고지방식을 섭취한 흰쥐의 체내지질함량에 대한 단백질 가수분해물의 섭취 효과 (Effects of Protein Hydrolysates on Blood and Liver Lipids in Rats fed Fat-enriched Diet)

  • The experiments were performed to investigate the effects of protein and protein hydrolysates on lipid metabolism in the hyperlipidemic/hypercholesterolemic rat model induced by feeding fat-enriched diet. In Except 1 male rats were fed four semi-purified high fat and cholesterol diets that contained different nitrogen source, casein(C), casein hydrolysate(CH), corn gluten(G) and corn gluten hydrolysate(GH), for 6 weeks. In Expt. 2 rats were fed high fat and cholesterol diet for 4 weeks to induce hyperlipidemia and hypercholesterolemia. Then the rats were divided into 4 groups and were fed the four kinds of above experimental diets for 4 weeks consecutively. The contents of total lipid , cholesterol and triglyceride in blood, liver and feces were determined. Serum lipid concentrations of CH, G and GH were significantly lower than that of C. Serum cholesterol concentrations of hydrolysate groups(CH and GH) were significantly lower than those of intact protein groups(C and G). Serum HDL -cholesterol concentration tended to increase by hydrolysate intake. The total lipid, cholesterol contents in liver showed similarity results as above. Fecal lipid excretions of CH, G, and GH groups were significantly higher than that of C group. These results indicate that hypolipidemic and /or hypocholesterolemic effect of corn gluten or protein hydrolysates were detected in the process of inducing hyperlipidemia by high-fat and cholesterol diet or after inducing hyperlipidemia.

  • Cholesterol acyltransferase (ACAT) catalyzes the acylation of cholesterol to cholesteryl ester with long chain fatty acids and ACAT inhibition is a useful strategy for treating hypercholesterolemia or atherosclerosis. Inhibitory effects on ACAT of the MeOH extracts prepared from 163 edible plants were evaluated. 15 species out of 163 species exhibited higher than 50% of inhibition on the hACAT-1 and 9 species exhibited higher than 50% of inhibition on the hACAT-2 activity at their concentration of $100\;{\mu}g/mL$.

  • 16세 여아에서 복통, 구토, 고혈압, 경련 및 고지혈증을 보였던 급성 간헐성 포르피린증 1예를 경험하였기에 보고하는 바이다. 소아에서 원인을 알 수 없는 복통에 고혈압, 경련 등 특징적 포르피리아 증상이 동반된 경우에 반드시 급성 간헐성 포르피린증을 고려해야 한다.

  • To evaluate the anti-hyperlipidemic effect of Brassica rapa L. and its major compound, $\beta$-sitosterol, the present study was undertaken, hypercholesterolemia was induced in rats with poloxamer-407, Triton WR-1339, 30% corn oil high cholesterol diet. The ethanol extract of Brassica rapa L. significantly decreased total cholesterol (TC), phospholipid, triglyceride at doses of 200 mg/kg, and $\beta$-sitosterol significantly exerted anti-hyperlipidemic activity at a dose of 15 mg/kg in rats with hyperlipidemia. Taken together, Brassica rapa L. and $\beta$-sitosterol can be useful agents for the prevention or treatment of hyperlipidemia.

동충하초 엑스의 고콜레스테롤혈증 대사효소 활성 변동에 관한 효과 (Studies of the Cordyceps militaris Extract Administration on the Metabolic Enzyme Activities in Hypercholesterolemia)

  • The objective of this study was to investigate the effects of the feeding Cordyceps militaris extract on the improvement of the free fatty acid, lipid peroxide, creatinine and enzyme (creatine phosphokinase; CPK, lactate dehydrogenase; LDH, alkaline phosphatase; ALP, lecithin cholesterol acyltransferase; LCAT) activities in the sera of dietary hypercholesterolemic rats (SD strain, male) fed the experimental diets for 5 weeks. Concentrations of free fatty acid, lipid peroxide and CPK, LDH, ALP activities in sera were fairly reduced in the Cordyceps militaris extract administration group (CHE) than in the hypercholesterolemic diet group (CHD). However, no significance was found in the effect of an creatinine concentration among the groups. The LCAT activity in serum was increased in the Cordyceps militaris extract administration (CHE) than in the hypercholesterolemic diet group (CHD). From these results, Cordyceps militaris extracts were effective on the improvement of the lipid components and metabolic enzyme activities in sera of dietary hypercholesterolemic rats.

도시 직장인의 건강 및 영양 상태 조사 (I) (The Health and Nutritional Status of Urban Area Workers in Korea)

  • This study was conducted to evaluate the workers' health conditions, nutritional status, food habits, and their needs for the nutrition programs at the worksite. Three hundred and fifty one(men 260, women 91) employees from 11 companies in urban area were recruited for the study. The results of this study were as follows : The average consumption of nutrients in the subjects' diets were found to be deficient such as total energy, calcium, iron, vitamin A and $B_2$. More than 42% of the subjects have at least one of the chronic diseases like obesity, diabetes mellitus, anemia, hypercholesterolemia, liver disease, and hypertension. Most subjects did not recognize their own blood cholesterol levels, blood pressure, or blood sugar level. Nutrition knowledge scores of the subjects were very low, which was only 52.4%. It should be noted that weight problems and other health problems of the employees at the worksite were frequently found among the employees who had relatively low nutrition knowledge scores. The nutrition programs for prevention of the obeses, hypercholesterolemia, diabetes mellitus, stress, and smoking were urgently needed by the subjects. Many subjects also wanted the nutrition education programs for their family.

Combined Lowering Effects of Rosuvastatin and L. acidophilus on Cholesterol Levels in Rat

  • Statins are a class of lipid-lowering drugs commonly used in the prevention of cardiovascular diseases. However, statin therapy presents many limitations, which have led to an increased interest in non-drug therapies, such as probiotics, to improve blood cholesterol levels. Indeed, probiotic strains such as Lactobacillus acidophilus have been found to improve blood lipid profiles, especially in reducing total cholesterol and LDL-C levels. In this study, we established a high-cholesterol rat model and studied the effect of Lactobacillus acidophilus administration alone or in combination with rosuvastatin. We were able to show that Lactobacillus exerts a cholesterol-lowering effect. Additionally, we observed that when administered together, rosuvastin and Lactobacillus exert a combined cholesterol-lowering effect. Altogether, our data advocate for the possibility of establishing probiotics as non-drug supplements for the treatment of hypercholesterolemia.

Increased Hepatic Lipogenesis Elevates Liver Cholesterol Content

  • Cardiovascular diseases (CVDs) are the most common cause of death in patients with nonalcoholic fatty liver disease (NAFLD) and dyslipidemia is considered at least partially responsible for the increased CVD risk in NAFLD patients. The aim of the present study is to understand how hepatic de novo lipogenesis influences hepatic cholesterol content as well as its effects on the plasma lipid levels. Hepatic lipogenesis was induced in mice by feeding a fat-free/high-sucrose (FF/HS) diet and the metabolic pathways associated with cholesterol were then analyzed. Both liver triglyceride and cholesterol contents were significantly increased in mice fed an FF/HS diet. Activation of fatty acid synthesis driven by the activation of sterol regulatory element binding protein (SREBP)-1c resulted in the increased liver triglycerides. The augmented cholesterol content in the liver could not be explained by an increased cholesterol synthesis, which was decreased by the FF/HS diet. HMG-CoA reductase protein level was decreased in mice fed an FF/HS diet. We found that the liver retained more cholesterol through a reduced excretion of bile acids, a reduced fecal cholesterol excretion, and an increased cholesterol uptake from plasma lipoproteins. Very low-density lipoproteintriglyceride and -cholesterol secretion were increased in mice fed an FF/HS diet, which led to hypertriglyceridemia and hypercholesterolemia in Ldlr-/- mice, a model that exhibits a more human like lipoprotein profile. These findings suggest that dietary cholesterol intake and cholesterol synthesis rates cannot only explain the hypercholesterolemia associated with NAFLD, and that the control of fatty acid synthesis should be considered for the management of dyslipidemia.

The Juhwangjam (JH) 3rd day of fifth instar silkworm exhibits antioxidative properties and prevents high-fat diet-induced hypercholesterolemia

  • The silkworm is a food material that can simultaneously ingest phytochemicals from mulberry leaves, proteins with essential amino acids, and fatty acids. They are known to have hypoglycemic properties; however, further functional investigation is needed. In this study, four varieties of 3rd day of fifth instar silkworm with different cocoon colors, namely Baegokjam (BG), Goldensilk (GS), Yeonnokjam (YN), and Juhwangjam (JH), were compared in terms of antioxidant properties and cholesterol-lowering effect. JH, which had the highest polyphenol content (+38% vs. GS, p<0.05) showed high antioxidant efficacy. Treatment with JH also resulted in the lowest cholesterol biosynthesis enzyme activity (28% vs. control, p<0.05). In the animal study, high-fat diet (HFD)-fed mice that were orally administered JH extract for 12 weeks showed lower body weight gain (-10.4% vs. HFD, p<0.05) and serum total cholesterol levels (-12.7% vs. HFD, p<0.05). Comparing the varieties, JH had the highest effect. In future studies, analysis of the active ingredients according to their variety should be done.

서울 일개 교회 여자 노인의 영양상태, 건강상태와 영양소 섭취에 관한 연구 (A Study on the Health and Nutritional Status and Nutrient Intakes in Elderly Korean Female)

  • Purpose: The purpose of this study was to investigate the health status, nutritional status, and nutrient intakes of Korean female elderly who are members of a local church in Seoul, Korea. Methods: The subjects were 59 female who are aged >65 yr. We investigated the nutritional status of the subjects using anthropometric and biochemical measurements. Interview was conducted to obtain the information regarding the health status. We also obtained dietary intake of these subjects to figure out the nutrient intakes. Results: The mean age of the subject was 74.0 yr old. Based on the WHO standard (obesity, body mass index, BMI ${\qeq}25$), we categorized the subjects to the obese and the non obese groups. The number of subjects in the obese group was 34 (57.6%). The mean Diastolic BP (p=.002), BMI (p<.001), waist circumference (p<.001), hip circumference (p<.001), blood level of transferrin (p=.038), and nutrient intakes of calcium (p=.026), potassium (p=.046), folate (p=.038) and vitamin E (p=.031) were higher in the obese group than in the non obese group. The subjects with hypertension was 67.5%. The presence of hypertension was correlated with BMI (p<.05). The number of subjects with hypercholesterolemia was 17 (28.8%). The level of cholesterol was correlated with BMI (p<.05) and wasit/hip ratio (p<.05). Conclusion: The female elderly in our study had higher prevalence of obesity which is a risk factor for hypertension and hypercholesterolemia. The nutrient intake of high sodium, high cholesterol, and low dietary fiber of these subjects suggests that there is an urgent need to develop an well planned nutritional education program for female elderly.
