New Cytotoxic Metabolites from a Marine Sponge Homaxinella sp.

  • Three new butenolides (1-3), and a new cyclopentenone derivative (4) were isolated from a marine sponge Homaxinella sp. by bioactivity guided fractionation. The gross structures were established on the basis of NMR and MS analyses. The stereochemistry of the butenolides and cyclopentenone derivative was defined on the basis of optical rotation and CD spectroscopy. The compounds were tested against a panel of five human solid tumor cell lines and displayed marginal to significant cytotoxicity.

Sponge-Specific Unknown Bacterial Groups Detected in Marine Sponges Collected from Korea Through Barcoded Pyrosequencing

  • The bacterial diversity of 10 marine sponges belonging to the species Cliona celata, an unidentified Cliona species, Haliclona cinerea, Halichondria okadai, Hymeniacidon sinapium, Lissodendoryx isodictyalis, Penares incrustans, Spirastrella abata, and Spirastrella panis collected from Jeju Island and Chuja Island was investigated using amplicon pyrosequencing of the 16S rRNA genes. The microbial diversity of these sponges has as of yet rarely or never been investigated. All sponges, except Cliona celata, Lissodendoryx isodictyalis, and Penares incrustans, showed simple bacterial diversity, in which one or two bacterial OTUs occupied more than 50% of the pyrosequencing reads and their OTU rank abundance curves saturated quickly. Most of the predominant OTUs belonged to Alpha-, Beta-, or Gammaproteobacteria. Some of the OTUs from the sponges with low diversity were distantly (88%~89%) or moderately (93%~97%) related to known sequences in the GenBank nucleotide database. Phylogenetic analysis showed that many of the representative sequences of the OTUs were related to the sequences originating from sponges and corals, and formed sponge-specific or -related clades. The marine sponges investigated herein harbored unexplored bacterial diversity, and further studies should be done to understand the microbes present in sponges.

주황해변해면(Hymeniacidon sinapium) 공생세균 군집의 계절적 차이 (Seasonal Differences of Bacterial Communities Associated with the Marine Sponge, Hymeniacidon sinapium)

  • ARDRA (amplified ribosomal DNA restriction analysis) 방법을 이용하여 주황해변해면(Hymeniacidon sinapium)의 배양 가능한 공생세균 군집에 대하여 봄과 여름의 계절에 따른 차이를 분석하였다. 공생세균의 배양은 변형된 Zobell 배지와 MA 배지를 사용하였다. 분리된 균주의 16S rDNA를 증폭하고 제한효소 HaeIII와 MspI을 이용하여 제한효소 type을 구별하였다. 그 결과 봄 해면인 경우 23개, 여름인 경우 28개의 ARDRA type을 구별할 수 있었다. 각 type 별로 1-3개의 분리균주를 선별하여 부분 염기서열 분석 결과, 알려진 세균 종과 94% 이상의 유사도를 나타내었다. 봄 해면으로부터 분리된 세균들은 Alphaproteobacteria, Gammaproteobacteria, Firmicutes, Actinobacteria, 4개의 문(phylum)에 속하였으며 여름 해면의 공생세균은 Alphaproteobacteria, Betaproteobacteria, Gammaproteobacteria, Firmicutes, Bacteroidetes, 5개의 문에 포함되었다. Gammaproteobacteria는 봄 해면에서 33.8%, 여름 해면에서 67.4%가 각각 관찰되어 두 계절에서 우점 하는 세균그룹으로 나타났으며 여름철에 증가하는 경향을 나타내었다. Firmicutes와 Actinobacteria의 경우 봄 해면에서 각각30.2%, 8.3%로 관찰된 반면 여름해면에서는 6.9%, 0%로 관찰되어 여름철에 감소하는 세균 그룹이었다. Betaproteobacteria(4.7%)와 Bacteroidetes (4.7%)는 여름 해면에서만 관찰되었다. H. sinapium 해면에서 봄철에 비해 여름철에 더 다양한 세균그룹을 발견할 수 있었으며 동일한 해면 종일지라도 계절에 따라 공생세균 군집에 차이를 나타냄을 알 수 있었다.

A new derivative of phorbaketals isolated from a Marine Sponge Phorbas species

  • A new sesterterpenoid, phorbaketal derivative, was isolated from the marine sponge Phorbas species. Its planar structures was completely determined from a combination of extensive 1D and 2D NMR experiments and MS data, and also the stereochemistry on the chiral centers were established by the ROESY experiment and the comparison with the $^1H$ and $^{13}C$ chemical shifts of the known phorbaketal compounds. This compound 1 moderately showed cytotoxicity effect against hepatoma cancer HepG2 cell.

Three Sterol Sulfates Isolated from a Marine Sponge Acanthodoryx Fibrosa

  • Three sterol sulfates were isolated from AMPK activity-guided fraction of a marine sponge Acanthodoryx fibrosa. Their structures were determined by an extensive NMR analysis, MS data, and two compounds were confirmed as unusual phosphorylated sterol sulfates by comparing with NMR data of the known compounds. Compound 3 was given to be a new dephosphated sterol sulfate derivative. Compound 1 moderately showed AMPK activation effect on L6 myoblast cell through Western blot analysis.

Scalaran-type sesterterpenes from a Marine Sponge Smenospongia species showing the AMPK activation

  • A new scalaran class sesterterpenoid with five known ones was isolated from a marine sponge Smenospongia species collected from the Gageo island, Korea. Chemical structure of all of compounds was determined on the basis of a combination of extensive 1D and 2D NMR experiments and MS data. The new compound exhibited a new functional group on a common scalaran sesterterpene skeleton, identified as 12-deacetoxy-23-deacetoxyscalarin. The compound 1 moderately showed the effect of the activation of AMP-activated protein kinase (AMPK) in L6 myoblast cell.

Polyketides from a Sponge-Derived Fungus, Aspergillus versicolor

  • Bioactivity guided fractionation of the cultured filtrates of Aspergillus versicolor, which was derived from a marine sponge Petrosia sp., yielded three polyketides: decumbenones A (1),B (2), and versiol (3). These compounds were identified on the basis of 1D and 2D NMR spectroscopic and MS analysis. The absolute configuration was defined by the modified Mosher's method. The isolated compounds were tested for cytotoxicity against a panel of five human solid tumor cell lines and antibacterial activity against twenty clinically isolated methicillin-resistant strains. This is the first report on the isolation of these compounds from a marine source.

  • Guided by the brine shrimp lethality assay, a new (4) and three known cyclitol derivatives (1-3) were isolated from the marine sponge Stelletta sp. Norsarcotride A (4) showed significant cytotoxicity against a small panel of five human tumor cell lines.

Cytotoxic Polyketides from the Marine Sponge Discodermia calyx

  • Bioassay-guided fractionation of the MeOH extract from the sponge Discodermia calyx collected off the coast of Jeju Island, South Korea, led to the isolation of a polyketide, icadamide C (1), along with previously reported theopederin K (3). Structure elucidation was performed by a combination of high resolution mass and 2D-NMR (principally COSY, HMBC, HSQC, and NOESY) spectroscopy. Stereochemistry of compound 1 was determined as 2R*, 3R*, 6R*, 10S*, 11S*, 12R*, 13S*, 15R* and 2'S by NMR data and Marfey analysis. Isolated metabolites displayed potent cytotoxic activity against a small panel of five human solid tumor cell lines with ED50 values of less than 0.1 μg/mL.

Sarcotrine G, a New Derivative Isolated from a Marine Sponge Sarcotragus Species

Their planar structures were determined from combination of extensive 1D and 2D NMR experiments and MS data, and the configuration at the chiral centers were assigned by comparison with the $^1H$ and $^{13}C$ chemical shifts of the known compounds. Among four compounds, compound 2 was found to be a new sarcotrine derivative. Though not strong, compounds 1-4 moderately showed AMPK activation effect on L6 myoblast cell through Western Blot analysis.
