Docosahexaenoic acid 함유 재구성지질의 효소적 합성 연구 (Lipase-Catalyzed Synthesis of DHA-Enriched Structured Lipid)

  • 조류(microalgae, from Schizochytrium sp.)로부터 DHA가 풍부하고 이취가 적은 지질을 획득하고 이를 옥수수유와 함께 반응기질로 이용하여 재구성지질을 합성하였다. 본 실험에 사용된 합성조건으로 oleic(17.3 mol%), linoleic(31.8 mol%), 그리고 docosahexaenoic acid(14.9 mol%)가 함유된 재구성지질을 합성하였다. ${\alpha}-,\;{\gamma}-$, 및 ${\delta}-tocopherol$의 총 함량은 0.059%로 분석되었고, 비누화 가(183.8) 및 요오드 가(201.8)를 측정하였다. 한편, 수행된 반응조건 및 반응시간동안 대부분의 반응은 6시간 내에 이루어 졌고, 그 상승폭은 3-6시간 사이에서 약 2.3배의 증가를 보이며 가장 크게 나타났다.

기수산 요각류 Paracyclopina nena의 배양을 위한 최적 미세조류의 선별 (Selecting the Optimal Microalgal Species for Culturing the Brackish Water Copepod Paracyclopina nana)

  • This study selected the optimal microalgal species for mass culture of a brackish water copepod Paracyclopina nana. Fifteen microalgal species were tested to examine nauplius production and the survival and maturation of brood females. Total and daily nauplius production were highest in P. nana fed Tetraselmis suecica followed Isochrysis galbana, Dunaliella tertiolecta, and Phaeodactylum tricornutum. With a monospecific microalgal diet, the total density was highest with P. nana fed I. galbana, at 63.3 inds./mL. With a mixed microalgal diet, the total density of P. nana fed T. suecica+I. galbana was higher than that fed other mixed diets, although there was no difference between a monospecific diet of I. galbana and a mixed diet of T. suecica+I. galbana. Examining the fatty acid composition of P. nana, the eicosapentaenoic acid (EPA) was highest in P. nana fed T. suecica at 5.4% while the docosahexaenoic acid (DHA) content was highest I. galbana diet at 31.9%. Although no DHA was detected in T. suecica, P. nana fed this microalgal species had a high DHA composition of 24.3%. We suggest that the optimal microalgal species for the mass culture of P. nana is T. suecica which is easy to culture on masse and has a high linolenic acid content.

  • Both docosahexaenoic acid (DHA, 22:6n-3) and eicosapentaenoic acid (EPA, 20:5n-3) have attracted increasing attention since the first epidemiological report on the importance of n-3 essential fatty acids. Lipids in microbial cells play various biological roles and, consequently, much research has been carried out on their role in cell physiology. The lipid composition of microorganisms can exhibit considerable variations depending on environment. The effects of culture conditions, temperature (15, 20, 24, 28, 32 and $36^{\circ}C$), salinity (10, 20, 30, 40 and 50 psu), pH (pH5, 6, 7, 8 and 9), rotation speeds (50, 100, 150 and 200 rpm), carbon sources, nitrogen sources and C/N ratio on the production of docosahexaenoic acid, fatty-acid profiles, and acids secreted to the broth culture by the oleaginous microorganism, Schizochytrium mangrovei (KCTC 11117BP), were studied. Temperature (initially $28^{\circ}C$), salinity (20 psu), pH (pH7), rotation speeds (100 rpm), organism fatty acids, and secreted acids in the broth were varied during cultivation of S. mangrovei. At pH 7.0, S. mangrovei was able to accumulate lipids up to 40% of its biomass, with 13% (w/w) DHA content. The monosaccharides glucose and fructose, and yeast extract were suitable carbon and nitrogen sources, respectively. The primary omega-3 polyunsaturated fatty acid produced was docosahexaenoic acid.

Dietary supplementation of finishing pigs with the docosahexaenoic acid-rich microalgae, Aurantiochytrium limacinum: effects on performance, carcass characteristics and tissue fatty acid profile

  • Objective: The aim of this experiment was to evaluate the effect of dietary supplementation with the docosahexaenoic acid (DHA)-rich microalgae, Aurantiochytrium limacinum (AURA) on pig performance, carcass traits, and the fatty acid composition of pork Longissimus lumborum (LL) and backfat. Methods: A total of 144 Pig Improvement Company (PIC)${\times}$Goland finishing pigs (72 females and 72 castrated males) of mean weight 117.1 (${\pm}13.1$) kg were blocked by sex and body weight and provided with 0% or 1% AURA in isonutritive and isocaloric diets. A total of 24 pens provided 12 replicates per treatment. Animals were weighed on day 0 and 28 with feed and water intake recorded per pen. After 31 days supplementation (28 days of study and 3 days until the slaughtering date) three animals per pen (n = 72) were slaughtered and the LL and backfat thickness, lean meat content and dressing percentage were recorded for the carcasses. The fatty acid (FA) profile of the LL and backfat was established by direct FA methyl ester synthesis. Results: No differences were observed for any performance parameters or carcass traits. Supplementation with AURA resulted in significant changes to the FA profiles of both the LL and backfat with male and female pigs responding differently to supplementation in terms of particular FAs. Overall, pork LL samples had significantly higher eicosapentaenoic acid (p<0.001) and DHA concentrations (p<0.001), and higher omega-3 (n-3) FAs (p<0.001), as well as an increased omega3:omega6 (n-3:n-6) ratio (p = 0.001). For backfat, supplementation resulted in significantly higher amounts of DHA (p<0.001) and n-3 FAs (p<0.001). Conclusion: These results indicate that dietary supplementation with 1% AURA over a 31 day period can increase the FA composition of pork LL and backfat, specifically the DHA, with no major impact on growth performance and carcass traits.

RELATIONSHIPS BETWEEN DAILY HERBAGE INTAKE OF GRAZING CATTLE WITH DAILY HERBAGE ALLOWANCE AND LEAFINESS

  • In order to investigate the relationship between daily herbage allowance (DHA) and daily herbage intake (DHI) on the points of leaf and stem masses, grazing experiments were carried out on the pasture of tall fescue (Festuca arundinacea Schreb.). The experiments consisted of four 3-days' grazing periods which were from August 14 to 17, August 17 to 20, October 12 to 15 and October 15 to 18 in 1990. The value of DHA was 32.1 to 84.3, and the value of DHI was 9.9 to 27.0. The index of leafiness (leaf to stem ratio, LSR) was 0.74 to 3.06. The relationship between DHI and DHA was quadratic. Both relationship of daily leaf intake (DLI) to daily leaf allowance (DLA) and that of daily stem intake (DSI) to daily stem allowance (DSA) were likely to be asymptotic forms, although DHI did not show any significant relationships to DLA nor to DSA. The relationship between DLI and LSR was quadratic, and a negative relationship was found between DSI and LSR. From the relationship of DLI to LSR and that of DSI to LSR, the relationship of daily intake of leaf and stem (DLSI) to LSR was derived as quadratic and the LSR value at which the maximum DLSI was attained was estimated. The DHA, DHI, DLA, DLI, DSA, DSI and DLAI were expressed as the same unit of g/kg live body weight/day.

반응표면분석에 의한 기능성 유지의 효소적 합성 조건 최적화 (Optimization of Enzymatic Synthesis Condition of Structured Lipids by Response Surface Methodology)

  • Omega-3 지방산인 DHA가 풍부하고 이취가 적은 조류(microalgae, Schizochytrum sp.)로부터 지질을 획득하여 이를 옥수수유와 기질로 이용, 고정화효소인 RM IM(from Rhizomucormiehei)을 촉매로 하여 interesterification 반응에 의해 고기능성 유지를 효과적으로 생성하기 위하여 합성조건을 반응표면분석에 의해 최적화하였다. 항온교반수조에서 소량 합성된 재구성 지질의 TG 지방산 조성 분석 결과, 재구성지질에 함유된 DHA의 함량은 15.06mol%를 나타내었고, DHA-enriched algae oil에 과량 함유된 palmitic acid(30.67mo1%), myristic acid(11.64mol%)의 함량은 각각 30, 60% 감소하여 21.70, 4.96 mol%를 보였으며, oleic과 linoleic acid의 함량은 급격히 증가하여 각각 20.20, 27.34 mo1%를 나타내었다. RP HPLC-ELSD system을 이용하여 재구성지질의 TG 형태를 분리한 결과, 두 기질에 존재하지 않은 다수의 새로운 TG peak를 확인한 수 있었으며, 이중 쉽게 식별이 가능한 3개의 peak를 선택, 이들 peak area% 총합을 반응변수로 하고 온도$(35-75^{\circ}C,\;X_1)$, 시간(2-42시간, $X_2$) 및 효소농도(2-14%, $X_3$)를 요인변수로 하여 중심합성계획에 의해 반응조건을 최적화하였다. 그 결과, 온도$(70.28^{\circ}C)$, 시간(28.74시간), 효소농도(11.30%)의 최적합성조건에서 최대값 6.97 area%로 예측되었다.

회분식 반응기를 이용한 DHA 함유 재구성지질의 효소적 합성 및 이화학적 특성 분석 (Analysis and Enzymatic Production of Structured Lipids Containing DHA Using a Stirred-Batch Type Reactor)

  • 미세조류(microalgae)인 Schizochytrium sp.로부터 유래된 조류유(250 g)와 식물성 유지인 대두유(250 g)의 혼합 유지를 기질로 하고, sn-1,3 위치에 특이적으로 작용하는 Lipozyme RM IM(Rhizomucor miehei, EC 3.1.1.3)을 이용한 interesterification에 의해 회분식 반응기$(65^{\circ}C,\;300 rpm,\;15hr)$에서 재구성 지질을 합성하였다. 합성 된 재구성 지질의 주요 지방산 조성은 DHA$(15.7\;mol\%)$와 linoleic acid$(31.1\;mol\%)$, palmitic acid$(20.2\;mol\%)$, oleic acid$(13.5\;mol\%)$$EPA(6.6\;mol\%)$로 분석되었으며, linoleic aicd와 DHA가 sn-2 위치에서 높은 함량을 차지하였다. Normal-phase HPLC 분석결과 재구성지질은 $87.1\;area\%$의 TAG와 $12.1\;area\%$의 DAG 형태와 같은 중성지질로 구성되어 있었다. Reversed-phase HPLC 분석 결과 재구성 지질의 $PN=20\~22(10.3\;area\%),\;PN=24\~32(53.1\;area\%)$$PN=36\~48(36.6\;area\%)$ peaks내에 각각 PN=20, PN=22, PN=28, PN=30, PN=34, PN=38, PN=42를 가지고 재구성 지 질 함량의 약 $50.5\;area\%$를 차지하며 반응기질과 구별된 새로운 peaks가 합성에 의해 생성되었음을 확인하였다 재구성지질의 요오드가는 고도의 불포화도를 가지는 조류유(274.0)보다 약 $25\%$ 낮아진 206.7을 나타내었으며, 분자량을 설명하는 비누화 값이 183.8, 산가는 반응기질과 같은 0.3 이하의 측정값을 나타내었다. 재구성지질의 Hunter color $L^{\ast}(\pm\;lightness/darkness)$ 값은 13.8로 측정되었으며, $b^{\ast}(\pm,\;yellowness/blueness)$ 값은 13.8로 측정되었으며, $b^{\ast}(\pm,\;yellowness/blueness)$ 값은 반응기질보다 유의적으로 가장 높은 55.1을 나타내었다(p<0.05).

냉장보관된 농축 먹이생물의 지방산 조성 변화와 농축먹이로 사육한 굴 유생의 생존율 (Fatty Acid Composition of Concentrated Phytoplanktons by Cold Storage and Their Effects on the Larval Survival of Pacific Oyster, Crassostrea gigas)

  • 패류 인공종묘 생산시에 먹이생물을 효율적으로 원활하게 공급하기 위한 일환으로 비종묘생산시기에 미리 생산하여 사용 가능한지 여부를 판단하기 위하여 패류 사육에 가장 일반적으로 사용되며 먹이효율도 우수한 것으로 알려진 Pavlova lutheli, Isochrysis galbana, Isochrysis aff, galbana Chaetoceros calcitrans를 농축 후, $4^{\circ}C$에서 보관하면서 보관기간에 따른 먹이생물의 생존율과 이들 먹이의 굴 유생에 대한 사육 효과를 생먹이와 비교하였으며 이들의 총지질 중 지방산 조성도 분석하였다. $4^{\circ}C$에서 30일간 냉장보관한 먹이생물의 생존율은 $23\~31\%$였다. 굴 유생에 4종 먹이생물을 생먹이와 농축먹이 형태로 공급하여 사육한 결과, 참굴 유생의 생존율은 농축먹이 공급구가 다소간 높았으며, 특히 농축한 I. aff. galbans를 공급한 실험구가 유생의 생존율이 가장 높았다. 먹이생물은 냉장보관 후 구성 지방산 중 EPA와 DHA가 유의적으로 증가하였고, 특히 I. aff. galbana는 냉장보관 후 조사한 먹이생물 중 DHA의 비율이 가장 높았다. 따라서 패류 유생 사육시 DHA는 먹이가치의 중요한 요인이므로 먹이생물을 농축 냉장 보관하여 공급함으로써 굴 유생의 생존율을 향상시킬 수 있는 것으로 조사되었고 특히 I. aff, galbana를 냉장보관하여 굴 유생에 공급하는 것이 바람직한 것으로 여겨진다.

Effects of Selected Fatty Acids Supplementation on Growth and Fecundity in Artemia franciscana

  • Effects of supplementing selected fatty acids on fatty acid incorporation (17 days) , and progeny production (14 days) in Artemia franciscana (Great Salt Lake, USA) were studied. To compare with the control four diets, which differed in fatty acid composition alone contain Dunalieia tertiolecta and an emulsion either rich in OA (oleic acid, 18: 1 n-9), ARA (arachidonic acid, 20:4n-6), EPA (eicosapentaenoic acid,20:5n-3), or DHA (docosahexaenoic acid, 22:6n-3). Each of these emulsions was supplemented at a ratio of 20 % of the daily dose of D. tertiolecta (% algal dry weight). The initial OA and ARA values were 33.5 and 1.7 mg/g DW of freshly-hatched nauplii, respectively. After 11 days of feeding, these values increased to 38.8 and 7.6 mg/g DW in Artemia receiving the fatty acid sup-plement rich in each of the respective fatty acids. After 14 days, the levels were almost doubled, reaching 62.8 and 13.4 mg/g respectively. On EPA supplementation, its level after 11 days of feeding was 14.3 and 17.3 mg/g in male and female, respectively and was 16.0 and 23.1 mg/g in the male and female after 14 days, respectively. The EPA accumulated more in the body (39.1 mg/g) than in ovisac (16.9 mg/g). In the DHA supplementation group also, DHA levels after 11 days of feeding were 3.1 and 5.5 mg/g in male and female, respectively. After 14 days, the DHA level continued to increase in male. but slightly decreased to 4.6 mg/g in female. It was not richer in ovisac (2.6 mg/g) than in the remaining body of female (4.6 mg/g). In conclusion, fatty acids supplied by a lipid emulsion as a supplement to the algal diet are well incorporated in the adult Artemia. Apart from being an extra source of energy, these emulsions may function as source of HUFA which may play an essential role for growth and progeny production (fecundity) of Artemia.

대사공학으로 제작된 재조합 Klebsiella pneumoniae를 이용한 아세토인 생산 (Acetoin Production Using Metabolically Engineered Klebsiella pneumoniae)

  • 아세토인(acetoin)은 식품과 화학산업에서 플랫폼 물질로 이용되며 산업적으로 다양한 응용이 가능한 물질이다. 본 연구에서는 대사공학(metabolic engineering)을 이용하여 아세토인의 생산량이 증가한 재조합 Klebsiella pneumoniae를 구축하였다. 우선 2,3-부탄디올(2,3-butanediol)생산을 위해 제작되었던 재조합 K. pneumoniae (KMK-05)에서 두 가지 2,3-butanediol dehydrogenase (budC, dhaD)를 유전체에서 제거하여 아세토인 생산량을 늘리고, 전사인자 중 하나인 AcoK를 제거하여 아세토인을 분해하는 효소의 발현량을 줄였다. 그리고 NADH oxidase를 발현시켜 세포 내 산화 환원 균형(redox balance)을 맞춰 대사흐름을 개선하였다. 이렇게 대사공학을 통해 구축된 재조합 Klebsiella pneumoniae(KJW-03-nox)로 아세토인 생산량과 수율을 높였고, 36시간 동안의 유가식 배양을 진행하여 51 g/L의 아세토인 농도와 최대 생산성 2.6 g/L/h을 달성하였다.
