Photoaging protective effects of BIOGF1K, a compound-K-rich fraction prepared from Panax ginseng

  • Background: BIOGF1K, a compound-K-rich fraction, has been shown to display anti-inflammatory activity. Although Panax ginseng is widely used for the prevention of photoaging events induced by UVB irradiation, the effect of BIOGF1K on photoaging has not yet been examined. In this study, we investigated the effects of BIOGF1K on UVB-induced photoaging events. Methods: We analyzed the ability of BIOGF1K to prevent UVB-induced apoptosis, enhance matrix metalloproteinase (MMP) expression, upregulate anti-inflammatory activity, reduce sirtuin 1 expression, and melanin production using reverse transcription-polymerase chain reaction, melanin content assay, tyrosinase assay, and flow cytometry. We also evaluated the effects of BIOGF1K on the activator protein-1 signaling pathway, which plays an important role in photoaging, by immunoblot analysis and luciferase reporter gene assays. Results: Treatment of UVB-irradiated NIH3T3 fibroblasts with BIOGF1K prevented UVB-induced cell death, inhibited apoptosis, suppressed morphological changes, reduced melanin secretion, restored the levels of type I procollagen and sirtuin 1, and prevented mRNA upregulation of MMP-1, MMP-2, and cyclo-oxygenase-2; these effects all occurred in a dose-dependent manner. In addition, BIOGF1K markedly reduced activator-protein-1-mediated luciferase activity and decreased the activity of mitogen-activated protein kinases (extracellular response kinase, p38, and C-Jun N-terminal kinase). Conclusion: Our results strongly suggest that BIOGF1K has anti-photoaging activity and that BIOGF1K could be used in anti-aging cosmeceutical preparations.

Antioxidant activity and anti-inflammatory effects of ethanol extract from Allium schoenoprasum (향부추 에탄올 추출물의 항산화 활성 및 항염증 효과)

  • This study evaluated the antioxidant and anti-inflammatory activity of ethanol extracts using three parts of the chives plant: the bulb, the leaf, and the flower. As a result of DPPH and ABTS radical scavenging ability, the scavenging activity of the flower extract was higher than that of the bulb and leaf. In addition, as a result of FRAP analysis, antioxidant activity increased in all extracts depending on the extract concentration. The total polyphenol content was high in the following order: flower (11.29±0.37 mgGAE/g) > leaf (6.61±0.14 mgGAE/g) > bulb (5.7±0.67 mgGAE/g) extract. The cytotoxicity of the three extracts against rat macrophage RAW264.7 cells and HaCaT cells, both of which are human cutaneous keratinocyte cell lines, was minimal. NO by LPS was generated as a result of examining the anti-inflammatory activity of each extract through the NO colorimetric analysis method and ELISA. TNF-α secretion was decreased to a significant level in the flower ethanol extract. Therefore, these results indicate that there is a high possibility that the ethanol extract of chives, a natural plant resource, can be used as a cosmetic raw material.

Studies on the Antioxidant and Whitening Effects of Chamaecyparis obtusa Extract (편백나무(Chamaecyparis obtusa) 추출물의 항산화, 미백효과에 관한 연구)

  • This study was conducted as follows in order to find out the possibility of using (Chamaecyparis obtusa) by part as a cosmetic material. Chamaecyparis obtusa was separated leaves, bark, wood, and root, and extracted with 99.9% ethanol, and the antioxidant and whitening effects of the sample were confirmed. For this, the following studies were conducted. Antioxidant evaluation was confirmed by 1 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, 2 2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS+) radical scavenging activity. To evaluate the whitening effect, mushroom tyrosinase inhibitory activity evaluation and cytotoxicity evaluation of Chamaecyparis obtusa extract through MTT assay were conducted, and cellular tyrosinase inhibition rate was measured and melanin contents was confirmed. As a result of antioxidant activity, Bark extract among Chamaecyparis obtusa parts showed the best efficacy, and bark in B16F10 melanoma cell showed tyrosinase activity inhibition and melanin contents inhibitory effects. Therefore, it was confirmed that the ethanol extract of Chamaecyparis obtusa was developed as a natural material for safe and excellent whitening functional cosmetics.

Antioxidative Effects and Component Analysis of Extracts of the Rumex acetosa L. (수영 전초 추출물의 항산화 활성 평가 및 성분 분석)

  • In this study, the antioxidative effects and component analysis for the extracts of Rumex acetosa L. were investigated. All experiments were performed with 50% ethanol extract, ethyl acetate fraction and aglycone fraction obtained from dried R. acetosa L.. Free radical scavenging activities (1,1-diphenyl-2- picrylhydrazyl) size of, in the order of aglycone fraction > ethyl acetate fraction > 50% ethanol extract, aglycone fraction ($45.10{\mu}g/mL$) showed the highest radical scavenging activity. Reactive oxygen species (ROS) scavenging activity (total antioxidant capacity, $OSC_{50}$) on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system was also, in the order of ethyl acetate fraction> aglycone fraction> 50% ethyl acetate fraction, ethyl acetate fraction ($2.68{\mu}g/mL$) was shown a great antioxidant capacity. The total antioxidant capacity of the ethyl acetate fraction was found to be greater than L-ascorbic acid, known as a typical hydrophilic antioxidant ($6.88 {\mu}g/mL$). The cellular protective effects of R. acetosa L. extracts on the $^1O_2$-induced cellular damage of human erythrocytes were exhibited at all concentration-dependent ($1{\sim}25{\mu}g/mL$). Especially, aglycone fraction (${\tau}_{50}$, 104.80 min) in $25{\mu}g/mL$ showed the most protective effect among extracts. Components of the ethyl acetate fraction obtained from R. acetosa L. extracts were analyzed by TLC, HPLC chromatogram, LC/ESI-MS/MS. As a result, the ethyl acetate fraction contained several flavonoids, such as orientin, isoorientin, vitexin, isovitexin. These results indicate that the R. acetosa L. extracts can be used as antioxidants in biological systems, particularly skins exposed to UV radiation by quenching and/or scavenging $^1O_2$ and other ROS. Thus, the extracts of R. acetosa L. could be applicable to new anti-aging cosmeceutical ingredients.

The Antimicrobial Activity against Helicobacter pylori and Antioxidant effect from the extracts of Mulberry leaves (Morus Alba L.) (품종이 다른 뽕잎추출물의 Helicobacter Pylori에 대한 항균활성 탐색과 항산화 효과)

  • The water extracts and 80% ethanol extracts from mulberry leaves of 108 kinds were tested their antimicrobial activities against Helicobacter pylori and antioxidant effects. The ABTS [2,2'-azino-bis(3-ethylbenzothiaznoline-6-sulfornic acid)] radical decolorization, electron donating ability (EDA), thiobarbituric acid reactive substance (TBARS) and antioxidant protection factor (PF) were determined for water extracts and 80% ethanol extracts from mulberry leaves. In the electron donating activity, the 13 kinds of water extracts showed high inhibition (>80%), whereas the 59 kinds of 80% ethanol extracts showed high inhibition. The inhibitory activities of water extracts from all kinds of mulberry leaves were higher than 90% in ABTS [2,2'-azinobis(3-ethylbenzothiaznoline-6-sulfornic acid)] radical decolorization. The thiobarbituric acid reactive substance (TBARS) and antioxidant protection factor (PF) of the 80% ethanol extracts were higher than that of water extracts. Antimicrobial activity against Helicobacter pylori showed high value in 80% ethanol extracts of 15 kinds mulberry leaves. The results implied that the mulberry leaves can be useful for natural antimicrobial medicine.

Enhancement of the Cosmeceutical Activity by Nano-encapsulation of Thiamine Di-lauryl Sulfate (TDS) with antimicrobial efficacy (항균 효능이 있는 비타민 B1 유도체(Thiamine Dilauryl Sulfate:TDS)의 나노입자화를 통한 기능성 향장 활성 증진)

  • This study was to improve cosmetical activity of thiamine di-lauryl sulfate (TDS) by encapsulation of nanoparticle with lecithin. Results showed that most of the nanoparticles containing the TDS were well formed in round shape with below 150 ~ 200 nm diameter as well as they were fairly stable in various pH ranges by measuring zeta potentials. The nanoparticles of TDS resulted in 85% cell viability of human normal fibroblast cells (CCD-986sk) when added at the highest concentration (1.0 mg/mL). The nanoparticles of Acer mono sap showed highest free radical scavengering effect as 88.1% in adding sample (1.0 mg/mL), compared to TDS solution of non-encapsulation (81.6%). The nanoparticles of TDS reduced the expression of MMP-1 on UV-irradiated CCD-986sk cells down to as 41.4%. The TDS solution and nanoparticles showed significant anti-microbial activities agaionst the salmonella typhimurium and listeria monocytogenes at 5 and 6 days as compared with control. Anti-microbial activities of TDS nanoparticles were similar to positive control. These results indicated that TDS nanoparticles may be a source for functional cosmetic agents capable of improving cosmetical activity such as antioxidant, whitening, and anti-wrinkling effects and can be further developed as natural preservative in cosmetics.

Biological Activity of Extracts from Garden Sage (Salvia officinalis L.) (가든 세이지(Salvia officinalis L.) 추출물의 생리활성 탐색)

  • The extracts from Salvia officinalis were studied for antioxidative activities and inhibitory activities against angiotensin converting enzyme(ACE) and xanthine oxidase (XOase). Total phenolic compounds were found as 22.28, 26.3, 24.63, and 28.22 mg/g in the water, 60% ethanol, 60% methanol and 60% acetone extracts, respectively. The antioxidant activities of Salvia officinalis extracts were measured as $64.4{\pm}1.5%$ at $200\;{\mu}g/ml$ on EDA, inhibition rate on ABTS of $96.9{\pm}0.2%$, antioxidant protection factor of $2.30{\pm}0.16$ PF and TBARS was $0.6{\pm}0.05$ (${\times}100\;{\mu}M$) in the control and $0.28{\pm}0.02$ (${\times}100\;{\mu}M$) in 60% ethanol extracts. Inhibitory activities was the ACE of 75.50% and XOase 100% in 60% ethanol extracts. The 60% ethanol extracts from Salvia officinalis exhibited antimicrobial activities against Helicobacter pylori such as 13 mm of clear zone and inhibition rate of 63.4% with $200\;{\mu}g/ml$ of phenolics content. Rosemarinic acid was the most abundant phenolic compounds as analyzed by HPLC. The results suggest that the 60% ethanol extracts from Salvia officinalis L. will be useful as natural antioxidants and functional foods.

Anti-melanogenesis and Anti-wrinkle Properties of Korean Native Dendrobium speciosum Ethanol Extract (Dendrobium speciosum 에탄올 추출물의 melanin 생성 억제 효능 및 주름개선 효과)

  • Melanin is produced by melanocytes of the melanoepidermic unit and other cell types. These cells secrete and distribute the melanin pigment, which provides protection from ultraviolet radiation. In this study, the inhibitory activity against tyrosinase and melanin biosynthesis in B16F10 melanoma cells and anti-wrinkling effects on human dermal fibroblasts of Dendrobium speciosum ethanol extract were investigated. The Dendrobium speciosum extract inhibited melanin biosynthesis and tyrosinase activity in a dose-dependent manner in comparison with an untreated control group. Treatment with the Dendrobium speciosum extract suppressed α-MSH-stimulated melanogenesis in B16F10 cells and the dendrite outgrowth of melanocyte/melanoma cells. The α-MSH-induced mRNA expression of tyrosinase-related protein-1 (TRP-1), tyrosinase-related protein-2 (TRP-2) and microphthalmia-associated transcription factor (MITF) was significantly attenuated in a concentration-dependent manner by Dendrobium speciosum treatment. In addition, Dendrobium speciosum treatment increased production of type I procollagen synthesis in human dermal fibroblasts. Dendrobium speciosum ethanol extract exhibited a potent inhibitory effect on melanin biosynthesis, tyrosinase activity and increased procollagen synthesis. These results indicate that Dendrobium speciosum shows promise as an ingredient in cosmeceutical products due to its whitening and anti-wrinkle effects.

Antioxidative Effects of Campanula takesimana Nakai Extract (섬초롱(Campanula takesimana Nakai) 용매 추출물의 항산화 효과)

  • The study analyzed the total polyphenolic contents, DPPH radical scavenging activities, ABTS radical scavenging activities, reducing power levels, SOD-like activities, and tyrosinase inhibition activities of fractions from Campanula takesimana Nakai extract. Total polyphenolic content of the ethyl acetate fraction from Campanula takesimana Nakai extract was the highest. The ethyl acetate fraction of Campanula takesimana Nakai extract showed the lowest $IC_{50}$ value (0.13 mg/mL) as well as greater than 98% ABTS radical scavenging activity, which was similar to ascorbic acid. Further, reducing power was significantly higher in the ethyl acetate fraction. The SOD-like activity of edible plant was 70.56% (n-butanol fraction) and 62.27% (ethyl acetate fraction) at 1 mg/mL. Tyrosinase inhibition activity of Campanula takesimana Nakai extract was reduced to 34.77% in the n-butanol fraction and 30.85% in the ethyl acetate fraction at 5 mg/mL. These results suggest that the ethyl acetate and n-butanol fractions of Campanula takesimana Nakai extract could be applicable as food additives or cosmeceutical ingredients.

Antioxidant Effect of the Fraction Isolated from Pyrus ussuriensis Leaves (산돌배나무(Pyrus ussuriensis) 잎 분획물의 항산화 효과)

  • This study was conducted to confirm the application as ingredients of cosmetics through an examination of the function for anti-oxidant activity of the fraction isolated from Pyrus ussuriensis leaves. The dried leaf of Pyrus ussuriensis were extracted with acetone-$H_2O$ (6:4, v/v), concentrated and fractionated with the upper layer of acetone on a separatory funnel. Each fraction was freeze dried, then a portion of acetone soluble powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol as eluents and also used the MIC-gel using a series of aqueous methanol as developing solvent. The isolated compounds were identified by silica-gel TLC. The concentration of total phenolic compound of Pyrus ussuriensis acetate soluble fraction was high, 914 mg/g. The results obtained from the analyses of the anti-oxidanat effects of Pyrus ussuriensis acetate fraction can be summarized as follows: In the result of DPPH scavenging radical activity, Pyrus ussuriensis acetate soluble fraction showed more than 80% at 100 ppm. SOD-like activity of one of Pyrus ussuriensis acetate soluble fractions was 77% at 1000 ppm. Xanthine oxidase inhibition of Pyrus ussuriensis acetate soluble fraction was 38% at 100 ppm. From these results, we confirmed that acetate fraction of Pyrus ussuriensis has a great potential as a natural ingredients with a natural antioxidant and antimicrobial source.
