Randomly Amplified Polymorphic DNA (RAPD) Analysis on Compatibility of Korean Native Cymbidium goeringii with Other Cymbidium Species (한국춘란(韓國春蘭)과 다른 Cymbidium간의 교배친화성(交配親和性)에 대한 RAPD 분석)

  • Cymbidium goeringii native to Korea and other orchid plants were pot-grown from spring to autumn under the greenhouse conditions, and were subjected to artificial pollination to elucidate the compatibility by revealing viable seed formation. A notable compatibility was found when Cym. goeringii was selfed and was crossed with either Cym. ensifolium, Cym. kanran, Cym. sinense, Cym. sinense for. albo-jucundissimum, Cym. 'Crystal Cherry Angel', or Cym. 'Anmitsu Hime'. Cym. goeringii, however, did not show such compatibility when crossed with either Cym. faberi, Cym. aloifolium, Dendrobium chrysotoxum, or Phalaenopsis spp. RAPD analysis indicated that taxa relationship between Cym. goeringii and either Cym. faberi or Cym. aloifolium (respective chromosome number, 2n=40) was distant, showing no compatibility, and even more distant in the case of cross-pollination between Cym. goeringii and either Dendrobium chrysotoxum or Phalaenopsis spp. having different chromosome number from all Cymbidium species.

Variation Analysis of Long-term in vitro Cultured Cymbidium goeringii Lindley and Cymbidium kanran Makino (장기간 기내 배양한 춘란(Cymbidium goeringii Lindley) 및 한란(Cymbidium kanran Makino)의 변이 비교)

  • RAPD (random amplified polymorphic DNA) analysis was examined to detect variation of in vitro cultured 30 rhizomes of Cymbidium goeringii Lindley and Cymbidium kanran Makino, with long-term (8 years) subculture, respectively. Out of 151 DNA bands detected, the 40 were polymorphic with a polymorphic rate 26.4% in the C. goeringii. Out of 155 DNA bands detected, the 56 were polymorphic with a polymorphic rate 36.1% in the C. kanran. Genetic similarity matrix (GSM) shows from 0.825 to 1.00 with an average of 0.944 in the rhizomes of C. goeringii and 0.812 to 1.00 with an average of 0.913 in the C. kanran. According to the clustering analysis, C. goeringii was divided into 1 group and 2 independent individuals and its structure of clustering was simple than that of C. kanran. The higher polymorphism and the decreased GSM were showed in the long-term in vitro cultured C. goeringii and C. kanran supplemented with growth regulators. The results provide as fundamental data to develop a new materials for plant breeding and resources plant.

Study on Germination of Seed and Growth of Rhizome in Cymbidium goerinii in vitro (Cymbidium goeringii 종자의 in vitro 에서의 발아에 관한 기초적 연구)

  • The study was conducted to determine the Ms orthogonaL modia and the concentration of plant growth regulator for seed matura-tion and growth of rhizome from Cymbidium goeringii Germination waswell in dark condition, but the growth of rhizome was better un-der dark than under light condition in MS orthoTonal . Sucrose con-centration( 3 %) gave better results than higher ones(6%), andthe use of NAA(0.1 PPm) effect significant difference of seed ge-rmination .But the growth of rhizome was best in medium Containingsucrose concentration(3%) Ippm NAA and 1 PPm BA.

  • Cymbidium goeringii REICHB. fil was extracted with 80% MeOH and solvent-fractionated with EtOAc, n-BuOH and $H_2O$, successively. From EtOAc fraction, three sterols were isolated on repeated silica gel and ODS column chromatographies. The chemical structures were determined as ${\beta}-sitosterol$ (1), daucosterol (2) and ergosterol peroxide (3) by NMR, MS and IR, which is the first to be isolated from Cymbidium goeringii REICHB. fil.

Effects of Prochloraz and Tebuconazole on Control of Fusarium Bulb and Root Rot of Oriental Orchid, Cymbidium goeringii (Prochloraz와 Tebuconazole의 Fusarium oxysporum에 의한 춘란(Cymbidium goeringii) 구경썩음병 방제효과)

  • Eight fungicides including prochloraz, tebuconazole, benomyl, mancozeb, thiophanate-methyl, azoxystrobin, and fluazinam were examined for their control effects on Fusarium bulb and root rot of oriental orchid, Cymbidium goeringii. Among the chemicals, prochloraz and tebuconazole were the most effective on suppression of the causal pathogen, F. oxysporum in vitro and on control of the disease in vivo. Prochloraz and tebuconazole inhibited mycelial growth of the fungi 95~100% at 10ppm a.i. and microconidial germination 75~100% at 100ppm a.i. Prochloraz and tebuconazole showed 80~92% and 84~88% protective control value on the disease, respectively. However, curative effects on infected orchid were relatively low. Other chemicals showed no or lower than 20% curative and 50% protective control value. Results indicated that prochloraz and tebuconazole can be used for the control of the Fusarium bulb and root rot of oriental orchids. However, the chemicals need to be applied prior to the disease development to achieve successful control efficacy.

Community Structure and Floristic Composition of Cymbidium goeringii Group in Korean Islets (한반도 도서지역 춘란집단의 종조성과 군락구조)

  • This text was analyzed and investigated the vegetation and floristic composition by ordination and classification of phytosociological method, to evaluate the species composition and community structure of Cymbidium goeringii group in Korean islets. In habitat of 33 plots, the mean altitude was 65.9m, the direction was the southeast slope, the mean slope was 7.9%. The coverage of Cymbidium goeringii was 4.5%. The appearing plants with the Cymbidium goeringii was the total 102 taxa, and it was the kind of trees 68 taxa (66.7%), herbs 34 taxa (33.3%), evergreen plants 36 taxa (35.3%) and deciduous plants 66 taxa (64.7 %) respectively. The frequency of appearing plant was the highest in the Eurya japonica (48.5%), followed by Pinus thunbergii (45.5%), Smilax china (36.4%), Carex lanceolata (33.3%), Hedera rhombea (33.3%), Machilus thunbergii (30.0%), Styrax japonicus (30.3%) and Pinus densiflora (27.3%), respectively. The vegetation of tree layer in Cymbidium goeringii group was classified into Pinus thunbergii community, Pinus densiflora community, Castanopsis sieboldii community and Quercus variabilis community. Pinus densiflora community showed a strong combination with Cymbidium goeringii group in Korean islets. Pinus thunbergii community among communities was combined with Castanopsis sieboldii community, and Pinus densiflora community and Quercus variabilis community were combined.

Identification of Orchid Mycorrhizal Fungi Isolated from Five Species of Terrestrial Orchids in Korea (한국에 자생하는 5종의 지생란에서 분리한 난균근균의 동정)

  • This study was performed to isolate and identify the orchid mycorrhizal fungi (OMF) from roots of five species of terrestrial orchids in Korea; Cymbidium goeringii, Spiranthes sinensis, Calanthe discolor, Bletilla striata, Pogonia minor. DNA was extracted from isolated OMF and ITS region was amplified using primers, ITS1-OF and ITS4-OF. Four species of OMF belong to Tulasnellaceae and Sebacinaceae were identified; Tulasnella calospora, Tulasnella irregularis, Tulasnella sp., Sebacina vermifera.

Days to Germination and Effect of Growth Regulator on Rhizome Growth in Cymbidium goeringii Hybrid

  • Germination efficiency, such as days to germination and conditions for the hybrid seeds, was tested after hybrid seeds were sown on the medium. Seeds were germinated from 67 to 126 days after seeding in all cross combinations, and germination condition was different among hybrid combinations. There was big difference on days to germination based on the different media, that is, days to germination in the hyponex medium was shorter than that in the Knudson C medium. Mutants such as MMS and NaN$_3$ were used to cause mutation. Germination occurred with 0.1 % NaN$_3$ and MMS. However, even though germination occurred in other treatments (0.01 % and 0.2% of NaN$_3$ and MMS), brownish phenomenon was intense, or did not proceed and got worse after 4-5 months of seeding. In addition, it was performed to choose appropriate medium for the growth of Cymbidium rhizome whose media adaptability is more different than other orchids. Different concentration of NAA and Kinetin was used. As a result, C. goeringii hybrid showed difference for the concentration of NAA and Kinetin combination.

Determination and Application of Combined Genotype of Simple Sequence Repeats (SSR) DNA Marker for Cultivars of Cymbidium goeringii (춘란(Cymbidium goeringii) 품종에 대한 Simple Sequence Repeats (SSR) DNA 마커의 복합 유전자형 결정과 적용)

  • Cymbidium goeringii is one of the most important and popular species in the orchid family in north-east Asia. In the present study, we prepared multiplex PCR system, and used it for the genotyping of eight simple sequence repeats (SSR) markers (CG409, CG415, CG709, CG722, CG787, CG1023, CG1210, and CG1281) in subject with 40 samples of cultivated varieties. All the analyzed samples showed different combined genotypes. The average combined power of discrimination was very high value of $7.14{\times}10^{-10}$, and observed heterozygosity (Ho = 0.466) was similar with two wild populations of C. goeringii, which may indicate no or little occurrence of genetic change after collection from wild habitats. The present study also developed a two-dimensional barcode to express information of genotype results of eight SSR markers (SSR DNA ID). The discrimination power of DNA ID between two individuals will be statistically more than 99.999999%. The SSR DNA ID and two-dimensional barcode may be very usefully applied for the discrimination and maintence of cultivars of C. goeringii.

Analysis of Genetic Relationship Among Collected Cymbidium goeringii Based on RAPD (RAPD를 이용한 춘란 수집종 20 품종의 유연관계 분석)

  • This research was performed for making data-base of cross-breeding between Cymbidium goeringii cultivars. Morphological characteristics were investigated and then genetic relationship was analyzed. Collected 20 Cymbidium goeringii cultivars were clustered into 2 groups. Seven cultivars were clustered into group I, and thirteen cultivars were clustered into group II. Group I doesn't have leaf pattern. Group have leaf pattern. The genetic relationship among collected 20 Cymbidium goeringii cultivars was anaylzed using RAPD with ten 10-mers random primer. Eighty-nine bands were generated by RAPD. Among the rest, three bands were monomorphic and eight-six bands were polymorphic. Overall similarity degree ranged from 0.521 to 0.862. The result of RAPD analysis was clustered into 2 groups, too. Sixteen cultivars were clustered into GroupX, and four cultivars were clustered into GroupY. Result of classification with morphological characteristics and RAPD showed different pattern, but 4 cultivars of GroupY by RAPD analysis were included in groupby morphological characteristics. Crossbreeding combination among low related coltivars in RAPD analysis may get more efficient result.
