말채나무 열수추출물의 항산화 활성을 알아보기 위해 HepG2 세포에 TBHP로 산화 스트레스를 유도한 뒤 말채나무 열수추출물의 세포 보호효과, ROS 생성억제, 지질과산화 억제 및 GSH 생성에 미치는 영향에 대해 살펴보았다. 말채나무 열수추출물은 HepG2 세포에 TBHP로 산화 스트레스를 유도한 뒤 나타나는 세포독성에 대해 농도 의존적으로 유의하게 세포 보호효과를 보였으며, ROS 생성과 과산화물에 대한 지표로서 측정한 MDA도 말채나무 열수추출물에 의해 효과적으로 억제되었다(P<0.05). 또한 항산화 성분으로 생체 내에서 산화 및 환원반응에 중요한 역할을 하며 항산화 효소인 GSH-Px, GST에 전자공여체로 작용하는 GSH의 생성촉진 효능에서도 말채나무 열수추출물은 산화 스트레스로 감소된 GSH의 생성을 농도 의존적으로 촉진시켰다(P<0.05). 산화 스트레스에 의해 활성이 증가된 항산화 효소(CAT, SOD, GSH-Px, GR)도 말채나무 열수추출물의 처리로 감소하는 경향을 보였다. 이러한 결과로 미루어 보아 말채나무 열수 추출물은 인체 내에서 질병과 노화를 일으키는 원인 물질인 활성산소에 대해 강한 항산화 활성을 나타냄에 따라 보다 다양한 형태의 소재로 활용될 수 있을 것으로 생각된다.

좌금환이 역류성 식도염 흰쥐에 미치는 영향 (Effects of Jwa Kum-Whan on Reflux Esophagitis in Rats)

  • Objectives: This study was designed to investigate the effects of Jwa Kum-Whan (JKW) on reflux esophagitis in rats.Methods: Forty rats were divided into five groups: a sham group (with no medication and only treated with ventrotoby); a group with reflux esophagitis (RE); a pantoprazole group (treated with 30 mg/kg pantoprazole per day for two weeks); a JKW280 group (treated with 280 mg/kg JKW per day for two weeks); and a JKW560 group (treated with 560 mg/kg JKW per day for 2 weeks). All rats fasted for 24 hrs and then were induced with RE by the oral administration of indomethacin and by a pylorus and forestomach ligation operation. After 8 hrs, the rats were sacrificed. We measured body weight, gastric juice pH, gastric volume, antioxidant activity, and cytokine and made a histologic examination of the esophagus and the stomach.Results: The weights of the rats in each group were not significantly different. The gastric juice pH significantly increased in the JKW560 group and the pantoprazole group compared with the RE group. Gastric volume significantly decreased in the JKW560 group compared with the RE group and the pantoprazole group. SOD activities significantly increased in the JKW280 and JKW560 groups compared with the RE group. Catalase activities significantly increased in the pantoprazole group and the JKW560 group compared with the RE group. TNF-α significantly decreased in the JKW280 and JKW560 groups compared with the RE group. IL-6 significantly decreased in the pantoprazole group and the JKW280 and JKW560 groups compared with the RE group. Histologic examination of the esophagus and the stomach showed significant improvements in the pantoprazole, JKW280, and JKW560 groups compared with the RE group.Conclusion: Based on these results, it is concluded that JKW can prevent reflux esophagitis.

유산균을 이용한 보리의 발효를 통한 항산화 및 미백 효과 (Antioxidant and Whitening Effects of the Fermentation of Barley Seeds (Hordeum vulgare L.) Using Lactic Acid Bacteria)

  • 보리(Hordeum vulgare L.)는 Poaceae/Gramineae 과에 속하며 주위에 흔히 있는 곡물이지만 고대 이집트에서는 보리가 의약품으로 사용되어 왔으며 오랜 전부터 많은 나라에서 기능성 식품, 향료 및 맥주로 사용되는 중요한 곡물이다. 보리의 효능에는 항염증, 항암 및 항당뇨병에 효과가 있는 것으로 알려져 있다. 현재까지 보리에 관한 많은 연구가 진행되어왔지만 유산균 발효를 이용한 발효 보리에 대한 연구는 많이 진행되지 않았다. 본 연구에서는 유산균을 이용하여 발효 후 에탄올로 추출한 보리 씨앗의 효과에 대하여 조사 하였다. 이를 위해서 유산균으로 발효한 보리 씨앗과 발효하지 않은 보리의 총 폴리페놀 함량, 총 플라보노이드 함량, DPPH 라디칼 소거능, SOD 유사활성 및 타이로시나제 저해 활성을 분석하였다. 결과적으로 유산균으로 발효한 보리 씨앗이 발효하지 않은 보리 씨앗보다 더 효과적인 항산화 효과 및 미백 활성을 나타내는 것으로 보였다. 따라서 우리는 유산균으로 발효한 보리 씨앗이 건강 기능 식품 및 화장품 개발의 기초가 될 수 있음을 제안한다.

찔레나무뿌리(Rosa multiflora root)의 항산화 및 항염증효과 (Anti-Oxidant and Antiinflammatory Effects of Rosa multiflora Root)

  • 찔레나무뿌리를 열수, 에탄올, 메탄올, 아세톤에서 추출하여 항산화 및 항염증 효능효과를 확인하였다. 그 결과 아세톤추출물에서 가장 많은 67.05${\pm}$0.56 mg/g의 폴리페놀 함량을 확인하였으며, SOD 유사활성능을 제외한 DPPH, ABTS, superoxide anion 라디컬소거능 확인 결과 우수한 항산화 효과를 확인하였다. 찔레나무뿌리 추출물의 HAase 저해능을 측정한 결과 에탄올, 메탄올, 아세톤 추출물 500 ${\mu}g/ml$ 에서 60% 이상의 높은 HAase 저해 효과를 확인할 수 있었다. NO 생성량을 확인한 결과, RAW 264.7 cell에서 LPS에 의해 유도된 NO 생성을 농도의존적으로 뚜렷하게 감소시키는 것을 확인하였으며, 에탄올 추출물에서 NO 생성억제 효과가 가장 우수한 것을 확인할 수 있었다. 위의 사실에 기초하여 NO 생성저해의 기전을 알아보기 위하여 iNOS의 발현을 분석한 결과, 에탄올추출물 100 ${\mu}g/ml$에서 40%의 iNOS protein 발현 감소를 확인하였으며, iNOS의 발현억제가 NO생성억제와 유사한 경향을 나타냄으로 NO생성억제는 iNOS의 발현저해를 경유한 것임을 확인할 수 있었다. 이러한 결과들은 찔레나무뿌리 추출물이 항산화 및 항염증 연구의 기초 자료로 활용될 것으로 예상된다. 또한 추후 산업적 응용도 가능함으로 기능성화장품의 가능성을 제시하고 있다.

Alloxan에 의한 HIT-T15 세포 손상에 대한 쑥갓주정추출물의 세포보호효과 (The Protective Effects of Chrysanthemum cornarium L. var. spatiosum Extract on HIT-T15 Pancreatic β-Cells against Alloxan-induced Oxidative Stress)

  • The objective of the present study was to evaluate the potential antidiabetic and antioxidant effect of the ethanol extract from Chrysanthemum cornarium L. var. spatiosum(CSE) against alloxan-induced oxidative stress in pancreatic ${\beta}$-cells, HIT-T15. In this study, the antidiabetic effect of CSE was examined using the 3-(4,5-dimethylthiazolyl-2)-2,5-diphenyltetrazoliu bromide(MTT) cell proliferation assay, lactate dehydrogenase(LDH) release assay, $NAD^+$/NADH ratio and insulin secretion. To further investigate whether CSE is involved in the antioxidant activity of alloxan-damaged HIT-T15 cells, its antioxidant effect against alloxan-induced oxidative stress was measured in HIT-T15 cells by determining the levels of antioxidant enzymes including superoxide dismutase(SOD), glutathione S-transferase(GST), glutathione reductase(GR) and glutathione peroxidase(GPx). The results of this analysis showed that alloxan significantly decreased cell viability, increased LDH leakage, and lowered $NAD^+$/NADH ratio and insulin secretion in HIT-T15 cells. However, CSE significantly increased the viability of alloxan-treated cells and lowered LDH leakage. The intracellular NAD+/NADH ratio and insulin secretion were also significantly increased by 1.7-fold and 1.3-fold, respectively, after treatment with 100 ${\mu}g/m{\ell}$ CSE. The HIT-T15 cells treated with alloxan showed significant decreases in the activities of antioxidant enzymes, while CSE significantly elevated the levels of antioxidant enzymes. These findings suggest that CSE could have a protective effect against cytotoxicity and dysfunction of pancreatic cells in the presence of alloxan-induced oxidative stress.

Effect of γ-Aminobutyric Acid (GABA) Producing Bacteria on In vitro Rumen Fermentation, Biogenic Amine Production and Anti-oxidation Using Corn Meal as Substrate

  • The effects and significance of ${\gamma}$-amino butyric acid (GABA) producing bacteria (GPB) on in vitro rumen fermentation and reduction of biogenic amines (histamine, methylamine, ethylamine, and tyramine) using corn meal as a substrate were determined. Ruminal samples collected from ruminally fistulated Holstein cows served as inoculum and corn was used as substrate at 2% dry matter (DM). Different inclusion rates of GPB and GABA were evaluated. After incubation, addition of GPB had no significant effect on in vitro fermentation pH and total gas production, but significantly increased the ammonia nitrogen ($NH_3$-N) concentration and reduced the total biogenic amines production (p<0.05). Furthermore, antioxidation activity was improved as indicated by the significantly higher concentration of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) among treated samples when compared to the control (p<0.05). Additionally, 0.2% GPB was established as the optimum inclusion level. Taken together, these results suggest the potential of utilizing GPB as feed additives to improve growth performance in ruminants by reducing biogenic amines and increasing anti-oxidation.

The influence of different durations of aerobic exercise on fuel utilization, lactate level and antioxidant defense system in trained rats

  • This study investigated the influence of different durations of aerobic exercise on fuel utilization, lactate levels and antioxidative status in trained rats. Forty rats underwent physical training (T, n = 20) or non- training (NT, n = 20) for 6 weeks. For physical training, animals exercised on a treadmill for 30 min 5 days per week. At the end of week 6, the animals in each group were subdivided into BE, DE-0.5, DE-1 and DE-2, which were sacrificed at the end of week 6 without having performed exercise or after exercise on a treadmill for 0.5h, 1h and 2h, respectively, immediately before being sacrificed. The plasma glucose level in DE-2 of the NT group was significantly lower than in the other groups. Muscle and liver glycogen levels were significantly lower in DE-1 and DE-2, but there were no significant differences between DE-1 and DE-2 in the T group. Liver protein in DE-2 of the NT group was significantly lower. Muscle TG levels were decreased in DE-0.5 of the T group, while those of the NT group were decreased in DE-1. FFA levels were increased in DE-0.5 of the T group and in DE-1 of the NT group. Lactate levels were increased in DE-0.5 of the NT group, while they were increased in DE-1 of the T group. Catalase activity of the T group was lower in BE but higher in DE-0.5, DE-1 and DE-2. SOD activities were higher in trained rats, while the GSH/GSSG ratios were higher in BE, DE-0.5 and DE-1 in the T group, and there was no difference in that of DE-2. There were no differences in MDA levels in BE and DE-0.5, but they were significantly lower in DE-1 and DE-2 of the T group. Overall, the results of this study, suggest that training may improve exercise performance by facilitating the mobilization and oxidation of fat and conserving limited carbohydrate storage, and that it may delay the onset of fatigue and enhance the antioxidative defense system, but cannot support two hours of vigorous exercise.

DSS로 궤양성 대장염 유발된 동물모델에서 의이아(薏苡芽) 열수 추출물의 개선 효과 (Improvement Effect of Sprout of Coix lacryma-jobi var. mayuen Stapf Water Extract on DSS-Induced Ulcerative Colitis in Mice.)

  • Objectives : The objective of this study was to investigate the improvement effect of Sprout of Coix lacryma-jobi var. mayuen Stapf water extract (SC) on the dextran sulfate sodium (DSS)-induced ulcerative colitis mice. Methods : The antioxidant activity of SC was measured through total polyphenol and total flavonoid content in vitro. The experiment was conducted with seven-week-old male Balb/c mice. After 1 week adaptation, acute colitis was induced by oral administration of 5% DSS dissolved in drinking water, for 7 days. And normal mice received drinking water without DSS throughout the entire experimental period. For each experiment, the mice were divided into 4 groups and 24 colitis mice were arbitrarily allocated into 3 groups (n = 8/group); Normal group, Control group, SC 100 mg/kg treated group (SCL), SC 200 mg/kg treated group (SCH). Serum and colon tissues were collected after one weeks of drug administration. Results : ROS levels, ONOO- levels, AST, and ALT in serum were decreased in SC treated groups compared to the control group. Western blotting measurements of Nrf2, HO-1, SOD, catalase, GPx-1/2, IL-4, IL-10, and Bcl2 showed that the SC treated groups was increased compared to the Control group. Also, western blot measurements of NF-κBp65, p-IκBα, COX-2, iNOS, TNF-α, IL-1β, Bax, and Caspase-3 showed that the SC treated groups was reduced compared to the Control group. Conclusion : Taken together, these results suggest that SC treatment can attenuate the DSS-induced colitis though inhibiting NF-κB pathway and enhancing Nrf2 pathway. Therefore, SC was the potential to be used as a natural therapeutic drug.

혈부축어탕(血府逐瘀湯) 추출물(抽出物)이 고지방식이(高脂肪食餌)로 유발(誘潑)시킨 고지혈증(高脂血症) 흰쥐의 혈액(血液) 및 간(肝) 기능(機能)에 미치는 영향(影響) (Effects of Hyolbuchukeo-tang Extracts on Blood and liver of Hyperlipidemia Rats Induced by High Fat Diet)

  • Objectives: The purpose of this study was to investigate the effects of Hyolbuchukeo-tang extracts on the hyperlipidemia rats induced by high fat diet. Methods and Materials: In vitro; The extracts prepared for Hyolbuchukeo-tang by hot water extraction (HH), fermentation(HF) and UMPM extraction(HU) method. The extracts were examined for levels of polyphenol compounds, antioxidant activities, and inhibitory potencies for HMG-CoA reductase. In vivo; Sprague-Dawley male rats of weighing $150{\pm}5g$ were randomly divided into six groups ; normal control diet(NC), and high fat diet(HC), high fat diet with treated lovastatin of 10mg/kg(PC), high fat diet with Hyolbuchukeo-tang extracts; HH, HF and HU treated extracts of 300mg/kg, respectively. Also, we compared the effects of the extracts of HH, HF and HU on rats fed high fat diet for four weeks. Results: 1. The content of polyphenol compounds and electron donating abilities(EDAs) was the HF higher than HH and HU. The superoxide dismutase(SOD)-like activities were proportionate in consistency and they appeared highly from all extracts. The HMG-CoA reductase inhibition activities was highest activities in the HU. 2. The activities of serum GOT and GPT were significantly lower in the HH and HF groups. The level of serum triglyceride was significantly decreased in the HF group. HH and HU groups were significantly decreased in the atherogenic index(AI). The total cholesterol concentration in liver was significantly decreased in the HF group, and HU showed more significantly decreased in the triglyceride than of the lovastatin. Also, photomicrographs of liver tissue showed higher fat accumulation in the HC group than in the HH, HF and HU groups. Conclusions: These result suggest that the hyper-lipidemia caused by a high fat diet was effectively inhibited the administration of HF and HU.
