Effects of different parts of Angelica gigas Nakai on brain damages and neuronal death in transient middle artery occlusion/reperfusion-induced ischemic rats (허혈성 뇌졸중 흰쥐에서 당귀의 부위에 따른 뇌신경보호효과 비교 연구)

  • Objectives : We compared with the effects of different parts (root head, root body and hairy root) of Angelica gigas Nakai (Angelicae Gigantis Radix, AG) with on middle cerebral artery occlusion(MCAO)-induced ischemic rats, and on LPS-induced inflammatory response in BV2 microglia. Methods : The 30% ethanol and water extracts of different parts of AG were prepared. Each extract (50 and 100 mg/kg) was administrated intraperitoneally once in MCAO-induced ischemic rats. We measured infarction volumes by TTC staining, and investigated the expression of iNOS, Bax, Bcl-2 and caspase-3 by Western blot. BV2 cells were treated with each extract for 30 min, and then stimulated with LPS. The levels of NO was measured by Griess assay. The expression of iNOS, Cox-2 and proinflammatory cytokines ($TNF-{\alpha}$, $IL-1{\beta}$, and IL-6) were determined RT-PCR and Western blot. The phosphorylation of ERK1/2 and JNK MAPK was determined by Western blot. Results : Among different parts of AG, the 30% ethanol and water extracts of hairy root significantly decreased infarction volume in ischemic brains and inhibited the expression of iNOS, bax and caspase-3. The extracts of hairy root significantly inhibited LPS-induced production of NO, $TNF-{\alpha}$ and IL-6 in BV2 cells, and suppressed the expression of iNOS and COX-2. The hairy root extracts attenuated LPS-induced phosphorylation of ERK1/2 and JNK MAPK in BV2 cells. Conclusions : Our results indicate that the root hairy of AG has a good neuroprotective and anti-inflammatory effects in ischemic stroke compared to other parts.

  • Effect of ginseng hairy and native roots on body absorption, fecal and urinary excretion of Ingested radiostrontium were investigated in the mouse(NIH-strain, male) treated with or without pre-feeding of each ginseng soluble fraction. The test groups were fed with basic diet supplemented with 1% each ginseng soluble fraction for 7 darts before the radiostrontium were administered by intragastric intubation. In the groups of treated with soluble fraction from ginseng hairy roots, the radioactivities of fecal and urinary excretion increased about 15% over than that of control groups and the whole body retention were about 38%. In the groups of treated with soluble fraction from native ginseng roots, the radioactivities of fecal and urinary excretion increased about 25% over than that of control groups and the whole body retention were about 28%. Also, the levels of radiostrontium accumulation retained significantly the higher percent in skeletons than in other organs.

  • Shikonin production was examined in a bubble column bioreactor system with the hairy roots of Lithosphermum erythrorhizon. The volumetric productivity was higher than those obtained from other reactor configurations with free or immobilized cells of the same cell line. The productivities of the bubble column reactor, with and without a product absorption trap, were 7.4 and 4.5 mg of shikonin/l/d, respectively. This indicated the importance of the product removal in the design and operation of the shikonin production system with hairy root culture.

  • New methods have been developed to transform Panax ginseng with Ri plasmids of Agrobacterium rhizogenes 15834 and A. rhizogenes A4. Modified leaf disc method was made feasible to establish hairy root culture even when an axonic plantlet was not available as in the case of P. ginseng. The contents of ginsenosides (Rgl, Rf, Rc, Rbl, and Rb2) in hairy roots. were determined by HPLC. Hairy root cultures, established as liquid culture in MS medium, was produced 0.34~1.19% ginsenosides on dry weight basis, and this result is significantly higher level than that of normal P. ginseng.

Influence of Different Strains of Agrobacterium rhizogenes on Hairy Root Induction and Rosmarinic Acid Production in Agastache rugosa Kuntze (Agrobacterium rhizogenes strains이 배초향 모상근 유도와 Rosmarinic acid 생산에 미치는 영향)

  • Rosmarinic acid, an ester of caffeic acid with 3,4-dihydroxyphenyl lactic acid, is one of the main active constituents of Agastache rugosa Kuntze and has an astringent property, antioxidant capacity, anti-inflammatory activity, antimutagenic ability, antimicrobial capacity, and an antiviral property. Five different strains of Agrobacterium rhizogenes differed in their ability to induce Korean mint (Agastache rugosa Kuntze) hairy roots and also showed varying effects on the growth and rosmarinic acid production in hairy root cultures. A. rhizogenes R1601 is the most effective strain for the induction (72.90%), growth (13.50 g/l) and rosmarinic acid production (22.60 mg/g) in hairy root of Korean mint. Our results demonstrate that use of suitable strains of A. rhizogenes may allow study of the regulation of rosmarinic acid biosynthesis in hairy root cultures of Agastache rugosa.

  • In order to elucidate the optimum medium on the growth and tropane alkaloid production of hairy root, hairy root were induced by inoculating leaf and stem of Datura stramonium var. tatula Torr. with Agrobacterium spp. Both Agrobacterium tumefaciens $\textrm{A}_{4}$ T and A, rhizogenes ATCC 15834 among tested strains were effective on hairy root formation. Among 23 clones selected in SH (Schenk and Hildebrandt, 1972) liquid medium, DTLA9 clone was shown fast growth of hairy root and DTLE6 clone was shown high level production of tropane alkaloids. When both DTLA9 and DTLE6 clones were cultured in the GD (Gresshoff and Doy, 1972) medium, alkaloid production was higher than in 8 tested media. It was elucidated that optimum medium for root proliferation and for tropane alkaloid production is SH, GD medium, respectively.

Growth Pattern and Content of Tropane Alkaloids of Metabolic Engineered Scopolia parviflora Hairy Root Lines

  • Hyoscyamine and scopolamine are two most common tropane alkaloids found in the Solanaceae. The pEB expression vector carrying Nspmt gene was transformed to Agrobacterium rhizogenes. The growth of transgenic hairy roots were approximately to 80% of the wild type root. Transgenic hairy roots are less developed on only their branch roots than those of the wild type root. The extracts from Nspmt transgenic hairy root lines, 3 and 5 contained between 3.52 and 4.23 mg/g dry weight as hyoscyamine and did between 5.23 to 6.40 mg/g dry weight as scopolamine. These results showed that the overexpression of the pmt gene enhanced tropane alkaloids production of S. parviflora transformed roots and this improvement affected both hyoscyamine and scopolamine production.

Upregulation of Isoprenoid Pathway Genes During Enhanced Saikosaponin Biosynthesis in the Hairy Roots of Bupleurum falcatum

  • In order to characterize saikosaponin biosynthesis in Bupleurum falcatum, the expression of five isoprenoid pathway genes and their relationship to saikosaponin accumulation in the hairy roots were analyzed. The hairy roots exhibited a rapid accumulation of saikosaponins when incubated in a root culture medium (3XRCM). Homology-based RT-PCR was used to isolate core fragments of five genes, HMGR, IPPI, FPS, SS, and OSC, from the hairy roots. The deduced amino acid sequences exhibited amino acid identities of more than 85% to previously reported genes. Using the fragments as probes, the expression of these five genes in the hairy roots during incubation in 3XRCM medium was examined. Expression of all five genes in the hairy roots increased soon after incubation. In particular, the SS and OSC genes were coordinately induced at 8 days of incubation, and their expression persisted throughout the incubation period. A quantitative HPLC analysis showed that the saikosaponin content of the hairy root culture also began to increase at 8 days of culture. The correlation between SS transcript level and saikosaponin content in the hairy roots suggests that transcriptional regulation plays a regulatory role in saikosaponin biosynthesis.

Optimal Culture Conditions for Transformed Root Growth and Trichosanthin Formation in Trichosanthes kirilowii Max. (하늘타리 형질전환근의 생장 및 Trichosanthin의 생합성을 위한 최적화)

  • Transformed hairy roots were induced from in vitro grown plantlets of Trichosanthes kirilowii by infection with Agrobacterium rhizogenes strain ATCC15834. Transformed hairy roots exhibited active growth with high branching of roots on plant growth regulators-free medium. Cloned line (TR-03) of hairy root was tested for its growth and extracellular protein accumulation in medium under various culture conditions. Among the culture media tested, a full-strength MS medium had a pronounced effect on root biomass and extracelluar protein accumulation in medium. The maximum root biomass (2.4 g DRW/flask) and extracellular total protein contents $(28.3ug/m\ell)$ in medium was obtained at inoculum size of 2 g (FRW) and in MS medium supplemented with 4% sucrose. In addition, the optimal shaking speed for root growth and extracellular protein accumulation in medium were 100 rpm. The total extracellualr protein concentration reached a maximum of $28.3ug/m\ell$ at 4 weeks and decreased thereafter. Protein translation inhibitory activity was observed in culture broths and reached levels of 21.3 unit. These studies demonstrate that the transformed hairy roots can be utilized for the in vitro production of ribosome-inactivating proteins.

Optimization of the Sucrose and Ion Concentrations for Saikosaponin Production in Hairy Root Culture of Bupleurum falcatum

  • Saikosaponin productivity was examined in a Bupleurum falcatum L. BFHR2 hairy root culture in response to changes in the sucrose content $(2{\sim}8%)$, nitrogen content $(0{\sim}250mM\;NH_4NO_3)$, phosphate content $(0{\sim}12mM\;NaH_2PO_4)$, and the potassium content $(0{\sim}87.2 mM\; KCl)$ of the culture media. We found that the conditions for maximal saikosaponin production differed from those for optimal root growth. Highest saikosaponin yield was achieved for 8% sucrose, 62mM $NH_4NO_3$ 1.2 mM $NaH_2PO_4$, and 0.5mM KCl.
