The purpose of the present study was to examine the effect of water extracts from Phellinus linteus on lipid composition and antioxidative system in rats fed high fat high cholesterol diet. Rats were divided into four experimental groups which are composed of normal diet group, high fat high cholesterol diet group, high fat high cholesterol diet with 50 mg/kg b.w water extracts from Phellinus linteus supplemented group (PA group), high fat high cholesterol diet with 100 mg/kg b.w water extracts from Phellinus linteus supplemented group (PB group). The serum TG and cholesterol contents of the water extracts from Phellinus linteus supplemented groups (PA and PB groups) were decreased compared to the high fat high cholesterol diet group. The serum HDL-cholesterol contents of the water extracts from Phellinus linteus supplemented groups were increased compared to the high fat high cholesterol diet group. The serum LDL-cholesterol and AI of the water extracts from Phellinus linteus supplemented groups were significantly decreased compared to the high fat high cholesterol diet group. Supplementation of water extracts from Phellinus linteus groups (PA and PB groups) resulted in increased activities of superoxide dismutase. Hepatic glutathione peroxidase (GSH-px) were increased compared to the high fat high cholesterol diet group. TBARS values in liver and plasma were reduced in water extracts from Phellinus linteus supplemented groups. These result suggest that supplementation of water extracts from Phellinus linteus may have a pronounced impact on lipid composition and antioxidative system in the rats fed high fat high cholesterol diet.
To examine the pharmacological activity of the polysaccharide extracted from phellinus linteus, we obtained the polysaccharide treated by the alkali and hot water, molecular weights of hot water was 10kD and 225 kD. Anticomplementary activity were highly observed in hot water fraction than alkali fraction. The tumor inhibition ratio of the polysaccharide extracted by hot water and alkali of Phellinus linteus against sarcoma 180 were 72.5% and 67.6%, respectively. These results suggest that protein bound polysaccharides extracted by a hot water and alkali of Phellinus linteus were similar to the pharmacological activities and chemical properties.
The biological and antioxidative activity of Phellinus linteus extracts from gradient ethanol concentrations were examined. The phenol contents of Phellinus linteus(28.36 mg/100 ml) was higher in the 80% ethanol extracts than other extracts. Electron donation ability on DPPH of 80% and 90% ethanol extracts(94.12% and 94.14% inhibition) from Phellinus linteus were the highest. The antioxidant activity against water soluble materials of Phellinus linteus ethanol extracts showed totally high inhibition rates above 80%, especially in 80% and 90% ethanol extracts, they showed each 94.12% inhibition and 94.14% inhibition. The inhibition against ABTS [2,2azinobis(3-ethylbenzothiazoline-6-sulfonic acid)] radical decolorization of 80% ethanol extracts was the highest as 96.2%. The antioxidant protection factor (PF) against lipid soluble materials was the highest in 80% ethanol extracts as 1.69 PF, and TBARS of 80% and 90% ethanol extracts were lower as $1.15{\times}100{\mu}M$ and $1.21{\times}100{\mu}M$ than control($1.95{\times}100{\mu}M$. Angiotensin converting enzyme and xanthine oxidase inhibitory activity of 80% ethanol extracts from Phellinus linteus was higher as 95.10%, 85.07% than other extracts. The results to analized of simple phenolic compounds of Phellinus linteus ethanol extrcts with HPLC showed that they were procatecuic acid, caffeic acid and coumaric acid.

This experiment was conducted to investigate the effects of molasses and $Phellinus$$linteus$ meal supplementation on the quality of Korean herbal medicine silage. Herbal medicine meal silages were produced by the addition of 0, 15, and 30% of $Phellinus$$linteus$ meal and 0, 1, and 2% of molasses ($3{\times}3$ factorial design) and stored for 40 days at room temperature. There were three replicates per treatment. Its quality such as chemical composition, pH, organic acids, the number of microorganisms and $in$$vitro$ dry matter disappearance were determined. As the $Phellinus$$linteus$ meal level increased, crude protein and crude fat contents increased significantly, but the acid detergent fiber (ADF) content decreased significantly. As the addition of $Phellinus$$linteus$ meal decreased, and molasses increased, pH tended to decrease, but the lactic acid content trended to increase. The acetic acid content was lower in a higher level (30%) of $Phellinus$$linteus$ meal, and higher in a higher level (2%) of molasses. As the $Phellinus$$linteus$ meal level increased, the butyric acid content decreased, and butyric acid of molasses treatment was higher than that of non-supplemented control. Molasses increased the number of Lactobacillus, but decreased the number of fungi in silage. A lower level of $Phellinus$$linteus$ meal increased the number of Lactobacillus, but decreased the number of fungi. $in$$vitro$ dry matter disappearance tended to increase by the addition of $Phellinus$$linteus$ meal and molasses. It is concluded that the addition of molasses and $Phellinus$$linteus$ to herbal medicine meal silage could enhance its nutritional quality. It is considered that proper levels of molasses and $Phellinus$$linteus$ meal in herbal medicine meal silage were 2 % and 15% respectively.
The skin of human is constantly being exposed to environmental irritants such as ultraviolet, smoke and chemicals. These irritants cause free radicals and reactive oxygen species which leave serious damages on the cells of skin. The water and ethanol extracts of Cambodian Phellinus linteus were investigated for the activities of anti-lipid peroxidation and anti-wrinkle effects to apply as a functional ingredient for cosmetic products. As the result of evaluation of liquid oxidation rate by add $Fe^{2+}$ and $Cu^{2+}$ to Cambodian Phellinus linteus extracts, Cambodian Phellinus linteus ethanol extracts were higher than Cambodian Phellinus linteus water extracts in the chealting ability of $Fe^{2+}$ and $Cu^{2+}$. The Cambodian Phellinus linteus ethanol extracts exhibited that anti-lipid peroxidation higher than butylated hydroxytoluene (BHT) at the concentration of 0.1 mg/ml, 0.5 mg/ml and 1 mg/ml. Cambodian Phellinus linteus water and ethanol extracts showed a higher inhibitory effect on $Fe^{2+}$-induced lipid peroxidation compared to $Cu^{2+}$-induced lipid peroxidation. In the case of anti-wrinkle effect, the elastase inhibition activity of Cambodian Phellinus linteus ethanol extracts was 50.7%, and it is higher than urosolic acid at the concentration of 0.01 mg/ml. Also, in collagenase inhibition activity, Cambodian Phellinus linteus water extract showed low effect, but Cambodian Phellinus linteus ethanol extract was about 50% at a 0.1 mg/ml. concentration. These results proved that Cambodian Phellinus linteus had anti-lipid peroxidation and anti-wrinkle effect. Therefore, Cambodian Phellinus linteus could be useful as an anti-wrinkle cosmetic ingredient.
Background : This study was to investigate existing studies on the mushroom Phellinus linteus for possible applications in treating hard to cure diseases and uses in herbal acupuncture as the above is known to have an anti-cancer effects and stimulating the immune system. Results : Based on the literary consideration, the following results were obtained : 1. Proteins and polysaccharides of the Class Basidiomycetes activate the host immune system for anti-cancer action without known side effects, thus it can be given for the prevention and treatment of cancer as a supplement. Phellinus linteus mushroom showed the most significant effects. 2. The Phellinus linteus mushroom is a fungi in the family Hymenochaetaceae and Phellinus, under the class Basidiomycetes. Phellinus linteus can be often found on the trunk of mulberry trees and other latifoliate trees. 3. The characteristics of Phellinus linteus are sweet, neutral in temperature, non-toxic, dissipate the stagnated blood, stop bleeding, invigorate the blood circulation and remove stasis, and nourish qi. It has been used for treating continuous menstrual discharge among the female patients and removing masses in the abdomen to name a few. 4. Phellinus linteus contains agaric acid, fatty acid, C23 and other numerous enzymes. 5. Phellinus linteus is known to have anti-cancer and anti-oxidant effects, stimulation of the immune system, as well as anti-inflammatory effects, inhibition of angiogenesis, and analgesic actions.
We have isolated an anti-complementary polysaccharide from the hot water extracts of Phellinus linteus mycelia. Anti-complementary polysaccharide, PL-5-IIIa, was purified by ultrafiltration, gel permeation chromatography using Sepharose CL-4B. GPC (Sepharose CL-4B) and its homogenicity was demonstrated by HPLC. Using gel permeation chromatography with standard dextrans, its molecular weight was determined as about 800,000 dalton. The purified PL-5-IIIa was identified as a protein bound polysaccharide comprising of 29.6% protein and 64.2% carbohydrate which was composed of fucose(15.8%), galactose(43.1%) and mannose(40.6%).
The fruiting bodies of Sanghwang mushrooms, Phellinus linteus HN1009K (PLH) and Phellinus linteus (Korea Sanghwang, PLK), and Phellinus baumii (Jangsu Sangwhang, PB), were extracted with 70% methanol. The methanolic extracts of different concentrations ($5-200{\mu}g/ml$) showed inhibitory effects of 20-95% on plated aggregation induced by collagen (2.5 ug/ml), ADP (10 uM), and thrombin (0.1 U/ml). The PLH, PLK, and PB extracts (200 ug/ml) reduced ATP release from ADP-activated platelets by 50-60%. These results suggest that the methanolic extracts from Sanghwang mushrooms inhibit platelet aggregation.
Background: The chemical characteristics of the exo-secretion from Phellinus linteus (referred to as exo-secretion) including the compositions of amino acids and monosaccharides were investigated. In addition, cytotoxicity of the exo-secretion on 5 tumor cell lines derived from human cancers and its antitumor activity against ascitic sarcoma-180 cells were examined. Methods: The antitumor activity of exo-secretion from Phellinus linteus was determined by measuring parameters including tumor weight, life span of mice, chemotatic activity of leukocytes, counts of immune cells, and activity of cytokines. Results: The exo-secretion from Phellinus linteus showed no direct cytotoxicity to the five tumor cell lines tested, but it had a strong antitumor activity against sarcoma-180 cells in ICR mice as measured by tumor weight and life span of mice. The exo-secretion stimulated the chemotaxis of leukocytes and production of immune cells and cytokines. Conclusion: These results suggest that the exo-secretion from Phellinus linteus do not act as a direct cytotoxic substance to cancer cells but as an immunomodulator.
