Various antibiotics, that could induce the appearance of resistant microorganisms, have been used for treatment or prevention of bacterial diseases in marine and ornamental fish. We determined and characterized the level of antibiotic-resistant bacteria and proportion of multi-drug resistant bacteria in intestinal microflora of both marine fish cultured in Korea and imported ornamental freshwater fish. For this the bacterial species and resistance to antibiotics were investigated in intestine of rock bream Oplegnathus fasciatus cultured in Korea and pearl gourami Trichogaster leeri imported from Singapore to characterise. Although the bacterial species were different, proportions of resistant bacteria to single antibiotics or multi-drug were higher in intestinal microflora of pearl gourami Trichogaster leeri imported from Singapore than in rock bream Oplegnathus fasciatus cultured in Korea. These results indicate that various antibiotics have been being used before trading without measures in the market of asian ornamental fishes, providing high risks for the emergence of multi-drug resistant bacteria.

Imported ornamental fish should be quarantined because they can have dangerous diseases depending on their habitat. The quarantine requires a lot of time because quarantine officers collect various information on the imported ornamental fish. Inefficient quarantine processes reduce its work efficiency and accuracy. Also, long-time quarantine causes the death of environmentally sensitive ornamental fish and huge financial losses. To improve existing quarantine systems, information on ornamental fish was collected and structured, and a server was established to develop quarantine performance support software equipped with a text search engine. However, the long names of ornamental fish in general can cause many typos and time bottlenecks when we type search words for the target fish information. Therefore, we need a technique that can correct typos. Typical typo character calibration compares input text with all characters in a calibrated candidate text dictionary. However, this approach requires computational power proportional to the number of typos, resulting in slow processing time and low calibration accuracy performance. Therefore, to improve the calibration accuracy of characters, we propose a fusion system of simple Artificial Neural Network (ANN) models and character preprocessing methods that accelerate the process by minimizing the computation of the models. We also propose a typo character generation method used for training the ANN models. Simulation results show that the proposed typo character correction system is about 6 times faster than the conventional method and has 10% higher accuracy.

Background: Live fish import may lead to the unintended introduction of pathogens. We examined the monthly distribution of microbial pathogens in ornamental finfish imported into South Korea over a 6-month period. Results: Vibrio alginolyticus was detected in one lemon damsel in June and July; V. vulnificus was detected in one lemon damsel, one caerulean damsel, and one pearl-spot chromis and one ocellaris clownfish in July, April, and May, respectively; Photobacterium damselae was detected in one ocellaris clownfish and one caerulean damsel in June and July, respectively; V. anguillarum was detected in one pearl-spot chromis in February; V. harveyi was detected in one ocellaris clownfish and two mandarin fish in February and April, respectively; Yersinia ruckeri was detected in a pearlscale goldfish group in June and July and in two colored carp groups in July; and Lactococcus garvieae was detected in a lemon damsel group and a sutchi catfish group in July and May, respectively. European catfish virus, the only viral pathogen detected, was found in two sutchi catfish groups in May. Conclusion: This study is the first to identify pathogenic species and the presence or absence of pathogens (non-quarantine diseases) in imported ornamental finfish. These results demonstrate that various pathogens with the potential to harm indigenous fish populations can accompany ornamental finfish imported into South Korea.
In this study, imported ornamental goldfish (Oranda and black moor) with high mortality was investigated with histopathological analysis. Fishes showed clinical symptoms characterized by gill necrosis and cumulative mortality ranging from 60% to 90% within a month after import from Thailand and China in 2021. CyHV-2 was detected in gill, kidney, spleen and liver of goldfishes. Histopathological analysis revealed epithelial necrosis, intranuclear inclusion bodies associated with viral symptoms in epithelial cell, macrophage infiltration and vacuolation in secondary lamella. In addition, disseminated granuloma formation was observed in various organs such as kidney, heart and mesenchymal membranes. These lesions refer to coinfections with CyHV-2 and other pathogens that cause chronic inflammation, which leads to mortality. Therefore, early disease monitoring needs to reduce the mortality when ornamental goldfish is imported.
In this study, the molecular structures of tet(M) and tet(G) carried by tetracycline (Tc) resistant bacteria in intestinal microflora from the imported ornamental fish were characterized and compared with each other depend on the imported countries. Of the total isolates, approximately 8.9% of the Ent-lac+(lactose fermentative bacteria on coliform media) Tc resistant isolates in fish from three different countries, Singapore, Taiwan and Brazil, were appeared to contain tet(M). Three representative isolates of different countries, Aeromonas spp. JSM-1 (Singapore), JTM-1 (Taiwan) and JBM-1 (Brazil), were isolated and analyzed the molecular structures of tet(M) gene. Interestingly, partial sequence of tet(M) genes (1099 bp) in JBM-1 (Brazil) showed 99.5% homology with the tet(M) found in the Vibrio spp. RV16 isolate, obtained from marine fish in Korea and known to carry Tn1545 parent type of tet(M). In contrast, tet(M) gene in JSM-1 and JTM-1 showed mosaic structure of Tn1545 and Tn916, and 100% homology with each other. It may suggest the presence of various characteristics in terms of tet(M) gene structure. The determined sequence of the tet(G) from Aeromonas spp. JSG-1 and JBG-1 isolated from Singapore and Indonesia ornamental fish respectively showed similar nucleotide sequence homology but revealed a few nucleotide changes in comparison with the sequence of the prototype tet(G) gene (S52437 in GenBank).
현재 국내에 유통되고 있는 담수관상어의 대부분은 수입에 의존하므로 외래 병원체 및 항생제 내성균의 유입 가능성이 크다. 본 연구에서는 담수에 상재하는 균으로 인수공통 감염이 가능하며 어류에 조건성병원균으로 작용하는 Aeromonas hydrophila를 다양한 수입 담수관상어로부터 분리하여 특성을 분석하고자 하였다. 수입 후 1일 이내의 관상어를 수족관으로부터 수집하여 0.5% NaCl을 포함하는 TSA 배지에 간, 신장, 비장을 도말하여 세균을 분리하였다. 세균의 형태, 생화학적 특성 및 starch- ampicillin agar (SA)에서의 clear zone 형성 유무에 근거하여 분리 균의 약 70%인 226 균주가 Aeromonas spp.로 확인되어 이들의 분리 빈도가 매우 높음을 알 수 있었다. 이들 중 9균주를 선택하여 16S rRNA gene의 sequence를 분석한 결과 모두 A. hydrophila로 동정되었으며 API 20E test에서 대부분의 분리 균주는 amygdalin, D-melibiose, D-sucrose, trisodium citrate, L-arabinose 이용에서 표준 균주와 차이를 보였다. 10종의 항균제에 대한 분리 균주의 감수성을 디스크 확산법으로 확인한 결과 amoxycillin, ampicillin에 대해서는 모든 균주가 저항성이었고 florfecicol에 대해서는 모두 감수성이었다. 그러나 7균주는 tetracycline 계통의 항균제와 erythromycin, nalidixic acid 등에 복합내성을 보였다. A. hydrophila로 동정된 9개 분리 균주를 금붕어에 인위 감염을 실시한 결과, 3 균주는 5일 이내에 60~80%의 폐사를 나타내었다. 이러한 결과는 다약제내성이면서 독력이 있는 Aeromonas가 수입관 상어를 따라 국내로 유입되어 전파될 수 있으므로 수입상에서 소비처로 판매되기 전의 방어적 조치가 필요하다는 것을 시사한다.
We report on mycobacteriosis in an imported tropical ornamental fish Macropodus opercularis commonly known as the paradise fish. Mass mortality occurred in paradise fish imported to Korea from Southeast Asia in 2008. The affected fish did not show any outward clinical signs, but enlargement of the spleen, kidneys, and liver was observed on dissection. Histopathological examination revealed numerous granulomas in the spleen, and acid-fast bacilli were observed in the centers of the granulomas. About 65% of spleen DNA samples were PCR positive using mycobacteria-specific primers targeting the 16S rRNA and hsp65 genes. The nucleotide identities of the 16S rRNA and hsp65 genes with those of Mycobacterium marinum were 99.5% and 99.4%, respectively. Although the bacterium was not cultured, the molecular diagnosis and histopathological findings were consistent with mycobacteriosis in paradise fish.
The presence of ISKNV-like viruses in various freshwater ornamental fish species imported from Asia was confirmed by polymerase chain reaction(PCR) amplification of the ATPase(adenosine triphosphatase) gene. Interestingly, molecular analyses of the Open Reading Frame 25(ORF25) region of these isolates based on the ISKNV(Infectious spleen and kidney necrosis virus) genome revealed the presence of various repetitive sequences. ORF25 repeat sequence length had no effect on cumulative mortality of rock bream Oplegnathus fasciatus challenged with tissue homogenates of infected pearl gourami, Trichogaster leeri; silver gourami, Trichogaster microlepis; blue gourami, or Trichogaster trichopterus. All isolates induce cumulative mortalities after 12 days of infection, confirming that ORF25 polymorphism did not affect the pathogenicity of ornamental fish megalocytiviruses that cross infect rock bream, a seawater fish. Also, no statistically significant differences in spleen index or viral copy number in infected tissues was detected between isolates with varying ORF25 repeat sequence lengths. However, further studies are necessary to fully characterize the functional characteristics of these polymorphisms in megalocytivirus disease in ornamental fishes.
Goldfish hematopoietic necrosis (GHN) affects hematopoietic organs and causes high mortality in goldfish. The causative agent of GHN is cyprinid herpesvirus 2 (CyHV-2). The purpose of this study was to detect CyHV-2 in ornamental cyprinid fish. CyHV-2 monitoring was conducted on a monthly basis for 1 year using goldfish Carassius auratus and pearlscale goldfish Carassius auratus that had been cultured at a domestic aquafarm and imported from Singapore, respectively. The results of polymerase chain reaction (PCR) assays using specific primers for the CyHV-2 helicase gene showed that 27.8% of goldfish and 33.3% of pearlscale goldfish were positive for CyHV-2. No cytophatic effects were detected in koi fin or common carp brain cells inoculated with tissue homogenates from either fish. Our data provides the useful data for establishing future quarantine and disinfection policies.
2004~2005년까지 수입산 pearl gourami (Trichogaster leeri), dwarf gourami (Colisa lalia), silver gourami (Trichogaster microlepis), blue gourami (Trichogaster trichopterus sumatranus) 그리고 freshwater angelfish (Pterophyllum scalare) 등 5 어종을 대상으로 iridovirus 감염 실태를 PCR 방법을 사용하여 조사하였다. 채집한 56 samples 중 11 samples에서 PCR 양성 반응이 관찰되었고, pearl gourami에서 31.8% (7/22)로 가장 높은 감염 빈도를 나타내었다. 그 다음으로 dwarf gourami, blue gourami, silver gourami 순으로 나타났으며, freshwater angelfish에서는 iridovirus가 검출되지 않았다. 자연 감염된 관상어종의 비장으로부터 iridovirus의 핵산을 분리하여 검출한계 PCR 방법으로 viral DNA의 양을 비교하였으며, dwarf gourami에 비해 pearl gourami에서 더 많은 viral DNA가 검출되는 것을 확인하였다. 자연 감염이 확인된 pearl gourami groups는 실험실 환경에서 100% 폐사하였고, 폐사어 중 57% 이상이 PCR 양성 반응을 나타내었다. PstⅠfragment의 염기서열 비교에서 pearl gourami와 dwarf gourami에서 분리한 iridoviruses는 중국의 mandarinfish (Siniperca chuatsi)에서 분리된 infectious spleen and kidney necrosis virus (ISKNV)와 99% 이상의 매우 높은 상동성을 나타내었다.
