Quantitative Changes of Polyphenolic Compounds in Mulberry (Morus alba L.) Leaves in Relation to Varieties, Harvest Period, and Heat Processing

  • Six polyphenolic compounds, such as chlorogenic acid (CA), rutin (RT), isoquercitrin (IQT), quercetin-3-O-(6-O-malonyl)-${\beta}$-D-glucoside (QMG), astragalin (AG), kaempferol-3-O-(6-O-malonyl)-${\beta}$-D-glucoside (KMG), were isolated from mulberry leaves by a series of isolation procedures, such as Diaion HP-20, silica-gel, Sephadex LH-20, and ODS-A column chromatographies. The chemical structures of the phenolic compounds were identified by UV and NMR spectral analyses. Levels of polyphenols in mulberry leaves from six different mulberry cultivars ranged from 1,042.16 to 1,871.97 mg% per dry weight; Guksang cultivar showed the highest levels of polyphenols, whereas Gaeryangdaehwa contained the least polyphenol contents. Generally, levels of polyphenols in mulberry leaves decreased with increasing harvest time, except for Yoolmok, but increased with heat processing time, except QMG and KMG. These results suggest that the heat processed mulberry leaves of Guksang cultivar harvested in early May can be potentially useful sources for production of high quality mulberry leaf teas.

Synergic Effect of Quercetin and Astragalin from Mulberry Leaves on Ani-inflammation (상엽 유래 퀘세틴과 아스트라갈린의 항염증에 대한 상승효과)

  • The leaf of mulberry (Morus alba L) has long been used in Oriental medicine for the prevention or treatment of several diseases. However, little is known about the inhibitory effects of a single compound isolated from the mulberry leaves on inflammatory response. We are isolate a single compound of quercetin (3,3',4',5,7-pentahydroxyflavone) and astragalin (kaempferol-3-O-glucopyranoside) from the mulberry leaves, and then investigate the anti-inflammatory effects of quercetin, astragalin or quercetin plus astragalin in lipopolysaccharide (LPS)-stimulated murine peritoneal macrophages. Each compound suppressed the production of inflammatory mediators (NO, $PGE_2$ and IL-6) in LPS-stimulated murine peritoneal macrophages in a dose-dependent manner. Especially, the cotreatment of quercetin (2.5 ${\mu}M$) and astragalin (2.5 ${\mu}M$) markedly suppressed the production and the expression of inflammatory mediators. These suppressive effects were synergistically increased by their combination. These results suggest that the combination of quercetin and astragalin from the mulberry leaves may be useful for therapeutic drugs against inflammatory immune diseases, probably by suppressing the production of inflammatory mediators.

Quality Characteristics of Oddi-Pyun prepared with various levels of mulberry fruit extract (오디 추출액 첨가비율에 따른 오디편의 품질특성에 관한 연구)

  • Oddi is a mulberry (Morus alba L) fruit which has antiinflammatory and antioxidative effects. This study was carried out to investigate the quality characteristics of Oddi-Pyun which was manufactured with various addition levels (0, 0.5, 1, 2, 4%) of Oddi extract according to the traditional Korean Kwaypun (a kind of jelly) methodology. Oddi-Pyun was made with various levels of Oddi extract, mungbean starch (9%) and sugar (30%). To establish the additional amount of mulberry fruit extract, sensory evaluation and physical test were carried out. From the results of sensory evaluation, the 2% of Oddi-Pyun was judged as the best in terms of color, flavor, hardness and elasticity. According to texture characteristics (hardness, adhesiveness, springiness, gumminess and chewiness), 1.0% and 2.0% of Oddi-Pyun were judged as the best. The contents of Ca (69.50, 74.75, 84.25, 100.60, 183.55ppm), Mg(27.37, 29.38, 34.20, 46.73, 97.45ppm) and Fe (320.23, 572.45, 680.50, 725.95, 906.50ppb) increased with increasing Oddi extract. Therefore, the optimal added amount of extract for the manufacture of Oddi-Pyun was proposed as 2% to the total weight. It was concluded that this Oddi-Pyun would be helpful to improve the health status of rheumatics and patients with similar diseases.

Anti-oxidative effects of Mori Folium, Mori Ramulus and Mori Cortex Radidus (상엽(桑葉), 상지(桑枝) 및 상백피(桑白皮)의 항산화 활성 연구)

  • Objective : This study was designed to investigate the anti-oxidative potentials to wide the usage of Morus alba L. derived from medicinal herbs on cosmoceutical fields. Methods : Anti-oxidative potentials were investigated by using several kinds of assays including electron donating ability, and the total contents of phenolic compounds were also measured. In some cases TLC (Thin Layer Chromatography) method were used to identify the patterns of phenolics in herbal extracts. Results : Aurones which are sub-types of flavonoids were observed in methanol extract of Mori Cortex Radidus, and total phenolic contents in Mori Cortex Radidus were more than in methanol extracts of Mori Folium and Mori Ramulus. Various kinds of phenolic compounds were observed in chloroform fraction of Mori Cortex Radidus. Conclusion : According to these results, the chloroform fraction of Mori Cortex Radidus could be the most possible resource that has potentials on anti-oxidant agents on cosmoceutical fields.

  • The present study was undertaken to investigate effect of water extracts of the mulberry leaves (MWE) on arterial blood pressure and respiration in cats. And also studied were depressor responses to intravenously administered MWE in the animals pretreated separately with atropine(2.5 mg/kg), propranolol(2 mg/kg), dibenamine (15 mg/kg), and hexamethonium (1.5 mg/kg) in order to find out the mechanism of depressor activity of MWE. The results obtained were as follows; 1) Following intravenous administration of 0.25 ml/kg and 0.5 ml/kg of MWE into the cat the maximum depressor responses observed were $60.2{\pm}2.3\;mmHg$ and $72.3{\pm}1.7\;mmHg$ respectively. 2) Since depressor responses to intravenously administered MWE were partially inhibited by hexamethonium and markedly by atropine, it is strongly suggested that depressor activity of MWE mainly results from its vagal effects. 3) After administration of MWE respiratory rate was invariably increased following a short period of apnea.

Antimicrobial Activity against Helicobacter pylori from the Korean Rice Liquor Fermented with Mulberry leaves(Morus Alba L.) (뽕잎발효주가 Helicobacter pylori에 대해 미치는 항균활성)

  • To develop a new traditional rice liquor by using Mulberry leaves, the optimal condition of fermentation was investigated by addition of different concentrations (1%, 3%, and 5%) of Mulberry leaves powder in the raw rice mash. The efficiency of ethanol production of all conditioned groups were not inhibited by addition Mulberry leaves. The acceptability and antimicrobial activity of the Mulberry leaves liquor with different concentrations of Mulberry leaves powder were tested. The 3% Mulberry leaves liquor showed the best acceptability in the sensory evaluation test and color test. The antimicrobial activity against Helicobacter pylori were increased according to added amount of Mulberry leaves. It was supposed that the antimicrobial activity were affected by phenolic compound extracted from Mulberry leaves with ethanol produced during the fermentation period. The results suggested that the Mulberry leaves can be useful source to develop a new traditional rice liquor.

  • The concentrations of ${\gamma}$-aminobutyric acid in some Korean plants and mushrooms were investigated. ${\gamma}$-amino-butyric acid concentrations in Morus alba leaves, Cudrania tricuspidate leaves and fruits were 134.03, 104.13 and 120.99 mg/100g dry matter basis, respectively, ${\gamma}$-aminobutyric acid concentrations in Pleurotus ostreatus, Flammulina velutipes and Auricularia auricula were 93.44, 72.17 and 66.48 mg/100g dry matter basis. respectivley.

Effect of Mulberry Leaf(Morus alba Linne) Powder Addition on Quality of Yukwa (유과 제조 시 뽕잎 분말 첨가가 품질에 미치는 영향)

  • The principal objective of this study was to evaluate the quality of Yukwa prepared with mulberry leaf (Morus alba Linne) powder. The moisture, crude protein, crude lipid, crude ash, dietary fiber, and rutin contents of used mulberry leaf powder were 6.65%, 19.50%, 3.26%, 18.24, 38.91 and 0.54%, respectively. The tested concentrations of mulberry leaf powder were 0, 3, 5, and 7%. The Yukwa were stored at $45^{\circ}C$ for 40 days. Yukwa dough prepared with mulberry leaf powder had a lower pH compared to the control group. As more mulberry leaf powder was added, the spread ratio, moisture content, and volume increased. As more mulberry leaf powder was added, L-value decreased while a and b values increased with increasing mulberry leaf powder content. The hardness of Yukwa made with 7% mulberry leaf powder was the highest among the samples. In the sensory tests, Yukwa with 5% mulberry leaf powder received the highest scores. The acid value and peroxide values were lower in Yukwa prepared with mulberry leaf powder than control. Yukwa prepared with mulberry leaf powder strongly inhibited oxidation of lipids. The thiobarbituric values were lower in Yukwa prepared with 5% mulberry leaf powder than in Yukwa prepared with 3% and 7% mulberry leaf powders and control Yukwa. These results suggest that mulberry leaf powder can be applied to Yukwa to achieve high quality and functionality.

Identification of Genetic Relationships Among Morus alba Genotypes Based on RAPD and ISSR Fingerprinting

  • Mulberries have importance in the sericulture industry as food for Bombyx mori, silkworm reared for its silk. Korean Morus alba have many cultivars and, for the protection of these cultivars and for utilization in plant-breeding programs, genetic information and the diversity among cultivars are essential. This study with 14 mulberry genotypes was undertaken using RAPD and ISSR fingerprinting to discover the genetic divergences between cultivars. Polymorphism rate among the cultivars produced by RAPD primer was found to be 64.48% and 66.29% relative to ISSR primer. The genetic relationships among the cultivars were identified using a dendrogram constructed with the UPGMA clustering method. Nei's method was used to calculate the genetic dissimilarity coefficients between each pair of genotypes, and the highest dissimilarity coefficient of 0.246 was exhibited between Suwon and Hwanggum cultivars. To determine the efficiency of each primer, a polymorphic index was calculated, and the robustness of the dendrogram was checked using cophenetic correlation coefficient. The results of this study can be utilized for the improvement of mulberry varieties in plant-breeding programs.

  • With extracts from Cheongmoknosang, anti-inflammatory effect was examined in LPS-stimulated Raw 264.7 cells. LPS (10 ng/ml) treatment increased the production of inflammatory cytokines, $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ but the ethanol extracts from Cheongmoknosang slightly decreased the production of $TNF-{\alpha}$ and also reduced the expression of iNOS and the production of COX-2. It seems that anti-inflammatory effects of ethanol extracts from Cheongmoknosang is partly due to the inhibition of iNOS and COX-2 expression by inhibiting nuclear translocation of $NF-{\kappa}B$ and AP-l in Raw 264.7 cells.
