사료 내 분말 청국장이 돌돔, parrootfish, Oplegnathus fasciatus의 간 내 superoxide dismutase 활성에 미치는 영향 (Effects of Dietary Cheongkukjang on Liver Superoxide Dismutase Activity of Parrotfish Oplegnathus fasciatus)

  • A four-week feeding trial was conducted to investigate the effects of dietary soybean meal (SBM) and powdered Cheongkukjang (CKJ) on non-specific immune responses of parrotfish Oplegnathus fasciatus. Three isonitrogenous (42% crude protein) and isocaloric (17.1 MJ/kg) diets were formulated to replace fish meal by 0, 25% SBM or 25% CKJ (designated as FM, 25SBM and 25CKJ, respectively). Ninety fish (initial body weight 122 g) were randomly allotted into nine 150 L tanks. One of the three experimental diets was fed to triplicate groups of fish for 4 weeks. After the feeding trial, no differences were observed in growth performances and feed utilization among fish groups. Liver superoxide dismutase activity of the fish fed CKJ containing diet was significantly higher than that of the control groups. DPPH radical scavenging and $Fe^{2+}-chelating$ activities of the experimental diets containing SBM or powdered CKJ were significantly higher than that of the control diet. The results of the present study suggest that dietary inclusion of powdered 25CKJ significantly increased liver superoxide dismutase activity and did not affect the growth performances, feed utilization, morphological parameters, as well as hematological values of parrotfish.

Genetic Variations of Aspergillus fumigatus Clinical Isolates from Korea

  • Fungal infections by human pathogenic fungi are increasing globally in elderly, children and immune suppressed or deficient patients. Aspergillus fumigatus is one of the well-known pathogenic fungi and causes aspergilloses in human world widely. However, current identification and classification methods based on its phenotypic characteristics still have limitations. Therefore, currently, molecular biological tools using their DNA sequences are used for genotype identification and classification. In the present study, in order to analyze genetic variations of A. fumigatus clinical isolates, a total of six housekeeping genes were amplified by PCR using specific primer pairs and multi-locus sequence typing (MLST) assay. Results from phylogenetic tree analysis showed that most A. fumigatus strains (88.9%) from respiratory specimens were classified into cluster A and B, and approximately half of A. fumigatus strains (46%) from non-respiratory specimens were classified into cluster C and D. Although the sample size was limited, genetic characteristics of A. fumigatus clinical isolates according to their origins were very similar and well-correlated with other clinical data.

  • Seborrheic dermatitis is characterized by the appearance of red, flacking, greasy lesions in regions rich in sebaceous glands. There are various opinions about the relation between seborrheic dermatitis and dandruff. Considering all available data, we consider seborrheic dermatitis and dandruff to be differing severity manifestations of similar origin. The known etiologies of seborrheic dermatitis are the abnormality of lipid and hormnes, Malassezia species, immune response. We recently focus on the relation between seborrheic dermatitis and HIV positive and AIDS patients, seborrheic dermatitis and neurological disease. The treatments of seborrheic dermatitis include topical treatments, oral treatments. There are non-specific agents, antifungal medication, corticosteroids in the topical treatments. Narrow band UVB and oral medications are known as the prophylaxis of seborrheic dermatitis but we need further study. In traditional korean medicine, the literature studies and case reports have been published. But we don't know what herbal medicine has antifungal effect against Malassezia species. So we should discriminate herbal medicines which have antifungal effect against Malassezia specieses. Then we could treat seborreic dermatitis by traditional korean medicine effectively.

넙치에 대한 b-용혈성 연쇄구균 불활화백신의 안정성과 효능 (Stability and efficacy of formalin-killed Streptococcus iniae vaccine for olive flounder, Paralichthys olivaceus)

  • 본 연구는 Streptococcus iniae 포르말린 불활화 백신을 냉장 보관한 후 보존 기간별로 넙치에 접종하여 면역 효능 및 안정성을 조사하였다. 그 결과 6, 12, 15개월 보존한 후에도 면역 효능의 감소가 확인되지 않아 백신으로서의 높은 안정성을 확인할 수 있었다. 또한, 1회 접종구에 비해 2회 접종구에서 높은 항체가가 유도되었으며 방어력도 증가하는 것으로 나타났다. 특히, 1회 접종만으로도 낮은 항체가에도 불구하고 60% 이상의 방어 효과가 나타남에 따라 항체 이외의 세포성 면역이 방어력 형성에서 중요한 역할을 하는 것으로 추정되었다.

Early Growth Response-1 Plays a Non-redundant Role in the Differentiation of B Cells into Plasma Cells

  • Early growth response (Egr)-1 is a $Cys_2-His_2-type$ zincfinger transcription factor. It has been shown to induce survival and proliferation of immature and mature B cells, respectively, but its role in the differentiation of B cells into plasma cells remains unclear. To examine the effects of Egr-1 deficiency on the activation of B cells, naive B cells from $Egr1^{-/-}$mice and their wild-type (WT) littermates were activated to proliferate and differentiate, and then assayed by FACS. Proportions of cells undergoing proliferation and apoptosis did not differ between $Egr1^{-/-}$ and WT mice. However, $Egr1^{-/-}$ B cells gave rise to fewer plasma cells than WT B cells. Consistently, $Egr1^{-/-}$ mice produced significantly lower titer of antigen-specific IgG than their WT littermates upon immunization. Our results demonstrate that Egr-1 participates in the differentiation program of B cells into plasma cells, while it is dispensable for the proliferation and survival of mature B cells.

Supplemental effects of biofloc powder on growth performance, innate immunity, and disease resistance of Pacific white shrimp Litopenaeus vannamei

  • An 8-week feeding trial was conducted to study the effect of dietary supplementation of a biofloc powder on growth performance and non-specific immune response of Litopenaeus vannamei. Seven experimental diets were prepared with supplementation of graded levels of dried biofloc powder by 0, 0.5, 1.0, 2.0, 4.0, 6.0, and 8.0% (designated as Con, BF0.5, BF1, BF2, BF4, BF6, and BF8, respectively). Triplicate groups of shrimp ($1.01{\pm}0.01g$) were hand-fed with one of the diets four times a day. At the end of the feeding trial, significantly (P<0.05) higher growth performance and feed utilization were obtained in BF4 groups compared to those fed the Con diet. The innate immunity of shrimp was improved by the dietary supplementation of biofloc. Dietary inclusion of biofloc at the level of 4.0% significantly increased disease resistance of shrimp against Vibrio harveyi. The results indicate that biofloc might be used as a dietary supplement for growth performance, innate immunity and disease resistance of Pacific white shrimp.

  • The purpose of this research was to investigate the effects of unripened fruits and ripened fruits of Rubus coreanus Miquel on murine peritoneal macrophages. The 70% ethyl alcohol extracts (20 or 100 mg/kg) of unripened fruits (RCE-I) and of ripened fruits (RCE-II) were administered p.o. once a day for 7 days to mice. RCE-I and RCE-II decreased the phagocytic activity of murine peritoneal macrophages and the production of nitric oxide. Also, RCE-I and RCE-II increased the production of tumor necrosis factor- a from peritoneal macrophages. In general, the immuno-suppressive action of RCE-I on macrophages was more potent than those of RCE-II. These results suggest that the fruits of Rubus coreanus Miquel regulates the non-specific immune response via decrease of phagocytic activity and increase of production of tumor necrosis factor- a from murine peritoneal macrophages.

  • Medication-related osteonecrosis of the jaw (MRONJ) has recently associated to the increase in antiresorptive and anti-angiogenic drugs prescriptions in the treatment of oncologic and osteoporotic patients. The physiopathogenesis of MRONJ remains unclear and available treatments are unsatisfactory. Newer pharmacological treatments have shown good results, but are not curative and could have major side effects. At the same time as pharmacological treatments, mesenchymal stem cells (MSCs) have emerged as a promising therapeutic modality for tissue regeneration and repair. MSCs are multipotential non-hematopoietic progenitor cells capable to differentiating into multiple lineages of the mesenchyme. Bone marrow MSCs can differentiate into osteogenic cells and display immunological properties and secrete paracrine anti-inflammatory factors in damaged tissues. The immunomodulatory, reparative, and anti-inflammatory properties of bone marrow MSCs have been tested in a variety of animal models of MRONJ and applied in specific clinical settings. The aim of this review is to discuss critically the immunogenicity and immunomodulatory properties of MSCs, both in vitro and in vivo, the possible underlying mechanisms of their effects, and their potential clinical use as modulators of immune responses in MRONJ, and to identify clinical safety and recommendations for future research.

  • Currently 18 monoclonal antibodies were approved by FDA for inj ection into humans for therapeutic or diagnostic purpose. And 146 clinical trials are under way to evaluate the efficacy of monoclonal antibodies as anti-cancer agents, which comprise 9 % of clinical trials in cancer therapy field. When considering a lot of disappointment and worries existed in this field during the past 15 years, this boom could be called as resurrection. Antibodies have several merits over small molecule drug. First of all it is easier and faster in development, as proper immunization of the target proteins usually raises good antibody response. The side effects of antibodies are more likely to be checked out in immunohistomchemical staining of whole human tissues. Antibody has better pharmacokinetics, which means a longer half-life. And it is non-toxic as it is purely a "natural drug. Vast array of methods was developed to get the recombinant antibodies to be used as drug. The mice with human immunoglobulin genes were generated. Fully human antibodies can be developed in fast and easy way from these mice through immunization. These mice could make even human monoclonal antibodies against any human antigen like albumin. The concept of combinatorial library was also actively adopted for this purpose. Specific antibodies can be screened out from phage, mRNA, ribosomal library displaying recombinant antibodies like single chain Fvs or Fabs. Then the coding genes of these specific antibodies are obtained from the selected protein-gene units, and used for industrial scale production. Both $na\ddot{i}ve$ and immunized libraries are proved to be effective for this purpose. In post-map arena, antibodies are receiving another spotlight as molecular probes against numerous targets screened out from functional genomics or proteomics. Actually many of these antibodies used for this purpose are already human ones. Through alliance of these two actively growing research areas, antibody would play a central role in target discovery and drug development.

Clinical Analysis of Stages of HBV Infection in 100 Cases of Lymphoma

  • Objective: HBV infection may cause damage to the immune system and induce lymphomas as a result. Some scholars have indicated that HBsAg(+) reflecting HBV infection may have a relationship with lymphoma development. This study was designed to find out the specific stage of HBV infection which may be related to lymphoma. Methods: HBV serum markers, including HBsAg, HBsAb, HBeAg, HBeAb, HBcAb were tested among 100 lymphoma patients and 100 other patients who were diagnosed with non-lymphoma diseases in the First Hospital of Jilin University from 2010.1.1 to 2012.12.31. Three subgroups were established depending on different combinations of HBV serum markers. Subgroup 1 was HBsAg(+) representing the early stage of HBV infection. Subgroup 2 was HbsAb(+) representing convalescence and Subgroup 3 was "HbsAg and HbsAb negative combined with other positive markers" representing the intermediate stage of HBV infection. Chi square tests were used to compare the rates of three subgroups in lymphoma and control groups. Results: The rates of Subgroup were 13% and 5% respectively, an association between HBsAg and lymphoma being found (P<0.05). There was no difference between rate of Subgroup 2 of lymphoma group (15%) and that of control group (16%). In lymphoma group and control group, the rate of Subgroup 3 was different (12% vs 4%). This evidence was not specific to T cell lymphoma, B cell lymphoma or Hodgkin's lymphoma. Conclusions: Among serum markers of HBV, the combination of serum markers representing the early stage and intermediate stage of HBV infection have a relationship with lymphoma. Convalescence from HBV infection appears to have no relationship with lymphoma.
