Studies on Antioxidant, Anti-Inflammation, and Collagenase Inhibitory Effects of Extracts from Plants of The Salix genus (버드나무 속 식물 추출물의 항산화, 항염 및 콜라게나제 저해 활성 연구)

  • This study was carried out to evaluate the possibility of willow plants (the genus Salix) as a cosmetic material. DPPH radical scavenging abilities of 70% ethanol extracts of S. gracilistyla, S. pseudolasiogyne, and S. koriyanagi were significantly increased compared to control. In addition, the treatment of three species of willow plant extracts significantly inhibited the production of nitric oxide (NO) in RAW 264.7 cells, indicating that they had anti-inflammatory activity, and all of them had collagenase inhibitory activity. Among them, the extracts of S. gracilistyla extracts exhibited the highest collagenase inhibitory activity. As a result of analyzing the collagenase inhibitory activity against the solvent fraction of S. gracilistyla extracts, water and butanol fractions showed the highest collagenase inhibitory activity. These results suggested that S. gracilistyla among the willow plants had high collagenase inhibitory activity, and thus it can be utilized for cosmetics as an effective functional cosmetic material in the future.

Screening of the Antibacterial Effects by Ethanol Extracts from Natural Plant in Jeju against Propionibacterium acnes (Propionibacterium acnes에 대한 제주 자생식물 에탄올 추출물의 항균효과 검색)

  • Propionibacterium acnes, a major pathogens bacterium of acne, is one of the microflora associated with the inflammation. In this study, the antibacterial effects of 70% ethanol extracts from the 61 natural plants in Jeju against P. acnes were investigated using the disc diffusion method. The 45 natural plant extracts showed antibacterial effects and the 16 natural plants extracts didn't show antibacterial effects. The Phyllanthus urinaria L. (stem and leaves) extract showed thr highest antimicrobial activity with $18.96{\pm}0.69mm$ clear zone of the growth of P. acnes, followed by Angelica gigas Nakai (roots), Gynostemma pentaphyllum (Thunb.) Makino, Angelica gigas Nakai (stem and leaves), Isodon inflexus (Thunb.) Kudo, camellia sinensis L. (flowers), Rosmarinus officinalis, etc. These results suggest that the natural plant in Jeju can be used as functional cosmetic biomaterials or antimicrobial agents against P. acnes.

Enhancement and Conversion of Ginsenoside Contents in Cultured Wild Ginseng Adventitious Root (산삼 부정배양근의 진세노사이드 함량 증진과 성분 변환)

  • Background: Culturing wild ginseng adventitious root using plant factory technology provides genetic safety and high productivity. This production technology is drawing attention in the fields of functional raw materials and product development. The cultivation method using elicitors is key technology for controlling biomass and increasing secondary metabolites. Methods and Results: Elicitor treatments using methyl jasmonate, pyruvic acid, squalene, β-sistosterol were performed to amplify total ginsenosides (Rb1, Rc, Rb2, Rb3, and Rd) of cultured wild ginseng adventitious root. Thereafter, fermentation and steaming processes were performed to convert total ginsenosides into minor molecular ginsenosides (Rg3, Rk1, and Rg5). The result indicated that methyl jasmonate minimizes the reduction in fresh weight of cultured wild ginseng adventitious root and maximizes total ginsenosides (sum of Rb1, Rc, Rb2, Rb3, and Rd). Ginsenoside conversion results showed a maximum degree of conversion of 131 mg/g. Conclusions: In this study, we demonstrated that the optimal elicitor treatment method increased the content of total ginsenosides, while the steaming and fermentation processing method increased the content of minor ginsenosides.

Preparation and application of the pine oil solution for wet tissues (물티슈용 소나무 오일 솔루션 제조 및 응용)

  • This study was prepared for a natural aroma solution for the purpose of improving depression, anxiety, and insomnia caused by stress in modern people, and applied the solution to wet tissues. Specifically, the pine oil solution was prepared by mixing Korean pine oil and black cherry oil. Heavy metal analysis, antibacterial analysis, psychological evaluation, and skin evaluation were performed on the prepared pine oil solution to examine the possibility of manufacturing functional aroma wet tissues for stress improvement. Furthermore, as a result of investigating whether pine oil solution wet tissues improved the skin, it was found that there was an improvement effect in terms of moisturizing and elasticity.

Analytical Method Development of Avicularin and Quercitrin in Astilbe chinensis Extract Using HPLC (HPLC를 이용한 노루오줌 추출물의 Avicularin 및 Quercitrin 분석법 개발)

  • This study attempted to eatablish a high performance liquid chromatography (HPLC) analysis method for the determination of avicularin, quercitrin as a part of the quality control for the development of functional cosmetic materials from Astilbe chinensis extract. HPLC was performed on a Capcell Pak C18 MGII column with a gradient elution of 0.05% (v/v) trifluoroacetic acid (TFA) and acetonitrile at a flow rate of 1.0 mL/min at 30 ℃. The analyte was detected at 254 nm. The HPLC method was performed in accordance with the international conference on harmonization (ICH) guideline (version 4, 2005) of analytical procedures with respect to specificity, precision, accuracy, and linearity. The limits of detection and quantitation of avicularin and quercitrin were 0.094 and 0.285 mg/mL, 0.031 and 0.095 mg/mL respectively. Calibration curves showed good linearity r2 > 0.99990 for avicularin and r2 > 0.99994 for quercitrin. Precision of analysis was satisfied with less than 0.59% for avicularin and 0.63% for quercitrin. Recoveries of quantified compounds ranged from 100.97 to 101.77% for avicularin and 100.18 to 100.32% for quercitrin. These result indicated that the established HPLC method is very useful for the determination of marker compounds in A. chinensis extracts.

Studies on Skin Whitening Effects of Resorcinol Dipentyl Ether (레조시놀다이펜틸에터의 피부 미백 효능 평가)

  • The objective of this study was to investigate skin whitening effect of Resorcinol dipentyl ether [1,3-di(pentyloxyl)benzene] by in vitro experiments. Resorcinol dipentyl ether was prepared by alkylation of resorcinol with 1-bromopentane. The reaction products were confirmed by NMR, MS and other analytical equipments. In order to evaluate the skin safety of resorcinol dipentyl ether, the cytotoxicity of the cells constituting the skin (keratinocyte, melanocyte, fibroblast) was analyzed and similar cell viability was observed in all cell lines as compared with the control group. Inhibition of extracellular melanin synthesis effect of resorcinol dipentyl ether was approximately 65.75% at $20{\mu}g/mL$ and inhibition of intracellular melanin synthesis effect of resorcinol dipentyl ether was approximately 53.89% at $20{\mu}g/mL$. The real-time PCR and western blot analysis of mRNA expression and protein expression of tyrosinase, TRP-1, and TRP-2 related to melanogenesis revealed that melanin inhibitory effect of resorcinol dipentyl ether was inhibited from the transcription stage respectively. Finally, this study suggested applicability of Resorcinol dipentyl ether [1,3-di(pentyloxyl)benzene] as a whitening functional cosmetic new material.

Biological Activity in Functional Cosmetic of Purple Sweet Potato Extracts (자색 고구마추출물의 기능성 화장품 활성)

  • The optimum condition for phenolics contents extraction from purple sweet potato was 12 hrs. in 50% ethanol. The electron donating scavenging activities (DPPH), ABTS radical cation decolorization (ABTS) and antioxidant protection factor (PF) of Jami, Yeonjami and Shinjami were higher than general sweet potato, and thiobarbituric acid reaction substance (TBARs) was below 30%. The minimum inhibitory concentration (MIC) against Staphylococcus aureus and Escherichia coli on skin were each 5,000 and 2,500 ppm in all purple sweet potatoes, and MIC of Jami was the lowest as 2,500 ppm against Staphlococcus epidermidis. The whitening (tyrosinase inhibitory) activity of purple sweet potatoes was the highest as 62.5% and 48.7% in Jami water and ethanol extracts. The anti-inflammation (hyaluronidase inhibitory) activity of purple sweet potatoes was the highest as 25.3% and 94.4% in Jami water and ethanol extracts. The safety of cosmetic with Jam; extracts was assessed by various safety profiles. pH and viscosity change of essence for 90 days was not detected. Cosmetic was stable to temperature and light for 90 days. The result to measure changes of skin color and pore size of the skin was that an aged skin was more effective than young skin.

Analytical Method Development of (-)-Epicatechin gallate in Penthorum chinense Pursh Extract using HPLC (HPLC를 이용한 낙지다리 추출물의 (-)-­Epicatechin gallate 분석법 개발)

HPLC was performed on a Unison $US-C_{18}$ column ($4.6{\times}250mm$, $5{\mu}m$) with a gradient elution of 0.05% (v/v) trifluoroacetic acid (TFA) and methyl alcohol at a flow rate of 1.0 mL/min at $30^{\circ}C$. The analyte was detected at 280 nm. The HPLC method was performed in accordance with the International Conference on Harmonization (ICH) guideline (version 4, 2005) of analytical procedures with respect to specificity, precision, accuracy, and linearity. The limits of detection and quantitation were 0.11 and 0.33 mg/mL, respectively. Calibration curves showed good linearity ($r^2$ > 0.9999), and the precision of analysis was satisfied (less than 0.6%). Recoveries of quantified compounds ranged from 99.51 to 101.92%. This result indicates that the established HPLC method is very useful for the determination of marker compound in P. chinense Pursh extracts.

Analytical Method Development of Isoscoparin in Silene seoulensis Extract Using HPLC (HPLC 를 이용한 가는장구채 추출물의 Isoscoparin 분석법 개발)

  • In this study, isoscoparin was selected as an indicator component to develop Silene seoulensis extract as a functional cosmetic material, and we developed an analysis method using high performance liquid chromatography (HPLC) for quality control. HPLC was performed on a Unison US-C18 with a gradient elution of 0.05% (v/v) trifluoroacetic acid (TFA) and methanol at a flow rate of 1.0 mL/min at 35 ℃, and the detection wavelength was 330 nm. The HPLC method was performed in accordance with the international conference on harmonization (ICH) guideline (version 4, 2005) of analytical procedures with respect to specificity, precision, accuracy, and linearity. The limits of detection and quantitation were 0.02 and 0.07 mg/mL respectively. Calibration curves showed good linearity (R2 > 0.99988), and the precision of analysis was satisfied (less than 0.46%). In addition, the recovery rate was in the range of 99.10 to 101.61%, it was shown to be accurate. This result indicated that the established HPLC method is very useful for the determination of marker compounds in Silene seoulensis extracts.

Anti-inflammation and Anti-aging Effect of Lactic Acid Bacteria Fermented Lycium chinense Extracts (젖산균 발효 구기자(Lycium chinense ) 추출물의 항염증 및 항노화 효능 연구)

  • In this study, fermented Lycium chinense (L. chinense) extract was used to analyze the efficacy of nitric oxide (NO) and anti-aging inhibition to verify its usefulness as a functional cosmetic material. L. chinense was extracted with hydrothermal and 70% ethanol as a solvent, fermented, concentrated, and freeze-dried to prepare a sample, followed by MTT assay, NO inhibition assay, western blot assay, and UPLC analysis.As a result of the analysis of NO inhibitory efficacy, fermented L. chinense water extract (FLW) and fermented L. chinense 70% ethanol extract (FLE) showed excellent NO inhibitory efficacy of 47.96% and 56.71%, respectively. As a result of measuring the expression patterns of the wrinkle-related proteins MMP-1, and TRPV-1 through Western blot, both FLW and FLE confirmed the inhibitory efficacy of MMP-1 and TRPV-1. Based on the results of the experiment, the fermented L. chinense extract is expected to have a high application value as a functional cosmetic material related anti-inflammatory, and anti-aging.
