HPLC Analysis and Extraction Methods of Decursin and Decursinol Angelate in Angelica gigas Roots (당귀(Angelica gigas) 중 Decursin 및 Decursinol Angelate 추출 방법과 HPLC 분석)

  • This paper is intended as an investigation of the analysis of high-performance liquid chromatography and the method of extraction of decursin and decursinol angelate in Angelica gigas roots. There are three kinds of extraction methods: distilled water, 50% EtOH and 100% EtOH. The condition of HPLC was obtained on a reversed-phase column $(Polarity\;dC_{18},\;4.6{\times}250 mm,\;5\;{\mu}m)$ using a phosphate buffer-acetonitrile-sodium lauryl sulfate as the mobile phase. Under these chromatographic conditions, UV detector was 230 nm, column temperature $30^{\circ}C$ and the speed of a current 1.0 ml/min, respectively. The results of extraction with distilled water, 50% EtOH and 100% EtOH in Angelica gigas roots were as follows. The concentrations of decursin and decursinol angelate were 182 and 153 ppm (distilled water), 3,142 and 2,547 ppm (50% EtOH) and 3,341 and 2,778 ppm (100% EtOH). There were high positive correlations between the concentrations of decursin and EtOH (r=0.8928, p<0.01) and decursinol angelate and EtOH (r=0.9009, p<0.01).

  • A survey on the traditional management of dietary life, seasonal diets, and frequency of using traditional fermented foods, was conducted from six hundred forty housewives living in rural areas of Yosu and Yocheon district, and the following results were obtained. 1. Majority of the subjects was in the range of 50-59 years old (43.1%), elementary schooling in education (38.1%), composition of family with parents and children (57.5%) with 3-4 family members (40.6%), and annual income of 5-8 million Won. 2. Korean style-house (36.7%), modernized kitchen (58.5%), and liquid fuel (69.7%) held the majority of the living environment. 3. Most of the households possessed refrigerator (98.9%), kitchenette range (98.4%) and electric rice pot (97.9%). 4. Access to traditional dishes was mostly prompted through elders or friends (84%). Preparing a meal was regarded to be a troublesome duty (41.5%). Husband's preference was the major factor for the meal preparation (53.7%) and about 30 min was spent for preparing dinner (53.7%). 5. Cooked rice was a main staple (99.5%) while three kinds of Kimchi (55.3%) were served as prime side dishes in the diets. Most Kimchi(94.7%) was prepared at home. To this basic menu, two or three side dishes were added at breakfast (77.2%), lunch (76.1%), and dinner (65.4%). MSG (mono-sodium glutamate) was consumed by 62.2% of the households. 6. Most rural households prepared traditional dishes on the traditional holidays in lunar calendar including New Year's Day (98.9%), First Full Moon of The Year (81.4%), and Autumn Full Moon (96.8%). 7 Traditional fermented foods prepared at home comprised Kimchi (87.2%), soybean paste (75.5%), red pepper soybean paste (73.9%), and soy sauce (70.7%).

A study on the reaction of carbonation in the preparation of lithium carbonate powders (탄산리튬 분말 제조에 있어서 탄산화 반응에 관한 연구)

  • In this study, we carried out the experiment to prepare lithium carbonate powder through gas-liquid reactions with a lithium-containing solution and $CO_2$ gas using lithium hydroxide, lithium chloride, and lithium sulfate. Thermodynamically, the carbonation reaction of a lithium-containing solution showed that aqueous reaction of lithium hydroxide occurs spontaneously, but aqueous reactions of lithium chloride and lithium sulfate does not occur spontaneously. In the case of lithium hydroxide solution, the recovery rate of lithium carbonate was 69.8 % at room temperature ($25^{\circ}C$), and increased to 89.4 % at $60^{\circ}C$. In the case of lithium chloride and lithium sulfate solution, lithium carbonate could be prepared using sodium hydroxide as an additive, but the recovery rates were 19.2 % and 16.7 %, respectively.

  • Pyroprocessing at KAERI (Korea Atomic Energy Research Institute) consists of pretreatment, electroreduction, electrorefining and electrowinning. SFR (Sodium Fast Reactor) fuel is prepared from the electrowinning process which is composed of LCC (Liquid Cadmium Process) and Cd distillation et al. LCC is an electrochemical process to obtain actinides from spent fuel. In order to recover actinides inert anodes such as carbon material are used, where chlorine gas ($Cl_2$) evolves on the surface of the carbon material. And, stainless steel (SUS) crucible should be installed in large-scale electrowinning system. Therefore, the effect of chlorine on the SUS material needs to be studied. LiCl-KCl-$UCl_3$-$NdCl_3$-$CeCl_3$-$LaCl_3$-$YCl_3$ salt was contained in 2 kinds of electrolytic crucible having an inner diameter of 5cm, made of an insulated alumina and an SUS, respectively. And, three kinds of electrodes such as cathode, anode, reference were used for the electrochemical experiments. Both solid tungsten (W) and LCC were used as cathodes. Cd of 45 g as the cathode material was contained in alumina crucibles for the deposition experiments, where the crucible has an inner diameter of 3 cm. Glassy carbon rod with the diameter of 0.3 cm was employed as an anode, where shroud was not used for the anode. A pyrex tube containing LiCl-KCl-1mol% AgCl and silver (Ag) wire having a diameter of 0.1cm was used as a reference electrode. Electrodeposition experiments were conducted at $500^{\circ}C$ at the current densities of $50{\sim}100mA/cm^2$. In conclusion, Fe ions were produced in the salt during the electrodeposition by the reaction of chlorine evolved from the anode and Fe of the SUS crucible and thereby LCC system using SUS crucible showed very low current efficiencies compared with the system using the insulated alumina crucible. Anode shroud needs to be installed around the glassy carbon not to influence surrounding SUS material.

  • Electro-membrane technology is a process for separating and purifying substances in aqueous solution by electric energy using an ion exchange membrane with selective permeability, such as electrodialysis (ED) and bipolar electrodialysis (BMED). Electro-membrane technology is attracting attention as an environmental friendly technology because it does not generate by-products during the process and the recovered base or acid can be reused during the process. In this paper, we investigate the principles of ED and BMED technologies and various characteristics and problems according to the cell configuration. In particular, by investigating and analyzing research cases related to the treatment of waste sodium sulfate (Na2SO4), which is generated in large amounts during the metal recovery process.

Development of Ternary Inorganic Binder System for Manufacturing High-Functional Ceramic Molds and Core (고기능성 세라믹 주형 및 중자 제작을 위한 3원계 무기 바인더 시스템 개발)

  • In existing ceramic mold manufacturing processes, inorganic binder systems (Si-Na, two-component system) are applied to ensure the effective firing strength of the ceramic mold and core. These inorganic binder systems makes it possible to manufacture a ceramic mold and core with high dimensional stability and effective strength. However, as in general sand casting processes, when molten metal is injected at room temperature, there is a limit to the production of thin or complex castings due to reduced fluidity caused by the rapid cooling of the molten metal. In addition, because sodium silicate generated through the vitrification reaction of the inorganic binder is converted into a liquid phase at a temperature of 1,000 ℃. or higher, it is somewhat difficult to manufacture parts through high-temperature casting. Therefore, in this study, a high-strength ceramic mold and core test piece with effective strength at high temperature was produced by applying a Si-Na-Ti three-component inorganic binder. The starting particles were coated with binary and ternary inorganic binders and mixed with an organic binder to prepare a molded body, and then heat-treated at 1,000/1,350/1,500 ℃ to prepare a fired body. In the sample where the two-component inorganic binder was applied, the glass was liquefied at a temperature of 1,000 ℃ or higher, and the strength decreased. However, the firing strength of the ceramic mold sample containing the three-component inorganic binder was improved, and it was confirmed that it was possible to manufacture a ceramic mold and core via high temperature casting.

Novel stability indicating high-performance liquid chromatography method for the separation and simultaneous quantification of acalabrutinib and its impurities in pharmaceutical formulation

  • This study reports for the first time about a stability indicating RP-HPLC method for qualitative and quantitative determination of acalabrutinib in bulk and dosage form and in presence its impurities 1, 2 and 3. The chromatographic separation was carried on Zorbax XDB-C18 (250×4.6 mm; 5 µ id) as stationary phase, Phosphate buffer pH 6.4 and methanol 80:20 (v/v) as mobile phase at a flow rate of 1.0 mL/min, UV detection was carried at wavelength of 238 nm and the analysis was completed with a run time of 15 min. In these conditions the retention time of acalabrutinib and its impurities 1, 2 and 3 was observed to be 3.50, 4.83, 8.40 and 9.93 min respectively. The method was validated for system suitability, range of analysis, precision, specificity, stability and robustness. Spiked recovery at 50 %, 100 % and 150 % was carried for both standard and impurities and the acceptable % recovery of 98-102 was observed for acalabrutinib and both impurities studied and the % RSD in each spiked level was found to be less than 2. Stability tests were done through exposure of the analyte solution to five different stress conditions i.e expose to 1N hydrochloric acid, 1 N sodium hydroxide, 3 % peroxide, 80 ℃ temperature and UV radiation at 254 nm. In all the degradation condition, standard drug acalabrutinib was detected along with both the impurities studied and the degradation products were successfully separated. In the formulation analysis there is no other chromatographic detection of other impurities and formulation excipients. Hence the developed method was found to be suitable for the quantification of acalabrutinib and can separate and analyse impurities 1 and 2.

Quantitative analysis of 3-MCPD in water using LC-MS (LC-MS를 이용한 수용액중의 3-MCPD 정량)

  • The analysis method of 3-Monochloropropane-1,2-diol (3-MCPD) compound in water was developed using liquid chromatography with mass spectrometric detection. Aqueous solution was controlled in strong basic condition with sodium hydroxide, and then $25{\mu}L$ of benzoyl chloride was added to the solution for the derivatization of 3-MCPD. The derivative was extracted using pentane and analyzed by the selected ion monitoring (SIM) method of LC-MS. The results of analyses showed that the calibration curves was in the range of 1.0 to $100{\mu}g/mL$ with a good linearity (correlation coefficient of $r^2=0.992$) and limit of detection was below $0.01{\mu}g/mL$. The recoveries of this analysis method by LC-MS were 92.3-98.0 %.

Titanium alloys: A closer-look at mechanical, gamma-ray, neutron, and transmission properties of different grade alloys through MCNPcode application

  • Titanium alloys play a vital role in optimizing the effectiveness and security of nuclear reactors, strengthening structural durability, and facilitating the effective handling of nuclear waste. The aim of this study is to investigate the gamma-ray, neutron, and transmission properties of four common titanium alloys through the examination of the deposited energy amount in the liquid sodium coolant material, in relation to the mechanical properties of these alloys. MCNP (version 6.3) is utilized for designing the titanium pipes. Next, the pipes were re-designed considering the elemental mass fractions and densities of the investigated titanium alloys. Grade 26 sample is reported with the highest values of mass attenuation coefficients and the lowest HVL values among those investigated alloys. Grade 26 is reported to have the lowest TF value, whereas Grade 12 demonstrated the highest TF value. The highest Effective Removal Cross Section (ΣR, 1/cm) value against fast neutrons is reported for Grade 26. The utilization of Grade 26 sample as pipe material resulted in the lowest deposited energy amount (MeV/g) and subsequent lowest contamination in the coolant material. Out of the alloys that were chosen for analysis, it has been determined that Grade 26 exhibits the highest level of strength. It can be concluded that the Grade 26 alloy exhibits desirable characteristics for applications in nuclear technologies that require superior gamma-ray and neutron absorption properties, as well as exceptional mechanical properties. Nevertheless, it is essential to emphasize the importance for ongoing studies to enhance the existing material properties of Grade 26, with the aim of achieving improved safety and efficacy in nuclear applications.

Large scale splitter-less FFD-SPLITT fractionation: effect of flow rate and channel thickness on fractionation efficiency (대용량 중력장 SPLITT Fractionation: 분획효율에 미치는 채널 두께와 유속의 영향)

  • SPLITT fractionation (SF) allows continuous (and thus a preparative scale) separation of micronsized particles into two size fractions ('fraction-a' and 'fraction-b'). SF is usually carried out in a thin rectangular channel with two inlets and two outlets, which is equipped with flow stream splitters at the inlet and the outlet of the channel, respectively. A new large scale splitter-less gravitational SF (GSF) system had been assembled, which was designed to eliminate the flow stream splitters and thus is operated by the full feed depletion (FFD) mode (FFD-GSF). In the FFD mode, there is only one inlet through which the sample is fed. There is no carrier liquid fed into the channel, and thus prevents the sample dilution. The effects of the sample-feeding flow rate, the channel thickness on the fractionation efficiency (FE, number % of particles that have the size predicted by theory) of FFD-GSF was investigated using industrial polyurethane (PU) latex beads. The carrier liquid was water containing 0.1% FL-70 (particle dispersing agent) and 0.02% sodium azide (used as bactericide). The sample loading rate was varied from about 4 to 7 L/hr with the sample concentration fixed at 0.01%. The GSF channel thickness was varied from 900 to $1300{\mu}m$. Particles exiting the GSF channel were collected and monitored by optical microscopy (OM). Sample recovery was monitored by collecting the fractionated particles on a $0.45{\mu}m$ membrane filter. It was found that FE of fraction-a was increased as the channel thickness increases, and FE of fraction-b was increased as the flow rate was increased. In all cases, the sample recovery has higher than 95%. It seems the new splitter-less FFD GSF system could become a useful tool for large scale separations of various types of micron-sized particles.
