Effects of Lycium chinense Miller Fruit and its Constituent Betaine on Immunomodulation in Balb/c Mice

  • BACKGROUND: The aim of this study is to investigate the effects of Lycium chinense Miller fruit and its representative constituent betaine on reduction of immobility time and blood parameters in balb/c mice. METHODS AND RESULTS: We investigated the immobility time and the changes in aspects of blood biochemical parameters by the administration of L. chinense Miller fruit and its representative constituent betaine, after the forced swimming test. The immobility time was significantly reduced about 41.3% and 53.6%, respectively, in the animal of L. chinense Miller fruit and its representative constituent betaine-administrated group for 7 days, in comparison with that of the control group. The level of blood urea nitrogen was significantly decreased in L. chinense Miller fruit and its representative constituent betaine-treated group compared with the control group (P < 0.05), respectively. In addition, the interlukin-2 levels of mice in L. chinense Miller fruit and betaine treated group was increased compared with the control group. CONCLUSION: These results indicate that L. chinense Miller fruit and betaine might be helpful in the immune function improvement, enhance physical stamina, and fatigue recovery.

Isolation of an Angiotensin Converting Enzyme Inhibitory Substance from Lycium chinense Miller

  • An angiotensin converting enzyme (ACE) inhibitory substance was isolated and purified from Lycium chinense Miller. A crude water extract of Lycium chinense Miller was prepared by adding it to water shaking at $25^{\circ}C$ for 1 hr, followed by centrifugation at 8000 ${\times}$ g for 30 min. The crude extract was then filtered using YM-3 and YM-1 membranes. An ACE inhibitor was isolated using consecutive chromatographic methods including: ion exchange chromatography, gel permeation chromatography, and FPLC. The inhibitor was identified to have a molecular mass of 862 daltons by mass spectrometry.

Evaluation of Antiaging Activity on Lycium chinense Miller. Extracts (구기자 추출물의 항노화 활성 평가)

  • The purpose of this study was to investigate the functionality of Lycium chinense Miller. as a functional cosmetic material. The total polyphenol content in the extract was 17.3 mg/mL, and as a result of confirming cytotoxicity through MTT assay before the enzyme experiment, it was confirmed that it did not affect cell growth at all concentrations below 1000 ㎍/mL. As a result of conducting DPPH radical scavenging tests as an antioxidant efficacy test, it showed 84.97% scavenging ability at a concentration of 1000 ㎍/mL, and showed very excellent antioxidant efficacy in SOD-like activity test with a result of 80.54% at 1000 ㎍/mL. The measured elastase inhibitory activity, the inhibition rate of 50.93% was observed at 1000 ㎍/mL, indicating that the extract of Lycium chinense Miller. was effective in inhibiting elastase. All these findings suggested that 70% ethanol extract of Lycium chinense Miller. was confirmed as a functional cosmetic material by confirming its antioxidant and anti-wrinkle efficacy.

The Washing Characteristics of Lycium chinense Miller with Different Washing Methods (구기자 세척기 개발을 위한 구기자의 세척특성)

  • This study was conducted to define the optimal Lycium chinense miller washing method for developing the Lycium chinense cleaner and we analyzed the Lycium chinense miller washing characteristics for removing pesticides and microorganism according to washing methods; habitual washing method, air bubble washing method and nozzle spray washing method. The results were summarized as follows; 1. In case of measuring physical properties according to the varieties, maximum yield strength of Hokwang was 2.562 kgf, minimum yield strength of Hokwang was 0.269 kgf and average yield strength was about 1 kgf. 2. In case of measuring change of bacteria according to washing methods, the number of bacteria of non-washing method was more than the number of bacteria of habitual washing method or mechanical washing method and the number of nozzle spray washing method was least. 3. Ahjoksiseuteurobin of 0.218 ppm was detected in the untreated sample, 0.051 ppm was detected in the habitual washing method, 0.047 ppm was detected in the air bubble washing method and 0.034 ppm was detected in nozzle spray washing method. Every amount detected were less than 2 ppm that is reference value and the detected amount was least in case of nozzle spay cleaning method. Cypermethrin of 0.772 ppm was detected in the non-cleaned sample, 0.089ppm was detected in habitual washing method, 0.26 ppm was detected in the air bubble washing method and 0.292 ppm was detected in the nozzle spray washing method. Every detected amount of Cypermethrin were less than 5 ppm that is reference value and the detected amount was least in case of habitual washing method.

Antimicrobial Property of $(+)-Lyoniresinol-3{\alpha}-O-\beta-D-Glucopyranoside$ Isolated From the Root Bark of Lycium chinense Miller Against Human Pathogenic Microorganisms

  • [ $(+)-Lyoniresinol-3{\alpha}-O-\beta-D-glucopyranoside$ ] (1) was isolated from an ethyl acetate extract of the root bark from Lycium chinense Miller, and its structure was determined using 1D and 2D NMR spectroscopy including DEPT, HMQC, and HMBC. $(+)-Lyoniresinol-3{\alpha}-O-\beta-D-glucopyranoside$ exhibited potent antimicrobial activity against antibiotic-resistant bacterial strains, methicillin-resistant Staphylococcus aureus (MRSA) isolated from patients, and human pathogenic fungi without having any hemolytic effect on human erythrocytes. In particular, compound 1 induced the accumulation of intracellular trehalose on C. albicans as stress response to the drug, and disrupted the dimorphic transition that forms pseudo-hyphae caused by the pathogenesis. This indicates that $(+)-Lyoniresinol-3{\alpha}-O-\beta-D-glucopyranoside$ has excellent potential as a lead compound for the development of antibiotic agents.

  • A new phenolic amide, dihydro-N-caffeoyltyramine (1) was isolated from the root bark of Lycium chinense Miller, along with known compounds, trans-N-caffeoyltyramine (2), cis-N-caffeoyltyramine (3), and lyoniresinol $3{\alpha}-Ο-{\beta}-D-glucopyranoside$ (4). Their structures were determined by spectroscopic analysis. A NBT superoxide scavenging assay revealed that three phenolic amides showed potent antioxidative activity.

Induction of Hariy Root and Bioreactor Culture of Lycium chinense (구기자나무 (Lycium chinense)의 모상근 유도 및 생물반응기 배양)

  • This article was conducted to induce the transgenic hairy roots and determine the effect of culture conditions on optimum growth of hairy roots by Agrobacterium rhizogenes strain, 15834 in Lycium chinense Miller. Hairy roots of L. chinense Miller. were induced from leaf segments by co-cultivation with A. rhizogenes. When the hairy roots were cultured in various MS medium strength and sucrose concentrations, the highest growth of hairy roots was observed in half-strength MS media supplemented with 3% sucrose, respectively. In air lift bioreactor cultures, the liquid medium contained with 1/2 MS and 3% sucrose was also the best for optimum growth of hairy roots.

  • This study was conducted to develop a functional feed additive for pig with spent $Lycium$ $chinense$ Mill fruit. We investigated the optimum conditions for the extraction of polyphenol from spent $Lycium$ $chinense$ Mill using methanol. Methanol concentration as a solvent for extraction, extraction time and the volume of solvent per a gram of solid (ground spent Lyceum chinense Mill) were selected as parameters. Three levels of parameters were configured according to Box Behnken experiment design, a fractional factorial design, and total 15 trials were employed. Total polyphenol concentration from each trial was used as response from experiment system and effects of parameters on total polyphenol extraction efficiency were determined using response surface model. As a result, all terms in analysis of variance, regression ($p$ = 0.001), linear ($p$ = 0.002), square ($p$ = 0.017) and interaction ($p$ = 0.047) was significant and adjusted determination coefficient ($R^2$) was 94.7%. Total polyphenol extraction efficiency was elevated along increased methanol content and decreased solvent to solid ratio. However extraction time did not affect the efficiency. This study provides a primary information for the optimum extraction conditions to maximize total polyphenol recovery from spent Lycium chinens Mill fruit and this result could be applied to re-use of argo-industrial by-products and to develop of functional feed additives in organic farming.

  • This experiment was conducted to breed the Boxthorn varieties with resistance to Anthracnose, good quality and high yielding potential in 1984 to 1986(3 years).The character of "Yu seong 1" with resistance to Anthracnose and "Yu seong 2"with good quality and high yielding potential are fellows;1, The plant type of "Yu seong 1" was semi-elect type but became to electtype after 60C" rediation and yield was decreased because of the number of flowers.2. The yield of "Yu seong 2" was the highest of all varieties.3. The quality(Extract and Betain contents) of "Yu seong 2" was the best ofall varieties and visible character was good.The results of this experinent showed that "Yu seong 1" should be promisinglines in south area because of resistance Anthracnose and "Yu seong 2" should bepromising lines in north area.hracnose and "Yu seong 2" should bepromising lines in north area.orth area.
