Anti-inflammatory effect of Crypsinus hastatus biorenovation extract (고란초 생물 전환 추출물의 항염증 효과)

  • Biorenovation is a method for converting materials using the enzyme properties of microorganisms. Natural products converted by that method increase physiological activity or reduce cytotoxicity. In this study, we investigated the anti-inflammatory activity of crypsinus hastatus prothallium (CH) and biorenovated CH prothallium (CHB) using RAW 264.7 cells stimulated with lipopolysaccharide (LPS). CHB inhibited the production of nitric oxide, prostaglandin E2 and cytokines (interleukin-6, interleukin-1β, tumor necrosis factor-α) compared to CH at a concentration of 50-200 ㎍/mL. In addition, CHB concentration of 200 ㎍/mL inhibited the expression of inducible nitric oxide synthase and cyclooxygenase-2 protein by LPS stimulation to the level of the untreated control group. These results indicate that CHB could be a novel anti-inflammatory agent for cosmetic and pharmaceutical ingredients.It also suggests that the application of biorenovation has potential usefulness in developing anti-inflammatory materials. We applied Biorenovation technology to Distylium racemosum extract (DR) to generate Distylium racemosum biorenovation product (DRB), and investigated the anti-inflammatory properties of DRB in lipopolysaccharide (LPS)-treated RAW264.7 macrophages.   • Distylium racemosum is an evergreen tree growing wild on Jeju Island, which has been reported to exert biological activity. Obesity is induced by genetic, metabolic, environmental, and other factors. Among these, certain bacterial and viral infections have been shown to cause obesity, which is known as infectobesity. Human adenovirus (HAdV)-36 is one of the viruses that are known to cause infectobesity in humans. Unlike extensive research on preventing obesity and developing anti-obesity drugs, little research has been conducted specifically on the prevention and treatment of infectobesity. Therefore, this study aimed to evaluate the effects of phytochemicals from D. racemosum on the replication of HAdV-36. A549 cells infected with HAdV-36 were treated with an ethyl acetate fraction of a D. racemosum leaf extract (DRE), and the virus titer was calculated based on the hemagglutination (HA) titer. The results showed a concentration-dependent inhibitory effect of DRE treatment on HA titers. DRE treatment was also found to inhibit the cytopathic effects of the virus and the expression of viral genes. Quercitrin was identified as the constituent of DRE exerting an inhibitory effect on HAdV-36 replication. This study shows that DRE can be used as a candidate substance for the development of treatment for HAdV-36 infections. In addition, this study provides a basis to further investigate DRE for the development of anti-infectobesity medication.

Antioxidant Effects of Plant Extracts on Free Radicals and Lipid Peroxidation

  • Reactive oxygen species damage biomolecules such as lipids, proteins, sugars and DNA, which can not only lead to various disease but also oxidative damage resulting aging. In order to search for antioxidants from plants, the antioxidant effects of the MeOH extracts from 182 plants were evaluated. The results showed that thirteen plant extracts exhibited antioxidant activity (>80%) in DPPH radical assay, seven plant extracts demonstrated antioxidant activity (>40%) in the hydroxyl radical assay and eighteen plant extracts were active (>80%) in the lipid peroxidation assay. In particular, the extracts of Distylium racemosum (Hamamelidaceae), Astilbe koreana (Saxifragaceae), Astilbe chinensis and Euphorbia supina (Euphorbiaceae) were identified as potent principles of antioxidant activity in all the assay systems.

Effects of Indoor Light Intensity on the Growth Characteristics of $Distylium$ $racemosum$, $Osmanthus$ $heterophyllus$ and $Damnacanthus$ $indicus$ (실내 광조건이 조록나무, 구골나무 및 호자나무의 생육에 미치는 영향)

  • This study was conducted to develope new indoor plants and to investigate the effects of light intensity on the growth of $Distylium$ $racemosum$, $Osmanthus$ $heterophyllus$ and $Damnacanthus$ $indicus$ under 100 lux, 1,000 lux and 2,500 lux fluorescent lighting for six months in the environment-controlled growth chambers. 1. $Distylium$ $racemosum$ : Most of $Distylium$ $racemosum$ under 100 lux light intensity were blighted in two months, whereas it was 100% of survival under 1,000 lux and 2,500 lux after six months. Plant height, number of leaf, leaf width and leaf length became higher as light intensity increased. The plants maintained under 2,500 lux showed the greatest plant height and leaf number. 2. $Osmanthus$ $heterophyllus$ : $Osmanthus$ $heterophyllus$ under 100 lux light intensity were blighted in two months, whereas it was 100% of survival under 1,000 lux and 2,500 lux after six months. However, under 1,000 lux, it paused plant height and was not increased in leaf number any more. A plant growth status showed the highest value under 2,500 lux in all conditions. 3. $Damnacanthus$ $indicus$ : $Damnacanthus$ $indicus$ was defoliated and blighted under 100 lux light intensity in two months, whereas it was grown properly with 1,000 lux or above. However, the growth under 2,500 lux of $Damnacanthus$ $indicus$ was superior to other treatments. But, $Damnacanthus$ $indicus$ under 1,000 lux after 6 months was more favorable chlorophyll contents, leaf length and leaf width than 2,500 lux. As increasing slightly of chlorophyll contents and leaf growth under 1,000 lux, $Damnacanthus$ $indicus$ could be utilized highly to the indoor ornamental plant.

  • Inhibitors of α-amylase are important for the treatment of diabetes and obesity. Using enzyme inhibitor's activity, ethanolic extracts of 87 species in 12 families were screened and compared their inhibitory effect on α-amylase, As a results, we can find that extracts of Distylium racemosum, Acer tegmentosum, Corylapsis veitchiana, Cornus walteri and Corylapsis spicata showed higher α-amylase inhibitory activities than the others and have potential possibility of using control agents for carbohydrate-dependent disease.

In vitro Screening of Jeju Medicinal Plants for Cosmeceutical Materials

  • One of the important functions of skin is protection from harmful environments. Many studies have explored how to prevent skin from wrinkling and the occurrence of pigmentation changes. Skin wrinkling and pigmentation changes could be caused by unusual disruption of connective tissue, the formation of free radicals and ultraviolet radiation. In this study, extracts obtained from 254 different kinds of Jeju medicinal plants were screened for inhibitory effects on tyrosinase and elastase, and for free radical scavenging effects. Four herbs, Phormium tenax, Morus bombycis, Morus alba, and Cudrania tricuspidata, were potent inhibitors of tyrosinase ($IC_{50}$ values 4.62, 5.46, 8.17, and 64.17 ${\mu}g$/mL, respectively). Aleurites fordii [$IC_{50}$: 5.29 ${\mu}g$/mL, 1,1-diphenyl-2-picrylhydrazyl (DPPH)], Distylium racemosum ($IC_{50}$: 6.14 ${\mu}g$/mL), Acer palmatum ($IC_{50}$: 5.44 ${\mu}g$/mL), and Spiraea salicifolia ($IC_{50}$: 5.25 ${\mu}g$/mL) showed good antioxidative effects. Furthermore, Distylium racemosum ($IC_{50}$: 7.51 ${\mu}g$/mL), Diospyros kaki ($IC_{50}$: 15.1 ${\mu}g$/mL), Cornus macrophylla ($IC_{50}:$ 16.59 ${\mu}g$/mL), and Psidium guajava ($IC_{50}$: 40.25 ${\mu}g$/mL) exhibited potent inhibitory effects on elastase. These results suggest that medicinal plants possessing several biological activities may be potent inhibitors of the processes involved in pigmentation increases and aging. Further investigations will focus on in vivo assays and on the chemical identification of the major active components responsible for whitening and anti-aging activity in the screened efficacious extracts.

Inhibitory Effects of Ninety Nine Korean Plants on Human Immunodeficiency Virus Type 1 Pretense Activity

  • Ninety nine extracts from Korean plants were screened for their inhibitory activities on human immunodeficiency virus (HIV) type 1 pretense by an HPLC method. The pretense inhibitory activities were determined by incubating the extracts in reaction mixtures containing pretense and substrate (His-Lys-Ala-Arg-Val-Leu-(p-NO$_2$- Phe)-Glu-Ala-Nle-Ser-NH$_2$) to perform proteolytic cleavage reactions. Of the extracts tested, the water extracts of Viburnum awabuki (stem and leaves) and Distylium racemosum (leaves) had the highest pretense inhibitory activities at a concentration of 100ug/mL. Activity-guided fractionation, revealed that the n-butanol fraction of the V. awabuki extract and the ethyl acetate fraction from the D. racemosum extract had the greatest inhibitory activity on HIV-1 pretense.

Protective Effect of Some Medicinal Plants on tert-Butyl Hydroperoxide-Induced Oxidative Stress in Human Keratinocytes

  • It is well known that excessive production of reactive oxygen species (ROS) leads to oxidative stress, loss of cell function, and ultimately apoptosis or necrosis. To search for natural antioxidants able to modulate cellular oxidative stress, we investigated the protective effect of ethanol extracts of 17 medicinal plants selected from the preliminary antioxidant screening on tert-butyl hydroperoxide (t-BuOOH)-induced oxidative stress in human keratinocytes. The result showed that extracts of the four plants, Distylium racemosum, Astilbe chinensis, Cercis chinensis and Sapium japonicum, exhibited significant cytoprotective activity (over 50% protection) against t-BuOOH-induced cellular injury.

  • This report observed for anatomical structure and identified species of small wood debris collected at Chungi-Hyankyo, Yunbuk-Chung, Jeju-Hyankyo, Teachung-Hyangkyo and Kwanduk-Chung in Jeju province constructed in a central period of Yi-dynasty, and now named as local cultral assets. 1. Sample of Chungi-Hyangkyo is gymnosperm with window-like pit, axial and radial cannal & dentate ray tracheid. This sample with identified with Pinus densiflora or Pinus thunbergii. 2. Samples of Yunbuk-Chung and Jeju-Hyangkyo are diffuse porous wood having small vessels with scalariform perforation plates scattered almost equally in annual ring, possess heterogenus ray parenchyma & banded apotracheal parenchyma. This sample is identified with Distylium racemosum. 3. Sample of Teachung-Hyangkyo is ring porous wood composed of big vessels of pore zone and compound vessels outside pore zone. It also has simple perforation plates, tylosis, homogenlls uniseriate ray parenchyma & diffuse parenchyma This sample is identified with Castanopsis cuspidata. 4. Sample of kwanduk-Chung is ring porous wood composed of big vessels of pore zone and compound vessels outside pore zone. It has simple perforation plates, heterogenus ray parenchyma, crystal, vasicentric parenchyma. This sample is identified with Zelkova serrata.

Attitudinal Distribution of Plant Communities at Donnaeko Valley in the Mt. Hallasan (한라산 돈내코계곡의 해발고별 식물군집분포)

  • To investigate the forest community structure ranging from 200 meters to 1,350 meters above sea level at Donnaeko valley of Mt. Hallasan in 2006, 15 plots were surveyed. According to the classification analysis by TWINSPAN, the plant communities were divided into five groups of Castanopsis sieboidii community, Castanopsis sieboldii - Quercus acuta community, mixed forest, Carpinus laxiflora - Quercus serrata community and deciduous broad-leaved forest. 22 species of evergreen broad-leaved trees such as Castanopsis sieboldii, Quercus acuta, Distylium racemosum, Camellia japonica, Eurya japonica, Ligustrum lucidum, Ilex crenata, Daphnipyllum macropodum, etc. were growing at Donnaeko valley. According to the attitudinal distribution of evergreen broad-leaved trees, Castanopsis sieboidii was a dominant species distributed from 200 meters to 350 meters above sea level, Castanopsis sieboldii and Quercus acuta were dominant species distributed from 400 meters to 600 meters above sea level and Quercus acuta was a dominant species distributed from 660 meters to 700 meters above sea level. Ilex crenata, Daphniphyllum macropodum, Elaeagnus glabra were distributed up to 1,350 meters above sea level in Donnaeko.
