Constituents of the Essential Oil from Eclipta prostrata L.

  • The volatile aroma constituents of Eclipta prostrata L. (leaves, stems, and flowers) were isolated by hydro-distillation extraction method and analyzed by GC/MS. The yield of Eclipta prostrata L. essential oil was 0.1% (v/w), and its color was yellow. Sixty-eight volatile flavor compounds, which make up 71.15% of the total volatile composition of the essential oil were tentatively characterized. It contained 35 hydrocarbons (56.25%) with sesquiterpene predominating, 12 alcohols (3.05%), 8 ketones (3.83%), 9 aldehydes (1.86%), 2 oxides (6.03%), and 2 esters (0.13%). ${\alpha}$-Humulene, 6,9-heptadecadiene, (E)-${\beta}$-farnesene, and ${\alpha}$-phellandrene were the major abundant aroma components in Eclipta prostrata L., aromatic and medicinal plant.

  • The plant Eclipta prostrata, a member of the Compositae family, has folkloric reputation of being used as a medicinal agent in Bangladesh. In the present investigation, attempt was taken to explore the antimicrobial potency and cytotoxicity of its extractives and purified compounds. The methanolic extract of the whole plant, its n-hexane, carbon tetrachloride, chloroform, aqueous soluble fractions and two purified compounds, eclalbasaponin I (1) and II (2), obtained from Eclipta prostrata were subjected to screening for inhibition of microbial growth by the disc diffusion method at 300 and 100 ${\mu}g$/disc for extracts and pure compounds, respectively. In this case, the carbon tetrachloride and chloroform soluble fractions of the methanolic extract appeared very potent in terms of both zone of inhibition and spectrum of activity. However, all the extractives were also subjected to brine shrimp lethality bioassay for preliminary cytotoxicity evaluation. Here, the carbon tetrachloride soluble fraction of methanolic extract revealed the strongest cytotoxicity having $LC_{50}$ of 1.318 ${\mu}g$/ml.

  • To study the effects of anti-cancer, anti-metastasis and immune response improvement of herbal-acupuncture with Eclipta prostrata diffuse herba infusion solution(ELP-HAS), we injected ELP-HAS into Chok-samni(St36) of C57BL/6 mice implanted intravenously with B16-F10 melanoma. We have reached the following conclusion through the effect on the number of $CD25^+/CD4^+$, $CD8^+/CD3e^+$, $CD69^+/B220^+$, $NK1.1+/CD3e^+$ cells in mouse PBMCs, the effect on the pulmonary colony number, and the effect on MST(Median Survival Time) and ILS(Increase of Life Span) of C57BL/6 mice implanted intravenously with B16-F10 melanoma The results were obtained as follows: 1. In the experiment groups treated with ELP(Eclipta prostrata) Herbal-Acupuncture, the spleen cell proliferation in Balb/c mouse was significantly increased compared with control group. 2. In the experiment groups treated with ELP(Eclipta prostrata) Herbal-Acupuncture, the percentage of $CD25^+/CD4^+$, $CD8^+/CD3e^+$, $CD69^+/B220^+$, $NK1.1^+/CD3e^+$ cells in C57Bl/6 mouse PBMCs was increased compared with control group. 3. In the experiment groups treated with ELP(Eclipta prostrata) Herbal-Acupuncture, the pulmonary colony number of C57BL/6 mice implanted intravenously with B16-F10 melanoma was decreased significantly compared with control group. 4. In the experiment groups treated with ELP(Eclipta prostrata)Herbal-Acupuncture, MST(Median Survival Time) and ILS(Increase of Life Span) of C57BL/6 mice implanted intravenously with B16-F10 melanoma were increased significantly compared with control group.

Suppressive Effect of a Carbohydrate Fraction from Eclipta prostrata on the Apoptosis of the Mouse Splenocytes in Culture

  • Eclipta prostrata grows abundantly in the tropical and the sub-tropical parts of the world including most part of the Korean Peninsula. The plant has been traditionally used for the treatment of a number of inflammatory diseases including hepatitis and enteritis but the nature of its immuno-modulating activity needs more studies. In this study, water-soluble sugar-containing fractions were purified from the herb and their effects on the culture of mouse splenocytes were examined. One of the fractions significantly suppressed apoptosis of the splenocytes in culture, which involves the gene expression regulation of a number of cytokines and cytokine receptors including MIP1-$\beta$. This study could explain an immunological activity of Eclipta prostrata and would lead to identify an immuno-active compound from the plant.

A Study of Melanin Synthesis Pathway of the Ethanol extract of Eclipta prostrata In Vitro Study (한련초 에탄올 추출물의 멜라닌 합성 경로에 대한 연구)

  • Objectives : This Study was conducted to investigate the melanin synthesis effect of Eclipta prostrata and to determine the relationship between melanin synthesis effect of Eclipta prostrata and cAMP/PKA melanin synthesis pathway. Methods : We measured melanin contents, tyrosinase activity, expression of TRPs, p-CREB in B16F10 cells cultured with Eclipta prostrata ethanol extracts(EEP). And after treatment with H89 and dibutyryl cAMP, which inhibit or promote the activation of PKA, we observed changes in melanin synthesis and tyrosinase activity stimulated by EEP. Results : EEP increased melanin synthesis by promoting the expression of tyrosinase and TRP-1. It also promoted expression of p-CREB. H89 suppresses melanogenic effect and expression of tyrosinase, TRP-1 in B16F10 stimulated by EEP. Dibutyryl cAMP promotes melanogenic effect of EEP. Conclusions : The results of this study suggest that melanin synthesis effect of EEP is related to induction of tyrosinase and TRP-1 expression through the cAMP/PKA pathway.

Antiproliferative Phenolics from Eclipta prostrata in the Activated Hepatic Stellate Cells

  • Activity-guided isolation to search for antifibrotic compounds from natural products using HSC-T6 cells afforded nine flavonoids or phenolics, luteolin (1), 3'-O-methylorobol (2), acactin 7-rutinoside (3), sedelolactone (4), 4-methoxyphenol (5), 4-hydroxyaldehyde (6), 4-hydoxyaldehyde (7), 4-hydroxy-3-methoxybenzoic acid (8), and ferulic acid (9) from the methanolic extract of aerial parts of Eclipta prostrata L.. Among the isolated compounds, luteolin (1) significantly inhibited the proliferation of HSCs in dose- and time-dependent manners. Antifibrotic activity of E. prostrata and its phenolic compounds might provide potential therapeutical choice in the treatment of hepatic fibrosis.

  • The major polysaccharide, named EAP, was purified from the aerial parts of Eclipta alba by Sepharose CL-2B ion exchange chromatography and recycling HPLC. The molecular weight of EAP was estimated to be 20911.9 D by MALDI-TOF MS. In addition, the sugar composition was determined to be arabinose (23.6%), mannose (24.8%), galactose (12.3%), and glucose (41.3%), respectively, by GC analysis. The EAP decreased the blood sugar level, which was induced by alloxan in rats, dose dependently.

  • Eclipta prostrata is an annual herb, belonging to Asteraceae family, and has been traditionally used to improve immunity and treat hepatitis and bacterial disease in Korea. In this study, a new coumestan glucoside (1) along with ten known compounds (2 - 11) was isolated from E. prostrata. The chemical structures of isolates were elucidated to be wedelolactone-9-O-β-D-glucopyranoside (1), wedelolactone (2), demethylwedelolactone (3), apigenin (4), apigenin-7-sulfate (5), luteolin (6), luteolin-7-sulfate (7), luteolin-7-O-β-D-glucopyranoside (8), pratensein-7-O-β-D-glucopyranoside (9), 3,4-di-O-caffeoylquinic acid (10) and 3,5-di-O-caffeoylquinic acid (11) based on the spectroscopic evidence.

  • As environmentally sound agriculture emphasize gradually as well as the importance of environment, many studies on weed and soil nematode control of new concept which does not affect adversely environment are in progress. For this purpose, plant seeds which are representative weed species in field were selected and treated with microwave having generating capacity of $17.73kw/m^3$, and investigated germination rates of weed seeds and the number of soil nematode according to soil moisture contents, soil depth and irradiation times. The microwave effect on the germination rates was extremely high in soil moisture content of 40% and irradiation time over 40 seconds for irradiation time, while soil depth did not affect germination rate. In view of the results so far achieved, it seems that universal dispersed microwave system is effective for the control of weed seeds.

Melanogenic Effect of Eclipta Prostrata (L.) L. (한련초의 멜라닌합성 촉진 효과)

  • This study was peformed to investigate the mechanism of ethanol extract of Eclipta Prostrata (L.) L. (EEP) induced melanogenesis. EEP enhanced tyrosinase activity and melanin contents of B16F10 cells. Moreover, EEP increased the protein expression of tyrosinase and tyrosinase-related protein 1 (TRP-1). But EEP did not increase the protein expression of tyrosinase-related protein 2 (TRP-2). These results suggest that melanogenesis-promoting effect of EEP was involved in regulation of tyrosinaase and TRP-1 protein, and EEP may be a potent pigmentation darkening agent in hypopigmentation condition.
