MiR-675 Promotes the Growth of Hepatocellular Carcinoma Cells Through Cdc25A Pathway

  • Background: MicroRNAs (miRNAs) have fundamental roles in tumorigenesis. MiR-675 is upregulated in hepatocellular carcinoma(HCC) cells. However, the roles of miR-675 in hepatocellular carcinogenesis are still not fully elucidated. In this study, we focus on investigating the effect and mechanism of miR-675 in proliferation of HCC cells. Materials and Methods: The cell proliferation was measured by MTT assays after transfection with miR-675 inhibitor and miR-675 mimics in HCC cells. The expression level of miR-675 was detected by real-time quantitative reverse transcription polymerase chain reaction. Protein expression of Cdc25A was measured by western blotting analysis. Results: In MTT assays, overexpression of miR-675 promoted the proliferation of HCC cells(P<0.05. at 48 hours, P<0.01. at 72 hours) compared with the miR-675mimics control group. Downexpression of miR-675 inhibited the proliferation of HCC cells(P<0.05. at 48 hours, P<0.01. at 72 hours) compared with the miR-675inhibitor control group. In western blotting analysis, the expression level of Cdc25A was significantly increased (p<0.05) after treatment with miR-675 mimics. The expression level of Cdc25A was significantly decreased (p<0.05) after treatment with miR-675 inhibitor. Conclusions: Our results indicate that miR-675 promotes the proliferation in human hepatocellular carcinoma cells by associating with Cdc25A signaling pathway.

Real-time modeling prediction for excavation behavior

  • Two real-time modeling prediction (RMP) schemes are presented in this paper for analyzing the behavior of deep excavations during construction. The first RMP scheme is developed from the traditional AR(p) model. The second is based on the simplified Elman-style recurrent neural networks. An on-line learning algorithm is introduced to describe the dynamic behavior of deep excavations. As a case study, in-situ measurements of an excavation were recorded and the measured data were used to verify the reliability of the two schemes. They proved to be both effective and convenient for predicting the behavior of deep excavations during construction. It is shown through the case study that the RMP scheme based on the neural network is more accurate than that based on the traditional AR(p) model.

An improved interval analysis method for uncertain structures

  • Based on the improved first order Taylor interval expansion, a new interval analysis method for the static or dynamic response of the structures with interval parameters is presented. In the improved first order Taylor interval expansion, the first order derivative terms of the function are also considered to be intervals. Combining the improved first order Taylor series expansion and the interval extension of function, the new interval analysis method is derived. The present method is implemented for a continuous beam and a frame structure. The numerical results show that the method is more accurate than the one based on the conventional first order Taylor expansion.

Identification and Characterization of Alternaria iridiaustralis Causing Leaf Spot on Iris ensata in China

  • In 2016, a severe leaf spot disease was found on Iris ensata Thumb. in Nanjing, China. The symptom was elliptical, fusiform, or irregularly necrotic lesion surrounded by a yellow halo, from which a small-spored Alternaria species was isolated. The fungus was identified as Alternaria iridiaustralis based on morphological characteristics. The pathogenicity tests revealed that the fungus was the causal pathogen of the disease. Phylogenic analyses using sequences of ITS, gpd, endoPG, and RPB2 genes confirmed the morphological identification. This study is the first report of A. iridiaustralis causing leaf spots on I. ensata in China.

Minimum BER Power Allocation for OFDM-based Cognitive Radio Networks

  • In this paper, the optimal power allocation algorithm that minimizes the aggregate bit error rate (BER) of the secondary user (SU) in a downlink orthogonal frequency division multiplexing (OFDM) based cognitive radio (CR) system, while subjecting to the interference power constraint and the transmit power constraint, is investigated under the assumption that the instantaneous channel state information (CSI) of the interference links between the secondary transmitter and the primary receiver, and between the primary transmitter and the secondary receiver is perfectly known. Besides, a suboptimal algorithm with less complexity is also proposed. In order to deal with more practical situations, we further assume that only the channel distribution information (CDI) of the interference links is available and propose heuristic power allocation algorithms based on bisection search method to minimize the aggregate BER under the interference outage constraint and the transmit power constraint. Simulation results are presented to verify the effectiveness of the proposed algorithms.

Asymmetrical Pulse-Width-Modulated Full-Bridge Secondary Dual Resonance DC-DC Converter

  • A full-bridge secondary dual-resonant DC-DC converter using the asymmetrical pulse-width modulated (APWM) strategy is proposed in this paper. The proposed converter achieves zero-voltage switching for the power switches and zero-current switching for the rectifier diodes in the whole load range without the help of any auxiliary circuit. Given the use of the APWM strategy, a circulating current that exists in a traditional phase-shift full-bridge converter is eliminated. The voltage stress of secondary rectifier diodes in the proposed converter is also clamped to the output voltage. Thus, the existing voltage oscillation of diodes in traditional PSFB converters is eliminated. This paper presents the circuit configuration of the proposed converter and analyzes its operating principle. Experimental results of a 1 kW 385 V/48 V prototype are presented to verify the analysis results of the proposed converter.

Roles for the lipid-signaling enzyme MitoPLD in mitochondrial dynamics, piRNA biogenesis, and spermatogenesis

  • Phospholipase D (PLD), a superfamily of signaling enzymes that most commonly generate the lipid second messenger Phosphatidic Acid (PA), is found in diverse organisms from bacteria to man and functions in multiple cellular pathways. A fascinating member of the family, MitoPLD, is anchored to the mitochondrial surface and has two reported roles. In the first role, MitoPLD-generated PA regulates mitochondrial shape through facilitating mitochondrial fusion. In the second role, MitoPLD performs a critical function in a pathway that creates a specialized form of RNAi required by developing spermatocytes to suppress transposon mobilization during meiosis. This spermatocyte-specific RNAi, known as piRNA, is generated in the nuage, an electron-dense accumulation of RNA templates and processing proteins that localize adjacent to mitochondria in a structure also called intermitochondrial cement. In this review, we summarize recent findings on these roles for MitoPLD functions, highlighting directions that need to be pursued to define the underlying mechanisms.

Trichodermamide A and Aspergillazine A, Two Cytotoxic Modified Dipeptides from a Marine-Derived Fungus Spicaria elegans

  • Two known modified dipeptides, trichodermamide A (1) and aspergillazine A (2), were isolated from an ethyl acetate extract of the metabolite of a marine-derived fungus Spicaria elegans, and were found to have a weak cytotoxic effect on three cancer cell lines P388, A-549, and HL-60 agreed. To our knowledge, this is the first report on the isolation of compounds 1 and 2 from the fungus Spicaria elegans and their cytotoxic effect.

A Compact Top-View Conformal Optical System Based on a Single Rotating Cylindrical Lens with Wide Field of Regard

  • A new compact conformal dome optical system was established, and the aberration characteristics of the dome were investigated using Zernike aberration theory. The aberrations induced by the dome at different fields of regard (FORs) from 30° to 100° were effectively balanced by a rotating cylindrical lens. This kind of optical system can be widely used in top-view optical searching devices. A design method was introduced and the optimization results analyzed in detail. The results showed that the Zernike aberrations produced by the conformal dome were decreased dramatically. Also, a complete conformal optical system was designed, to further illustrate the aberration correction effect of the rotating cylindrical lens. Using a cylindrical lens not only provided a large FOR, but also simplified enormously the structure of the conformal optical system.

  • In this paper a new control approach of detecting load voltage is proposed. It can directly detect the as side harmonic voltage of a voltage source load for the series APE to do dynamic compensation for harmonic voltages. In addition, the control methods of PWM inverter and its dc side voltage are discussed. According to these, a prototype of the series APF is manufactured and corresponding experiment studies are done.
