Optimization of Submerged Culture Conditions for Exo-biopolymer Production by Paecilomyces japonica

  • Optimization of submerged culture conditions for the production of exo-biopolymer from Paecilomyces japonica was studied. Maltose, yeast extract and potassium phosphate were the most suitable sources of carbon, nitrogen, and inorganic salt, respectively, for both production of the exo-biopolymer and mycelial growth. The optimal culture conditions in flask culture were pH 5.0, $25^{\circ}C$ and 150 rpm in a meidum containing of 30 g maltose, 6 g yeast extract, 2 g polypeptone, 0.5 g $K_2HPO_4$, 0.2 g $KH_2PO_4$, 0.2 g $MnSo_4\;{\cdot}\;5H_2O$, 0.2 g $MgSO_4\;{\cdot}\;7H_2O$ in 1-L distilled water. Exo-biopolymer production and mycelial growth in the suggested medium were significantly increased in a 2.5-L jar fermentor, where the maximum biopolymer concentration was 8 g/1.

길항미생물 Bacillus lentimorbus G-74의 배양을 위한 여뀌즙액 배지개발

  • Persicaria hydropiper juice was applied to develop a novel meidum for the mass cultivation of an antifungal bacterium, Bacillus lentimorbus G-74. In this research, a optimized culture condition of B. lentimorbus G-74 were detenninecl. Persicaria hyclropiper juice was contained 1.76% nitrogen, 0.15% phosphorus, 18,230 ppm potassium, 13,753 ppm calcium and 4,844 ppm magnesium. The optimal growth was obtained when the juice was a 30%(v/v) level. The optimum temperature and pH for the bacterial growth were $35^{\circ}C$ and 5.0, respectively.

  • A wireless LAN medical information transmission system is useful for patients who need mobility in a local area environment. This paper proposes a method using WLAN(wireless local area network) and implements a stand-alone system with MAC(meidum access control) layer protocol referenced IEEE 802.11 draft standard. The system consists of a 8bit-microprocessor which handles media access control protocol and a WL100(GEC Plessey) chip which takes care of phsical layer specific routines and uses a RF module DE6003(GEC Plessey). The major features of the implemented system are the CSMA/CA protocol used a consecutive DATA-ACK trasmission method which yields more effective bandwidth allocation for asyncronous traffic transmission and the modified PCF protocol for time-bounded traffic transmission, which operates in ad-hoc network topology apart from IEEE 802.11 draft standard confirm PCF mode operate in infrastruture topology.

Study on Development of In Vitro Culture Medium for Rabbit Embryos (토끼 수정란 체외 배양액의 개발에 관한 연구)

  • This experiment was carried out to improve in vitro development of rabbit one-cell embryos to the blastocyst stage. One-cell rabbit embryos were collected at 19\ulcorner20hr after superovulation induction and incubated at 39\ulcorner in 5% CO2 for 72hr. In order to find optimum conditions in medium that affects the rabbit embryo's development in vitro, RDH medium which mixed with RPMI1640, DMEM and Ham's F10 was compared with the previously reported mediums (Ham's F10 and RD) for embryo development and cell numbers. Three additives (BSA, taurine and glucose) were tested for the development of rabbit one-cell embryos in vitro. When the embryos were cultured in RDH medium, their development was markedly promoted as compared with Ham's F-10 or RD alone. Glucose exhibited no significant effects on embryo development and cell numbers. BSA a, pp.ared to promote transition from morula to blastocyst stage and taurine increased cell numbers of cultured embryos markedly regardless of medium. BSA and taurine together in RDH medium showed the additive effects on embryos development and cell number.

Studies on the Effects of Co-Culture with Cumulus Cells, Oviduct Epithelial Cells and Uterine Endometrial Cells on in-vitro Fertilization and Cleavage Rate of Porcine Oocytes (난구, 난관 상피세포 및 자궁 내막세포와의 공동배양이 돼지 난포란의 체외수정 및 분할율에 미치는 영향에 관한 연구)

  • The studies were carried out to investigate the effects of co-culture with cumulus cell, oviduct epithelial cells and uterine endometrial cells on the in-vitro fertilization and cleavage rate of porcine follicular oocytes. The ovaries were obtained from slaughtered swine. The follicular oocytes surrounded with cumulus cells were recovered by aspirating follicular fluids from the visible follicles of diameter 3~5 mm. The follicular oocytes were cultured in TCM-199 medium containing hormones and 10% FCS for 24~48 hrs in a incubator with 5% CO2 in air at 38.5$^{\circ}C$ and then matured oocytes were again cultured for 12~18 hrs with motile capacitated sperm by preincubation. The results obtained in these experiments were summarized as follows : 1. The in-vitro maturation and fertilization rate of porcine oocytes co-cultured with cumulus cells in TCM-199 meidum were 64.6%~74.5% and 37.5%~55.3%, respectively. And in-vitro fertilization rate of cumulus-enclosed oocytes(51.5%) were significantly(p<0.05) higher than cumulus-denuded oocytes(21.7%). 2. The in-vitro maturation and fertilization rate of porcine oocytes co-cultured with 1$\times$104 cells/ml, 1$\times$106 cells/ml, 1$\times$108 cells/ml and 1$\times$1015 cells/ml oviduct epithelial cells in TCM-199 medium were 53.5% and 37.2%, 61.7% and 46.8%, 54.5% and 31.8%, 42.2% and 26.7%, respectively. 3. The in-vintro maturation and fertilization rate of porcine oocytes co-cultured with 1$\times$106/ml, 1$\times$108/ml, 1$\times$1015/ml uterine endometrial cells in TCM-199 medium were 54.3% and 39.1%, 58.3% and 43.8%, 55.5% and 33.3%, and 45.7% and 30.4%, respectively. 4. When the in-vitro fertilized oocytes were co-cultured with porcine cumulus cells, ovdiduct epithelial cells and uterine endometrial cells, the development rate to the blastocyst stage was 9.5%, 10.7% and 11.8%, respectively and the rates were higher than that of control, 2.1%(p<0.05).

Culture Optimization for Bacillus lentimorbus G-74 by Using a Miscanthus purpurascens Juice Medium (억새즙액 배지를 이용한 Bacillus lentimorbus G-74 균주의 배양 최적화)

  • Miscanthus purpurascens Juice containing potassium (37,952 mg/L), nitrogen (14,000 mg/L), phosphorus (6,800 mg/L), magnesium (5,969 mg/L), calcium (5,910 mg/L), etc., was investigated to develop a novel meidum far the mass cultivation of useful microorganisms. For this research, we first isolated an antagonistic bacterium G-74 from soil, which showed strong growth inhibition against two phytopathogenic fungi, Rhizoctonia solani and Botrytis cinerea, and identified as Bacillus lentimorbus G-74 based on the morphological characteristics and MIDI analysis. Culture conditions for G-74 strain in the M. purpurascens juice medium were optimized. Dilution rate of the medium, temperature and initial pH for the optimum growth of G-74 strain were 30% (V/V), $35^{\circ}C$ and 5.0, respectively. It was found that additions of 2.0% (W/V) corn starch as a carbon source and 1.0% (W/V) yeast extract as a nitrogen source in this medium increased B. lentimorbus G-74 growth to 66% more efficient than Luria Bertani medium.

Characterization of Crude $\alpha-fucosidase$ from Bacteroides fragilis Roid8 (Bacteroides fragilis Roid8의 $\alpha-fucosidase$ 조효소 특성)

  • Bacteroides is the most predominant bacteria in the human large intestinal tract. Bacteroides fragilis RoidB isolated from a Korean faecal sample showed high $\alpha-fucosidase$ activity compared to other bacterial species. The optimized meidum for the production of $\alpha-fucosidase$ from 8. fragilis contained BHI 37g, hemin 10mg, cysteine 0.5g, resazurin 1mg, vitamin K1mg, and starch 5g per 1 L. The crude $\alpha-fucosidase$ obtained through DEAE-sepharose and CM- cellulose chromatography showed optimum temperature and pH at $40^{\circ}C$ and 7.0, respectively. Among several metals, $Co^{++}\;and\;Zn^{++}$ showed strong inhibition on enzyme activity.

The Spheroplast Formation and Regeneration of Pseudomonas spp. (Pseudomonas spp.의 원형질체 생성과 재생)

  • The optimal conditions for the formation and the regeneration of Pseudomonas spheroplast were measured. Pseudomonas spp. cells were transformed to spheroplasts from 99.0% to 99.9% by treatment of $100{\mu}g/ml$ lysozyme and 10mM EDTA at room temperature. The optimal pH for the spheroplast formation was pH 8.0. Magnecium chloride, calcium chloride and streptomycin were effective on the stabilization of Pseudomonas spheroplast, while $Mg^+\;and\;Na^+$ ions were effective on the formation of Pseudonomas spheroplast. Rich Regeneration Medium was used for the regeneration of Pseudonomas spheroplast. To improve regeneration frequency, Bovin Serum Albumine and cationic ions were added to the spheroplast dilution beffer and regeneration environment respectively. Treatment of 20mM calcium chloride in ehr Rich Regeneration Medium could improve the yield of regenerants as much as 28-fold. Treatment of 1% Bovin Serum Albumine in the spgeroplast formation and dilution buffer increased the yield of regenerants to 10-fold. Also, the regeneration frequency was improved to 14-fold shen Rich Regeneration Meidum containing 0.5% Gelatin was used for regeneration as well as 1% Bovin Serum Albumine.

SOME PHYSIOLOGICAL STUDIES ON THE UTILIZATION OF ORGANIC SUBSTRATES BY EUGLENA GRACILIS VAR. BACILLA 10616 IN LIGHT AND IN DARKNESS ("유-그레나"의 명암배양에 따르는 유기질의 이용과 호흡 및 생장에 대하여)

  • 1) The comparative studies of the quantitative measurement of growth characteristics and utilization of substrates by Euglena gracilis var. bacilla 10616 in the light and in darkness have been carried out. Eodogenous respiration, effect of respiratory inhibitors and responses to the added substrates for the exogenous respiration are also investigated. 2) All cultures are grown in the open air under the continuous illumination of fluorescent light of 3500 lux at room termperature, the growth rate of the culture in the basal medium added 0.5% lactate is found to be the highest. The growth rate decreases successively for the cultures of 0.5% sucinate, 0.5% Na-acetate, 0.5% malate, and control. There is no growth in the basal meidum added 0.5% butyrate and 0.5% hydroquinone. The similar results are obtained for the mentioned cultures in the darkness. However, the growth rate in basal medium added 0.5% glucose and 0.5% sucrose does seem to increase in the darkness unlike the illumination. 3) The endogenous rate of respiration for the organism cultured photosynthetically is about 12.94ul 02/mg/hr, in basal medium and the respiratory quotient is about 0.84. The rate is decreased by starvations to 6.5ul 02/mg/hr, about to a half, but the respiratory quotient does net change. 4) The oxygen consomption during initial 2 hours in suspending solution ranging from pH 4.5 to pH 9.3 is highest at pH 4.5 in which the algae had grown, at pH 5.5 and at pH 6.9. 5) Endogenous respiration of the cells is strongly inhibited by 0.1M of potassium cyanide, malomic acid, sodium fluoride and iodo-acetic acid. It is also strongly inhibited by 0.01M of potassium cyanide. 6) The respiratory response to added substrates for the exogenous respiration in the organism is coincided with the rate in the basal medium added the substrate in light and in darkness, whether the cells are fed or starved. 7) According to the results of this study, there seems to be the flexibility of the interconversion between photosynthesis and chemosynthesis, heterotropic mode of metabolism, in Euglena gracilis var. bacillaris, and that this organism utilizes the lactate most. It also may be suggested that the enayme systems linked in the each steps of Embden-Myerhof-Parnas path way and TCA cycle seem to exist in this organism.

Development of New Mushroom Substrate of Pleurotus Ostreatus Using Foodwaste and Wormcast (남은 음식물과 지렁이 분변토를 이용한 느타리버섯의 대체 배지개발)

  • This study was investigated to develop an alternative medium for the cultivation of oyster mushrooms. The cultivation characteristics for major cultivated varieties of oyster mushrooms were campared with the food waste medium. In addition, water control & a gap formation of food waste is being pointed out as a problem of the medium material. Therefore, certain amounts of earthworm casting were supplied to the food waste medium to overcome the water and compaction phenomenon. This study investigated the use of the total 6 cultivars of oyster mushrooms, the Suhan(PL.1), Chiak No. 5(PL.2), Chiak No. 7(PL.3), Samgu No. 5(PL.4), Samgu No. 8(PL.5), Jangan No. 5(PL.6). In terms of medium, the optimum growth of the mycelium of the oyster mushroom was observed in the beet pulp medium but in food waste, the mycelium's growth was observed as insufficient in the bulk of cultivars. The Jangan No. 5(PL.6) was superior to other cultivars relative to the mycelium's growth. The optimum concentration of earthworm casting added the foodwaste medium was 30%. When Jangan No. 5(PL.6) was incubated in the foodwaste meidum containing 30% earthworm casting, the early days to germination were long, almost double the amount of time than that of the beet pulp medium. However, it was found that the fruit body size was larger than that of the beet pulp by up to 20.5%, and the length of the fruit body was up to 6.2% longer than that of the beet plup. The total yields increased by 60.5%. The analysis results of the oyster mushroom's nutritional contents cultured using the food waste medium, shows that the protein, vitamin A and vitamin E increased 70.6%, 2.4%, 0.8% respectively, the fat decreased by 12.6% and the oleic acid increased in a very small amount compared to that of the beet pulp medium.
