Inhalation Toxicity of Particulate Matters Doped with Arsenic Induced Genotoxicity and Altered Akt Signaling Pathway in Lungs of Mice

  • In the workplace, the arsenic is used in the semiconductor production and the manufacturing of pigments, glass, pesticides and fungicides. Therefore, workers may be exposed to airborne arsenic during its use in manufacturing. The purpose of this study was to evaluate the potential toxicity of particulate matters (PMs) doped with arsenic (PMs-Arsenic) using a rodent model and to compare the genotoxicity in various concentrations and to examine the role of PMs-Arsenic in the induction of signaling pathway in the lung. Mice were exposed to PMs $124.4{\pm}24.5\;{\mu}g/m^3$ (low concentration), $220.2{\pm}34.5\;{\mu}g/m^3$ (middle concentration), $426.4{\pm}40.3\;{\mu}g/m^3$ (high concentration) doped with arsenic $1.4\;{\mu}g/m^3$ (Low concentration), $2.5\;{\mu}g/m^3$ (middle concentration), $5.7\;{\mu}g/m^3$ (high concentration) for 4 wks (6 h/d, 5 d/wk), respectively in the whole-body inhalation exposure chambers. To determine the level of genotoxicity, Chromosomal aberration (CA) assay in splenic lymphocytes and Supravital micronucleus (SMN) assay were performed. Then, signal pathway in the lung was analyzed. In the genotoxicity experiments, the increases of aberrant cells were concentration-dependent. Also, PMs-arsenic caused peripheral blood micronucleus frequency at high concentration. The inhalation of PMs-Arsenic increased an expression of phosphorylated Akt (p-Akt: protein kinase B) and phpsphorylated mammalian target of rapamycin (p-mTOR) at high concentration group. Taken together, inhaled PMs-Arsenic caused genotoxicity and altered Akt signaling pathway in the lung. Therefore, the inhalation of PMs-Arsenic needs for a careful risk assessment in the workplace.

Two Cases of Sodium Bicarbonate Inhalation Therapy in Chlorine Gas Intoxication (염소 가스 중독에서 나트륨 중탄산염의 흡입치료 2례)

  • A chlorine gas is a common irritant and when exposed, it result in mild occular, oropharyngeal, or respiratory symptoms. In severe case, however, it may result in pulmonary edema, interstitial pneumonia, or respiratory failure. We report the case of 29-year-old and 46-year-old men is accidentally exposed to chlorine gas during cleaning water. The patients complained dyspnea, chest tightness, cough and both eye pain. During hospitalization, they were treated with inhalation of humidified oxygen, beta-adrenergic agonist and $2\%$ sodium bicarbonate. After several days, patients were discharged without respiratory symptoms and complication. In treatment of chlorine gas toxicity the inhalation of sodium bicarbonate is a possible initial therapy can improve respiratory symptoms in spite of lack of evidence.

  • According to the epidemiological studies in chromium workers, hexavalent chromium is associated with the risk of lung cancer. Reactive oxygen species produced by hexavalent chromium exposure may play an important role in the carcinogenesis process. We measured one kind of oxidative DNA damages, 8-hydroxydeoxyguanosine (8-OHdG) and its repair activities as an indicators of cellular oxidative stress in rat lungs by the repeated inhalation exposure to sodium chromate.(omitted)

  • From the harmfulness expectation test conducted through a toxicity anticipation program, methylcyclohexane turned out to be harmful and simulative, but no carcinogenicity was anticipated. In a four-hour acute inhalation toxicity test, the result showed that lethal concentration ($LC_{50}$) was 3,750 ppm (15,054 mg/L), which was identified as a harmful substance on the basis of the harmful substance classification standard $2 of the Industrial safety and health law. methylcyclohexane fell under the category $4(2,500 substance from the GHS standard acute toxicity harmfulness classification. Also, from subchronic inhalation toxicity test that included 6 hours a day, five days a week, and for 13 weeks, we could observe weight, activity, long term weight, blood and blood biochemical influence from the exposure of test substance. No-observed effect level (NOEL) was determined below $100{\sim}400ppm$ inboth male and female. This material falls under the Category 2 ($50{\sim}250ppm/6hours/90days$) in the GHS (Globally Harmonized System) standard trace long-term whole body toxicity repeated exposure, and can be classified as a harmful substance in accordance with the Industrial Safety and Health Law harmful substance standard $NOEL{\leq}0.5mg/L/6hr/90day$ (rat).

Genome-Wide Transcriptional Response During the Development of Bleomycin-Induced Pulmonary Fibrosis in Sprague-Dawley Rats

  • Pulmonary fibrosis is a common consequence of many lung diseases and a leading cause of morbidity and mortality. The molecular mechanisms underlying the development of pulmonary fibrosis remain poorly understood. One model used successfully to study pulmonary fibrosis over the past few decades is the bleomycin-induced pulmonary fibrosis model. We aimed to identify the genes associated with fibrogenesis using an Affymetrix GeneChip system in a bleomycin-induced rat model for pulmonary fibrosis. To confirm fibrosis development, several analyses were performed, including cellular evaluations using bronchoalveolar lavage fluid, measurement of lactate dehydrogenase activity, and histopathological examinations. Common aspects of pulmonary fibrosis such as prolonged inflammation, immune cell infiltration, emergence of fibroblasts, and deposition of extracellular matrix and connective tissue elements were observed. Global gene expression analysis revealed significantly altered expression of genes ($\geq$ 1.5-fold, p < 0.05.) in a time-dependent manner during the development of pulmonary fibrosis. Our results are consistent with previous results of well-documented gene expression. Interestingly, the expression of triggering receptor expressed on myeloid cells 2 (Trem2), secreted phosphoprotein 1 (Spp1), and several proteases such as Tpsab1, Mcpt1, and Cma1 was considerably induced in the lung after bleomycin treatment, despite little evidence that they are involved in pulmonary fibrogenesis. These data will aid in our understanding of fibrogenic mechanisms and contribute to the identification of candidate biomarkers of fibrotic disease development.

Comparison of Distribution and Inflammatory Response by Diameter and Shape of Silver Nanoparticles (은나노 입자의 입경 크기 및 형태에 따른 체내 동태 및 염증 반응)

  • The market size of engineered nanoparticles is rapidly increasing due to the fast application of nanotechnologies into different industries and consumer products. The development of new technology and materials has improved human's quality of life, but it also entails the possibility of exposure to new materials. In this study, we compared the distribution in the body by the inflow of silver nanoparticles having another diameter and shape at 1 h or 24 h after injection via the tail vein. And, we compared the cell composition and cytokine concentration in BAL fluid, and histopathological changes. As results, discharge of silver nanoparticles having small diameter and sphere shape was more rapid than that of big diameter or plate shape. It is estimated that the toxicity in liver and lung was proportional to accumulation level. The persistence of inflammation was also longer in mice treated with plate shape. Consequently, we suggest that the first choice of silver nanoparticles having small diameter and sphere shape in applying is desirable.

Assessment of Inhalation Exposure to Volatile Disinfection By-products Associated with Household Uses of Chlorinated Tap Water (가정에서의 수돗물 사용과 관련된 휘발성 염소소독부산물에 대한 흡입노출 평가)

  • Volatile disinfection by-products (DBPs) contained in chlorinated tap water are released into household air during indoor activities (showering, cooking, dish -washing, etc.) associated with tap water uses and may cause adverse health effects on humans. Twenty seven subjects were recruited and their homes were visited during the winter of 2002. Tap water, household air, and exhaled breath samples were collected and analyzed for five volatile DBPs (chloroform, bromodichloromethane, dichloroacetonitrile, 1,1 -dichloropropanone and 1,1,1 trichloropropanone). Chloroform was a major DBP found in most samples. Tap water chloroform concentrations were not statistically correlated with its household air concentrations, probably due to individual variability in indoor activities such as showering, cooking, and dish - washing as well as household ventilation. Correlation of breath chloroform concentration with household air chloroform concentration showed its possible use as a biomarker of exposure to household air chloroform. Exposure estimates suggested that inhalation during household stay be a major route of exposure to volatile DBPs and that ingestion of tap water be a trivial contributor to the total exposure in Koreans.

  • Cyanides and Hydrogen cyanide are used in production of chemicals, electroplating, photographic development, making plastics, fumigating ships, and some mining processes. We experienced of Hydrogen cyanide inhalation injury. A 45-year-old man worked at electroplating. As soon as he mixed sodium cyanide with acid, cyanide produced hydrogen cyanide, occasionally he inhaled Hydrogen cyanide and he lost his consciousness. He was moved to near hospital and took emergency treatment and then was transferred to our hospital. On arrival he had severe dyspnea, metabolic acidosis, and tachycardia. After he was treated with supportive method he recovered his consciousness in 20 hours at emergency room.

Prioritization of Chemicals for Chronic/Carcinogenic Inhalation Testing (만성발암성 흡입독성시험 우선순위 물질 선정 연구)

  • Objectives: In this study, we seek to perform a priority selection for test substances for chronic inhalation toxicity studies, including acute and subchronic inhalation toxicity studies, which are to be performed after the construction of a chronic/carcinogenicity inhalation toxicity study facility and enactment of pertinent legislation. Methods: Through this study, qualitative and quantitative priority evaluation of test substances according to acute, subchronic and chronic categories were respectively performed and priorities were suggested by expert group review, redundancy and other methods. Meanwhile, a draft on test substance selection criteria, procedures and methods referring to the National Toxicology Program (NTP) system was proposed. Results: This study selected priorities for candidate substances for chronic inhalation toxicity studies to be conducted from 2016. Conclusions: In the future, by assessing in advance the toxicological effects of chemicals to which workers can be potentially exposed in the workplace via long-term inhalation, expected health disturbances among workers will be reduced and it is anticipated that occupational disease induced by chemicals will be effectively prevented.
