Inorganic nanomaterial-based biocatalysts

  • Over the years, nanostructures have been developed to enable to support enzyme usability to obtain highly selective and efficient biocatalysts for catalyzing processes under various conditions. This review summarizes recent developments in the nanostructures for enzyme supporters, typically those formed with various inorganic materials. To improve enzyme attachment, the surface of nanomaterials is properly modified to express specific functional groups. Various materials and nanostructures can be applied to improve both enzyme activity and stability. The merits of the incorporation of enzymes in inorganic nanomaterials and unprecedented opportunities for enhanced enzyme properties are discussed. Finally, the limitations encountered with nanomaterial-based enzyme immobilization are discussed together with the future prospects of such systems.

Affinity Immobilization of Dextransucrase on Dextran-based Support and the Production of Leucrose

  • A simple and convenient method of immobilizing dextransucrase via an affinity interaction is described, along with the use of this system to synthesize leucrose. Dextransucrase was produced in sucrose-free medium by fermenting a constitutive mutant of Leuconostoc mesenteroides NRRL B-512F and was separated using an ultrafiltration membrane. The purified enzyme was free of dextran polymer, which previously was always found with the sucrose-induced enzyme. Therefore, it was possible to immobilize the enzyme on dextran-based resins using an affinity interaction. Sephadex G-200 was the best resin for immobilizing the dextransucrase and gave a fast flow rate through the packed column. The immobilized dextransucrase retained more than 80% of its specific activity after immobilization ($K_m\;=\;18.1\;mM$ and $k_{cat}\;=\;450\;sec^{-1}$ vs. 13.1 mM and $640\;sec^{-1}$, respectively, for the free enzyme). The immobilized dextransucrase showed improved stability over a pH range of 4.0 to 6.5 and at moderately high temperatures over $40^{\circ}C$. When immobilized dextransucrase was used to synthesize leucrose via the transfer reaction with sucrose and fructose, about 74% of the sucrose was converted into leucrose after one day, and the half-life of the enzyme activity was 15 days. Regeneration of the resin by supplementation with dextransucrase enabled the recovery of the initial activity of the system, but both the reaction and the flow rate were lower, probably owing to the accumulation of dextran inside the resin.

  • $TiCl(O-i-Pr)_3/SiO_2$ and $Ti(O-i-Pr)_4/SiO_2$ were prepared by immobilizing chlorotitanium (IV) isopropoxide ($TiCl(O-i-Pr)_3$) and titanium (IV) isopropoxide ($Ti(O-i-Pr)_4$), to pretreated silica. The effect of the polymerization reaction conditions on the catalytic activity and characteristics of the resulting PLA were investigated. The catalytic conversion, molecular weight and polydispersity index (PDI) of the PLA produced on the titanium alkoxide supported catalysts increased proportionally with the reaction temperature. When the PLA was synthesized in bulk polymerization, the PLA produced with the supported catalysts had higher molecular weight than those with homogeneous catalysts. The melting temperature of the polymer produced with silica supported alkoxide catalysts was approximately $170-180^{\circ}C$.

  • Biochips are a powerful emerging technology for biomedical, environmental applications. Especially, making use of bioseonors in the evaluation of toxicity becomes increasingly important. For biosensor as a toxicity detection, biomolecules like antibodies or aptamers have been developed to specifically capture the toxic target molecules. In addition, the development of optimal chip materials capable of maintaining the activity of embedded biomolecules such as proteins or aptamers has proven challenging. Here, using sol-gel materials, new chip material, whose ability for immobilizing the embedded aptamers and maintaining the ability of embedded aptamers is optimal, was searched. We used sol-gel formulation screening methods previously developed and found the best formulation which shows high sensitive and specific interactions of aptamers. This study results will support the technological advancement for diagnosis and environmental sensor.

  • A self-assembled monolayer of protein G was fabricated to develop an immunosensor based on surface plasmon resonance (SPR), thereby improving the performance of the antibodybased biosensor through immobilizing the antibody molecules (lgG). As such, 11-mercaptoundecanoic acid (11-MUA) was adsorbed on a gold (Au) support, while the non-reactive hydrophilic surface was changed through substituting the carboxylic acid group (-COOH) in the 11-MUA molecule using 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrocholide (EDAC). The formation of the self-assembled protein G layer on the Au substrate and binding of the antibody and antigen were investigated using SPR spectroscopy, while the surface topographies of the fabricated thin films were analyzed using atomic force microscopy (AFM). A fabricated monoclonal antibody (Mab) layer was applied for detecting E. coli O157:H7. As a result, a linear relationship was achieved between the pathogen concentration and the SPR angle shift, plus the detection limit was enhanced up to 10$^2$ CFU/mL.

방사선중합체로 고정화된 Rhizopus oryzae의 유산생성 (Production of L(+)-Lactic Acid by Rhizopus oryzae after Immobilization in Polymer Supports with Gamma-ray Induced Polymerization)

  • 방사선 중합체로 고정화한 Rhizopus oryzae의 유산생성 방사선 공중합 고분자로 Rhizopus oryzae를 흡착 고정화하였다. 이 고정화 균체는 고정화하지 않은 생균체보다 반응시간도 짧았으며 제품수율도 아주 높았다. 즉 글루코스를 주 탄소원으로 한 경우 고정화 하지 않은 균체 배양은 반응후 96시간에 탄소원을 거의 소비하고 유산생성 수율이 약60%에 다다랐으나 고정화 균체는 반응후 48시간 안에 수율이 약 88%이상으로 되었다. 또 중합체 중 모노머의 비율이 5-10%의 것이 고정화에 좋은 결과를 보였다.

광촉매 반응기용 세라믹 막에의 TiO2 층 형성과 성능평가 (In-situ TiO2 Formation and Performance on Ceramic Membranes in Photocatalytic Membrane Reactor)

  • 메조포러스 공극구조를 갖는 광촉매 멤브레인은 다양한 환경기술에 적용될 수 있다. 본 연구에서는 $TiO_2$ 층을 형성시킨 광촉매 반응기용 세라믹 멤브레인을 개발하고 이를 염색용액 처리에 적용하였다. 높은 공극률과 균질성을 지닌 $TiO_2$ 광촉매층을 그라프트 공중합체를 사용하여 제조하였다. 멤브레인은 광촉매 반응기와 멤브레인 여과를 결합시킨 하이브리드 광촉매 반응기에 성공적으로 적용하였다. 실험결과 정렬된 구조의 $TiO_2$ 층이 $Al_2O_3$ 지지체에 형성되었다. $TiO_2$ 층 형성 후 제조된 세라믹 분리막의 순수 투과도는 형성된 광촉매 층 저항으로 감소하였다. 정렬된 구조의 $TiO_2$ 층은 UV 결합 시 5시간 안에 완벽한 염색용액 분해를 달성시킬 수 있었다. 광촉매 멤브레인의 염색용액 분해는 Langmuir-Hinshelwood 흡착 모델로 잘 설명할 수 있었다. 또한 $TiO_2$ 층이 고정화된 세라믹 멤브레인의 model Congo Red에 대한 1차 속도상수는 $Al_2O_3$ 지지체 단독인 경우에 비해 약 6배 정도 큰 값을 나타내었다(0.0081 vs. $0.0013min^{-1}$).

치주수술후 치주포대의 사용유무에 따른 임상적 효과에 대한 비교논문 (A Comparative Study of Clinical Sffects Following Periodontal Surgery with and without Dressing)

  • Since they were introduced by Ward in 1923, periodontal dressing have been routinely used following the periodontal surgery to avoid pain, infection, desensitizing teeth, inhibiting food impaction of the surgical areas, and immobilizing injured areas. Recently, however, the value of periodontal dressings and their effects on periodontal wound healing have been questioned, several authors have been reported that the use of dressing has little influence on healing following periodontal surgical procedures. In addition, there is evidence that when good flap adaptation is achieved, the use of a periodontal dressing does not add to patient comfort nor promote healing. The purpose of this study was to evaluate patient postoperative pain experience and discomfort with and without the use of periodontal dressing following periodontal surgery. Twenty-eight patients, 11 male and 17 female. were selected for this study; The age range was 31 to 56, with an average of 40.2years. Patient selection was based on existence of two bilateral sites presenting similar periodontal involvement, as determined by clinical and radiographic assessment, and requiring comparable bilateral surgical procedures. Using a splitmouth dressing. one site received a periodontal dressing while the other site did not. Pain assessment was made according to a horizontal, rating scale(0-10). After at least a two-week period, the second surgical precedure was performed using the alternate postoperative treatment. At the conclusion of the trial, a self-administered questionnaire on postoperative experience was administered, and were asked of their preference of either, dressed or undressed. The results were as follows: 1. A similar trend for mean pain and discomfort scores as assessed by patients both dressed and salinetreated procedures was evident during 7-day postsurgical period. 2. Statistical analysis of differences between the dressed side and salinetreated side with respect to pain, discomfort and patient's experiences revealed that both treatment sides behaved similarly at any postoperative day(P>0.05). 3. Considering the patient's preference, on the basis of pain and discomfort experienced, 43% preferred the saline-mouthwash and 32% preferred the dressing, 25% showing no preference for either the dressing or the saline-mouthwash. There is evidence to support the use of a periodontal dressing in retention of an apically positioned flap by preventing coronal displacement, or its use to provide additional support to stabilize a free gingival graft. However, there will always be a use for periodontal dressing although routine use of dressings may decrease because of better surgical techniques and the use of antibacterial mouth rinses.

Chlorella pyrenoidosa에 의한 납 흡.탈착시 고정화 방법의 영향 (Effect of Immobilization Method in the Biosorption and Desorption of Lead by Algae, Chlorella pyrenoidosa)

  • 본 연구는 조류 Chlorella pyrenoidosa에 의한 Pb 흡착시 고정화 방법이 흡착특성에 미치는 영향에 관한 것으로, 고정화는 Ca-alginate, K-carrageenan, and polyacrylamide를 이용하여 실시하였다. 연구결과, 고정화에 따라 흡착평형 도달시간의 지연이 나타났으며, Pb의 최대 흡착능(mg/g)은 중금속 농도와 pH가 높을수록, 고정체 투입량이 적을수록 높게 나타났다. 고정화 방법별 흡착능은 Ca-alginate로 고정화한 경우 가장 높은 흡착능을 나타낸 반면, 흡착량이 pH에 가장 민감하게 영향을 받았다. Pb 흡착현상은 Freundlich 등온흡착식에 의해 잘 해석되었고, 흡착속도는 2차 모형식을 따르는 것으로 나타났다. FT-IR분석으로 Chlorella pyrenoidosa에 의한 Pb의 생물흡착 메커니즘이 주로 carbo-acid와 amide 그룹에 Pb가 결합되어 일어남을 알 수 있었다. 흡착된 Pb의 탈착은 탈착제 농도가 높을수록 탈착 효율도 높았다. 실험에 사용되어진 여러 탈착제(NTA, HCl, EDTA, $H_2SO_4,\;Na_2CO_3$)중 Ca-alginate과 K-carrageenan로 고정화한 경우 NTA가 가장 높게 나타났으며, polyacrylamide로 고정한 경우 EDTA를 사용한 경우 가장 효과적인 것으로 나타났다. 고정화 방법별 최대 탈착효율은 각각 90.0, 83.0, 80.0% 이었다.
