Isolation and Characterization of the Plant Growth Promoting Rhizobacterium, Arthrobacter scleromae SYE-3 on the Yam Growth (식물성장촉진근권미생물 Arthrobacter scleromae SYE-3의 분리 및 Yam (Dioscorea japonica Thunb.) 성장에 미치는 영향 연구)

  • In this study, Arthrobacter scleromae SYE-3, which was isolated from indigenous plant in a subtropical region, Neigeria, with plant growth promoting activity was evaluated to determine the optimal culture condition. A bacterial strain SYE-3 had the IAA productivity ($89.15{\pm}0.36mg/L$) and ACC deaminase activity ($0.20{\pm}0.06$ at 72 hours). Also, optimal culture conditions such as temperature and pH of strain SYE-3 were $20^{\circ}C$ and 10 in LB medium, respectively. Strain SYE-3 had up to 3% salt tolerance in the LB medium. Plant growth promoting ability of strain SYE-3 using yam (Dioscorea japonica Thunb.) was evaluated. As a result, strain SYE-3 had showed very powerful effect on the increase of the shoot length and root biomass of yam (190.0% and 282.41% increase for 112 days, respectively). These results indicated that Arthrobacter scleromae SYE-3 can serve as a promising microbial resource for the biofertilizers of subtropical crops.

Lignin Removal from Barley Straw by Ethanosolv Pretreatment (Ethanosolv 전처리에 의한 보릿짚의 리그닌 제거)

  • Lignocellulose represents a key sustainable source of biomass for transformation into biofuels and bio-based products. Unfortunately, lignocellulosic biomass is highly recalcitrant to biotransformation, both microbial and enzymatic, which limits its use and prevents. As a result, effective pretreatment strategies are necessary. The vast majority of pretreatment strategies have focused on achieving a reduction of lignin content. In this work, an ethanosolv pretreatment has been evaluated for extracting lignin from barley straw. 75% ethanol was used as a pretreatment solvent to extract lignin from barley straw. The influence on delignification of three independent variables are temperature, time, catalyst (1 M $H_2SO_4$) dose. The best pretreatment condition observed was $180^{\circ}C$, 120 min, 0.2% $H_2SO_4$ and delignification was 38%. A combined roasting and ethanosolv, 2-step pretreatment, was developed in order to improve the delignification. Roasting didn't increase the delignification but reduced the pretreatment time. X-ray diffraction results indicated that these physical changes enhance the enzymatic digestibility in the ethanosolv treated barley straw. The cellulose in the pretreated barley straw becomes more crystalline without undergoing ethanosolv.

Seasonal Variations and Species Composition of Planktonic Ciliates in the Southern Coastal Waters of Jeju Island, Korea (제주도 남부해역의 부유성 섬모충류의 종 조성과 계절 변동)

  • As part of study on the microbial food web in the southern Jeju Island of Korea, Planktonic tintinnids and aloricate oligotrichs were investigated from July 1998 to June 2000 to understand seasonal variation and water mass indication with environmental factors. 39 species of tintinnids were identified representing 1 order, 11 families, 20 genera. Tintinnid abundance ranged from 100 to 5,400 cells.1$^{-1}$ (mean 314 cells.1$^{-1}$ ), and oceanic species were mainly dominant in fall and winter season, while neritic species were a little pre- dominant in spring and summer season. 15 species of aloricate oligotrichs were identified representing 1 order, 5 families, 7 genera, and many belonged to genus Strombidium which was most abundant and most frequently occurred in many months. Abundance of aloricate oligotrichs ranged from 140 to 21,000 cells.1$^{-1}$ (mean 2,356 cells.1$^{-1}$ ). Species diversity and standing crops of tintinnids were quite different according to seasons, but few seasonal variations were detected in aloricate oligotrichs. In terms of water mass indication tintinnids represented obvious characteristics which were affected by marine environmental factors, but alor- icate oligotrichs had no such apparent indication as tintinnids. Ciliates were more abundant and more diverse in inshore than in offshore station. Total carbon biomass of ciliates ranged from 0.01 to 136.06 $\mu\textrm{g}$C.1$^{-1}$ (mean 5.01 $\mu\textrm{g}$C.1$^{-1}$ ). The carbon biomass, however, did not coincided with seasonal variations of abun- dance Vertical profiles of mean abundance of both tintinnids and aloricate oligotrichs were similar, and had same trend as those of mean chlorophyll o concentration. It suggests that phytoplankton and ciliates reflected the prey-predator relationship in the study area.

Soil Microbial Diversity of the Plastic Film House Fields in Korea (우리나라 중부지방 시설재배지 토양 미생물의 다양성에 관한 연구)

  • Although biological metabolism in soil is very important for evaluating the soil properties, most of researches have concerned mainly about physical and chemical sides. In this study, biological characteristics were examined to demonstrate the biota in the plastic film house soils. Contents of organic matter and phosphate in soil were increased with cultivation period. ECs of soil cultivated spinach and melon were $3.59dS\;m^{-1}$ and $3.46dS\;m^{-1}$ respectively: these values were higher than that of rose and flower, which were $1.23dS\;m^{-1}$ and $1.32dS\;m^{-1}$ respectively. The population of fluorescent Pseudomonas strains of the soil cultivated flowers: $113.8{\times}10^4{\sim}129.7{\times}10^4cfu\;g^{-1}$ was higher than that of leafy vegetables: $40.7{\times}10^4{\sim}97.9{\times}10^4cfu\;g^{-1}$ and fruiting vegetables: $25.0{\times}10^4{\sim}91.7{\times}10^4cfu\;g^{-1}$. However the number of Fusarium strains of the soil cultivated with flowers: $3.8{\times}10^2{\sim}4.0{\times}10^2cfu\;g^{-1}$ was lower than that of leafy vegetables: $4.3{\times}10^2{\sim}16.3{\times}10^2cfu\;g^{-1}$ and fruiting vegetables: $7.6{\times}10^2{\sim}30.0{\times}10^2cfu\;g^{-1}$. In relation to the cultivation period, the habitation density of aerobic bacteria, mesophilic Bacillus, thermophilic Bacillus, and fluorescent Pseudomonas strains was the highest in the soil cultivated over 11 years, but diversity index showed negative correlation with cultivation period. Microbial biomass C in these soils had positive correlation with each number of microorganisms including aerobic bacteria, actinomycetes, and strains of mesophilic Bacillus as well as the total number of these microorganisms.

Effect of Culture Conditions on Characteristics of Growth and Production of Docosahexaenoic acid (DHA) by Schizochytrium mangrovei (배양조건에 따른 Schizochytrium mangrovei의 성장 및 Docosahexaenoic acid의 생산특성)

  • Both docosahexaenoic acid (DHA, 22:6n-3) and eicosapentaenoic acid (EPA, 20:5n-3) have attracted increasing attention since the first epidemiological report on the importance of n-3 essential fatty acids. Lipids in microbial cells play various biological roles and, consequently, much research has been carried out on their role in cell physiology. The lipid composition of microorganisms can exhibit considerable variations depending on environment. The effects of culture conditions, temperature (15, 20, 24, 28, 32 and $36^{\circ}C$), salinity (10, 20, 30, 40 and 50 psu), pH (pH5, 6, 7, 8 and 9), rotation speeds (50, 100, 150 and 200 rpm), carbon sources, nitrogen sources and C/N ratio on the production of docosahexaenoic acid, fatty-acid profiles, and acids secreted to the broth culture by the oleaginous microorganism, Schizochytrium mangrovei (KCTC 11117BP), were studied. Temperature (initially $28^{\circ}C$), salinity (20 psu), pH (pH7), rotation speeds (100 rpm), organism fatty acids, and secreted acids in the broth were varied during cultivation of S. mangrovei. At pH 7.0, S. mangrovei was able to accumulate lipids up to 40% of its biomass, with 13% (w/w) DHA content. The monosaccharides glucose and fructose, and yeast extract were suitable carbon and nitrogen sources, respectively. The primary omega-3 polyunsaturated fatty acid produced was docosahexaenoic acid.

Soil Chemical Property and Microbial Community under Organic and Conventional Radish Farming Systems (무 유기재배와 관행재배 토양의 화학성과 미생물 군집 비교)

  • This study was conducted to investigate the responses of soil properties and microbial communities to different agricultural management and soil types, including organic management in Andisols (Org-A), organic management in Non-andisols (Org-NA), conventional management in Andisols (Con-A) and conventional management in Non-andisols (Con-NA) by using a pyrosequencing approach of 16S rRNA gene amplicon in Radish farms of volcanic ash soil in Jeju island. The results showed that agricultural management systems had a little influence on the soil chemical properties but had significant influence on microbial communities. In addition, soil types had significant influences on both the soil chemical properties and microbial communities. Organic farming increased the microbial density of bacteria and biomass C compared to conventional farming, regardless of soil types. Additionally, Org-NA had the highest dehydrogenase activity among treatments, whereas no difference was found between Org-A, Con-A and Con-NA and had the highest species richness (Chao 1) and diversity (Phyrogenetic diversity). Particularly, Chao 1 and Phyrogenetic diversity were increased in organic plots by 12% and 20%, compared with conventional plots, respectively. Also, regardless of agricultural management and soil types, Proteobacteria was the most abundant bacterial phylum, accounting for 21.9-25.9% of the bacterial 16S rRNAs. The relative abundance of putative copiotroph such as Firmicutes was highest in Org-NA plot by 21.0%, as follows Con-NA (13.1%), Con-A (6.7%) and Org-A (5.1%.), respectively and those of putative oligotrophs such as Acidobacteria and Planctomycetes were higher in Con-A than those in the other plots. Furthermore, LEfSe indicated that organic system enhanced the abundance of Fumicutes, while conventional system increased the abundance of Acidobacteria, especially in Non-andisols. Correlation analysis showed that total organic carbon (TOC) and nutrient levels (e.g. available P and exchangeable K) were significantly correlated to the structure of the microbial community and microbial activity. Overall, our results showed that the continuous organic farming systems without chemical materials, as well as the soil types made by long-term environmental factors might influence on soil properties and increase microbial abundances and diversity.

Design, Optimization and Verification of 16S rRNA Oligonucleotide Probes of Fluorescence in-situ Hybridization for Targeting Clostridium spp. and Clostridium kluyveri

  • Fluorescence in-situ hybridization (FISH) is a common and popular method used to investigate microbial communities in natural and engineered environments. In this study, two specific 16S rRNA-targeted oligonucleotide probes, CLZ and KCLZ, were designed and verified to quantify the genus Clostridium and the species Clostridium kluyveri. The optimal concentration of hybridization buffer solution for both probes was 30% (w/v). The specificity of the designed probes was high due to the use of pellets from pure reference strains. Feasibility was tested using samples of Chinese liquor from the famed Luzhou manufacturing cellar. The effectiveness of detecting target cells appears to vary widely in different environments. In pit mud, the detection effectiveness of the target cell by probes CLZ and KCLZ was 49.11% and 32.14%, respectively. Quantitative analysis by FISH technique of microbes in pit mud and fermented grains showed consistency with the results detected by qPCR and PCR-DGGE techniques, which showed that the probes CLZ and KCLZ were suitable to analyze the biomass of Clostridium spp. and C. kluyveri during liquor fermentation. Therefore, this study provides a method for quantitative analysis of Clostridium spp. and C. kluyveri and monitoring their community dynamics in microecosystems.

Soil Carbon Storage in Upland Soils by Biochar Application in East Asia: Review and Data Analysis (바이오차를 이용한 밭 토양 탄소 저장: 동아시아 지역 연구 리뷰 및 데이터 분석)

  • BACKGROUND: Biochar is a solid material converted from agricultural biomass such as crop residues and pruning branch through pyrolysis under limited oxygen supply. Biochar consists of non-degradable carbon (C) double bonds and aromatic ring that are not readily broken down by microbial degradation in the soils. Due to the recalcitrancy of C in biochar, biochar application to the soils is of help in enhancing soil carbon sequestration in arable lands that might be a strategy of agricultural sector to mitigate climate change. METHODS AND RESULTS: Data were collected from studies on the effect of biochar application on soil C content conducted in East Asian countries including China, Japan and Korea under different experimental conditions (incubation, column, pot, and field). The magnitude of soil C storage was positively correlated (p < 0.001) with biochar application rate under field conditions, reflecting accumulation of recalcitrant black C in the biochar. However, The changes in soil C contents per C input from biochar (% per t/ha) were 6.80 in field condition, and 12.58 in laboratory condition. The magnitude of increment of soil C was lower in field than in laboratory conditions due to potential loss of C through weathering of biochar under field conditions. Biochar production condition also affected soil C increment; more C increment was found with biochar produced at a high temperature (over 450℃). CONCLUSION: This review suggests that biochar application is a potential measures of C sequestration in agricultural soils. However, as the increment of soil C biochar was affected by biochar types, further studies are necessary to find better biochar types for enhanced soil C storage.

Nutrient Balances and Soil Properties Affected by Application of Soybean and Barley Residues

  • An accurate analysis of nutrient balance in different cropping systems is necessary for improving soil fertilities, causing higher crop yields and quality. This study was carried out to investigate the nutrient balance, changes in soil properties, and their effects on crop yield in long-term field cultivation under mono- and rotation-cropping systems (MCS and RCS, respectively). The analytical results of the soil properties showed that the application of mineral fertilizers alone in the MCS leads the reduction of soil CEC, exchangeable Ca, and microbial biomass C and N. Compared with the MCS of soybean, the RCS of soybean and barley significantly improved the soil properties, which increased crop yield. It might be due to the barley residue added to the RCS soil. Mean nutrient balances for 4 years were -55.9 kg N, +34.7 kg $P_2O_5$, and -0.3 kg $K_2O$ $ha^{-1}$ for the MCS and +19.7 kg N, +107.4 kg $P_2O_5$, and -48.6 kg $K_2O$ $ha^{-1}$ for the RCS, respectively. These nutrient imbalances mean that conventional fertilizer recommendations were inadequate for maintaining soil nutrient balance. From these results, we can conclude that the crop rotation may change comprehensive physical, chemical, and biological soil properties. These changes could affect the nutrient balance and then the crop yield.

Treatment of Acid dye Using Microbial Immobilization (미생물 고정화를 이용한 산성염료의 처리)

  • Strains degrading and decolorizing acid dyes, Nylosan red E-BL 150%. were isolated from natural system, was named as ARK3. The optimal culture conditions of temperature and pH were $35^\circ{C}$, 7.0, respectively. Growth rate of cells in conditions of aerobic shaking more than standing culture conspicuously increased, and optical density of those to strain ARK3 were found as 1.38 and 0.25 after 42 hrs. Decolorization efficiency in batch culture which used as immobilization media to natural zeolite was 15% after 6 hrs, while suspension culture was 5%, also its of immobilization and suspension culture were 90% and 85% after 48 hrs, respectively. Decolorization efficiency of air-lift bioreactor was more than 90% to a dilution rate of $0.038hr^{-1}$, but that was decreased as 70%, when the dilution rate was $0.05hr^{-1}$. Even though at maximum dilution rate of this study, there was not appeared "wash out" phenomienon of biomass. Decolorization efficiency was 97.7% at a dilution rate of $0.025hr^{-1}$, when influent dye concentration was $100mg/\ell$. But if influent dye concentration increased as $150mg/\ell$, even though MLVSS increased, that of treatment water decreased as 93%. Also, when influent dye concentration increased as $200mg/\ell$ and $300mg/\ell$, decolorization efficiencies of treatment water abruptly decreased as 85% and 63%, respectively. Decolorization efficiency was more than 92% to the limit volumetric loading rate of $3.75mg/\ell\cdot{hr}$hr, without regard to variation of influent dye concentration or hydraulic retention time. if volumetric loading rate was more than $3.80mg/\ell\cdot{hr}$, at same condition, decolorization efficiency was lower decrease of retention time than increase of influent dye concentration.entration.
