  • Free radical scavengers in the bisdemethoxycurcumin (BDMC), demethoxycurcumin (DMC) and curcumin of turmeric (Curcuma longa) were screened, identified, quantified and isolation using coupled off-line-2,2'-azino-bis(3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) and on-line screening high-performance liquid chromatography (HPLC)-ABTS assay. There was a very small margin of error between the off-line-ABTS method and the on-line screening HPLC-ABTS method.

  • This study employed high performance liquid chromatography (HPLC) coupled to an on-line $ABTS^+$ radical scavenging detection (RSD) system along with HPLC-electro spin impact/mass spectrometry (ESI/MS), to rapidly determine and identify antioxidant compounds occurring in blueberry extract. The extract was separated by HPLC, and then the radical scavenging activities of the separated compounds were evaluated by the on-line coupled $ABTS^+$-RSD system. The negative peaks of the $ABTS^+$-RSD system, which indicates the presence of antioxidant activity, were monitored by measuring the decrease in absorbance at 734 nm. The active components in the blueberry extract were identified by HPLC-ESI/MS using their MS spectra and retention times. According to the data acquired from the on-line HPLC-$ABTS^+$-based assay and HPLC-ESI/MS systems, the antioxidant compounds detected in the blueberry extract were identified as chlorogenic acid and 11 anthocyanins.

  • In this work, we describes analysis of the antioxidant potential of Korean green tea phenolics using an high-performance liquid chromatography (HPLC) on-line $ABTS^{+}$ antioxidant screening method. In conjunction with the analysis of their 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS+) radical scavenging ability, the extraction of catechine compounds from Korean green tea were performed by various temperature and time. The optimum operating conditions were experimentally determined to analyze the catechine compounds in the pretreatment extracts. From the results, the extraction temperature $60^{\circ}C$, time 3 min was selected as an optimal antioxidant activity condition. The analysis by $C_{18}$ column was performed, the flow rate of mobile phase and UV wavelength was fixed at 1.0 ml/min and 254 nm, respectively. the mobile phase was composed from acetonitrile and water, and the gradient elution mode were applied.

  • The Peanut sprouts have been used as food for long time in many Asian countries. Among them, resveratrol have bioactivity effects as antioxidant and anticancer. In this work, resveratrol was extracted from peanut sprouts by dipping(extraction time 3h, $25^{\circ}C$) method. And extraction efficiency and amount was identified using on-line screening HPLC-ABTS. From the experimental results, it is evident that the amount of resveratrol($0.310-0.346{\mu}g/mL$) by 50% aqueous EtOH was higher than any 100% water and EtOH composition. Also, the 100% water extracts sample was low yield 2.040-2.145%. The shown results can be applied as sources for pharmaceuticals and functional material.

  • Hyphenated-HPLC techniques combine the separation power of HPLC with the structural and bioactivity information provided by NMR, ESI/MS, and an on-line antioxidant screening system. The major advantages over the traditional off-line techniques are rapidity and efficiency. In this study, we used hyphenated HPLC techniques including online HPLC-ABTS, LC-NMR, and LC-MS todirectly identify the major antioxidants of safflower (Carthamus tinctorius L.) seeds. The results demonstrated that the major antioxidant compounds from on-line HPLC-ABTS analysis were identified as 8'-hydroxyarcgenin-4'-O-$\beta$-D-glucoside, N-(p-coumaroyl) serotonin, and N-feruloylserotonin. Among them, N-feruloylserotonin accounted for almost 50% of the ABTS radical scavenging activity of the total extract. The results demonstrate that HPLC hyphenated techniques can be used to rapidly screen and structurally identify antioxidants from crude plant extracts.

  • In an attempt to evaluate licorice quality based on its biological activity, we grafted an on-line high-performance liquid chromatography (HPLC) bioassay method into the previously established HPLC analysis method. The common antioxidant peaks in licorices of various origin were observed through an on-line HPLC/DPPH system leading to a decrease in absorbance at 517 nm for 2,2-diphenyl-1-picrylhydrazyl (DPPH). Among them, the licorice from Youngju possessed the highest activity. Therefore, three active standard compounds from the dehydroglyasperin C, dehydroglyasperin D, and isoangustone A, were isolated and elucidated by medium pressure liquid chromatography (MPLC) and instrumental analysis such as nuclear magnetic resonance (NMR), respectively. On-line HPLC/ABTS analysis method with the simultaneous determination of three standard compounds and their radical scavenging activity was established for the quality evaluation of licorices. 2,2'-azinobis-3-ethylbenzothiazoline-6-sulfonic acid radicals (ABTS) which is stable and effective was used in replace of DPPH. The radical scavenging activity of three standards is compared with that of Trolox, known as antioxidant, showing a negative peak with a decrease in absorbance at 734 nm for ABTS. This on-line HPLC/ABTS analysis method was validated for specificity, linearity, precision and accuracy in compliance with international conference on harmonization (ICH) guideline.
