잠재적 사료첨가제로서 Pediococcus acidilactici SRCM102607의 생균제 특성 및 면역활성 효과 (Probiotic Properties and Immunomodulator Evaluation of the Potential Feed Additive Pediococcus acidilactici SRCM102607)

  • 본 연구에서는 가축의 면역증강용 생균제 개발을 위하여 Pediococcus acidilactici SRCM102607의 프로바이오틱스 특성 및 면역활성을 조사하였다. 전통발효식품으로부터 유산균을 분리 하였고, 분리 유산균을 대상으로 가축유해 미생물 5종에 대한 항균활성을 측정하였다. 우수한 결과를 나타내는 5종의 유산균을 1차 선별하였고, 이를 대상으로 생균제 소재 활용 가능성을 확인하기 위해 용혈성, 담즙산염 분해효소, 항산화 활성 분석을 실시하여 최종적으로 SRCM102607을 선별하였으며, 16S rRNA 유전자 염기서열 분석을 통해 Pediococcus acidilactici SRCM102607로 명명하였다. SRCM102607의 pH 2 조건에서 내산성은 1.54×105 CFU/ml의 생균수를 보였으며, 0.5% 이상의 oxgall이 포함된 조건에서도 105 CFU/ml 이상의 높은 생균수를 나타내었다. 또한, 선발균주의 산업적으로 활용할 수 있는 가능성을 검토하기 위해 항생제 내성 및 분해 효소능을 측정하였고 다양한 항생제에 대한 내성과 유해 효소를 생성하지 않음을 확인하였고, 최종적으로 면역 증강제로서의 활용 여부를 확인하기 위해 TNF-α 생성능(171.86±4.00 ng/ml)을 확인하였다. 본 연구를 기반으로 SRCM102607은 가축 생균제 소재로 활용 가능성과 면역활성이 뛰어난 유산균으로 생균제 산업에서의 잠재적 적용 가능성을 확인하였다.

  • The objective of this study was isolation of halotolerant lactic acid bacteria for fermentation of food wastes. 5 strains of lactic acid bacteria were isolated from fermented foods. Among isolated strains, the strain 5-2 was selected according to the growth characteristics in food wastes containing medium. The selected strain 5-2 was identified as Pediococcus acidilactici based on its biochemical characteristics.

백합(Meretrix meretrix)식해에서 분리한 Pediococcus pentosaceus SH-10의 생균제적 특성 (Probiotic Properties of Pediococcus pentosaceus SH-10 Isolated from the Hard Clam Meretrix meretrix Shikhae)

  • This study examined the suitability of characteristics of potential strains of probiotic bacteria. Among 25 lactic acid bacteria isolated from Korean traditional fermented food, the Hard Clam Meretrix meretrix Shikhae, the SH-10 strain, which exhibited superior resistance to low pH and bile salts, was selected as a potential probiotic bacteria. By examining carbohydrate utilization, morphological properties, and the 16S rRNA gene sequence, the SH-10 strain was identified as Pediococcus pentosaceus (hereafter, P. pentosaceus SH-10). P. pentosaceus SH-10 was resistant to amikacin, cefotetan, ciprofloxacin, gentamicin, kanamycin, nalidixic acid, streptomycin, and vancomycin. Tests of antimicrobial activities against pathogens such as Bacillus cereus, Listeria monocytogenes, Salmonella choleraesuis, and Staphylococcus aureus, indicated that P. pentosaceus SH-10 inhibited the growth of pathogenic bacteria. These results suggest that P. pentosaceus SH-10 can be developed as a probiotic bacteria.

  • 김치발효 중 Gram 양성세균 또는 유산균의 분리와 동정을 시도하였다. 종의 다양성은 분리배지와 온도에 영양을 받았고, 다양성은 온도가 낮아질수록 감소하는 경향이었다. MRS는 세균의 분리에 KM(김치재료로 만든 자연배지)은 종의 수를 파악 하는데 각각 적합하였다. 분리균의 동정은 Bergey's manual of Systematic Bacterio]ogy (1986)를 기초로 하여 작성한 이 분농생표(dichotomous Identification scheme)에 의하여 선행하였다. 각 온도(5, 15, $25^{\circ}C$)에서 동정된 Gram 양성세균은 Leuconostoc 5종, Streptococcus 4종, Pediococcus 3종, Bacillus 2종 그리고 Ltobacillms 18종이였다. 각 온도에서 출현 빈도가 높은 종은 $25^{\circ}C$에서 LactobaIlus ptantanmz, Streptococcµs faffinolactis, Leuconostoc maιnteroid, subsp mlsentιroides 이었고, $15^{\circ}C$에서 L. mesenteroides Lactobacillus fructosus, L. maen teroid,l.I subsp. mesent,roid, 이였고, $5^{\circ}C$ 에서 Leuconostoc sp.(65.2 빈도)에 의하여 이루어졌다. 그리고 각 온도에 따른 김치발효 중 지금까지 알려진 Pediococcus cerevisiae 와 Streptococcus faecalis는 분리되지 않았다.

Detection and Characterization of a Lytic Pediococcus Bacteriophage from the Fermenting Cucumber Brine

  • Of the twelve lytic bacteriophages recovered from five different fermenting cucumber tanks that were inoculated with Pediococcus sp. LA0281, a lytic phage, ${\phi}ps05$, was characterized in the present study. The plaques were mostly clear and round-shaped on the lawn of starter strain, indicating lytic phage. Overall appearance indicated that it belongs to the Siphoviridae family or Bradley's group B1, with a small isometric head and a flexible noncontractile tail with swollen base plate. The average size was found to be 51.2 nm in head diameter and 11.6 nm wide ${\times}$ 129.6 nm long for the tail. The single-step growth kinetics curve showed that the eclipse and the latent period were 29 min and 34 min, respectively, and an average burst size was calculated to be 12 particles per infective center. The optimum proliferating temperature ($35^{\circ}C$) was slightly lower than that of cell growth ($35\;to\;40^{\circ}C$). The structural proteins revealed by SDS-PAGE consisted of one main protein of 33 kDa and three minor proteins of 85, 58, and 52 kDa. The phage genome was a linear double-stranded DNA without cohesive ends. Based on the single and double digestion patterns obtained by EcoRI, HindIII, and SalI, the physical map was constructed. The overall size of the phage genome was estimated to be 24.1 kb. The present report describes the presence of a lytic phage active against a commercial starter culture Pediococcus sp. LA0281 in cucumber fermentation, and a preliminary study characterizes the phage on bacterial successions in the process of starter-added cucumber fermentation.

Comparison of Treatment Effect of Domestically Distributed Major Silage Inoculant

  • Silage inoculants, crucial in modern silage production, comprise beneficial microorganisms, primarily lactic acid bacteria (LAB), strategically applied to forage material during ensiling. This study aimed to compare the effectiveness of various inoculants produced by different companies. Five treatments were evaluated, including a control group: T1 (Lactobacillus plantarum), T2 (Lactobacillus plantarum + Pediococcus pentosaceus), T3 (Lactobacillus plantarum + Pediococcus pentosaceus + Lactobacillus buchneri), T4 (Lactobacillus plantarum + Lactobacillus acidophilus + Lactobacillus bulgaricus), and T5 (Lactobacillus plantarum + Pediococcus pentosaceus + Enterococcus faecium). Italian ryegrass was harvested at the heading stage and treated with these silage inoculants. Samples were collected over a 60-day ensiling period. Co-inoculation with L. plantarum and P. pentosaceus (T2) resulted in significantly higher CP compared to the control group co-inoculation exhibited with resulted in Lactobacillus plantarum and Pediococcus pentosaceus in the T2 treatment exhibited higher CP content of 106.35 g/kg dry matter (DM). The T3 treatment, which included heterofermentative bacterial strains such as Lactobacillus buchneri, exhibited an increase in acetic acid concentration (11.15 g/kg DM). In the T4 treatment group, which utilized a mixed culture of Lactobacillus acidophilus and Lactobacillus bulgaricus, the NH3-N/TN content was observed to be the lowest (20.52 g/kg DM). The T5 containing Enterococcus faecium had the highest RFV (123) after 60 days. Expanding upon these findings, the study underscores not only the beneficial effects of particular inoculant treatments on silage quality but also underscores the potential of customized inoculation strategies in maximizing nutrient retention and overall silage preservation.

Bacteriocin Produced by Pediococcus sp. in Kimchi and Its Characteristics

  • A bacteriocin-producing strain identified as Pediococcus acidilactici was isolated from kimchi. The bacteriocin was identified to belong to the pediocin family and exhibited bactericidal activity against most Gram-positive bacteria as well as some Gram-negative bacteria. The bacteriocin was stable up to $80^{\circ}C with wide pH ranges (5.0-10.0). The bactericidal activity remained unchanged after treatment with nonproteolytic enzymes such as nuclease and ${\alpha}$-amylase, however, it was destroyed after treatment with protease. The bacteriocin was effectively extracted by the pH-mediated adsorption-desorption method and purified effectively by semi-preparative RP-HPLC. The molecular weight of the bacteriocin was 4,622, as determined by electrospray mass spectrometry. The amino acid sequence consisted of 44 amino acid residues with four cysteines. The high solubility and pH stability of the isolated pediocin provide definite advantages over nisin and other bacteriocins in regards to its potential applications.

Probiotic Potential of Pediococcus acidilactici and Enterococcus faecium Isolated from Indigenous Yogurt and Raw Goat Milk

  • Probiotics are live microorganisms that, when administered in adequate amounts, confer health benefits to the host. This study was conducted for the isolation of potential lactic acid bacteria (LAB) with probiotic properties from goat milk and yogurt. Several tests were conducted in vitro using the standard procedures for evaluating the inhibitory spectra of LAB against pathogenic bacteria; tolerance to NaCl, bile salt, and phenol; hemolytic, milk coagulation, and bile salt hydrolase activities; gastrointestinal transit tolerance; adhesion properties; and antibiotic susceptibility. Among 40 LAB strains screened according to culture characteristics, five isolates exhibited antagonistic properties. Three were identified as Pediococcus acidilactici, and two were identified as Enterococcus faecium, exploiting 16S rRNA gene sequencing. All the isolates succeeded in the gastrointestinal transit tolerance assay and successively colonized mucosal epithelial cells. Based on the results of these in vitro assays, both P. acidilactici and E. faecium can be considered as potential probiotic candidates.

Morphological Changes Induced in Listeria monocytogenes V7 by a Bacteriocin Produced by Pediococcus acidilactici

  • Pediococcus acidilactici produces bacteriocin, which kills Listeria monocytogenes. The bactericidal mode of action of the bacteriocin against L. monocytogenes V7 was investigated by transmission electron microscopy. The bacteriocin was purified partially from the cell-free extract using Micro-Cel and cation-exchange chromatography, and the specific activity was increased 1,791 fold. The bacteriocin (6,400 AU/ml) was inoculated with L. monocytogenes V7 and incubated for 0.5h, 1h, 3h, and 6h. The bacteriocin was found to destroy most of the cell wall and released most of the inclusions in the cells after 6 h of incubation. These results suggest that the bactericidal effect of the bacteriocin was due to bacterial lysis.

Optimization of Carbon Sources to Improve Antioxidant Activity in Solid State Fermentation of Persimmon peel Using Lactic Acid Bacteria

  • The present study was conducted to develop persimmon peel, a by-product of dried persimmon manufacturing, as a feed additive via lactic acid bacteria fermentation. Pediococcus pentosaceus, Lactobacillus plantarum, and three strains of Leuconostoc mesenteroides were used as a starter culture in the solid state fermentation of persimmon peel, and antioxidant activity and total polyphenol content were assessed. Leuconostoc mesenteroides KCTC 3100 showed high antioxidant activity (p<0.05), whereas Pediococcus pentosaceus showed high total polyphenol content (p<0.05). These two strains were thus selected as starter culture strains. Glucose, sucrose and molasses were used as variables for optimization and a total 15 experimental runs were produced according to Box-Behnken design. Regarding significant effects of variables, molasses showed linear and square effects on antioxidant activity of persimmon peel fermentation (p<0.05). In conclusion, optimum concentrations of glucose, sucrose, and molasses were determined to be 4.2, 3.9 and 5.3 g/L, respectively, using a response surface model. Antioxidant activity was also improved 2.5 fold after optimization.
