Breeding of Cold Resistant Mulberry Cultivar “Hansungppong” (Morus alba L.) (내동다수성 뽕품종 “寒盛뽕” 육성)

  • New mulberry cultivar, Hansungppong, was bred among selected lines from F1 seedlings obtained by crossing the female, Yongcheonppong, and the male, Kugsang 21. Local adaptability test has been carried out at two places (Suwon and Chunchun) for four years since 1995. Leaf quality was tested by silkworm rearing for two silkworm rearing seasons (spring and autumn) at Suwon in 1998. Hansungppong was recognized to be superior in productivity and cold resistance to control cultivar, Yongcheonppong. The new cultivar, Hansungppong, has following agronomic characteristics as compared with Yongcheonppong, which was strongest leading cultivar against cold injury in Korea. It is belonging to Morus alba L. with the ovate leaf shape, sprouting in spring nearly the same to the Yongcheonppong (Morus alba L.), high leaf yielding of 10% compared to Yongcheonppong (Morus alba L.) and the high degree of cold resistance. It is adaptable to everywhere in Korea. Hansungppong is recommended to rear silkworms not only in spring but also in autumn besides for young silkworms.

Fertility and Mulberry Fruit Characteristics of Three Korean Indigenous Mulberry Species (한반도에서 자생하는 뽕나무 3종이 자연교잡된 때의 稔性과 오디의 과실특성)

  • Three indigenous mulberry species of Morus tiliaefolia Makino, Morus mongolica C. K. Shneider and Morus bombycis Koidz. were naturally crossed and their fertilities and fruit characteristics were investigated during the years of 1996 and 1997. M. tiliaefolia Makino and M. mongolica C.K. Schneider showed low fertility, but M. bombycis Koidz. was medium in fertility. The fruits of M. tiliaefolia Makino were red-purple and midium in size and high sugar content, but those of M. mongolica C.K. Schneider were light red and small but extremely high sugar content. The total acidity of fruits was 0.73, 0.50 and 0.36-0.94% in M. tiliaefolia Makino, M. mongolica C.K. Schneider and M. bombycis Koidz. respectively. Regarding ripenening stage, M. mongolica C.K. Schneider was early and M. tiliaefolia Makino was medium, while M. bombycis Koidz was early or medium.

Analysis of ITS Nucleotide Sequences in Ribosomal DNA of Morus Species (뽕나무속 식물의 ITS 영역 염기서열 분석)

  • Nucleotide sequence in internal transcribed spacer (ITS) regions of ribosomal DNA among mulberry varieties (Morus species) were analyzed in order to identify the possibility of classification for the species. The variations in the ITS regions were compared among 9 mulberry varieties and one variety of Cudrania species as an outgroup. ITS 1 region of the varieties ranging from 219 to 220 bp in length was 49-50 bp shorter than ITS 2 region. Of 510 sites in the ITS 1 and 2 regions, 148 sites were potentially variable, of which 52% and 48% sites were distributed in ITS 1 and ITS 2 regions, respectively. By pairwise comparisons on the nucleotide sequences in the ITS 1 and 2 regions among 9 mulberry varieties, they were classified into 5 groups. Divergence values of the sequences, however, were considerably low ranging from 0 to 1.3%. Especially, there was no divergence among Backasipmunja, Chungilppong and Milsungpong and Jungyasang, Ssarigol II and Yulbon, respectively.

Properties of the Proteolytic Enzymes from Mulberry Tree Barks(Morus alba Linne) (상백피에서 추출한 단백질 분해효소의 특성)

  • Water extract of mulberry tree barks(Morus alba Linne) was studied for its proteolytic activity. Protein content of the extract was 1.12mg/ml and its specific activity was 5.14U/ml. The enzyme was active on various proteins : the relative acitities were 100 for casein, 63 for albumin, 58 for collagen, 45 for hemoglobin and 36 gelatin, respectively. There suggested that the ability of the enzyme to hydrolyze meat was relatively high since those are major meat proteins. Optimum pH and temperature for proteolytic activity were : pH 6.0 and 6$0^{\circ}C$. And the enzyme was stable at the pH range of 6.0 to 7.0 and temperature between 50 and 8$0^{\circ}C$. Apparent proteolytic activities could support some scientific grounds of traditional application of mulberry tree barks to home cooking for meat tenderization.

The Inhibitory Activity on Monoamine Oxidase of the Fruit of Morus alba (상심자의 모노아민산화효소 저해활성)

  • We examined the inhibitory activities against monoamine oxidase (MAO) of Morus alba in vitro and in vivo methods. Methanolic extract of M. alba showed significantly inhibitory activities on MAO-A and MAO-B that were prepared from rat brain and liver in vitro. The inhibitory activities were measured by serotonin and benzylamine as substrates, respectively. MAO-A and MAO-B activities were potently inhibited by ethylacetate extracts of M. alba in vitro tests. Those activities in vivo tests have different tendency each other. MAO-A activity was increased by the oral administration of methanolic extract of M. Alba, while, MAO-B activity was decreased. Consequently, we can suggest that M. alba may have the effects on the inhibitory activities against MAO both in vitro and in vivo.

  • The root-bark of the mulberry tree has been used as a medicine with antitussive, antiphlogistic, diuretic etc. in Korea. Among many kinds of mulberry trees, Morus Lhou has been widely cultivated in our country. In this experiment, three substances were obtained from the root-bark of Morus Lhou removed its cork layer by column chromatography of methanol extract. One of the substances was supposed as ${\alpha}$-amyrin acetate, another was alyceride, the other was confirmed as ${\beta}$-sistosterol by chemical and physical methods.

Inhibitory Effects of Steppogenin and Oxyresveratrol from Morus alba L. against Yeast ${\alpha}$-Glucosidase (뽕나무에서 분리한 Steppogenin과 Oxyresveratrol의 효모 ${\alpha}$-Glucosidase의 억제효과)

  • [ ${\alpha}$ ]Glucosidase inhibitor is a target in the treatment of type II diabetes through the mainly inhibition of glucose levels after meals. In this study, we purified steppogenin and oxyresveratrol from the stem of Morus alba L. and examined their inhibitory activity against yeast ${\alpha}$-glucosidase. Steppogenin and oxyresveratrol were inhibited yeast ${\alpha}$-glucosidase in a dose dependent manner. The $IC_{50}$ activities (50% inhibition) were 34.4 and 9.3 ${\mu}M$, respectively. The kinetic inhibition of steppogenin showed noncompetitive inhibition ($K_m:1.1{\times}10^{-3}M$; $K_i:1{\times}10^{-5}M$), meanwhile oxyresveratrol showed competitive inhibition ($K_m:4.3{\times}10^{-3}M$; $K_i:3.4{\times}10^{-6}M$) against yeast ${\alpha}$-glucosidase. These results indicate that steppogenin and oxyresveratrol are noncompetitive and competitive inhibitors, respectively, against yeast ${\alpha}$-glucosidase.

  • The MeOH extract of the twig of Morus alba L. (Moraceae) inhibited strong lipid peroxidation activity. In order to find out active principle from the plant, acivity-guided fractionation was performed and five antioxidant compounds were isolated. Their chemical structures were identified as 6-geranylapigenin (1), 6-geranylnorartocarpetin (2), resveratrol (3), oxyresveratrol (4) and quercetin (5) by physicochemical and spectrometric methods. Compounds 1-5 significantly inhibited lipid peroxidation in rat brain homogenate $(IC_{50}\;values\;of\;3.37,\;3.74,\;0.23,\;0.29\;and\;0.06\; {\mu}M,\;respectively)$.

  • This study aimed to compare the chemical composition of Morus nigra leaves extracts, obtained by maceration, accelerated solvent (ASE) and supercritical fluid extraction (SFE) under different extraction conditions. With regards to chemical composition, mainly phenolic acids and flavonoids were identified. HPLC-ESI-QTOF-MS allowed the identification of 13 new compounds reported in M. nigra leaves for the first time. ASE as a fast, green and innovative approach, seems to be the best choice for extracting compounds of different polarities within the shortest extraction time. The present study also highlights the potential application of M. nigra extracts as constituents of new added-value formulations.

  • The crossabilities among 3 wild mulberry species and 3 cultivars growing in the Korean Peninsula had been studied. The result of the crossabilities of Moms tiliaefolia with Moms bombycis and 3 cultivars were possible, but with Morus mongolica was impossible. Those of Moms mongolica with Moms bombycis, Morus tiliaefolia and 2 cultivars except Daeryukppong were possible too. And those of Morus bombycis with Moms titiaefolia, Moms mongolica were impossible, but with 3 cultivars were possible moderately.
