A Simple, Single Triplex PCR of IS6110, IS1081, and 23S Ribosomal DNA Targets, Developed for Rapid Detection and Discrimination of Mycobacterium from Clinical Samples

  • Tuberculosis (TB) is the most common mycobacterial infection in developing countries, requiring a rapid, accurate, and well-differentiated detection/diagnosis. For the rapid detection and discrimination of Mycobacterium tuberculosis complex (MTC) from non-tuberculous mycobacteria (NTM), a novel, simple, and primer-combined single-step multiplex PCR using three primer pairs (6110F-6110R, 1081F-1081R, and 23SF-23SR; annealing on each of IS6110, IS1081, and 23S rDNA targets), hereafter referred to as a triplex PCR, has been developed and evaluated. The expected product for IS6110 is 416 bp, for IS1081 is 300 bp, and for 23S rDNA is 206 bp by single PCR, which was used to verify the specificity of primers and the identity of MTC using DNA extracted from the M. tuberculosis H37Rv reference strain (ATCC, USA) and other mycobacteria other than tuberculosis (MOTT) templates. The triplex PCR assay showed 100% specificity and 96% sensitivity; the limit of detection for mycobacteria was ~100 fg; and it failed to amplify any target from DNA of MOTT (50 samples tested). Of 307 blinded clinical samples, overall 205 positive M. tuberculosis samples were detected by single PCR, 142 by conventional culture, and 90 by AFB smear methods. Remarkably, the triplex PCR could subsequently detect 55 positive M. tuberculosis from 165 culture-negative and 115 from 217 AFB smear-negative samples. The triplex PCR, targeting three regions in the M. tuberculosis genome, has proved to be an efficient tool for increasing positive detection/discrimination of this bacterium from clinical samples.

Top Emission Organic EL Devices Having Metal-Doped Cathode Interface Layer

  • Top emission organic EL devices were fabricated by using metal-doped cathode interface layer to achieve low drive voltages. Also, facing-targets-type sputtering was used to sputter indium-tin oxide layer on top of organic active layer. The devices fabricated in this study showed reasonably high external quantum efficiency of about 1 % which is comparable to that of bottom-emission-type devices.

  • Mathematical analysis is made on a mesh free method for the compressible Euler equations. In particular, the Moving Least Square Reproducing Kernel (MLSRK) method is employed for space approximation. With the backward-Euler method used for time discretization, existence of discrete solution and it's $L^2-error$ estimate are obtained under a regularity assumption of the continuous solution. The result of numerical experiment made on the biconvex airfoil is presented.

  • Generally, biped walking is difficult to control because a biped robot is a nonlinear system with various uncertainties. In this paper, we propose a hybrid sliding-mode control method using a WNN uncertainty observer for stable walking of the 5-link biped robot with model uncertainties and the external disturbance. In our control system, the sliding mode control is used as main controller for the stable walking and a wavelet neural network(WNN) is used as an uncertainty observe. to estimate uncertainties of a biped robot model, and the error compensator is designed to compensate the reconstruction error of the WNN. The weights of WNN are trained by adaptation laws that are induced from the Lyapunov stability theorem. Finally, the effectiveness of the proposed control system is verified through computer simulations.

A Pair Dipole Antenna with Double Tapered Microstrip Balun for Wireless Communications

  • In this paper a printed pair dipole antenna with double tapered microstrip balun for wireless communications is proposed. The proposed antenna consists of a pair arm of different sizes that is branched microstrip line and microstrip line with the ground plane on opposite side of the dielectric substrate plane. The proposed antenna is matched between the ground plane to the microstrip line by double tapered microstrip balun. This antenna obtains multi-band radiation frequency band. The impedance bandwidths for a reflection coefficient of VSWR ≤ 2 are about 1.01 GHz (2.35~3.336 GHz), 1.56 GHz (4.7~6.26 GHz) and 1.15GHz (6.85~8.0[GHz]). Additionally, the measurement peak gain is about 3.6 dBi. The proposed antenna is able to support wireless communication applications.

  • The measurement and control device which is for controlling the best mixture rate is needed to produce the best quality concrete in Batch Plant. Present the workers do the measurement and control with each equipped devices such as a indicator, PLC and a scale. That is difficult to use especially for beginner in field. In this paper, we developed the integrated measurement and control system which is composed of hardware with PCB and software. It is easy to use for beginner worker in plant field.

  • In this paper, a steady axisymmetric MHD flow of two dimensional incompressible fluids is studied under the influence of a uniform transverse magnetic field. The governing equations are reduced to nonlinear boundary value problem by applying the integribility conditions. Optimal Homotopy Asymptotic Method (OHAM) is applied to obtain solution of reduced fourth order nonlinear boundary value problem. For comparison, the same problem is also solved by Variational Iteration Method (VIM).

  • The phenomena called "combustion instabilities" in a solid-propellant rocket motor may be viewed as sustaining or amplifying pressure waves. Energy is supplied by combustion processes near the surface of the burning propellant. T-burner method is used to determine the response function of the propellant to the pressure wave. But initial tests were failed because of the Helmholtz resonation inside the T-burner. Acoustic analysis of the original T-burner is carried out and suppression techniques for the Helmholtz oscillation are introduced.ntroduced.

  • The purpose of this work is to investigate the working limit of carbon steel and forging condition of cold forging. The fracture criteria which was proposed by Oyane and Osakada was used. Compression test, hardness study and tension test by Oyane's creteria, microstructure study by Osakada's were carried out. The results were compared with each other. It was found that working limit on compressive hydrostatic stress is increased in comparison with that of tensile stress field and can be forecasted the fracture limit of closed cold forging.
