Antioxidative Effects of Water-Soluble Chitinous Compounds on Oxidation of Low Density Lipoprotein in Macrophages (대식세포에서 지단백 산화에 대한 수용성 Chitinous Compounds의 항산화 효과에 대한 연구)

  • It has been proposed that oxidative modification of LDL (oxLDL) plays a significant role in the pathogenicity of atherogenesis. We tested the hypothesis that chitin and chitosan may function as antioxidants with respect to 0.1 mg cholesterol/ml LDL incubated with 5 $\mu$ M Cu$^2$$^{+}$alone or in the P338Dl mouse macrophage system using L-ascorbic acid as a standard classical antioxidant. The degree of oxLDL formation was ascertained by the relative electrophoretic mobility (rEM) in the combination of thiobarbituric acid reactive substances (TBARS) levels, and the cytotoxicity of oxLDL was detected by macrophage viability. The oxLDL uptake and foam cell formation of macrophages were measured by Oil Red O staining. Incubation with Cu$^2$$^{+}$and macrophages increased rEM of LDL and stimulated TBARS formation. Culture of macrophages with LDL in the presence 5 $\mu$ M Cu$^2$$^{+}$induced macrophage death. In cell-free system 200 $\mu$g/ml water-soluble chitosan and chitosan-oligosaccharide blocked oxLDL formation. Water-soluble chitosan and chitosan-oligosaccharide blocked oxLDL formation near-completely relative to L-ascorbic acid, whereas water-soluble chitin and chitin-oligosaccharide had no measurable antioxidant effect. In macrophage system water-soluble chitosan and chitosan-oligosaccharide blocked oxidation of LDL with a significant increase in cell viability, and decreased TBARS in medium. As for the inhibitory effect on macrophage foam cell formation, chitosan and its oligosaccharide, but not watersoluble chitin, revealed the effectiveness. The endothelial expression of lectin-like oxLDL receptor-1 (LOX-1) was tested by Western blot analysis, and chitosan, chitosan-oligosaccharide and chitin-oligosaccharide blocked LOX-1 expression. These results indicate that water-soluble chitosan and its oligosaccharide showed the inhibitory effect on Cu$^2$$^{+}$-induced LDL oxidation of macrophages, and chitosan, chitosan-oligosaccharide and chitin-oligosaccharide had blocking effect on oxLDL receptor expression in the human umbilical vein endothelial system. Thus, water-soluble chitosan and its oligosaccharides possess anti-atherogenic potentials possibly through the inhibition of macrophage LDL oxidation or endothelial oxLDL receptor expression depending on chemical types.l types.

Stem Rot on Ligularia fischeri Caused by Sclerotium rolfsii in Korea (Sclerotium rolfsii에 의한 곰취 흰비단병)

  • In June 2012 and 2013, a destructive stem rot symptoms of Ligularia fischeri occurred sporadically in Hoengseong-gun and Pyeongchang-gun Gangwon-do, Korea. The typical symptom included water-soaking on the main stem, rotting, wilting and blighting, which eventually leads to death of the plant. White mycelial mats were spread over lesions and brown sclerotia were formed on stems and near soil surface. The sclerotia were white to brown, spherical or irregular, 1-3 mm in size on potato dextrose agar (PDA), The optimum temperature range of hyphal growth was $25-30^{\circ}C$ and the hyphal diameter was $4-10{\mu}m$. The typical clamp connections were observed in the hyphae of the fungus grown on PDA. The resulting sequence of 695 bp was deposited in GenBank. A BLAST search revealed that sequences of the this isolates showed >99% identity with those of Sclerotium rolfsii. On the basis of the morphological characteristics and phylogenetic analyses of molecular markers ITS rDNA, the fungi were identified as S. rolfsii. A pathogenicity test was carried out to fulfill Koch's postulates. To our knowledge, this is the first report of S. rolfsii on Ligularia fischeri in Korea.

Characterization of Brugmansia mosaic virus Isolated from Brugmansia spp. in Korea (국내 브루그만시아에서 분리한 Brugmansia mosaic virus의 특성)

  • In May 2013, an angel's trumpet leaves showing mosaic and malformation symptoms were collected from Suwon city, Gyeonggi-do. An analysis of the collected sample by transmission electron microscopy observation showed filamentous rod particles of 720-800 nm in length. On the basis of the these observations, we performed PCR against three reported Potyviruses (Brugmansia mosaic virus, Colombian datura virus and Brugmansia suaveolens mottle virus), and the sample was positive for BruMV. Pathogenicity and host range test of BruMV was determined by mechanical inoculation. Solanaceae (tobacco, tomato and eggplant) and Amaranthaceae (ground cherry) appeared typical virus symptoms. To determine coat protein of this virus, we designed specific primer pairs, and performed PCR amplification, cloning, and sequencing. Phylogenetic analysis showed that BruMV-SW was most closely related to BruMV isolate SK. Comparison of the BruMV-SW coat protein nucleotide sequences showed 92% to 99% identities to the other BruMV isolates.

Identification of Pathogens Causing Grape Rot on 'Red Glove' Variety Imported from Chile (칠레산 수입포도 '레드글로브' 품종에 발생한 부패병 병원균의 동정)

  • Post-harvest rot of grape causes a severe economic loss and lower of the grape quality. It is also one of the important limiting factors for grape export. Grape rots and their casual agents on 'Red Globe' variety imported from Chile were identified. Grapes shown rotting symptom were collected from the storages near the import harbor. The 3 different rots were identified on the imported 'Red Globe'; melting decay, gray mold, and blue mold. A bacterium that isolated from a typical melting decay symptom was identified as Gluconobacter cerinus on basis of its nucleotide sequence of 16S rDNA and fatty acid profile. By inoculation on grape, it caused cracking and dissolution of epidermis of grape which were the characteristics of melting decay. Botrytis cinerea and Penicillium expansum were isolated from grapes showing gray mold and blue mold. The 2 fungal isolates were identified on basis of their morphological characteristics and nucleotide sequence of their beta-tubulin genes. They showed strong pathogenicity on 'Campbell Early' variety that is a major table grape in Korea.

Isolation and Characterization of Pepper mottle virus Infecting Tomato in Korea

  • A peculiar virus-like disease of tomato showing yellow mosaic and necrotic spots on leaves and necrosis on veins, petioles and stems was observed at the Tomato Experimental Station (TES), Buyeo, Chungcheongnamdo, Korea. The disease incidence at TES fields ranged from 21 to 35% infecting different tomato cultivars. For this reason, to identify the virus infecting tomato and to characterize the virus based on biology, serology, cytology and at molecular level. Here, leaf samples were randomly collected from different infected tomato cultivars at TES fields and greenhouses and tested by ELISA using Pepper mottle virus (PePMoV) and Tomato mosaic virus (ToMV) antisera. Infected saps were mechanically inoculated in different host plants to test for pathogenicity, symptomatology and host ranges. Infected tissues and ultrathin sections were examined by electron microscopy. Finally, putative coat protein and 3'-untranslated region (CP/3'-UTR) fragment was amplified and cloned for sequence determination and analyzed its genetic relationship to existing PepMoV and PVY sequences at the Genbank. Results showed 69% of the samples were positive with PepMoV, 13% with ToMV and 19 % were doubly infected with PepMoV and ToMV. Symptoms greatly varied from different host plants inoculated with tomato leaf sap infected with PepMoV alone and discussed in detailed in this paper. Electron microscopy from infected tissues showed filamentous particles of 720-750nm in length, a typical morphology and size of PepMoV. In addition, cylindrical inclusion bodies, pinwheels, scrolls and laminates with masses of fibrillar inclusions were also found in ultrathin sections. Alignment of the sequences of the CP/3'-UTR revealed >96% sequence identity with PepMoV and only <61% with PVY. Taken together, all these evidences presented clearly indicated that the causal agent infecting tomato at TES was PepMoV and we designated this PepMoV infecting tomato as Tom-sd2 strain in this study.

  • Rice blast is one of the most serious disease threatening stable production of rice. Breeding of resistant cultivars has been used as the most effective and useful method to controll rice blast caused by Magnaporthe oryzae. To collect rice blast isolates in fields and test their pathogenicity on new cultivars are important for establishment of new resistant cultivars breeding program of rice. Pathotypes of Korean rice blast isolates have been categorized to Korean differential race system developed in 1985. However, it is little known about genetic background of Korean differential cultivars, so that it is hard to understand for relationship between each pathogen and each host plant at genetic level. In this study, we suggested necessity of a new differential system by analyzing pathogenic responses between 24 monogenic rice lines and 200 Korean rice blast isolates. In addition, we determined the nine representative resistant genes based on the resistance responses of the monogenic lines to rice blast isolates, indexed resistant responses of the monogenic lines to ten representative rice blast isolates and selected 30 Korean representative rice blast isolates proper to Korean system. We think the newly developed differential race system can be broadly used to select resistant cultivars to rice blast in Korea.

Variation of Disease Severity by Mixed Inoculation of Compatible and Incompatible Races of Bacterial Blight in Rice (비친화적 및 친화적 레이스의 혼합접종에 따른 벼흰잎마름병 발병도의 변화)

  • Compatible and incompatible interactions of near-isogenic lines containing one of Xa1, Xa3, and Xa7 resistance genes with Japanese bacterial blight isolates (T7174, T7147, and T7133) were examined in order to determine the variation of bacterial blight resistance and the stability of resistance gene. IRBB 101 line having a Xal gene was compatible (host susceptible) with T7147 and T7133 isolates but incompatible (host resistant) with T7174 isolate at all the tested rice growth stages. IRBB 103 line having a Xa3 gene was susceptible or moderately resistant to the three isolates at seedling and maximum tillering stage but resistant at heading stage. IRBB 101 line having a Xa7 gene was semi-compatible with the three isolates at seedling stage but incompatible at the other growth stages. Overall there were clear differences between compatible and incompatible interactions of rice with Xanthomonas oryzae pv. oryzae races. In the mixed inoculations of compatible and incompatible isolates, the lesion length from near-isogenic lines decreased as the ratios of incompatible races increased. When the distinction between compatible and incompatible isolates was unclear, there was almost no variation of lesion length regardless of mixed ratios. The pathogenicity of the mixed races in the incompatible Interactions increased rather than the individual inoculation whereas the lesion length of compatible interactions was similar to that of the individual inoculation. These data indicate the incompatible races inhibit the virulence of a compatible race but compatible races increase the disease occurrence due to incompatible races. Furthermore, IRBB 107 line that showed resistance to all the isolates at all the tested growth stages was considered as a good parent f3r breeding of resistant variety.

Insecticidal Effect of an Entomopathogenic Fungus, Beauveria bassiana ANU1 to Spodoptera exigua and Plutella xylostella by Different Temperature and Humidity Conditions (파밤나방과 배추좀나방에 대한 곤충병원성 곰팡이 Beauveria bassiana ANU1의 온도와 습도조건에 따른 살충효과)

  • Entomopathogenic fungi have been studied to develop for biological control agents as an alternative to chemical control agents in insect pest management. Two Lepidopteran insects, Spodoptera exigua and Plutella xylostella, are serious insect pests infested various crops, but not effectively controlled by commercial chemical pesticides due to its high insecticide resistance. A fungal isolate was isolated from S. exigua larvae collected from green onion field in Andong, Korea. To identify the fungal isolate, 18srRNA sequence for internal transcribed spacer (ITS) and ${\beta}$-tubulin regions were sequenced. The ITS and ${\beta}$-tubulin sequence were highly matched to Beauveria bassiana and morphological characteristics also was fit to known B. bassiana. Finally, isolated fungus has identified as B. bassiana and named B. bassiana ANU1. The result of bioassay, median lethal concentrations were $2.7{\times}10^3$ and $0.9{\times}10^3conidia/ml$ and medial lethal times were 65.6 and 60.8 h to S. exigua and P. xylostella, respectively. B. bassiana ANU1 showed high pathogenicity to two insect pests from $20^{\circ}C$ to $30^{\circ}C$ at 50% relative humidity (RH) and more than 40% RH at $25^{\circ}C$ with $10^7conidia/ml$ of concentration.

Pink Mold Rot on Apple (Malus pumila var. dulcissima Koidz.) Caused by Trichothecium roseum (Pers.) Link ex Gray in Korea (Trichothecium roseum에 의한 사과 분홍빛열매썩음병 발생)

  • In 2012, a pink mold rot was observed on apple (Malus pumila var. dulcissima Koidz.) at the Wholesale Market for Agricultural Products, Jinju, Korea. The first symptom of pink mold rot on apple fruit is a water-soaked appearance of the affected tissue and surface fruit formed pink mold rot, then became brown and produced a mass of powdery pink conidia spores. Colony was fast growing colonies, pinksh, zonate in diurnal rhythm, powdery from conidia. Optimum temperature for mycelial growth was $25^{\circ}C$. Conidia were hyaline, smooth, two-celled, thick-walled conidia with truncate bases, ellipsoidal to pyriform, and characteristically held together zig-zag chains and $12-26{\times}8-12{\mu}m$ in size. Conidiophore was erect, colorless, unbranched, and $4-5{\mu}m$ wide. On the basis of mycological characteristics, pathogenicity test, and molecular identification with the ITS region, the causal fungus was identified as Trichothecium roseum (Pers.) Link ex Gray.

  • Korean entomopathogenic nematode strain were evaluated against white grubs, Popillia quadriguttata, Ectinohoplia rufipes and Phyllopertha diversa in laboratory. In addition we examined the efficacy of entomopatogenic nematode, Heterorhabditis sp. KCTC 0991BP strain against white grubs in Seaside Golf Club in Gimpo, Gyeonggii and persistence of entomopatogenic nematode in fairway of Anyang Benest Golf Club in Gunpo, Gyeonggii. In laboratory Heterorhabditis bacteriophora Hamyang strain showed 95% mortality against Popillia quadriguttata. H. bacteriophora Hamyang strain and Heterorhabditis sp. KCTC 0991BP induced 80% mortality against 3rd instar of Ectinohoplia rufipes. However, the letter showed only 60% mortality against Phyllopertha diversa. White grub density was reduced to 39% by the application of Heterorhabditis sp. KCTC 0991BP at the rate of $2.24{\times}10^9$ infective juveniles $ha^{-1}$ compared with control in Seaside Golf Club. Heterorhabditis sp. KCTC 0991BP persisted for two months at $5{\sim}10$ cm soil depth but not persisted at $0{\sim}5$ cm soil depth for the same periods when applied at the rate of $2.47{\times}10^9$ infective juveniles $ha^{-1}$.
