Development of Disposable Immunosensors for Rapid Determination of Sildenafil and Vardenafil in Functional Foods

  • We introduced disposable amperometric immunosensors for the detection of Sildenafil and Vardenafil (SDF/VDF) based on screen printed carbon electrodes. The developed immunosensors were used as a non-competitive sandwich-type enzyme immunoassay with a horseradish peroxidase label. The sensors were constructed on screen printed carbon electrodes by the simple electrochemical deposition of a reduced graphene oxide and chitosan (ErGO-CS) composite. To evaluate the sensing chemistry and optimize the sensor characteristics, a series of electrochemical experiments were carried out including electrochemical impedance spectroscopy, cyclic voltammetry and amperometry. The sensors showed a linear response to SDF/VDF concentrations in a range from 100 pg/mL to 300 ng/mL. The lower detection limit was calculated to be 55 pg/mL, the sensitivity was calculated to be $1.02{\mu}Ang/mL/cm^2$, and the sensor performance exhibited good reproducibility with a relative standard deviation (RSD) of 7.1%. The proposed sensing chemistry strategy and the sensor format can be used as a simple, cost-effective, and feasible method for the in-field analysis of SDF/VDF in functional or health supplement food samples.

Highly sensitive and selective enzymatic detection for hydrogen peroxide using a non-destructively assembled single-walled carbon nanotube film (탄소나노튜브 대면적 어셈블리를 통한 고감도-고선택성 과산화수소 센서 개발)

  • This study presents a simple approach for the assembly of a free-standing conductive electronic nanofilm of single-walled carbon nanotubes (SWNTs) suitable for enzymatic electrochemical biosensors. A large-scale SWNT electronic film was successfully produced by the dialysis of p-Terphenyl-4,4''-dithiol (TPDT)-treated SWNTs. Furthermore, Horseradish peroxidase (HRP) was immobilized on the TPDT-SWNT electronic film, and the enzymatic detection of hydrogen peroxide (H2O2) was demonstrated without mediators. The detection of H2O2 in the negative potential range (-0.4 V vs. Ag/AgCl) was achieved by direct electron transfer of heme-based enzymes that were immobilized on the TPDT-SWNT electronic film. The SWNT-based biosensor exhibited a wide detection range of H2O2 from 10 µM to 10 mM. The HRP-doped SWNT electronic film achieved a high sensitivity of 342 ㎛A/mM·cm2 and excellent selectivity against a variety of redox-active interfering substances, such as ascorbic acid, uric acid, and acetaminophen.

Amperometric Morphine Detection Using Pt-Co Alloy Nanowire Array-modified Electrode

  • Pt-Co alloy nanowire array was directly synthesized by electrochemical deposition with polycarbonate template at -1.0V and subsequent chemical etching of the template. The use of Pt-Co alloy nanowire array-modified electrode (Pt-Co NAE) for the determination of morphine (MO) is described. The morphology of the Pt-Co alloy nanowire array has been investigated by scanning electron microscopy (SEM) and energy disperse X-ray spectroscopy (EDS) analysis), respectively. The resulting Pt-Co NAE offered a linear amperometric response for morphine ranging from $2.35\times10^{-5}$ to $2.39\times10^{-3}$ M with a detection limit of $7.83\times10^{-6}$ M at optimum conditions. This sensor displayed high sensitivity and long-term stability.

Electrochemical oxidation-reduction and determination of urea at enzyme free PPY-GO electrode

  • This manuscript explains the effective determination of urea by redox cyclic voltammetric analysis, for which a modified polypyrrole-graphene oxide (PPY-GO, GO 20% w/w of PPY) nanocomposite electrode was developed. Cyclic voltammetry measurements revealed an effective electron transfer in 0.1 M KOH electrolytic solution in the potential window range of 0 to 0.6 V. This PPY-GO modified electrode exhibited a moderate electrocatalytic effect towards urea oxidation, thereby allowing its determination in an electrolytic solution. The linear dependence of the current vs. urea concentration was reached using square-wave voltammetry in the concentration range of urea between 0.5 to $3.0{\mu}M$ with a relatively low limit of detection of $0.27{\mu}M$. The scanning electron microscopy was used to characterize the morphologies and properties of the nanocomposite layer, along with Fourier transform infrared spectroscopy. The results indicated that the nanocomposite film modified electrode exhibited a synergistic effect, including high conductivity, a fast electron-transfer rate, and an inherent catalytic ability.

Glucose Sensing Properties of Electrospinning-Synthesized ZnO Nanofibers (전기방사로 합성된 산화아연 나노섬유의 Glucose 감응특성)

  • The development of glucose biosensors has been attracting much attention because of their importance in monitoring glucose in the human body; such sensors are used to diagnose diabetes and related human diseases. Thanks to the high selectivity, sensitivity to glucose detection, and relatively low-cost fabrication of enzyme-immobilized electrochemical glucose sensors, these devices are recognized as one of the most intensively investigated glucose sensor types. In this work, ZnO nanofibers were synthesized using an electrospinning method with polyvinyl alcohol zinc acetate as precursor material. Using the synthesized ZnO nanofibers, we fabricated glucose biosensors in which glucose oxidase was immobilized on the ZnO nanofibers. The sensors were used to detect a wide range of glucose from 10 to 700 M with a sensitivity of $10.01nA/cm^2-{\mu}M$, indicating that the ZnO nanofiber-based glucose sensor can be used for the detection of glucose in the human body. The control of nanograins in terms of the size and crystalline quality of the individual nanofibers is required for improving the glucose-sensing abilities of the nanofibers.

Assembly of Laccase over Platinum Oxide Surface and Application as an Amperometric Biosensor

  • Laccase could be successfully assembled on an amine-derivatized platinum electrode by glutaraldehyde coupling. The enzyme layer formed on the surface does not communicate electron directly with the electrode, but the enzymatic activity of the surf ace could be followed by electrochemical detection of enzymatically oxidized products. The well-known laccase substrates, ABTS (2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid)) and PPD (p-phenylenediamine) were used. ABTS can be detected down to 0.5 ${\mu}M$ with linear response up to 15 ${\mu}M$ and current sensitivity of 75 nA/ ${\mu}M.$ PPD showed better response with detection limit of 0.05 ${\mu}M$, linear response up to 20 ${\mu}M$, and current sensitivity of 340 nA/ ${\mu}M$ with the same electrode. The sensor responses fit well to the Michaelis-Menten equation and apparent $K_M$ values are 0.16 mM for ABTS and 0.055 mM for PPD, which show the enzymatic reaction is the rate-determining step. The laccase electrode we developed is very stable and more than 80% of initial activity was still maintained after 2 months of uses.

Detection of Volatile Alcohol Vapors Using Silicon Quantum Dots Based on Porous Silicon (다공성 실리콘을 근거한 실리콘 양자점을 이용한 휘발성 알콜 증기의 감지)

  • Silicon quantum dots base on photoluminescent porous silicon were prepared from an electrochemical etching of n-type silicon wafer (boron-dopped<100> orientation, resistivity of 1~10 ${\Omega}-cm$) and used as a alcohol sensor. Silicon quantum dots displayed an emission band at the wavelength of 675 nm with an excitation wavelength of 480 nm. Photoluminescence of silicon quantum dots was quenched in the presence of alcohol vapors such as methanol, ethanol, and isopropanol. Quenching efficiencies of 21.5, 32.5, and 45.8% were obtained for isopropanol, ethanol, and methanol, respectively. A linear relationship was obtained between quenching efficiencies and vapor pressure of analytes used. Quenching photoluminescence was recovered upon introducing of fresh air after the detection of alcohol. This provides easy fabrication of alcohol sensor based on porous silicon.

Simultaneous Electrochemical Determination of Hydroquinone, Catechol and Resorcinol at Nitrogen Doped Porous Carbon Nanopolyhedrons-multiwall Carbon Nanotubes Hybrid Materials Modified Glassy Carbon Electrode

  • The nitrogen doped porous carbon nanopolyhedrons (N-PCNPs)-multi-walled carbon nanotubes (MWCNTs) hybrid materials were prepared for the first time. Combining the excellent catalytic activities, good electrical conductivities and high surface areas of N-PCNPs and MWCNTs, the simultaneous determination of hydroquinone (HQ), catechol (CC) and resorcinol (RE) with good analytical performance was achieved at the N-PCNPs-MWCNTs modified electrode. The linear response ranges for HQ, CC and RE are 0.2-455 ${\mu}M$, 0.7-440 ${\mu}M$ and 3.0-365 ${\mu}M$, respectively, and the detection limits (S/N = 3) are $0.03{\mu}M$, $0.11{\mu}M$ and $0.38{\mu}M$, respectively. These results are much better than that obtained on some graphene or CNTs-based materials modified electrodes. Furthermore, the developed sensor was successfully applied to simultaneously detect HQ, CC and RE in the local river water samples.

Bismuth Coated Carbon Fiber Microelectrode with Gallic Acid n-Propyl Ester for Trace Copper Analysis (비스무스코팅 탄소섬유전극과 갤릭산 착물을 사용한 구리 이온의 흔적량 분석)

  • A bismuth-coated carbon fiber microelectrode was prepared using cyclic voltammetry (CV). An analytical application was performed for the copper analysis with Square Wave Stripping Voltammetry (SWSV). Gallic acid n-propyl ester (PG) was used for the complex formation with a copper ion, and electrochemical measurements were performed with a pre-amplifier of a low-current module for nano am per detection. The effects of various parameters on the response were optimized. Analytical working ranges of $0.03-25.9\;{\mu}gl^{-1}$ and $0-25\'mgl^{-1}$ Cu(II) were obtained. The relative standard deviation at $13\;mgl^{-1}$ Cu was 0.9% (n = 12) in optimum conditions. The detection limit was found to have been $0.019\;{\mu}gl^{-1}$, with a 30-sec accumulation time. The developed methods were applied to a copper assay in water samples.

Propectives of Environmental Colorimetric-Sensors (환경색센서에 관한 기술 전망)

  • The electrochemical or optical sensors for environmental pollutants are developed over the past several years. Nowadays, the development of colorimetric sensing is particularly challenging since it requires no equipment at all as color changes can be detected by the naked eye. Visual detection can give immediate qualitative information and is becoming increasingly appreciated in terms of quantitative analysis. In addition, simple colorimetric-sensor have shown useful in the detection, identification, and quantification of volatile organic compounds(VOC) in gas phase or heavy metal ion in aqueous phase. In this review, we investigated the wide applications and some drawbacks of colorimetric-sensors. And thus, we try to suggest the methodologies of development approach of multi-functional and reversible colorimetric-sensor.
