Conformational Preferences of Glycerol in the Gas Phase and in Water

  • The conformational study of glycerol has been carried out using the M06-2X/cc-pVTZ level of theory in the gas phase and the SMD M06-2X/cc-pVTZ level of theory in water in order to understand its conformational preferences and solvation effects. Most of the preferred conformers of glycerol have two $C_5$ hydrogen bonds in the gas phase, as found by the analysis of calorimetric data. It has been known that the solvation drove the hydrogen bonds of glycerol to be weaker and its potential surface to be fatter and that glycerol exists as an ensemble of many feasible local minima in water. The calculated populations of glycerol in the gas phase and in water are consistent with the observed values, which are better than the previously calculated ones at the G2(MP2), CBS-QB3, and SM5.42 HF/6-31G(d) levels of theory.

  • High oil price and biodiesel expansion lead the surplus of glycerol in the market. Glycerol has been used as a raw material itself at petroleum chemistry, paint, tobacco, household products and cosmetics in the conventional market. Recently, many research to find new applications of glycerol as a low-cost feedstock for functional derivatives have led to the introduction of a number of selective processes for converting glycerol into commercially value-added products. The recent studies on the development of new value-added glycerol derivatives will be reviewed.

Effect of Glycerol and Ethylene Glycol on Post-Thawed Sperm Function in Jeju Horse (제주마의 동결정액 제조에 있어 Glycerol과 EthyleneGlycol이 동결 융해 후 정자의 기능에 미치는 영향)

  • Cryopreservation induces sublethal damage to the spermatozoa, which leads to their reduced fertile life. This study was designed to determine effect of glycerol and ethylene glycol as cryoprotectant in extender on improve the freezability of Jeju horse semen. The semen was cryopreserved with glucose-EDTA extender containing each 5% glycerol, 5% ethylene glycol, 8% glycerol or 8% ethylene glycol, respectively. Post-thawed sperm were evaluated motility, viability, Membrane integrity and acrosome integrity. Post-thawed sperm motility were not significantly differences among treatments. However, sperm viability were significantly higher (p<0.05) in 8% glycerol ($39.85%{\pm}11.41$) than in 5% glycerol treatment ($18.08%{\pm}1.61$). In membrane integrity, swelling sperm ratio was significantly higher (p<0.05) in 8% glycerol ($34.12%{\pm}11.02$) than other treatments. In the percentage of capacitated sperm assessed by CTC staining, F pattern was significantly higher in 8% ethylene glycol than 5% glycerol and 5% ethylene glycol (p<0.05). B pattern ratio was significantly increased in 5% ethylene glycol compared with 8% glcerol and 8% ethylene glycol (p<0.05). Moreover, 8% ethylene glycol treatment was significantly decreased AR pattern ratio compared with other treatments (p<0.05). It is concluded that treatment of 8% glycerol was improved the sperm viability and 8% ethylene glycol was improved the sperm ascrosome integrity after thawing. However, they were not significantly difference between 8% glycerol and 8% ethylene glycol on post-thawed sperm viability. Therefore, 8% ethylene glycol was more effective sperm cryoprotectant than 8% glycerol in Jeju Horse.

Co-digestion of Waste Glycerol with Swine Manure (폐 글리세롤과 돈분의 혼합 소화)

  • Production of crude glycerol from biodiesel industry is expected to exceed the commercial demand for purified glycerol in the near future. This study aimed to evaluate the feasibility of co-digestion of crude glycerol with swine manure. Crude glycerol up to 13.8 g/L was regarded as a good co-substrate for swine manure digester. It improved methane production and productivity by 90% and 120%, respectively. Methane yield of crude glycerol at the condition was estimated to be 232 mL/g. However, it inhibited methanogenic activity at above 27.5 g/L. Optimum concentration of crude glycerol for co-digestion with swine manure would be near to 13.8 g/L.

  • Pichia pastoris, a methylotrophic yeast widely used for the production of heterologous proteins, was used to produce Hepatitis B surface antigen (HBsAg) under the control of the strong, tightly-regulated alcohol oxidase promoter. It is highly induced during the growth on methanol, but the presence of non-methanol carbon sources such as glycerol and glucose repressed fully the expression of alcohol oxidase. In this study, glycerol and sorbitol feedings for the expression of the recombinant HBsAg were compared to examine the potential of sorbitol as a less repressive carbon source in fed-batch fermentation. The sorbitol feeding enhanced the production yield by 12% compared to that in glycerol feeding, although the cell concentration was lower.

  • The glycerol carbonate was synthesized by glycerol and urea using metal oxide catalysts. The physical properties of the prepared metal oxide catalysts were investigated by X-ray diffraction (XRD), specific surface area analysis (BET), field emission scanning electron microscopy (FE-SEM) and temperature programmed desorption (TPD). In addition, we confirmed the conversion of the glycerol and the yield of the glycerol carbonate according to characteristics of metal oxide catalysts. From XRD and FE-SEM analysis, the crystallite size and crystallinity of metal oxide catalysts decrease with addition of Al. In addition, the Zn-Al mixed metal oxide had higher catalytic activity than the pure ZnO due to decreased side reaction in the synthesis of glycerol carbonate.

  • New technology-driven biocatalysts are revolutionizing the biochemical industries. With maximum utilization of renewable feedstock, biocatalysts have been the basis for a major breakthrough. Lipases are the most widely established catalysts used for hydrolysis, esterification and transesterification reactions. In this research, a biochemical process that combines extraction of lipase enzyme from germinated wheat seeds and its application to valorize glycerol to acetins by esterification is presented. Acetins are among highly rated, value-added products derived from glycerol. The favorable conditions for the enzymatic conversion of glycerol were observed as glycerol to acetic acid molar ratio (1:5), reaction temperature ($40^{\circ}C$) and the amount of enzyme (20% v/v). 65.93% of glycerol conversion was achieved for duration of 15 h with the use of tert-butanol solvent. This method proposes to explore the viability of a biological route to convert glycerol derived from biodiesel industry to acetins with further streamlining.

Bioethanol Production Based on Crude Glycerol Using Enterobacter aerogenes (Enterobacter aerogenes를 이용한 crude glycerol 기반의 바이오에탄올 생산)

  • The effects of pH, glycerol concentration and salt on cell growth and ethanol production using Enterobacter aerogenes KCTC 2190 were evaluated in the anaerobic culture condition. In condition of initial pH 5, cell growth and ethanol production were highest. An initial concentration of 10 g/L of pure glycerol gave the highest cell growth and ethanol production. However, in case of over 15 g/L of pure glycerol, they decreased. The cell growth and ethanol production decreased with the increase of salt concentration. When 10 g/L of crude glycerol was used as the carbon source, the cell growth and ethanol production were $1.32\;OD_{600}$ and 3.95 g/L, respectively, which were about 94.4% and 88.5% compared to those of pure glycerol. These result indicates that the crude glycerol produced in the biodiesel manufacturing process maybe useful as a potential carbon source for ethanol production form Enterobacter aerogenes KCTC 2190.

  • Glycerol kinase deficiency (GKD) is an X-linked recessive enzyme defect characterized biochemically by hyperglycerolaemia and glyceroluria. GK gene is located on the short arm of X chromosome 21.3 region tandemly with AHC gene, and DMD gene and there is a long deletion resulting in contiguous gene deletion syndrome. In Korea there was a report of contiguous gene deletion syndrome of adrenal hypoplasia congenita, glycerol kinase deficiency and Duchenne muscular dystrophy but no isolated glycerol kinase deficiency. This is the first case of isolated glycerol kinase deficiency confirmed by organic acid analysis and gene analysis in Korea.
