오카다익산 군 독소: 독성, 분석법 및 관리 동향 (Okadaic Acid Group Toxins: Toxicity, Exposure Routes, and Global Safety Management)

  • Okadaic acid (OA) 군 독소는 설사성 패류중독(diarrhetic shellfish poison, DSP)을 유발하는 해양생물독소이다. 설사성패독은 오카다익산(OA)과 그 유사체인 디노피시스톡신(DTX)으로 구성되어 있으며, 주로 와편모조류에서 생성되어 이매패류의 체내에 축적되어 독화된다. 이에 EFSA, WHO에서는 안전관리를 위하여 수산물 내 OA군 독소 함량 기준을 설정하였다. 최근 우리나라 연안에서도 원인조류인 Dinophysis sp.의 출현이 보고된 바 있으며, 국내 수산물 생산과 소비에도 영향을 미치고 있다. 또한 국제적인 움직임에 발맞추어 국내에서도 2022년 설사성 패독 기준 관리 대상 물질에 DTX2를 추가하여 관리하고 있다. 본 연구는 OA군 독소의 이화학적 특성, 분석법, 인체 노출 사례와 국내외 관리 현황 등의 자료를 검토하여 OA군 독소의 체계적인 모니터링과 안전관리의 기반을 마련하고자 수행되었다.

해조류인 뜸부기의 추출물이 지질대사에 미치는 영향 (Effects of the Extracts from the Marine Algae Pelvetia siliquosa on Hyperlipidemia in Rats)

  • The effects of the MeOH and water extracts from the marine algae Pelvetia siliquosa were evaluated on hyperlipidemic rats induced by cholesterol rich diet or poloxamer-407. The MeOH and water extracts, when administered orally for 3 consecutive days in hyperlipidemic rats induced by poloxamer-407 (1 ml of 30%), were found to cause a significant decrease in plasma cholesterol and triglyceride concentrations. Both extracts also showed a significant inhibition of serum total cholesterol and triglyceride level in rats treated with cholesterol rich diet. HDL-cholesterol, however, was increased significantly.

Bioluminescent Assay of ${\alpha}$-Oxidase from Cucumis sativus using Bacterial Luciferase-Coupled Reaction

  • A new assay method of ${\alpha}-Oxidase$ (fatty acid : oxygen dioxygenase, 1-decarboxylating) was developed using a bioluminescence reaction system of marine luminous bacterium, Photobacterium phosphoreum. ${\alpha}$-Oxidase was isolated from a cucumber (Cucumis sativus). Pentadecanoic acid was used as a substrate, and the product, tetradecanal, was analyzed with a bacterial luciferase-coupled reaction. Initial light intensity was directly related to the concentration of tetradecanal in the range of 1 nM to 10 ${\mu}M$. Optimal pH and temperature were 7.5 and $25^{\circ}C$, respectively. Optimal pentadecanoic acid concentration in a standard assay of ${\alpha}$-oxidase was 0.1 mM. The Km value of pentedecanoic acid was $85{\mu}M$. This method is straightforward, rapid, convenient, and easy. Its needs no treatment or extraction of reaction mixture.

해양동물 눈알고둥 렉틴의 림프구 분열효과 및 면역화학적 특성 (Lymphocytes Mitogenic and Immunochemical Properties of the Lectins from Marine Animal Lunella coronata coreensis)

  • Developing new substance for immunosuppressor or immunomodulator from natural products is extremely important in the present biomedicine. In this paper, we focused our efforts on the mitogenicity and immunochemical properties of the two lectins (LCC-I, LCC-II) obtained from marine animal Lunella coronata coreensis. Immunochemical techniques were employed to elucidate the structural and/or functional similarities between the LCC lectins. Molecular weight of the LCC lectins, LCC-I and LCC-II were estimated to be around 60 KD and 66-70 KD, respectively. LCC lectins were mitogens for murine splenic lymphocytes, and the optimum mitogenic doses were 31.25 $\mu\textrm{g}$/ml and 3.91 $\mu\textrm{g}$/ml, respectively. LCC-II lectin was a good mitogen toward human peripheral lymphocytes at a concentration about 31.25 $\mu\textrm{g}$/ml.

  • The marine sponge Petrosia sp. is known for various bioactive compounds including the recently reported polyacetylenic alcohols. In our continuous survey of bioactive compounds from the Petrosia sp. collected from Korean waters, a cyclopentanepentol which rarely occurs in natural products has been isolated. It was found to inhibit DNA replication at the initiation step.

Bioluminescent Assay of Bovine Liver Riboflavin Kinase Using a Bactreial Luciferase Coupled Reaction

  • For the demonstration of a novel riboflavin kinase assay method based on the bacterial bioluminescence, partially purified riboflavin kinase was prepared from bovine liver through ammonium sulfate precipitation and DEAE-cellulose ion exchange chromatography. Using bacterial luciferase from Photobacterium phosphoreum and the dithionite reduction method, and easy, safe, and fast assay method was established. The optimal temperature, pH, Km values form riboflavin and ATP of boving liver riboflavin kinase determined with this luminescence method were 35$^{\circ}C$, pH 7, 15.3${\mu}$M and 8.3.${\mu}$M, respectively. The detection limit of FMN produced by riboflavin kinase was in the range of 200 pM to 4${\mu}$M which is comparable to the HPLC-fluorescence detection method, while the detection time for each assay was less than 15 sec compared to the HPLC method which requeires at least 10 min for completion.

Production of Oleamide, a Functional Lipid, by Streptomyces sp. KK90378

  • Oleamide (cis-9-octadecenamide) is endogenous primary amide of fatty acid that is produced in small amounts in animal brains. It is known to induce sleep and to lower temperature by destroying the lipid plasma membrane structure of cells, thereby disclosing gap junction channels. To develop a new biological production method for oleamide, a screening program was conducted to isolate a microorganism producing oleamide. Among 1,500 soil microorganisms tested, KK90378 exhibited a potent positive reaction with Dragendoff`s reagent, used to detect the primary amide of oleamide. KK90378 was identified as a Streptomyces species based on cultural and morpohological characteristics, the presence of diaminopimelic acid in the cell wall, and the sugar patterns for the whole-cell extrat. Streptomyces sp. KK90378 produced oleamide 3 days after culture at $28^{\circ}C$, pH 7.2 A series of purification steps, including hexane extraction, silica gel column, and preparative thin layer chromatographies, were performed for the purification of oleamide. A spectrophotometric analysis using $^1H$, $^13C$-NMR, and GC-MS confirmed that the chemical structure of the purified oleamide was identical to that of authentic oleamide.

  • Korea, China and Japan are countries located in Northeast Asia territory sharing similar natural environment. Countries are also using rice as a main diet material accompanied with vegetables, marine products and bean or fermented bean food. Yet, at the same time each country's food culture appeared as to be unique in food space due to their diverse food materials from different natural, cultural and religious background. This research is probing a way to recover subjective food culture and accomplish food environment which appropriates to modern globalized era by developing simple and economical repast tools meeting public's needs and distributing unique repast tools becomes Korean food culture and can be used in the life pattern of modernized nuclear families.

Determination of Silybin B in the Different Parts of Silybum marianum using HPLC-UV

  • Silymarin is the standardized extract from Silybum marianum which consists mainly of flavonoids and polyphenols. It is highly regarded for its hepatoprotective ability. Silybin B is a flavonolignan and one of the active components of silymarin. The content of silybin B in various parts of S. marianum was analyzed by HPLC-UV. Results show that the extract of seeds contain the highest amount of silybin B (7.434 mg/g DW). The petioles of S. marianum showed a low content of silybin B. This study revealed that seeds of S. marianum contain high amount of silybin B and could be a good source of the compound.

Microbial Metabolism of Yangonin, a Styryl Lactone from Piper methysticum (Kava)

  • Microbial metabolism studies of yangonin (1), a major styryl lactone from Piper methysticum, have resulted in the production of three hydroxylated metabolites (2-4). The chemical structures of these compounds were elucidated to be 4-methoxy-6-(12-hydroxystyryl)-2-pyrone (2),4-methoxy-6-(11,12-dihydroxystyryl)-2-pyrone (3),and 4,12-dimethoxy-6-(7,8-dihydroxy-7,8-dihydrostyryl)-2-pyrone (4) on the basis of the chemical and spectroscopic analyses. The compounds 3 and 4 are reported herein as microbial metabolites of yangonin for the first time.
